Muscarinic receptor binding and behavioral effects of atropoine following chronic catecholamine depletion or acetylcholinesterase inhibition in rats.

Rats were subjected to one of two experimental treatments: (1) intraventricular infusion of the catecholamine neurotoxin 6-hydroxydopamine (6-OHDA), known to permanently reduce brain dopamine and norepinephrine levels, or (2) chronic administration of the irreversible acetylcholinesterase inhibitor diisopropylfluorophosphate (DFP). Both treatments are believed to produce relative overactivity of cholinergic systems and to suppress forward locomotion. The anticholinergic agent atropine sulfate yielded excessive forward walking in otherwise chronically akinetic 6-OHDA-treated rats, whereas atrop

Effect of variations in adrenocortical function on dopamine beta-hydroxylase and norepinephrine in the brain of the rat.

The effect of adrenalectomy and corticosterone treatment on dopamine beta-hydroxylase (DBH) activity, catecholamine content and norepinephrine formation and metabolism were studied in the hypothalamus and other parts of the brain of male rats. Two days after adrenalectomy, there was a decrease in DBH activity in the hypothalamus and the brain stem but no change in norepinephrine or dopamine content. Conversion of intraventricularly administered tritiated dopamine to tritiated norepinephrine was slightly increased and norepinephrine was metabolized at a more rapid rate than normal. Corticostero

Protective effect of caffeine against neurodegeneration in a model of Parkinson's disease in rat: behavioral and histochemical evidence.

Epidemiological studies have consistently demonstrated an inverse association between coffee consumption and Parkinson's disease (PD). This study was designed to investigate the beneficial effect of caffeine at a dose comparable to that of human exposure in a model of PD. For this purpose, unilateral intrastriatal 6-hydroxydopamine (6-OHDA)-lesioned rats were pretreated with caffeine (20 mg/kg; i.p.) 1 h before surgery and treated twice a day (10 mg/kg) for 1 month. Apomorphine-induced rotations and number of Nissl-stained neurons of substantia nigra pars compacta (SNC) were counted. The resul

Hypothalamic norepinephrine mediates limbic effects on adrenocortical secretion.

The purpose of this study was to elucidate the role of norepinephrine (NE) in the mediation of adrenocortical responses following limbic stimuli. The effects of stimulation of the dorsal and ventral hippocampus and the midbrain reticular formation on the plasma corticosterone (CS) levels was studied in rats with vehicle or 6-hydroxydopamine (6-OHDA) injected bilaterally into the paraventricular nucleus of the hypothalamus (PVN). The injection of 6-OHDA caused a very significant reduction in the concentration of PVN NE and blocked the rise in plasma CS following the stimulation of the above thr

Studies on thyrotropin-releasing hormone-induced micturition in cats.

In unanaesthetized cats micturition produced by thyrotropin-releasing hormone (TRH) was investigated after its injection into the cerebral ventricles through chronically implanted cannulae. TRH in doses from 0.1 to 1.0 mg evoked dose-dependent micturition. In cats treated with intracerebroventricular (i.c.v.) reserpine and 6-hydroxydopamine, but not with i.c.v. 5,6-dihydroxytryptamine and hemicholinium, the micturition caused by i.c.v. TRH was abolished. Chlorpromazine and antazoline injected into the cerebral ventricles prevented the micturition induced by i.c.v. TRH. On the other hand, mecam

Methamphetamine-induced behavioral alterations following repeated administration of methamphetamine.

Repeated administration of a large dose of methamphetamine (MA) (25 mg/kg, i.p. twice daily for 4 days) to mice enhanced locomotor activity and decreased stereotyped behavior following a subsequent injection of MA. Simultaneous determinations of catecholamines revealed a depletion of brain dopamine. The moderate doses of haloperidol significantly enhanced MA-induced locomotor activity in mice. A significant enhancement of MA-induced locomotor activity was observed in the rats pretreated with 6-hydroxydopamine into the striatum, and this effect correlated negatively with the striatal dopamine l

Meclizine-induced enhanced glycolysis is neuroprotective in Parkinson disease cell models.

