Development of multilayered artificial urethra graft for urethroplasty.

To enhance the treatment of patients' urethral defects, such as strictures and hypospadias, we investigated the potential of using artificial urethral tissue. Our study aimed to generate this tissue and assess its effectiveness in a rabbit model. Two types of bioprinted grafts, based on methacrylated gelatin-silk fibroin (GelMA-SF) hydrogels, were produced: acellular, as well as loaded with autologous rabbit stem cells. Rabbit adipose stem cells (RASC) were differentiated toward smooth muscle in the GelMA-SF hydrogel, while rabbit buccal mucosa stem cells (RBMC), differentiated toward the epit

Development and characterization of a smart obstetric dressing with a visual indication function.

Excessive adhesion between dressings and the wound bed often causes secondary tissue damage and delays healing during dressing removal. This study aimed to develop and evaluate a multifunctional moisture-containing nonwoven dressing composed of silk fibroin protein (SF), polycaprolactone (PCL), and ellagic acid (EA) to improve wound healing while allowing visual assessment of moisture. A composite nonwoven fabric integrating SF, PCL, and EA was fabricated and characterized using spectroscopic analysis to evaluate its fluorescence properties. In vitro cytocompatibility was assessed in the four

Orb-weaving spiders possess three types of aqueous silk glue genes with pervasive variation in repeat structure, organization, and length.

The evolutionary success of orb-web spiders has been attributed to the use of a liquid glue produced by the aggregate glands that coats the web's capture threads to secure prey. Remarkably, the primary protein components within these glue droplets are derived from the same family of genes, spidroins, that encode silk fibers. The complete gene sequence of the aggregate spidroins, AgSp1 and AgSp2, have been described in a few species, but the molecular variation across species is not well characterized. Here, we provide a comparative analysis of AgSp genetic diversity among thirteen orb-web and

A silk-based high impact composite for the core decompression of the femoral head.

Core decompression of the femoral head is a recommended head-conserving strategy for early-stage osteonecrosis of the femoral head. However, no ideal filling material has been found so far. In this study, we fabricated a "solid core-porous coating" composite scaffold, which is a silk fibroin/hydroxypropyl methylcellulose (SF/HPMC) scaffold, by a "two-step" process. The solid core scaffold possesses a sufficient compression modulus (860 MPa) for support, while the porous coating scaffold with controllable pore size and porosity provides a suitable microenvironment for the osteoblast cell to adh

Scalable Mie-Silk textile for dual-mode personal cooling indoors and outdoors.

Radiative cooling textiles are typically designed for specific indoor or outdoor conditions, limiting their environmental adaptability. Here, we report a continuously spun molecular interface-engineered silk (Mie-Silk) that integrates radiative and conductive cooling for effective body temperature regulation in diverse settings. The Mie-Silk has a natural silk yarn core with high mid-infrared (MIR) emissivity and a surface layer of ZrO2 nanoparticles (NPs) enabling strong solar scattering based on the Mie scattering principle. Natural silk fibroin acts as a robust binder, anchoring the NPs on

Polyphenol mediated zinc-oxygen synergistic hydrogel remodels senescent microenvironment for periodontal tissue regeneration.

Senescent mesenchymal stem cells residing in an inflammatory, dysbiotic, and hypoxic microenvironment pose a barrier to periodontal regeneration. Here we introduce a hydrocaffeic acid (HCA)-mediated silk fibroin hydrogel incorporating Mn/HCA-modified calcium peroxide (Mn-hCaO₂) and HCA-modified zeolitic imidazolate framework-8 (hZIF8) to rejuvenate this environment. The strategy imbues the hydrogel with enhanced adhesion and adaptability in periodontal pockets. The Mn-HCA complex acts catalytically to reduce oxidative stress and correct hypoxia. Simultaneously, Zn²⁺ and HCA restore microbial b

BRD9-mediated chromatin remodeling suppresses osteoclastogenesis through negative feedback mechanism.

Bromodomain-containing protein 9 (BRD9), a component of non-canonical BAF chromatin remodeling complex, has been identified as a critical therapeutic target in hematological diseases. Despite the hematopoietic origin of osteoclasts, the role of BRD9 in osteoclastogenesis and bone diseases remains unresolved. Here, we show Brd9 deficiency in myeloid lineage enhances osteoclast lineage commitment and bone resorption through downregulating interferon-beta (IFN-β) signaling with released constraint on osteoclastogenesis. Notably, we show that BRD9 interacts with transcription factor FOXP1 activati

Silk Nanococoons: Bio-Nanoreactors for Enzymatic Catalytic Reactions and Applications to Alcohol Intoxication.

Enzymatic reactions are often terminated by severe aqueous environments. Inspired by bio-protection of silk cocoons, a system of silk fibroin (SF) nanococoons is developed, which is capable of providing a bio-protective environment for enzymatic activities on one hand, and facilitating the process of cascade enzymatic reactions on the other hand. This gives rise to the significant enhancement in enzymatic stability and efficacy. For instance, the protected glucose oxidase and horseradish peroxidase enzymes exhibit a 17.5- and 7.0-fold increase when stored at 60 °C (>50% activity; ≈28 h) and 25

Fabrication of 3D porous silk scaffolds by particulate (salt/sucrose) leaching for bone tissue reconstruction.

