Disease-associated amyloid and misfolded protein aggregates activate the inflammasome.

The pathogenesis of type 2 diabetes, Alzheimer's disease and amyotrophic lateral sclerosis continues to be debated. Recently, the inflammasome protein complex has been shown to be a key regulator of IL-1β, a cytokine implicated in each of these diseases. In all three cases, it is now apparent that unique protein aggregates caused by inappropriate oligomerization or misfolding are sensed by the inflammasome, providing a unifying mechanism for this IL-1β production. What evolved as an innate defense against infection-related particles, therefore, now seems to be a driving force for inflammation

Inflammasome-activating nanoparticles as modular systems for optimizing vaccine efficacy.

Innate immune system activation is a critical step in the initiation of an effective adaptive immune response; therefore, activation of a class of innate pathogen receptors called pattern recognition receptors (PRR) is a central feature of many adjuvant systems. It has recently been shown that one member of an intracellular PRR, the NLRP3 inflammasome, is activated by a number of classical adjuvants including aluminum hydroxide and saponins [Eisenbarth SC, Colegio OR, O'Connor W, Sutterwala FS, Flavell RA. Crucial role for the Nalp3 inflammasome in the immunostimulatory properties of aluminium

The caspase 1 inflammasome is not required for control of murine Lyme borreliosis.

The contribution of the inflammasome to the development of immune responses and disease during infection with the Lyme disease spirochete, Borrelia burgdorferi, is not well defined. Host defense against the spirochete is severely impaired in mice deficient in the adaptor molecule myeloid differentiation antigen 88 (MyD88), which is required not only for Toll-like receptor-mediated responses but also for the production of the proforms of interleukin 1beta (IL-1beta) and IL-18. These cytokines are released in active forms after cleavage by the inflammasome-associated enzyme caspase 1. To investi

Gout-associated uric acid crystals activate the NALP3 inflammasome.

Development of the acute and chronic inflammatory responses known as gout and pseudogout are associated with the deposition of monosodium urate (MSU) or calcium pyrophosphate dihydrate (CPPD) crystals, respectively, in joints and periarticular tissues. Although MSU crystals were first identified as the aetiological agent of gout in the eighteenth century and more recently as a 'danger signal' released from dying cells, little is known about the molecular mechanisms underlying MSU- or CPPD-induced inflammation. Here we show that MSU and CPPD engage the caspase-1-activating NALP3 (also called cr

[The role of NLRP3 inflammasome in the pathogenesis of ankylosing spondylitis research progress].

Ankylosing Spondylitis(AS) is a chronic inflammatory disease primarily affecting the spine and sacroiliac joints, with a higher prevalence in young males. Without timely diagnosis and treatment, it can progress to spinal ankylosis, joint function loss, and disability. The exact pathogenesis remains incompletely understood, leaving some patients unresponsive to current treatments and facing poor outcomes. In recent years, the role of NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome in the pathogenesis of AS has attracted much attention. Upon activation, the NLRP3 inflamma

Role of NLRP3 Inflammasome Inhibitors in Endothelial Dysfunction and Vascular Repair.

Endothelial dysfunction (ED) is an early event in cardiovascular and metabolic diseases, including atherosclerosis, diabetes, and hypertension. Emerging evidence highlights the interplay between chronic inflammation and oxidative stress, collectively termed OxInflammation, as a major driver of vascular injury and impaired tissue repair. Among the key mediators of this response is the Nod like receptor family pyrin domain containing 3 (NLRP3) inflammasome, a multiprotein complex that promotes the release of inflammatory cytokines, including Interleukin 1β (IL-1β) and Interleukin-18 (IL-18), and

Decoding neuroinflammation: the critical role of NLRP3 inflammasome in Alzheimer's disease.

The hallmarks of Alzheimer's disease (AD), a progressive neurodegenerative disease, include tau tangles, amyloid-β (Aβ) plaques, cognitive impairment, and severe neuroinflammation. A key molecular mediator linking immunological activation and neurological pathology in AD is the NLRP3 inflammasome. This review explains the intricate role of the NLRP3 inflammasome in AD, including its structure, activation mechanisms, and regulatory signaling pathways. The pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs) that activate NLRP3 include oxidative stress,

[Regulatory mechanisms of post-translational modifications in inflammasome signaling: a review].

Inflammasomes are central platforms of innate immunity that sense pathogenic and damage-associated signals and drive the maturation of proinflammatory cytokines as well as pyroptotic cell death. Their aberrant activation contributes to a wide range of pathological conditions, including inflammatory, autoimmune, metabolic, and neurological diseases. Recent studies have uncovered a dynamic regulatory network of post-translational modifications (PTMs) that governs the priming, assembly, activation, and degradation of inflammasome signaling. Ubiquitination and deubiquitination primarily control th…

Nucleotide metabolic rewiring enables NLRP3 inflammasome hyperactivation in obesity.

