The causal relationship between psoriasis and chronic obstructive pulmonary disease: A bidirectional mendelian randomization study.

Источник пометил публикацию как отозванную

PURPOSE: Although many studies have investigated the association between psoriasis and chronic obstructive pulmonary disease (COPD), the causal relationship between psoriasis and COPD is still unknown. METHODS: We employed bidirectional Mendelian randomization to investigate the causal relationship between psoriasis and COPD. Genetic instruments for exposure were selected from two distinct genome-wide association study databases. Single nucleotide polymorphisms associated with exposures at the genome-wide significance level (p < 5 × 10^-8 ) and exhibiting low linkage disequilibrium (r^2  < 0.

Exploring genetic associations between vitiligo and mental disorders using Mendelian randomization.

Although a large number of existing studies have confirmed that people with vitiligo are prone to mental disorders, these observational studies may be subject to confounding factors and reverse causality, so the true causal relationship is inconclusive. We conducted a bidirectional Mendelian randomization (MR) analysis to assess the causality between vitiligo and mental disorders, namely depression, anxiety, insomnia, schizophrenia, bipolar disorder, obsessive-compulsive disorder (OCD) and attention-deficit hyperactivity disorder (ADHD). Summary statistics from large available genome-wide asso

Circulating antioxidants and Alzheimer disease prevention: a Mendelian randomization study.

BACKGROUND: Higher circulating antioxidant concentrations are associated with a lower risk of late-onset Alzheimer disease (AD) in observational studies, suggesting that diet-sourced antioxidants may be modifiable targets for reducing disease risk. However, observational evidence is prone to substantial biases that limit causal inference, including residual confounding and reverse causation. OBJECTIVES: In order to infer whether long-term circulating antioxidant exposure plays a role in AD etiology, we tested the hypothesis that AD risk would be lower in individuals with lifelong, genetically

Genetic exploration of the relationship between liability to psychiatric disorders and acne vulgaris.

Observational epidemiology suggests a link between the dermatological disorder acne vulgaris and several psychiatric disorders. However, the biological mechanisms that underlie the relationship between acne and mental health are poorly characterised. Here, we employed a genetic approach using large-scale genome-wide association studies of acne and ten psychiatric disorders to both estimate causal effects and uncover potential shared genetic risk factors. Multiple psychiatric disorders displayed evidence of small-to-moderate genetic correlations with acne. However, only genetic liability to sch

MRBEE: A bias-corrected multivariable Mendelian randomization method.

Mendelian randomization (MR) is an instrumental variable approach used to infer causal relationships between exposures and outcomes, which is becoming increasingly popular because of its ability to handle summary statistics from genome-wide association studies. However, existing MR approaches often suffer the bias from weak instrumental variables, horizontal pleiotropy and sample overlap. We introduce MRBEE (MR using bias-corrected estimating equation), a multivariable MR method capable of simultaneously removing weak instrument and sample overlap bias and identifying horizontal pleiotropy. Mendelian randomization study on the causal relationship between chronic hepatitis B/C virus infection and idiopathic pulmonary fibrosis.

BACKGROUND: The pathogenesis of idiopathic pulmonary fibrosis (IPF) is not well understood. Given the known role of hepatitis C virus (HCV) in inducing cirrhosis, the virus has also received attention in the study of IPF. An earlier retrospective study found an increased incidence of IPF in patients with HCV, supported by evidence in the alveolar lavage fluid of the patients, whereas another set of observational studies did not find an association, which prompted us to explore a causal relationship. It is well known that HCV and hepatitis B virus (HBV) have some similarities: both are RNA viru

Causal association between multiple sclerosis and trigeminal neuralgia: A Mendelian randomization study with meta-analysis.

OBJECTIVE: This study utilized a meta-analysis of two-sample Mendelian randomization (MR) to investigate the causal link between multiple sclerosis (MS) and trigeminal neuralgia (TN). METHODS: Using genetic summary data from the OpenGWAS database (European descent), researchers identified eight MS GWAS studies as exposures and one TN study as the outcome. Ten MR approaches, including IVW and MR-Egger, were employed. Pooled estimates were calculated using a fixed-effects inverse-variance model, with significance set at P < 0.05. RESULTS: Seven of the eight MS exposures showed a positive causa

Sequence variants associated with BMI affect disease risk through BMI itself.

Mendelian Randomization studies indicate that BMI contributes to various diseases, but it's unclear if this is entirely mediated by BMI itself. This study examines whether disease risk from BMI-associated sequence variants is mediated through BMI or other mechanisms, using data from Iceland and the UK Biobank. The associations of BMI genetic risk score with diseases like fatty liver disease, knee replacement, and glucose intolerance were fully attenuated when conditioned on BMI, and largely for type 2 diabetes, heart failure, myocardial infarction, atrial fibrillation, and hip replacement. Sex-specific genetics underlie increased chronic pain risk in women: genome-wide association studies from the UK Biobank.

