A new accumulation assay of Schistosoma mansoni miracidia using square capillary glass tubes.

Current control measures for schistosomiasis have only been partially successful in endemic areas due to socioeconomic constraints. One possibility for controlling the disease is to aim at the miracidial stage of the trematode to avoid infecting intermediate snail hosts by introducing more attractive substances for miracidia in the environment. Here, we introduce an accumulation assay of Schistosoma mansoni miracidia using a square glass tube for analysis of the positive responses of miracidia toward several substances, including snail-conditioned water of Biomphalaria glabrata, Bulinus globos

Exploring the paradox of defense between host and Leishmania parasite.

Leishmaniasis, a neglected tropical disease, still remains a global concern for the healthcare sector. The primary causative agents of the disease comprise diverse leishmanial species, leading to recurring failures in disease diagnosis and delaying the initiation of appropriate chemotherapy. Various species of the Leishmania parasite cause diverse clinical manifestations ranging from skin ulcers to systemic infections. Therefore, host immunity in response to different forms of infecting species of Leishmania becomes pivotal in disease progression or regression. Thus, understanding the paradox

Comparative proteomics reveals Cryptosporidium parvum manipulation of the host cell molecular expression and immune response.

Cryptosporidium is a life-threating protozoan parasite belonging to the phylum Apicomplexa, which mainly causes gastroenteritis in a variety of vertebrate hosts. Currently, there is a re-emergence of Cryptosporidium infection; however, no fully effective drug or vaccine is available to treat Cryptosporidiosis. In the present study, to better understand the detailed interaction between the host and Cryptosporidium parvum, a large-scale label-free proteomics study was conducted to characterize the changes to the proteome induced by C. parvum infection. Among 4406 proteins identified, 121 protein

Toxoplasma gondii Mechanisms of Entry Into Host Cells.

Toxoplasma gondii, the causative agent of toxoplasmosis, is an obligate intracellular protozoan parasite. Toxoplasma can invade and multiply inside any nucleated cell of a wide range of homeothermic hosts. The canonical process of internalization involves several steps: an initial recognition of the host cell surface and a sequential secretion of proteins from micronemes followed by rhoptries that assemble a macromolecular complex constituting a specialized and transient moving junction. The parasite is then internalized via an endocytic process with the establishment of a parasitophorous vacu

How costly are metacercarial infections in a bivalve host? Effects of two trematode species on biochemical performance of cockles.

Bivalve stocks have been decreasing in the last decades largely due to emergent diseases and consequent mass mortality episodes. Cerastoderma edule (the edible cockle) is one of the most exploited bivalves in Europe and is among the most common hosts for trematodes, the most prevalent macroparasites in coastal waters but yet poorly studied. Therefore, in the present study, this bivalve species was used as host model to determine if trematode infection exerts a negative effect on bivalve energy metabolism and balance and if the tissues targeted by different trematodes influence the metabolic co

Vickermania gen. nov., trypanosomatids that use two joined flagella to resist midgut peristaltic flow within the fly host.

BACKGROUND: The family Trypanosomatidae encompasses parasitic flagellates, some of which cause serious vector-transmitted diseases of humans and domestic animals. However, insect-restricted parasites represent the ancestral and most diverse group within the family. They display a range of unusual features and their study can provide insights into the biology of human pathogens. Here we describe Vickermania, a new genus of fly midgut-dwelling parasites that bear two flagella in contrast to other trypanosomatids, which are unambiguously uniflagellate. RESULTS: Vickermania has an odd cell cycle,

Fasciola Species Introgression: Just a Fluke or Something More?

The threats posed by a range of viral and bacterial zoonotic diseases inevitably receive renewed attention in the wake of global pandemic events due to their overt and devastating impacts on human health and the economy. Parasitic zoonoses, however, many of which affect millions of people each day, are frequently ignored. In the case of fasciolosis, caused by infection with Fasciola hepatica or Fasciola gigantica, this oversight has allowed for the expansion of areas of parasite sympatry and thus increased the incidence of hybridization and possible introgression betweenthe two species. Here w

Toxoplasma and Dendritic Cells: An Intimate Relationship That Deserves Further Scrutiny.

Toxoplasma gondii (Tg), an obligate intracellular parasite of the phylum Apicomplexa, infects a wide range of animals, including humans. A hallmark of Tg infection is the subversion of host responses, which is thought to favor parasite persistence and propagation to new hosts. Recently, a variety of parasite-secreted modulatory effectors have been uncovered in fibroblasts and macrophages, but the specific interplay between Tg and dendritic cells (DCs) is just beginning to emerge. In this review, we summarize the current knowledge on Tg-DC interactions, including innate recognition, cytokine pr

Counting Ticks (Acari: Ixodida) on Hosts Is Complex: A Review and Comparison of Methods.

Locating and counting parasites on a host is a fundamental aspect of ecological research and basic veterinary and clinical practice. Variability in the biology and behavior of both hosts and parasites creates many methodological, logistical, and ethical considerations that must be made to collect this deceptively simple measurement. We identified methods that are used to count ticks (Acari: Ixodida, Leach 1815) on hosts by reviewing the methods sections of relevant published studies. Unfortunately, there is no best method agreed upon by scientists to collect ticks from hosts. In general, we su

A latent ability to persist: differentiation in Toxoplasma gondii.

A critical factor in the transmission and pathogenesis of Toxoplasma gondii is the ability to convert from an acute disease-causing, proliferative stage (tachyzoite), to a chronic, dormant stage (bradyzoite). The conversion of the tachyzoite-containing parasitophorous vacuole membrane into the less permeable bradyzoite cyst wall allows the parasite to persist for years within the host to maximize transmissibility to both primary (felids) and secondary (virtually all other warm-blooded vertebrates) hosts. This review presents our current understanding of the latent stage, including the factors

Beyond Blood: African Trypanosomes on the Move.

