Prenatal and Postnatal Maternal Psychological Distress and the Metabolomic Profile of Human Milk.

BACKGROUND: Human milk is the gold standard of infant nutrition, providing nutrients and bioactive metabolites that shape infant immune, cognitive, and metabolic development. Maternal psychological distress, including stress, anxiety, and depression, is prevalent during the perinatal period and may influence milk composition. Emerging evidence suggests that these alter metabolite classes, with potential long-term consequences for infant health. OBJECTIVES: This study aims to identify whether maternal stress, depressive, and anxiety symptoms are associated with differential metabolomic signatu

Indoxyl sulfate as a potential tubular function marker across kidney disease models.

Kidney tubular damage is a strong predictor of chronic kidney disease progression. Tubular function involves nutrient reabsorption and active secretion via transporters, such as the organic anion transporters (OATs), to eliminate waste and metabolites, including protein-bound uremic toxins (PBUTs). In tubular dysfunction, PBUTs accumulate in plasma, which has been associated with many comorbidities. We hypothesized that PBUT plasma concentration and clearance are sensitive markers for tubular dysfunction. We evaluated this in experimental models of chronic [rat nephrectomy and mouse ischemia-r

ALternatives To prophylactic Antibiotics for the treatment of Recurrent urinary tract infection in women (ALTAR): study protocol for a multicentre, pragmatic, patient-randomised, non-inferiority trial.

BACKGROUND: At least half of all adult women will experience infective cystitis (urinary tract infection: UTI) at least once in their life and many suffer from repeated episodes. Recurrent urinary tract infection (rUTI) in adult women is usually treated with long-term, low-dose antibiotics and current national and international guidelines recommend this as the 'gold standard' preventative treatment. Although they are reasonably effective, long-term antibiotics can result in bacteria becoming resistant not only to the prescribed antibiotic but to other antimicrobial agents. The problem of antim

Mammary nutrient uptake in multiparous sows fed supplementary arginine during gestation and lactation.

Arginine is the precursor for the synthesis of nitric oxide and may increase mammary plasma flow (MPF), which may in turn increase mammary nutrient uptake. Quantifying mammary nutrient uptake improves our understanding of mammary nutrient metabolism and may potentially allow identification of limiting nutrients for colostrum and milk production. Thus, the objectives of the present study were 1) to study the impact of 25 g/d of crystalline Arg (ARG) on MPF and uptake of nutrients by the mammary glands compared with an isonitrogenous supply of Ala (51 g/d; control [CON]) fed to a total of 8 sows

Comprehensive Biological Monitoring to Assess Isocyanates and Solvents Exposure in the NSW Australia Motor Vehicle Repair Industry.

Urethane products that contain isocyanates are extensively used in the motor vehicle repair (MVR) industry and other industries such as furniture and cabinet-making as two-pack spray paints, clears, and adhesives. Attention has recently been refocussed on isocyanate-containing chemicals, particularly in paints. The spray painters in the MVR industry had a propensity to develop industrial asthma at a rate 80 times higher than the general public, which was previously reported in the UK. To track workers exposure to isocyanates, urine samples were collected from 196 spray painters who worked main

Meconium metabolomic profiling dysregulation and neonatal brain injury in selective fetal growth restriction.

BACKGROUND: Meconium serves as a valuable biological matrix for characterizing fetal metabolic signatures throughout gestation. Selective fetal growth restriction (sFGR) is associated with adverse neurological outcomes, potentially mediated by underlying metabolic perturbations. However, the specific relationship between dysregulated meconium metabolome and brain injury in sFGR remains poorly understood. METHODS: Untargeted metabolomics analysis was performed on meconium samples from sFGR (n = 20) and monochorionic diamniotic twins with birth weight concordance (MCDA-C, n = 13) to quantify me

Determination of metabolic markers for lipid metabolism disorders and liver injury recovery via PXR/FXR-CYP7A1 regulation by Folium Ginkgo.

