Role of the hypophysis in regulation of pancreatic and gastric somatostatin.

Since hypophysectomy and GH deficiency are associated with decreases in hypothalamic content and release of SRIF, it was of interest to determine whether these hormonal alterations also affect peripheral tissue levels of SRIF. Hypophysectomized (hypox) rats were studied at various times after surgery and compared with age-matched controls. Pancreatic, gastric, and hypothalamic SRIF levels were measured by RIA and expressed as nanograms per mg protein or nanograms per organ. Decreased levels of hypothalamic SRIF were observed in hypox animals at all time periods after surgery. In contrast, panc

Hypothalamic insulin signaling is required for inhibition of glucose production.

Circulating insulin inhibits endogenous glucose production. Here we report that bidirectional changes in hypothalamic insulin signaling affect glucose production. The infusion of either insulin or a small-molecule insulin mimetic in the third cerebral ventricle suppressed glucose production independent of circulating levels of insulin and of other glucoregulatory hormones. Conversely, central antagonism of insulin signaling impaired the ability of circulating insulin to inhibit glucose production. Finally, third-cerebral-ventricle administration of inhibitors of ATP-sensitive potassium channel

Alpha 1 noradrenergic antagonism decreases hormonally-induced and hormonally-independent lordosis.

Manipulations of the alpha noradrenergic (NE) system affect both lordosis and the concentration of hypothalamic steroid receptors. The present studies explored whether NE affects lordosis in guinea pigs via changes in hypothalamic estrogen or progestin receptors or through some other mechanism. The alpha 1 NE antagonist prazosin blocked lordosis which was induced with estradiol benzoate (EB) followed by progesterone (P), lordosis which was induced by EB alone and lordosis which is not dependent upon gonadal hormones for its display. These results suggest that NE modulation of lordosis in the g

Luteinizing hormone responses to luteinizing hormone releasing hormone, and growth hormone and cortisol responses to insulin induced hypoglycaemia in functional secondary amenorrhoea.

Luteinizing hormone (LH) responses to luteinizing hormone releasing hormone (LHRH), and growth hormone (GH) and cortisol responses to insulin induced hypoglycaemia were studied in 56 women classified into 4 distinct groups of functional secondary amenorrhoea. The groups were: I, self-induced weight reduction (20 patients); II, post pill amenorrhoea (14 patients); III, anorexia nervosa (10 patients); and IV, idiopathic secondary amenorrhoea (12 patients). Only patients with no overlapping anamnestic factors were included. Group I patients had the most heavily impaired LHRH-LH responses, and the

Neuronal Dynamics of Menstrually Related Migraine: A Longitudinal Task-Based fMRI Study.

BACKGROUND AND OBJECTIVES: Recent fMRI research has identified hypothalamic activation in the preictal phase of migraine, thereby suggesting that the hypothalamus plays a pivotal role in subsequent headaches. However, menstrual migraine (MM) attacks differ from nonmenstrual attacks because of prominent hormonal changes, which are suggested to be the cause of such attacks. This raises questions as to whether MM has different neuronal attack generators. We aimed to use longitudinal daily task-based fMRI to investigate the central changes before MM attacks. METHODS: Patients with pure menstrual

Taurine Attenuates Repeated Heat Stress-Induced Male Reproductive Dysfunction in Mice: Associations with HPG Axis Homeostasis and Hypothalamic Microglial Changes.

Heat stress (HS)-induced male reproductive dysfunction is usually interpreted within a testis-centered framework, whereas the contribution of central neuroendocrine disturbance remains less well defined. In the present study, a repeated HS mouse model was used to examine whether taurine pretreatment attenuates reproductive dysfunction by integrating local testicular endpoints with HPG-axis-related and hypothalamic inflammatory readouts. Male C57BL/6J mice were assigned to control, HS + vehicle, and HS + taurine groups. Mice in the HS groups were exposed to 40 ± 1 °C for 1 h daily for 14 consec

Neonatal treatment with a pegylated leptin antagonist has a sexually dimorphic effect on hypothalamic trophic factors and neuropeptide levels.

It is clear that the prenatal and early neonatal environments are important for determining the metabolic equilibrium in the adult animal, with prenatal/neonatal leptin levels being at least one of the factors involved. Leptin modulates hypothalamic development and, in particular, the neuronal circuits involved in metabolic control. We have recently reported that maternal deprivation (MD) for 24 h on postnatal day (PND) 9 modifies trophic factors and markers of cell turnover and neuronal maturation in the hypothalamus, as well as body weight and circulating leptin levels at PND13, with long- t

Simultaneous occurrence of SIADH, secondary hypogonadism and alopecia universalis in a woman with IDDM.

Syndrome of inappropriate secretion of antidiuretic hormone (SIADH), hypothalamic hypogonadism and alopecia universalis occurred in a 31-year-old female with insulin-dependent diabetes mellitus (IDDM). Despite various clinical investigations and careful observation for 20 months, the cause and pathogenesis of SIADH and hypothalamic hypogonadism were not elucidated. The complex of these disorders had not been described. The presence of IDDM and alopecia universalis, in which an autoimmune process has been assumed to be involved, is interesting in considering the pathogenesis of the SIADH and hy

Contraception as prevention and therapy: sex steroids and the brain.