While the African trypanosomes are among the best-studied parasites, almost everything we know about them is based on the brucei group, which includes the human-infective sleeping sickness parasites and the causative agent of the cattle plague nagana. The past decades have seen an ever-more detailed molecular dissection of Trypanosoma brucei, which today is an accepted cell biological model system. Therefore, recent work on some fundamental aspects of trypanosome biology surprises, as we realise that our knowledge about parasite motility and tropism in the changing host microenvironments is fa

Clearance of schistosome parasites by resistant genotypes at a single genomic region in Biomphalaria glabrata snails involves cellular components of the hemolymph.

Schistosomiasis is one of the most detrimental neglected tropical diseases. Controlling the spread of this parasitic illness requires effective sanitation, access to chemotherapeutic drugs, and control over populations of the freshwater snails, such as Biomphalaria glabrata, that are essential intermediate hosts for schistosomes. Effectively controlling this disease, while minimising ecological implications of such control, will require an extensive understanding of the immunological interactions between schistosomes and their molluscan intermediate hosts. Here we histologically characterise t

The effects of fungal infection and physiological condition on the locomotory behaviour of the ant Myrmica scabrinodis.

Parasite infection often results in alterations in host behaviour. These changes vary greatly in their magnitude, from slight shifts in the time spent by the host performing a given activity to the appearance of novel behaviours. The effects of parasites can differ with the age and the physiological condition of the host. Rickia wasmannii is an ectoparasitic fungal symbiont in Myrmica ants that covers the whole body surface of the host and reduces its lifespan. The fungus is present in both young and old individuals, making it an optimal subject for the study of age-related parasitic effects.

A review of Sarcoptes scabiei: past, present and future.

The disease scabies is one of the earliest diseases of humans for which the cause was known. It is caused by the mite, Sarcoptes scabiei, that burrows in the epidermis of the skin of humans and many other mammals. This mite was previously known as Acarus scabiei DeGeer, 1778 before the genus Sarcoptes was established (Latreille 1802) and it became S. scabiei. Research during the last 40 years has tremendously increased insight into the mite's biology, parasite-host interactions, and the mechanisms it uses to evade the host's defenses. This review highlights some of the major advancements of ou

Consistency of trematode infection prevalence in host populations across large spatial and temporal scales.

Parasites can impart heavy fitness costs on their hosts. Thus, understanding the spatial and temporal consistency in parasite pressure can elucidate the likeliness of parasites' role as agents of directional selection, as well as revealing variable environmental factors associated with infection risk. We examined spatiotemporal variation in digenetic trematode infection in 18 populations of an intertidal host snail (Littorina littorea) over a 300 km range at an 11-yr interval, more than double the generation time of the snail. Despite a complete turnover in the snail host population, the avera

Cryptosporidium within-host genetic diversity: systematic bibliographical search and narrative overview.

Knowledge of the within-host genetic diversity of a pathogen often has broad implications for disease management. Cryptosporidium protozoan parasites are among the most common causative agents of infectious diarrhoea. Current limitations of in vitro culture impose the use of uncultured isolates obtained directly from the hosts as operational units of Cryptosporidium genotyping. The validity of this practice is centred on the assumption of genetic homogeneity of the parasite within the host, and genetic studies often take little account of the within-host genetic diversity of Cryptosporidium. Now you see it, now you don't: flushing hosts prior to experimentation can predict their responses to brood parasitism.

Brood parasitic birds lay their eggs in other birds' nests, leaving hosts to raise their offspring. To understand parasite-host coevolutionary arms races, many studies have examined host responses to experimentally introduced eggs. However, attending parents often need to be flushed from their nests to add experimental eggs. If these birds witness parasitism events, they may recognize and reject foreign eggs more readily than parents who did not. We found that, after being flushed, female blackbirds, Turdus merula, remained close to their nests. Flushed females were more likely to eject foreig

Schistosome feeding and regurgitation.

Schistosomes are parasitic flatworms that infect >200 million people worldwide, causing the chronic, debilitating disease schistosomiasis. Unusual among parasitic helminths, the long-lived adult worms, continuously bathed in blood, take up nutrients directly across the body surface and also by ingestion of blood into the gut. Recent proteomic analyses of the body surface revealed the presence of hydrolytic enzymes, solute, and ion transporters, thus emphasising its metabolic credentials. Furthermore, definition of the molecular mechanisms for the uptake of selected metabolites (glucose, certai

Nutritional performance of the tomato fruit borer, Helicoverpa armigera, on different tomato cultivars.

The development and cultivation of tomato cultivars that are resistant to the tomato fruit borer, Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), are very limited in Iran and other parts of the world because of the lack of information about resistant tomato cultivars to minimize the use of insecticides. Therefore, the present study was carried out to identify alternative methods to chemical control. Nutritional performance of the larval stages (fourth, fifth, and sixth instars) of H. armigera on fruit of eight tomato cultivars, including SUN 6108 f1, Rio grande UG, Korral, Super strain

Examining the tsetse teneral phenomenon and permissiveness to trypanosome infection.

Tsetse flies are the most important vectors of African trypanosomiasis but, surprisingly, are highly refractory to trypanosome parasite infection. In populations of wild caught flies, it is rare to find mature salivarian and mouthpart parasite infection rates exceeding 1 and 15%, respectively. This inherent refractoriness persists throughout the lifespan of the fly, although extreme starvation and suboptimal environmental conditions can cause a reversion to the susceptible phenotype. The teneral phenomenon is a phenotype unique to newly emerged, previously unfed tsetse, and is evidenced by a p