The therapeutic effect of Folium Ginkgo (GK) in treating hyperlipidemia is well known and has been widely investigated. However, a precise and microcosmic biological evidence is lacking to support and validate this effect. To better understand the therapeutic mechanisms of GK, the regulatory activity of the Pregnane X receptor (PXR)/ Farnesoid X receptor (FXR)- Cytochrome P450 family 7 subfamily A polypeptide 1 (CYP7A1) axis was hypothesized as a potential pathway and validated through multiple dimensions, including pharmacology, molecular biology, and metabolomics experiments. Biochemical and Clinical Impact of Organic Uremic Retention Solutes: A Comprehensive Update.

In this narrative review, the biological/biochemical impact (toxicity) of a large array of known individual uremic retention solutes and groups of solutes is summarized. We classified these compounds along their physico-chemical characteristics as small water-soluble compounds or groups, protein bound compounds and middle molecules. All but one solute (glomerulopressin) affected at least one mechanism with the potential to contribute to the uremic syndrome. In general, several mechanisms were influenced for each individual solute or group of solutes, with some impacting up to 7 different biolo

Nitrofurantoin vs other prophylactic agents in reducing recurrent urinary tract infections in adult women: a systematic review and meta-analysis.

BACKGROUND: The clinical and financial burden from bladder infections is significant. Daily antibiotic use is the recommended strategy for recurrent urinary tract infection prevention. Increasing antibiotic resistance rates, however, require immediate identification of innovative alternative prophylactic therapies. This systematic review aims to provide guidance on gaps in evidence to guide future research. OBJECTIVE: The objective of this review was to provide current pooled estimates of randomized control trials comparing the effects of nitrofurantoin vs other agents in reducing recurrent u

Streptococcus group B isolates in a regional hospital area.

A survey of the frequency of group B streptococcal infections in the Sharon area (Israel) was done in this laboratory. In the female genital tract streptococcus group B was found in 11.8%. This microorganism was recovered in lower frequencies (1.6%-7.4%) in other infection sites (CSF, wounds, throat, blood, and urine). The streptococci were identified as belonging to group B by biochemical properties such as resistance to bacitracin and capability to hydrolyze hippurate. Later the isolates were classified serologically. Serotypes Ib, Ic, and II were predominant in the vaginal smears (25%-28% e

Derivation and elimination of uremic toxins from kidney-gut axis.

Uremic toxins are chemicals, organic or inorganic, that accumulate in the body fluids of individuals with acute or chronic kidney disease and impaired renal function. More than 130 uremic solutions are included in the most comprehensive reviews to date by the European Uremic Toxins Work Group, and novel investigations are ongoing to increase this number. Although approaches to remove uremic toxins have emerged, recalcitrant toxins that injure the human body remain a difficult problem. Herein, we review the derivation and elimination of uremic toxins, outline kidney-gut axis function and relati

Genomic analysis of virulence factors and antimicrobial resistance of group B Streptococcus isolated from pregnant women in northeastern Mexico.

INTRODUCTION: Group B Streptococcus (GBS) causes infections in women during pregnancy and puerperium and invasive infections in newborns. The genes lmb, cylE, scpB, and hvgA are involved with increased virulence of GBS, and hypervirulent clones have been identified in different regions. In addition, increasing resistance of GBS to macrolides and lincosamides has been reported, so knowing the patterns of antibiotic resistance may be necessary to prevent and treat GBS infections. This study aimed to identify virulence genes and antibiotic resistance associated with GBS colonization in pregnant w

Longitudinal characterization of plasma amino acids, biogenic amines, and amino acid-related metabolites in heifers from birth to first calving in response to colostrum and transition milk feeding.

This study presents the first comprehensive longitudinal metabolomic profiling of plasma AA, biogenic amines, and AA-related metabolites in Holstein heifers from birth to first calving, with a focus on developmental dynamics and the influence of colostrum and transition milk (TM) feeding. Thirty female Holstein calves were enrolled. Newborn calves were fed ∼3.8 L of colostrum within 2 h of birth, followed by 1 to 1.5 L at 11.5 h postnatum. Calves were then assigned to receive either TM (6 L twice daily from dam) or milk replacer (MR; 6 L twice daily, 140 g/L) for 5 d, followed by feeding MR (1
