Antimicrobial Peptides Derived from Bacteria: Classification, Sources, and Mechanism of Action against Multidrug-Resistant Bacteria.

Antimicrobial peptides (AMPs) are short, usually cationic peptides with an amphiphilic structure, which allows them to easily bind and interact with the cellular membranes of viruses, bacteria, fungi, and other pathogens. Bacterial AMPs, or bacteriocins, can be produced from Gram-negative and Gram-positive bacteria via ribosomal synthesis to eliminate competing organisms. Bacterial AMPs are vital in addressing the increasing antibiotic resistance of various pathogens, potentially serving as an alternative to ineffective antibiotics. Bacteriocins have a narrow spectrum of action, making them hi…

Tannic acid-derived selective capture of bacteria from apple juice.

Given the enormous burden pathogens pose on human health, rapid capture and removal of bacteria for sterilization or further bacterial detection is essential. Herein, tannic acid-functionalized virus-like Fe3O4 (vFe3O4-TA) was established for bacterial enrichment. We investigated the ability of vFe3O4-TA to capture Gram-negative bacteria (E. coli, S. flex and S. typhi) and Gram-positive bacteria (S. aureus, MRSA and LM), respectively. Compared to the capture efficiency of <15 % for Gram-negative bacteria, vFe3O4-TA showed excellent selectivity and efficiency in isolating Gram-positive bacteria…

Are dental diseases examples of ecological catastrophes?

Dental diseases are among the most prevalent and costly diseases affecting industrialized societies, and yet are highly preventable. The microflora of dental plaque biofilms from diseased sites is distinct from that found in health, although the putative pathogens can often be detected in low numbers at normal sites. In dental caries, there is a shift towards community dominance by acidogenic and acid-tolerant Gram-positive bacteria (e.g. mutans streptococci and lactobacilli) at the expense of the acid-sensitive species associated with sound enamel. In contrast, the numbers and proportions of …

In vitro activity of BMY-28142 in comparison with those of other beta-lactam antimicrobial agents.

The in vitro susceptibility of 406 clinical isolates to BMY-28142, a new semisynthetic cephem, was evaluated and compared with cefpimizole, HR 810, ceftazidime, cefotaxime, moxalactam, and cefoperazone in a broth microdilution assay. On a weight basis, the activity of BMY-28142 against Escherichia coli, Proteus species, and Klebsiella-Enterobacter-Serratia was superior to the other cephems. Against gentamicin-susceptible and -resistant Pseudomonas aeruginosa, BMY-28142 was more active than the other cephems, except ceftazidime and HR 810. A total of 74% of gram-negative and 56% of gram-positiv

In-vitro studies of meropenem.

Meropenem, a new carbapenem, was compared with imipenem and seven other broad-spectrum antimicrobial agents against approximately 1000 clinical isolates. Meropenem was two- to four-fold more active than imipenem against Gram-negative organisms and its spectrum of antimicrobial activity was wider than those of all other drugs tested. However, imipenem was more potent than meropenem against the staphylococci, Streptococcus spp. and enterococci. Many rarely isolated organisms were more susceptible to the carbapenems than to other comparison compounds. All anaerobic bacteria were inhibited by mero

Gram-Positive and Gram-Negative Probiotic Extracellular Vesicles: Mechanisms of Immune Modulation and Therapeutic Opportunities in Atopic Dermatitis.

Atopic dermatitis (AD) is a prevalent chronic inflammatory skin disease characterized by epidermal barrier dysfunction, type 2 immune skewing, and microbiome dysbiosis. Although live probiotics can modulate the gut-skin axis, their clinical use is limited by variable efficacy and safety concerns. Probiotic extracellular vesicles (PEVs) have emerged as cell-free nanotherapeutics that retain microbial bioactivity while circumventing the risks associated with live bacteria. However, the comparative immunological roles of vesicles from Gram-positive and Gram-negative probiotics remain incompletely

Flavonoids as Potential Therapeutic Agents Against Gastrointestinal Pathogen Growth and Their Mechanisms of Action: A Comprehensive Review.

The increasing prevalence of gastrointestinal infections, along with growing resistance to conventional antibiotics, has driven the search for alternative antibacterial agents including flavonoids, which are widely recognised for their antimicrobial properties. Flavonoids exhibit broad-spectrum antimicrobial activity against both Gram-positive and Gram-negative bacteria through multifactorial mechanisms, disrupting multiple bacterial cellular processes, including energy metabolism, membrane integrity, and protein synthesis. Several studies have highlighted the antimicrobial potential of flavon

Low-Cytotoxicity, Broad-Spectrum Corn Starch-Based Antibacterial Particles that Inhibit Multidrug-Resistant Bacteria.

Antimicrobial resistance is a serious problem in biomedical applications that seriously increases the risk of medical failure. Therefore, developing highly efficient antibacterial agents that inhibit the growth of multidrug-resistant bacteria is a long-standing research goal. In this report, a low-cytotoxicity and highly efficient alternative to antibiotics was designed and prepared using edible corn starch as the scaffold and 2-hydroxypropyl-trimethylammonium chloride chitosan (HTCC) as the antimicrobial agent. The HTCC/starch particles were found to have a positively charged surface over a w

Immunological functional differentiation of two transmembrane variants of Dscam in Chinese mitten crab.

Down syndrome cell adhesion molecule (Dscam), also called hypervariable Dscam (Dscam-hv), is an important player in arthropod alternative splicing that connects neurons and immune regulation, acting as a pathogen-specific recognition molecule. Dscam-hv has two forms: transmembrane (TM) Dscam (mDscam) and soluble Dscam (sDscam). Herein, we investigated two transmembrane variants of mDscam resulting from alternative splicing of the transmembrane domain, focusing on differences in their immune regulation. We characterized the Dscam[TM1] and Dscam[TM2] genes of Chinese mitten crab (Eriocheir sinen

Antimicrobial Activity of Gallium Compounds on ESKAPE Pathogens.

ESKAPE bacteria are a major cause of multidrug-resistant infections, and new drugs are urgently needed to combat these pathogens. Given the importance of iron in bacterial physiology and pathogenicity, iron uptake and metabolism have become attractive targets for the development of new antibacterial drugs. In this scenario, the FDA-approved iron mimetic metal Gallium [Ga(III)] has been successfully repurposed as an antimicrobial drug. Ga(III) disrupts ferric iron-dependent metabolic pathways, thereby inhibiting microbial growth. This work provides the first comparative assessment of the antiba

Ultrasound Assisted Cu-catalyzed Synthesis of 1,2-Disubstituted Benzimidazoles as Potential Antibacterial Agents.

BACKGROUND: The ultrasound assisted synthesis and anti-bacterial evaluation of a series of compounds based on 1,2-disubstituted benzimidazole framework is reported here. The approach leading to the synthesis of these compounds involved the Cu-catalyzed coupling-cyclization of N-substituted 2-iodoanilines with benzamide derivatives in the presence of 2-aminoethanol as a ligand. CONCLUSION: A number of compounds were prepared using this methodology in acceptable yield. Two of these compounds showed promising antibacterial activities when tested against both Gram-positive and Gram-negative bacte
