Aripiprazole prevents stress-induced anxiety and social impairment, but impairs antipredatory behavior in zebrafish.

Environmental pollution caused by antipsychotic residues is a relevant ecological problem. Studies revealed that residues of these drugs are present in a wide range of different ecosystems and can have adverse effects on non-target organisms even in low environmental concentrations. Among these antipsychotic drugs, aripiprazole (APPZ) is a second-generation atypical antipsychotic that is a partial agonist of dopaminergic and serotoninergic receptors. APPZ is used to treat schizophrenia, bipolar disorder, autism, obsessive-compulsive disorder, and anxiety or panic disorders. Thus, in this study

Interactive effects of diclofenac and copper on bioconcentration and multiple biomarkers in crucian carp (Carassius auratus).

Diclofenac (DCF), a non-steroidal anti-inflammatory drug, is widespread in aquatic environments and coexists with heavy metals to form combined pollution. However, the interactive effects of DCF and heavy metals on aquatic organisms remain unknown. This study aimed to investigate the interactive effects of DCF and copper (Cu) on the bioconcentration, oxidative stress status and detoxification-related gene expression in crucian carp (Carassius auratus). Fish were exposed to Cu (100 μg L-1) and DCF (1, 10, 100 and 1000 μg L-1) alone or in combination for 7 days. Results obtained showed that the

IFNa of triploid hybrid of gold fish and allotetraploid is an antiviral cytokine against SVCV and GCRV.

Triploid hybrid of gold fish (Carassius auratus red var., ♀, 2n = 100) and allotetroploid (♂, 2n = 100) displays much improved disease resistance than its parents. In this paper, one of the type I IFNs of triploid hybrid (3nIFNa) has been cloned and characterized. The full-length cDNA of 3nIFNa consists of 740 nucleotides and the predicted 3nIFNa protein contains 183 amino acids. The mRNA transcription of 3nIFNa was detected in all the selected tissues of triploid hybrid and was obviously enhanced after SVCV or GCRV infection. bcIFNa was detected in both whole cell lysate and supernatant media

Protective immunity in gibel carp, Carassius gibelio of the truncated proteins of cyprinid herpesvirus 2 expressed in Pichia pastoris.

Cyprinid herpesvirus 2 (CyHV-2) infection is a newly emerged infectious disease of farmed gibel carp (Carassius gibelio) in China and causes huge economic losses to the aquaculture industry. In this study, the three membrane proteins encoded by genes ORF25, ORF25C, and ORF25D of CyHV-2 were truncated and expressed in yeast, Pichia pastoris. Screening of the recombinant yeasts was done by detecting the truncated proteins using Western blot. Through immunogold labeling, it was shown that proteins binding the colloidal gold were presented on the surface of cells. In the experiment of inhibition o

New surgical approach to overcome the inability of injured mammalian axons to grow within their environment.

We present a new method for creating conditions conductive to axonal growth in injured optic nerves of adult rabbits. The surgical approach consists of making a cavity in the adult rabbit optic nerve, into which a piece of nitrocellulose soaked with conditioned medium originating from regenerating fish optic nerves is implanted. In addition, daily irradiation (10 days, 5 min, 35 mW) with low energy He-Ne laser is carried out. Such a combined treatment may open a door to neurobiologists and clinicians, hoping to unravel the enigma of mammalian CNS regeneration.

High substance P-like immunoreactivity (SPLI) in the olfactory and electrosensory cells of gymnotid fish.

Substance P has been proposed as a candidate neurotransmitter or neuromodulator in the nociceptive system. Using a light microscopial immunohistochemical peroxidase-anti-peroxidase technique we have detected high substance P-like immunoreactivity (SPLI) in several types of sensory organs of 4 species of gymnotiform teleost fish: olfactory epithelium, vestibular, lateral line and electrosensory organs. The olfactory nerve and its endings within the olfactory bulb and the telencephalon were also strongly labelled. At these sites no SPLI was revealed in other teleosts (Carassius auratus, Gnathone

Deep evolutionary origin of limb and fin regeneration.