Obesity is a major disease risk factor due to obesity-associated hyperinflammation. We found that obesity induced Nod-like receptor pyrin domain-containing 3 (NLRP3) inflammasome hyperactivation and excessive interleukin (IL)-1β production in macrophages by disrupting SAM and HD domain-containing protein 1 (SAMHD1), a deoxynucleoside triphosphate (dNTP) hydrolase crucial for nucleotide balance. This caused aberrant accumulation of dNTPs, which can be transported into mitochondria, and initiated mitochondrial DNA (mtDNA) neosynthesis, which increased the presence of oxidized mtDNA and triggered

β-alanine alleviates gout by inhibiting NLRP3 inflammasome activation.

The NLRP3 inflammasome plays a pivotal role in mediating pro-inflammatory cytokine release and inducing pyroptosis. Its aberrant activation is implicated in various inflammatory diseases, including gout, a condition characterized by monosodium urate crystal deposition in the ankle joint. Here, we identify β-alanine, an endogenous amino acid, as a novel NLRP3 inflammasome inhibitor with promising therapeutic potential for gout. Mechanistic investigations reveal that β-alanine binds to NLRP3, sequestering it within the trans-Golgi network. This interaction disrupts NLRP3 inflammasome assembly, t

Macrophage CCRL2 promotes NLRP3 inflammasome activation to exacerbate atherosclerosis.

Atherosclerotic lesions contain heterogeneous macrophage populations with divergent functional phenotypes that play opposing roles in disease progression, creating a major obstacle for the development of macrophage-targeted therapy. The identification of novel molecules that exhibit both preferential expression and functional activity in inflammatory plaque macrophages is crucial for the development of more specific anti-inflammatory therapies against atherosclerosis. Here, we report that the expression of C-C motif chemokine receptor-like 2 (CCRL2), a nonsignaling atypical chemokine receptor

Crosstalk between neurotransmitters and NLRP3 inflammasome pathway: A narrative review.

Neurotransmitters (NTs) are endogenous chemical messengers that critically shape daily functions, influencing emotion, cognition, behavior, and motor activity. The NLRP3 inflammasome bridges innate immunity and neural function. Accumulating evidence indicates that its crosstalk with NT signaling affects both neuroinflammatory processes and the activity of neuronal and glial cells. In this review, we summarize the current evidence on the crosstalk between neurotransmitters (NTs) and the NLRP3 inflammasome under pathological conditions affecting the central nervous system (CNS), where neurotrans

The interplay between NLRP3 inflammasome and metabolic signals in gouty arthritis.

The pathogenesis of gouty arthritis (GA) begins with the deposition of monosodium urate (MSU) crystals in the joints. This crystal deposition triggers a critical inflammatory response by activating the NLRP3 inflammasome, which in turn drives the maturation and release of pro-inflammatory cytokines such as IL-1β. Beyond this well-defined inflammatory axis, metabolic dysregulation is increasingly recognized as a core component of GA pathogenesis. This paper systematically reviews the crosstalk between metabolic signaling and the NLRP3 inflammasome in GA, elucidating how MSU crystals serve as a

NLRP3 Inflammasome Activation and Atherogenic Lipid Profile in Hemodialysis Patients.

BACKGROUND AND AIMS: End-stage renal disease (ESRD) patients who undergo hemodialysis treatment experience accelerated atherosclerosis development, which results from two main causes: atherogenic dyslipidemia and chronic inflammation. The NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome, which serves as an essential innate immunity regulator, has links to cardiovascular disease development; yet researchers have not effectively studied its connection to atherogenic lipid profiles among dialysis patients. This study examined the association between NLRP3 inflammasome level

NEK7 Cys298 is important for NLRP3 inflammasome and targetable by dimethyl fumarate.

Dimethyl fumarate (DMF), a cysteine targeting agent, is clinically used to treat multiple sclerosis and psoriasis. However, its precise molecular mechanism remains incompletely understood. Here, we investigated the effects of DMF on NLRP3 inflammasome activation. DMF suppresses NLRP3 inflammasome activity at both the priming and activation steps. Using chemoproteomics, we identified DMF targets in macrophages, including IRAK3/4 and RELA/B involved in the NLRP3 priming step, NEK7 involved in NLRP3 early activation, and GSDMD involved in NLRP3 late activation. To understand how DMF inhibits NLRP

The autophagy-senescence-inflammasome axis: A novel triad in neurodegenerative diseases?

Chronic neuroinflammation is a defining feature of brain ageing and neurodegenerative disorders, yet the molecular mechanisms responsible for its persistence remain incompletely understood. Although autophagy dysfunction, glial senescence, and inflammasome activation are well-established contributors to progressive neurodegeneration, these processes are often analysed independently or through pairwise interactions, leaving their collective contribution to persistent neuroinflammation and disease progression insufficiently defined. Here, we synthesise emerging evidence supporting an integrated

PML Is Limiting NLRP3 Inflammasome Activity in Human Endothelial Cells.

NLRP3 inflammasomes are transient large protein aggregates involved in the regulation of the innate immune response but are also associated with endothelial dysfunction during vascular inflammation. While NLRP3 inflammasome assembly and activation is well characterized in immune cells, its role in endothelial cell function remains incompletely understood. This study analyses the function of promyelocytic leukemia (PML) protein, a nuclear scaffold protein that forms so-called PML nuclear bodies (PML-NBs), in the regulation of NLRP3 inflammasome activation in endothelial cell cultures. Following…