Meclizine is a well-tolerated drug routinely used as an anti-histamine agent in the management of disequilibrium. Recently, meclizine has been assessed for its neuroprotective properties in ischemic stroke and Huntington disease models. We found that meclizine protected against 6-hydroxydopamine-induced apoptosis and cell death in both SH-SY5Y cells and rat primary cortical cultures. Meclizine increases the level of 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (PFKFB3), which activates phosphofructokinase, a rate-determining enzyme of glycolysis. This protection is therefore mediated

Subtype-selective N-methyl-D-aspartate receptor antagonists: benzimidazalone and hydantoin as phenol replacements.

Previous work in our laboratories investigating compounds with structural similarity to ifenprodil (5) and 6 (CP101,606) resulted in compound 7 as a potent and selective antagonist of the NR1/2B subtype of the NMDA receptors. Replacement of the phenol group of 7 with a benzimidazalone group tethered by a three-carbon chain to 4-benzylpiperidine resulted in a slightly less active, but selective, compound. Lengthening the carbon tether resulted in a decrease in NR1/2B potency. Replacement of the phenol ring with a hydantoin resulted in weak antagonists. Compound 11a was one of the most potent NR

The role of the mesolimbic dopaminergic system in the desipramine effect in the forced swimming test.

The effects of sulpiride isomers injected into the nucleus accumbens on the anti-immobility activity of desipramine in rats were studied to clarify the role of dopamine receptors. (-)-Sulpiride 100 ng/0.5 microliters injected bilaterally into the nucleus accumbens 5 min before testing reduced the effect of a 7-day treatment with 10 mg/kg per day desipramine in the swimming test but 5 micrograms/0.5 microliter (+)-sulpiride injected in the same area had no effect. Fluphenazine, 5 micrograms/0.5 microliter, or 6-hydroxydopamine-induced depletion of dopamine in the nucleus accumbens also reduced

Influence of chemical stimulation of central dopaminergic system on the open field behaviour of rats.

Influence of some substances known to selectively modify the brain levels of dopamine, was studied on the Open Field behaviour in rats. Of the agents studied, I-dopa, benztropine and amantadine showed some similarity with amphetamine in exhibiting a high degree of complex stereotyped behaviour in rats. The complex stereotyped behaviour due to amphetamine and the similarity shown by these antiparkinsonian agents has been explained on the basis of brain dopamine levels. On the other hand apomorphine depressed the Open Field behaviour and produced a typical stereotyped biting in rats. Since 6-hyd

Behavioral outcome measures for the assessment of sensorimotor function in animal models of movement disorders.

Animal models have and continue to contribute to our understanding of the neurobiology many types of disorders. In movement disorders such as Parkinson's disease (PD), animal models have directly led to various therapeutic treatments such as deep brain stimulation. To facilitate the development of potential therapeutics, sensitive and reliable outcome measures in animal models are necessary to maximize their benefit. In this chapter, behavioral outcome measures, sensitive to varying degrees of sensorimotor dysfunction, are reviewed in rats and mice.

[Experimental cerebral vasospasm induced by oxyhemoglobin and the sympathetic nervous system (Part 2): An investigation into denervation supersensitivity].

UNLABELLED: The pathological mechanism of cerebral vasospasm following subarachnoid hemorrhage has not been well clarified until now. There have been reports that denervation supersensitivity in cerebral vessels occurring after subarachnoid hemorrhage may possible play a role in the appearance of cerebral vasospasm. On the other hand, we have accepted that oxyhemoglobin (Oxy-Hb) is an important spasmogenic substance. In the present study, we investigated the effect of chemical denervation (reserpine or 6-hydroxydopamine: 6-OHDA) and surgical denervation (bilateral superior cervical ganglionect

Nucleus accumbens opiate-dopamine interactions and locomotor activation in the rat: evidence for a pre-synaptic locus.