Silk fibroin is a biomaterial being actively studied in the field of bone tissue engineering. In this study, we aimed to select the best strategy for bone reconstruction on scaffolds by changing various conditions. We compared the characteristics of each scaffold via structural analysis using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), the swelling ratio, water uptake, porosity, compressive strength, cell infiltration and cell viability (CCK-8). The scaffolds had high porosity with good inter pore connectivity and showed high compressive strength and mod

Hybrid 3D bioprinting for advanced tissue-engineered trachea: merging fused deposition modeling (FDM) and top-down digital light processing (DLP).

In this present study, we introduce an innovative hybrid 3D bioprinting methodology that integrates fused deposition modeling (FDM) with top-down digital light processing (DLP) for the fabrication of an artificial trachea. Initially, polycaprolactone (PCL) was incorporated using an FDM 3D printer to provide essential mechanical support, replicating the structure of tracheal cartilage. Subsequently, a chondrocyte-laden glycidyl methacrylated silk fibroin hydrogel was introduced via top-down DLP into the PCL scaffold (PCL-Sil scaffold). The mechanical evaluation of PCL-Sil scaffolds showed that

Clickable, Thermally Responsive Hydrogels Enabled by Recombinant Spider Silk Protein and Spy Chemistry for Sustained Neurotrophin Delivery.

The ability to deliver protein therapeutics in a minimally invasive, safe, and sustained manner, without resorting to viral delivery systems, will be crucial for treating a wide range of chronic injuries and diseases. Among these challenges, achieving axon regeneration and functional recovery post-injury or disease in the central nervous system remains elusive to most clinical interventions, constantly calling for innovative solutions. Here, a thermally responsive hydrogel system utilizing recombinant spider silk protein (spidroin) is developed. The protein solution undergoes rapid sol-gel tra

Adelmidrol: A novel osteogenic mineralization regulator for bone defect repair via activating the Notch pathway with drug-loaded hydrogel.

Bone defect, irreversible bone loss impairing physiological functions, raises complication risks, lowers life quality and burdens healthcare, demanding effective novel repair strategies. This study aimed to screen candidate drugs promoting osteoblast (OB) osteogenic mineralization for bone defect repair. High-throughput screening identified adelmidrol from a phase 3 clinical drug library. In vitro experiments showed adelmidrol significantly upregulated RUNX2 and OSX expression, enhancing OB osteogenic mineralization by activating the Notch signaling pathway. In vivo, a methacrylated silk fibro

3D-Printed Porous Titanium Alloy Scaffolds Incorporated With Tetramethylpyrazine-Loaded Composite Hydrogels Enhance Angiogenesis and Bone Repair for Femoral Head Osteonecrosis.

Osteonecrosis of the femoral head (ONFH) is a debilitating condition caused by compromised local blood supply to the femoral head, which leads to bone tissue death. The primary therapeutic goals are to restore blood perfusion and reconstruct the damaged bone structure. In this study, we present an advanced three dimensional printed multifunctional composite scaffold that integrates structural biomimicry, sustained drug release, and microenvironment regulation to enable the sequentially coordinated augmentation of ONFH repair. The scaffold features Voronoi-designed porous titanium alloy biomime

DFO biofunctionalized CaTiO₃@CS/SF-DFO scaffolds for regulating Schwann cells and dorsal root ganglion behaviors.

Peripheral nerve injury (PNI) repair faces great hurdles. Autologous nerve grafts, the clinical gold standard, suffer from donor shortage and secondary injury, while traditional nerve conduits yield unsatisfactory regeneration. Herein, we fabricated an electrospun anisotropic chitosan/silk fibroin scaffold embedded with calcium titanate nanoparticles (CTO@CS/SF), modified by dopamine to conjugate deferoxamine (DFO) for PNI treatment. We fully characterized the scaffold's microstructure, wettability, mechanical properties and degradability, and evaluated its bioactivity via Schwann cells (SCs),

Epithelialized chimeric repairing patch supports in situ healing of tracheal fistula.

Tracheal fistula (TF) is a severe thoracic disease characterized by high mortality rates, and current treatment methods present substantial risks while presenting challenges in reepithelialization. In this study, we developed a chimeric repairing patch (CRP) featuring a double-disc structure, designed to seal TF defects and promote organized tissue regeneration. The CRP incorporates a double-cross-linking silk fibroin network and hierarchical micropores, resulting in a flexible, hydrophilic, and biocompatible patch with tissue-matching mechanical properties. The CRP could effectively seal defe

Effect of degumming degree on the structure and tensile properties of RSF/RSS composite films prepared by one-step extraction.

Regenerated silk fibroin (RSF) and regenerated sericin (RSS) have attracted much attention for tissue engineering due to excellent biocompatibility and controllable degradation. However, pure RSF films prepared by existing methods are brittle, which limits applications in the field of high-strength and/or flexible tissues (e.g. cornea, periosteum and dura). A series of RSF/RSS composite films were developed from solutions prepared by dissolving silks with different degumming rates. The molecular conformation, crystalline structure and tensile properties of the films and the effect of sericin c

Bioinspired Injectable Hydrogels Dynamically Stiffen and Contract to Promote Mechanosensing-Mediated Chondrogenic Commitment of Stem Cells.

Developing stiff and resilient injectable hydrogels that can mechanically support load-bearing joints while enabling chondrogenic differentiation of stem cells is a major challenge in the field of cartilage tissue engineering. In the present work, a triple-network injectable hydrogel system was engineered using Bombyx mori silk fibroin, carboxymethyl cellulose (CMC), and gelatin. The developed hydrogel demonstrated a simultaneous increase in both stiffness and contraction over time, thereby imparting a four-dimensional (4D) evolving niche to the cells. While resilience was provided by CMC, the