The brain is one of the specific target tissues for sex steroid hormones. Estrogens, progestins and androgens are able to induce several effects in brain areas of the central nervous system (CNS), through the binding with specific receptors. Specific receptors for gonadal steroids have been identified in the amygdala, hippocampus, basal forebrain cortex, cerebellum, locus ceruleus, midbrain rafe nuclei, glial cells, pituitary gland, hypothalamus and central gray matter. At the hypothalamic level, the principal target for sex steroids is those neurons producing the pulsatile release of the gona

Avian reproductive endocrinology.

Hypothalamic-releasing factors regulate the secretion of anterior pituitary hormones. The anterior pituitary gland secretes the same six hormones as found in mammals: FSH, LH, prolactin, GH (somatotropic hormone), ACTH, and TSH, plus the melanotropic hormone. The endocrine hormones of the avian posterior pituitary gland concerned with reproduction are mesotocin and AVT. The pineal gland, through the secretion of the hormone melatonin, modulates the periodic autonomic functions of the central nervous system. The ovary produces estrogens, progestogens, and androgenic compounds. The testes produc

The physiology of functional hypothalamic amenorrhea associated with energy deficiency in exercising women and in women with anorexia nervosa.

An energy deficiency is the result of inadequate energy intake relative to high energy expenditure. Often observed with the development of an energy deficiency is a high drive for thinness, dietary restraint, and weight and shape concerns in association with eating behaviors. At a basic physiologic level, a chronic energy deficiency promotes compensatory mechanisms to conserve fuel for vital physiologic function. Alterations have been documented in resting energy expenditure (REE) and metabolic hormones. Observed metabolic alterations include nutritionally acquired growth hormone resistance an

Endocrine consequences of brain irradiation.

Cranial radiation is routinely used to manage pituitary tumours, craniopharyngiomas, primary brain tumours, tumours of the head and neck and, in the past, for the prophylaxis of intracranial disease in patients with acute lymphoblastic leukaemia. If the hypothalamic-pituitary axis falls within the radiation fields, the patient is at risk of developing hypopituitarism. The effect of radiation is determined by the dose and the time that has elapsed since treatment. Classically, growth hormone (GH) is the most sensitive of the anterior pituitary hormones to irradiation, followed by gonadotrophins

Endocrine responses to growth hormone releasing hormone and corticotropin releasing hormone in early-onset Alzheimer's disease.

In a study of the hypothalamic-pituitary-somatotropic (HPS) and the hypothalamic-pituitary-adrenal (HPA) systems in early-onset Alzheimer's disease (AD), 10 drug-naive patients and matched controls were given 50 micrograms growth hormone releasing hormone (GHRH) at 9 a.m. and 100 micrograms corticotropin releasing hormone (CRH) at 6 p.m. as an i.v. bolus dose. Compared with controls, patients with AD showed attenuated GHRH-induced growth hormone (GH) responses and decreased adrenocorticotropic hormone (ACTH) but normal cortisol secretion following CRH. GH responses to GHRH were negatively corr

Developmental patterns of the hypothalamic-pituitary axis in the euthyroid and hypothyroid male rat.

The purpose of this study was to determine the effect of rendering newborn male Sprague-Dawley rats hypothyroid with 131I on development of the hypothalamic-pituitary axis. Hormonal responses to thyrotropin-releasing hormone (TRH), haloperidol (H), and luteinizing hormone-releasing hormone (LHRH) were determined at 9, 15, 30 and 75 days of age in hypothyroids and controls. Basal and TRH-stimulated thyroid-stimulating hormone (TSH) levels were higher in the hypothyroids, and the developmental patterns of basal TSH secretion and TSH reserve were parallel in the two groups, with maximal TSH secre

Melanin-concentrating hormone and melanin-concentrating hormone receptors in mammalian skin physiopathology.

To date, there is a dearth of evidence to support functions for melanin-concentrating hormone (MCH) and melanin-concentrating hormone receptors (MCH-R) in mammalian skin physiology including pigmentation, inflammation and immune responses and skin cell proliferation. Much research is therefore still needed to define the roles of the hormone and its receptors in mammalian skin. This will be a crucial step to identifying pathogenic mechanisms that may involve the MCH/MCH-R system in the context of inflammatory and autoimmune skin diseases as well as skin cancers. The following review summarizes

Transition to the adult endocrine clinic: testing pituitary function - what tests and when?

The preferred method(s) and timing for retesting of the growth hormone (GH) axis in young adults with childhood-onset GH deficiency during the transition period are less well characterized than those for children and older adults. Retesting of other hypothalamic-pituitary axes for which replacement therapy had (e.g., thyroid hormone) or had not been (e.g., glucocorticoids and sex steroids) initiated in childhood also requires periodic reassessment. The transition period in children with hypopituitarism also represents the time for initiation of plans for transfer of care to an internist-endocr

Neural connections between the hypothalamus and the liver.

After receiving information from afferent nerves, the hypothalamus sends signals to peripheral organs, including the liver, to keep homeostasis. There are two ways for the hypothalamus to signal to the peripheral organs: by stimulating the autonomic nerves and by releasing hormones from the pituitary gland. In order to reveal the involvement of the autonomic nervous system in liver function, we focus in this study on autonomic nerves and neuroendocrine connections between the hypothalamus and the liver. The hypothalamus consists of three major areas: lateral, medial, and periventricular. Each