Salamanders and lungfishes are the only sarcopterygians (lobe-finned vertebrates) capable of paired appendage regeneration, regardless of the amputation level. Among actinopterygians (ray-finned fishes), regeneration after amputation at the fin endoskeleton has only been demonstrated in polypterid fishes (Cladistia). Whether this ability evolved independently in sarcopterygians and actinopterygians or has a common origin remains unknown. Here we combine fin regeneration assays and comparative RNA-sequencing (RNA-seq) analysis of Polypterus and axolotl blastemas to provide support for a common

Quantifying the effect of light activated outer and inner retinal inhibitory pathways on glutamate release from mixed bipolar cells.

Inhibition mediated by horizontal and amacrine cells in the outer and inner retina, respectively, are fundamental components of visual processing. Here, our purpose was to determine how these different inhibitory processes affect glutamate release from ON bipolar cells when the retina is stimulated with full-field light of various intensities. Light-evoked membrane potential changes (ΔVm ) were recorded directly from axon terminals of intact bipolar cells receiving mixed rod and cone inputs (Mbs) in slices of dark-adapted goldfish retina. Inner and outer retinal inhibition to Mbs was blocked w

An element in the alpha1-tubulin promoter is necessary for retinal expression during optic nerve regeneration but not after eye injury in the adult zebrafish.

We have shown previously that a 1.696 kb upstream fragment of the goldfish alpha1-tubulin promoter was capable of driving green fluorescent protein (GFP) expression in the developing and regenerating zebrafish CNS in a pattern closely mimicking the endogenous alpha1-tubulin gene. Comparison of fish and rat alpha1-tubulin promoters identified a 64 bp region with a conserved repetitive homeodomain (HD) consensus sequence core (TAAT) and a nearby basic helix-loop-helix binding E-box sequence (CANNTG), which led us to speculate that it could be of importance for regulating alpha1-tubulin gene tran

Exploration of novel ecological niches for pigmented bacteria with dye bioremediation potential.

INTRODUCTION: This study explores the comparative bioremediation potential of pigmented bacteria isolated from diverse ecological niches. Three distinct isolates were identified: Shewanella xiamenensis (U2f, from goldfish gills), Paracoccus sp. (THN1, a rose thorn endophyte), and Kocuria sp. (YC, an incidental laboratory isolate). METHODS: The isolated bacterial pigments were characterized using FTIR spectroscopy. The isolates and a developed bacterial consortium were evaluated for their ability to tolerate, decolorize, and degrade various synthetic dyes (Congo red, malachite green, bromophen

Sox9a and amh indirectly cooperate to promote spermatogonia differentiation and gonadal development in Cyprinus carpio var. koi(koi carp).

In teleost fish, sex differentiation is co-regulated by genetic factors and environmental signals. Genetically, Wnt4, Rspo1, Foxl2, and cyp19a dominate female differentiation, while dmrt1, Sox9, SF-1, and amh govern male differentiation. Among these, amh (TGF-βsuperfamily domain) and Sox9a (HMG-box domain) are conserved male-specific genes critical for testicular development in zebrafish and goldfish. Notably, their functional interactions under long-term artificial selection in koi carp remains unelucidated at the molecular level. Additionally, female koi carp have higher market value in aquac

Eat, Heal, Repeat: How zebrafish master regeneration.

For eukaryotic organisms the maintenance and regulation of cellular homeostasis is essential. This process requires continuous elimination of cellular debris, including misfolded, oxidized, and aggregated proteins, and damaged/obsolete organelle. It also requires the remodelling of the cytoplasm compartments, and the provision of nutrients to support basic cellular functions. One of the central molecular mechanisms that maintain this precarious homeostasis is the recycling program of the cell: autophagy. Dysregulation of autophagy has a relationship with several diseases, including neurodegene

Light-evoked lateral GABAergic inhibition at single bipolar cell synaptic terminals is driven by distinct retinal microcircuits.

Inhibitory amacrine cells (ACs) filter visual signals crossing the retina by modulating the excitatory, glutamatergic output of bipolar cells (BCs) on multiple temporal and spatial scales. Reciprocal feedback from ACs provides focal inhibition that is temporally locked to the activity of presynaptic BC activity, whereas lateral feedback originates from ACs excited by distant BCs. These distinct feedback mechanisms permit temporal and spatial computation at BC terminals. Here, we used a unique preparation to study light-evoked IPSCs recorded from axotomized terminals of ON-type mixed rod/cone B

Fiber connections of the central nucleus of semicircular torus in cyprinids.

