Evaluation of the substantivity of chlorhexidine in association with sodium fluoride in vitro.

The efficacy of the fluoride-chlorhexidine association in the prevention of gingivitis and caries has been advocated for a number of years. The objective of the association of these therapeutic agents is a synergistic action. The aim of the present study was to determine the substantivity of chlorhexidine associated or not to sodium fluoride at different intervals of time, in vitro. Bovine enamel surfaces were treated with 0.12% chlorhexidine gluconate (Periogard--Colgate) or 0.05% sodium fluoride with 0.12% chlorhexidine (Duplak--Dentsply) solutions for one minute. Fragments were placed in di

Treatment of osteoporosis in men with fluoride alone or in combination with bisphosphonates.

Alendronate has been approved as a first antiresorptive drug for treatment of osteoporosis in men. Except for fluoride, in some countries so far there has been no approved anabolic substance for the treatment of male osteoporosis. From small studies in men and male patients included in studies on postmenopausal osteoporosis there is sufficient evidence that fluoride has the same osteoblast-stimulating potency in men and women. In our own study on 64 men with idiopathic osteoporosis without prevalent fractures, a low-dose intermittent fluoride regimen (15 mg fluoride ions 3 months on, 1 month o

[Fluorosis in primary dentition in a region with endemic water fluoride].

OBJECTIVE: The aim of the present study was to estimate the prevalence of dental fluorosis in primary dentition of a San Luis Potosi children population, and its association to fluoride concentration in drinking water and urine. An additional objective was, to develop, validate, and test a specific index for dental fluorosis in primary dentition. MATERIAL AND METHODS: From May 1997, to January 1999, we conducted a cross-sectional study to assess the prevalence of dental fluorosis in primary dentition. Study subjects were 100 children aged 3-6 years, selected at random from three kindergartens

Tissue necrosis after subgingival irrigation with fluoride solution.

Irrigation of periodontal pockets with fluoride solution after scaling and root planing is occasionally recommended to inhibit the growth of pathogenic bacteria in the periodontal pocket. At the same time, irrigation enables mechanical removal of loosely adhering plaque and debris. Due to its toxicity, fluoride solution deposited in the periodontium may lead to tissue damage. We report in this paper, a case of extensive periodontal tissue necrosis and permanent loss of alveolar bone after irrigation of periodontal pockets with stannous fluoride solution. The literature on the toxic effects of

Toxicology of hydrogen fluoride in relation to major accident hazards.

A major industrial accident involving the release of hydrogen fluoride has the potential to cause serious injury and possibly death in the surrounding human population. Given the potential for such serious and large-scale effects, there is a need for scientifically based risk assessments for determining appropriate on-site risk control measures, as well as for informing off-site emergency plans and land-use development decisions. Within Great Britain, the Health and Safety Executive (HSE) has the statutory responsibility for providing land-use planning advice in the vicinity of major accident

Heterotrimeric G proteins as fluoride targets in bone (review).

Fluoride is an acknowledged bone anabolic agent. Nevertheless, a narrow therapeutic window and the adverse effects at higher therapeutic doses prevent broad clinical application of fluoride for treatment of diseases of bone loss, such as osteoporosis. The cellular and molecular mechanisms of fluoride action are poorly understood. recent advances in the elucidation of signal transduction pathways induced by fluoride in osteoblastic cells are reviewed. Fluoride and traces of aluminum form a complex, fluoroaluminate, which stimulates cellular heterotrimeric G proteins. Such complex can form in fo

Experimental intra-oral caries models in fluoride research.

The use of experimental intra-oral caries models has increased in fluoride research. This paper focuses on the pre-clinical intra-oral models, the in situ and in vivo models, the various types, their benefits and disadvantages. Both preparation and sterilization of the hard tissue substrates can affect the substrates and therefore the results. Care needs to be taken that dentine samples are not exposed to drying and consequently shrinking during preparation and evaluation. Sterilization by gamma-radiation is at present the least tissue-damaging method. The most realistic experimental model is

Fluoride profiles of perikymata in enamel surfaces of human premolars.

Twenty-five premolars (from Nagoya, Japan < 0.1 parts/10(-6)F in drinking water) were sampled to determine the fluoride content in imbrication lines of Retzius between the grooves and ridges of perikymata on the enamel surface. Eight small windows were formed on each surface in groove and ridge regions using an etched microsampling technique. By using a regression curve, y = ax(-b), fluoride concentrations were compared at depths of 1, 3, 5, 10, 20, 30 and 50 microm in the perikymata regions. Fluoride concentrations increased gradually from the age of 10 to 12 years on the premolar surfaces, b

Fluorosis risk from early exposure to fluoride toothpaste.

UNLABELLED: Swallowed fluoride toothpaste in the early years of life has been postulated to be a risk factor for fluorosis, but the epidemiological evidence is weakened by the fact that most of the relevant studies were done in developed countries where an individual is exposed to multiple sources of fluoride. OBJECTIVES: To quantify the risk of fluorosis from fluoride toothpaste in a population whose only potential source of fluoride was fluoride toothpaste. METHODS: Case-control analyses were conducted to test the hypothesis that fluoride toothpaste use before the age of 6 years increased

[Cariostatic mechanisms of fluoride and its effects on human beings].