BACKGROUND: Chronic pain disproportionately affects women, but the reasons for this disparity are unclear. METHODS: We investigated this disparity from a genetic perspective using data from the UK Biobank, focusing on multisite chronic pain, which is highly heritable and manifests a sex bias. RESULTS: Genome-wide association studies (GWAS) revealed that women have approximately 4500 sex-specific causal loci for overlapping pains, four times more than men, accounting for their higher heritability. Heritability partitioning indicated that pain-related loci are primarily enriched in specific br

Shared biological pathways linking ADHD and cortisol variability are related to externalizing behaviors.

Altered cortisol regulation is implicated in Attention-Deficit/Hyperactivity Disorder (ADHD), but causality remains debated. While meta-analyses suggest that lower basal cortisol levels, especially in the morning, correlate with ADHD, study heterogeneity prompts further inquiry. Leveraging post-genome-wide association approaches, we examined morning cortisol levels (n = 25,314) and ADHD (n = 225,543). We employed seven Mendelian Randomization methods for causal inference, assessed global and local genetic correlations, and applied the conjFDR method. Additionally, we conducted polygenic score

Pleiotropic Effects of Common and Rare GCKR Exonic Mutations on Cardiometabolic Traits.

BACKGROUND: The common non-synonymous mutation of the glucokinase regulator (GCKR) gene, namely rs1260326, is widely reported to have pleiotropic effects on cardio-metabolic traits and hematological parameters. OBJECTIVE: This study aimed to identify whether other GCKR variants may have pleiotropic effects independent of the rs1260326 genotypes. METHODS: In total, 81,097 Taiwan Biobank participants were enrolled for the regional plot association studies and candidate variant analysis of the region around the GCKR gene. RESULTS: The initial candidate variant approach showed the significant a

Mendelian randomization study of coffee consumption and age at onset of Huntington's disease.

BACKGROUND & AIM: The association between habitual coffee or caffeine consumption and age at onset (AAO) of Huntington's disease (HD) is unclear. We employed Mendelian randomization to investigate the causal relationship between coffee consumption and AAO of HD. METHODS: The instrumental variable including 14 independent genetic variants associated with coffee consumption was selected from a genome-wide association study (GWAS) meta-analysis of 375,833 individuals of European ancestry. Genetic association estimates for AAO of HD were obtained from the Genetic Modifiers of Huntington's Disease

Rare genetic causes of complex kidney and urological diseases.

Although often considered a single-entity, chronic kidney disease (CKD) comprises many pathophysiologically distinct disorders that result in persistently abnormal kidney structure and/or function, and encompass both monogenic and polygenic aetiologies. Rare inherited forms of CKD frequently span diverse phenotypes, reflecting genetic phenomena including pleiotropy, incomplete penetrance and variable expressivity. Use of chromosomal microarray and massively parallel sequencing technologies has revealed that genomic disorders and monogenic aetiologies contribute meaningfully to seemingly comple

Leveraging multimodal neuroimaging and GWAS for identifying modality-level causal pathways to Alzheimer's disease.

The UK Biobank study has produced thousands of brain imaging-derived phenotypes (IDPs) collected from more than 40,000 genotyped individuals so far, facilitating the investigation of genetic and imaging biomarkers for brain disorders. Motivated by efforts in genetics to integrate gene expression levels with genome-wide association studies (GWASs), recent methods in imaging genetics adopted an instrumental variable (IV) approach to identify causal IDPs for brain disorders. However, several methodological challenges arise with existing methods in achieving causality in imaging genetics, includin

The Causal Relationship between Immune-Mediated Inflammatory Diseases and Aortic Aneurysm: A Bidirectional Two-Sample Mendelian Randomization Study.

METHODS: We sourced genetic association data from public genome-wide association study databases for populations of European ancestry. Adhering to MR principles, we identified valid instrumental variables from genetic variants. A range of statistical methods were applied for MR analysis, with the inverse variance weighted (IVW) method emerging as the most reliable estimator of causality in this context. RESULTS: The causal estimates obtained using the IVW method revealed a significant association between genetically predicted AA and rheumatoid arthritis (RA; OR = 1.06, 95% CI = 1.01-1.12, P=0

Genetic testing in dementia - utility and clinical strategies.

Techniques for clinical genetic testing in dementia disorders have advanced rapidly but remain to be more widely implemented in practice. A positive genetic test offers a precise molecular diagnosis, can help members of an affected family to determine personal risk, provides a basis for reproductive choices and can offer options for clinical trials. The likelihood of identifying a specific genetic cause of dementia depends on the clinical condition, the age at onset and family history. Attempts to match phenotypes to single genes are mostly inadvisable owing to clinical overlap between the dem

Analysis of multi-trait evolution across independently evolved cavefish populations reveals shared and independent evolution of suites of cave-associated traits.

Environmental perturbations often lead to the evolution of multiple traits. Determining whether shared genetic factors underlie multi-trait evolution is a central question in evolutionary biology. In the Mexican tetra, Astyanax mexicanus, cave-dwelling populations have repeatedly evolved multiple traits. The repeated evolution of these traits, paired the robust environmental differences between the surface and cave habitats, provide an opportunity to investigate the genetic basis of multi-trait evolution. Here, we investigate the extent to which shared genetic mechanisms underlie the repeated