Locomotor activation produced by the indirect dopamine (DA) agonist amphetamine is reversed by the opiate-receptor antagonist naloxone. Since amphetamine-stimulated locomotion results from the release of DA within the nucleus accumbens (N.Acc.), it is possible that these effects of naloxone result either from a decrease in the pre-synaptic release of DA within the N.Acc. or from a disruption of the effects of DA at, or distal to, the post-synaptic DA receptor. In the present study, we investigated the effects of naloxone on the locomotor-activating properties of dopamine injected directly into

Proliferation of arteriovenous anastomoses in the developing rabbit ear is enhanced after denervation.

The innervation of the rabbit ear vasculature by noradrenergic and substance P-immunoreactive (SP-IR) nerves was investigated in both young and adult animals. All vascular segments were supplied by both noradrenergic and SP-IR nerves. In the ear margins, the arteriovenous anastomoses (AVAs) were more densely innervated than other vessels. In general, the density of both noradrenergic and SP-IR nerves increased with age until 10-12 weeks postnatum. The ear vasculature was denervated in young rabbits to test whether the normal proliferation of AVAs in the growing ear was altered in the absence o

Central depletion of catecholamines--importance of hypertension and anesthesia.

The importance of the brain noradrenergic sympathetic nervous system in blood pressure control during anesthesia with halothane and enflurane was investigated in normotensive and hypertensive rats. Central noradrenergic neurons were destroyed by instillation of 6-hydroxydopamine (6-OHDA) into the cerebral ventricle of both normotensive Wistar rats and spontaneously hypertensive rats (SHR). One week later, arterial and venous lines were placed in the rats under anesthesia; the rats were allowed to recover; and blood pressure and plasma renin activity were measured while the rats were awake. Brain noradrenaline and anaesthesia: further characterization of the beta-receptor.

The sleeping time induced by thiopentone in rats was markedly prolonged by the (-)-isomer of propranolol while the (+)-isomer was virtually without effect. Since the two isomers are equipotent in their membrane-stabilizing effects but the (-)-isomer is about seven to ten times more potent than the (+)-isomer in beta-blockade this suggests that the potentiation of barbiturate sleeping time is due to blockade of beta-adrenergic receptors. The centrally active beta-agonist, clenbuterol, shortened thiopentone-induced sleeping time in a dose-dependent fashion while the beta-agonist, salbutamol, whi

Evidence for a contribution by purines to the neurogenic response of the guinea-pig urinary bladder.

In order to determine if ATP contributes as an excitatory transmitter in the guinea-pig bladder, experiments were conducted with ANAPP3, a photoaffinity analogue of ATP, which is an antagonist of adenine nucleotides in several other smooth muscles. With or without photoactivation with visible light, ANAPP3 antagonized contractile responses of in vitro strips of bladder to exogenous ATP. The antagonism was specific in that responses to acetylcholine and KCl were not affected by ANAPP3. Responses of strips of bladder to transmural electrical stimulation were not antagonized by ANAPP3 and were re

Effects of CDP-choline on striatal dopamine level and behavior in rats.

To further assess the effects of CDP (cytidine diphosphate)-choline on Parkinsonian symptoms, striatal dopamine (DA) was measured fluorometrically in rats after injection of CDP-choline. CDP-choline (300 mg/kg, i.p.) increased the DA content in the striatum (p less than 0.05) one hour after injection. The behavioral effect of CDP-choline was then tested in rats in which the unilateral nigro-striatal DA neurons had degenerated following an intranigral injection of 6-hydroxydopamine (6-OHDA). CDP-choline alone did not produce behavioral changes in these rats. However, pretreatment with a single

Differential development of renal, DOCA-salt, and spontaneous hypertension in the rat after neonatal sympathectomy.

The development of hypertension was studied in rats after neonatal sympathectomy by s.c. injection of 6-hydroxydopamine HCl. Three different types of hypertension were investigated: renal hypertension in the two-kidney Goldblatt model, steroid hypertension produced by deoxycorticosterone (DOCA) implantation and saline as drinking fluid, and genetic hypertension in the spontaneous hypertensive rat (SHR). Blood pressure was measured directly in conscious animals via the iliac artery. Mean blood pressure of conscious sympathectomized (SX) normotensive rats was not significantly different from tha