Fluorine is ubiquitous in man's environment, rocks, soil, water, air, etc, and mainly is in the form of inorganic fluoride (compounds of fluorine) and utilized extensively for industrial and medical purposes. It is also known that fluoride has both beneficial and toxic effects in man. One of the most beneficial effects of fluorine on human health is the application of fluoride solution for the prevention or reduction the incidence of dental caries. The major cariostatic mechanisms is believed to be that fluoride reduces dental caries through influencing the morphology of teeth by reducing the

The effects of fluorides and/or trace elements on the solubilities of enamel and cementum.

The purpose of this study is to determine the effects of fluorides and trace elements applied alone or in combination at different concentrations on the solubilities of enamel and cementum surfaces of the same teeth. The study has been performed on enamel and cementum surfaces of the impacted third molars extracted by surgical operation. Aqueous solutions of sodium fluoride, aluminum potassium phosphate, strontium chloride and titanium tetrachloride at different concentrations were applied to the surfaces. The solubilities of enamel and cementum and the depth of etchings have been calculated b

The effects of pregnancy and fluoride on orthodontic tooth movements in rats.

The present investigation was undertaken in order to study the velocity of orthodontic tooth movement in rats and the effect of the hormonal changes that occur during pregnancy or with fluoride, which is one of several trace elements that affect hard tissue metabolism. Adult, female Sprague-Dawley rats were separated into three groups: non-pregnant, pregnant, and non-pregnant NaF-supplied. All rats were treated with a fixed orthodontic appliance which moved the upper first molars in a buccal direction during 21 days. The appliances delivered an initial force of 150 mN. In vitro effects of fluoride and bromide on pseudocholinesterase and acetylcholinesterase activities.

The in vitro effects of two metabolites of inhalational anaesthetics, fluoride and bromide, on pseudocholinesterase (PCHE) and acetylcholinesterase (ACHE) activities in the blood samples of seven healthy patients were studied. The PCHE and ACHE activities were determined by kinetic spectrophotometric methods. Fluoride at the levels achieved with clinical concentrations of enflurane and sevoflurane (25-75 microM.L-1) inhibited PCHE activity by 28-65 per cent (P less than 0.01) and ACHE activity by less than five per cent (P greater than 0.05). Enamel defects in a fluoridated south-east Asian community.

The prevalence and distribution patterns of enamel defects in maxillary incisors was assessed in 194 Singaporean children aged 11-15 years and belonging to three different ethnic groups. All were born and continuously resident in Singapore, which has a tropical climate. The water supply was fluoridated in 1957 at a level of 0.7 ppm. The mouth prevalence of defects was 71.5 per cent and the tooth prevalence was 55.9 per cent; 82 per cent of all affected teeth demonstrated white lesions of various forms. Although there was no sex difference in the prevalence and distribution pattern of defects,

Effect of a high dose of fluoride on resorbing osteoclasts in vivo.

Histomorphometric analysis of osteoclasts resorbing bone above the erupting first rat molar tooth germ was made at 1, 2, 6, 24, and 48 h following systemic administration of a single high dose of sodium fluoride. Compared to the number of osteoclasts in control rats, a significant reduction was observed in the number of actively resorbing osteoclasts in fluoride-injected rats at 2 and 6 h after dose administration, after which a gradual increase to control levels was observed. A locally increased number of inactive osteoclasts, not attached to bone surfaces, accompanied the reduced number of a

Reversible uranyl fluoride nephrotoxicity in the Long Evans rat.

Severity and duration of renal injury produced by low levels of uranyl fluoride (UO2F2) were examined in the rat. Rats received multiple ip injections of UO2F2 (cumulative dose: 0.66 or 1.32 mg U/kg body wt). Renal injury was characterized histologically by cellular and tubular necrosis of pars recta of proximal tubule (S2 and S3), with less severe cellular injury to thick ascending limb of loop of Henle and collecting tubule. Injury was evident when renal uranium levels were between 0.7 and 1.4 micrograms U/g wet kidney and was most severe when renal uranium burden was between 3.4 and 5.6 mic

Nephrotoxicity of uranyl fluoride in uninephrectomized and sham-operated rats.

The aim of the present study was to determine whether the nephrotoxicity of the uranium-containing compound uranyl fluoride (UO2F2) is enhanced after unilateral nephrectomy. Unilaterally nephrectomized (NPX) and sham-operated (SO) rats were given single intravenous injections of UO2F2 at doses delivering 100 or 250 micrograms U/kg 16 days after surgery. Between the second and third day after the administration of either dose of UO2F2, the urinary excretion of the cellular enzymes lactate dehydrogenase (LDH) and aspartate aminotransferase (AST) and the plasma solute albumin began to increase si

Toxic effects of food-borne fluoride in silver foxes.

Chronic ingestion of excessive amounts of fluoride from commercial fox food is associated with agalactia in vixens resulting in the starvation deaths of large numbers of kits in three fox herds. Evidence of infectious disease or poor management could not be found and a causal relationship between fluoride and high kit mortality is suggested.