Fiber connections of the presumed auditory portion of the semicircular torus or the central nucleus (TSc) as well as other central and peripheral auditory structures were studied by tract-tracing methods in carp and goldfish. Major ascending projections to the TSc originated from the descending octaval nucleus (DO) and medullary secondary octaval population (SO). Toropetal neurons of the DO were within the saccular terminal zone. A small number of toropetal DO neurons might receive inputs from other inner ear and lateral line endorgans as well. Fibers from the DO terminated in a deep zone of t

Endocrine disruption and altered gonadal development in white perch (Morone americana) from the lower Great Lakes region.

High prevalences of gonadal intersex have been observed in wild fish populations in areas affected by domestic and industrial effluents. For this study, fish were collected in 1998 from the Cootes Paradise region of Hamilton Harbour in western Lake Ontario, Canada, to determine whether gonadal abnormalities, including intersex, were present in young of the year (YOY) fish. No gonadal abnormalities were observed in goldfish (Carassius auratus), common carp (Cyprinus carpio), gizzard shad (Dorosoma cepedianum), brown bullhead (Ictalurus ameiurus), pumpkinseed (Lepomis gibbosus), and bluegill (Le

Atypical properties of release and short-term depression at a specialized nicotinic synapse in the Mauthner cell network.

Many synapses exhibit temporally complex forms of activity-dependent short-term synaptic plasticity. The diversity of these phenomena reflects the evolutionary specialization of synapses within networks. We examined the properties of transmission and plasticity, in vivo, at an identified, specialized axo-axonic nicotinic synapse between the goldfish Mauthner cell and one of its targets, the cranial relay neuron (CRN), using intracellular paired recordings and low frequency (0.33-2 Hz) train stimulations. Depression of successive excitatory postsynaptic potentials (EPSPs), which dominates short

Involvement of calcium and calmodulin in the regulation of ovarian steroidogenesis in Atlantic croaker (Micropogonias undulatus) and modulation by Aroclor 1254.

The involvement of calcium-dependent signal transduction pathways in the regulation of ovarian steroidogenesis was investigated in Atlantic croaker. Treatment with the calcium ionophores A23187 and ionomycin caused a 2- to 5-fold increase in basal steroid accumulation by croaker ovarian tissue in vitro. A23187 potentiated human chorionic gonadotropin (hCG)-induced testosterone (T) accumulation, whereas it inhibited accumulation of estradiol-17beta (E(2)) and the conversion of T to E(2), suggesting that intracellular calcium modulates aromatase enzyme activity. Gonadotropin stimulation of ovari

Establishment of a novel clonal GFP-expressing transgenic ginbuna crucian carp.

The clonal triploid ginbuna crucian carp Carassius auratus langsdorfii, a naturally occurring gynogenetic fish, is suitable for cell transplantation studies to reveal the roles of stem cells and immune cells. To ensure long-term traceability of donor cells within recipient fish, we have established a transgenic ginbuna line that expresses green fluorescent protein (GFP). The Xenopus laevis ef1a promoter was introduced for regulating GFP expression. Tol2 transposon-based transgenesis to ginbuna embryos resulted in producing a putative founder fish (F0) in a mosaic fluorescent fashion; the frequ

Differential polyvalent passive immune protection of egg yolk antibodies (IgY) against live and inactivated Vibrio fluvialis in fish.

Egg yolk antibodies (IgY) can be prepared in large quantities and economically, and have potential value as polyvalent passive vaccines (against multiple bacteria) in aquaculture. This study prepared live and inactivated Vibrio fluvialis IgY and immunized Carassius auratus prior to infection with V. fluvialis and Aeromonas hydrophila. The results showed that the two IgY antibodies hold effective passive protective rates against V. fluvialis and A. hydrophila in C. auratus. Further, the serum of C. auratus recognized the two bacteria in vitro, with a decrease in the bacteria content of the kidn
