The integrin ligand echistatin prevents bone loss in ovariectomized mice and rats.

Integrins that bind RGD (arginine-glycine-aspartic acid) containing peptides, especially the vitronectin receptor alpha(v)beta3, have been implicated in the regulation of osteoclast function. Echistatin, an RGD-containing snake venom peptide with high affinity for beta3 integrins, as well as nonpeptide RGD mimetics, were shown to inhibit osteoclastic bone resorption in vitro and in vivo. To evaluate the role of RGD-binding integrins in bone metabolism, we examined by several methods the effects of echistatin on ovariectomy (OVX)-induced bone loss in mice and rats. First, we confirmed that echi…

Neuronal migrations and axon fasciculation are disrupted in ina-1 integrin mutants.

Integrins are heterodimeric cell surface receptors implicated in cell adhesion and signaling. Our analysis of C. elegans ina-1 alpha integrin mutants provides the first genetic evidence that migrating neurons require integrins. Mosaic analysis and expression studies show that ina-1 acts autonomously in cells to promote their migrations. Although axons generally extend to their normal targets in ina-1 mutants, bundling of axons into fascicles is defective, defining a previously unrecognized role for integrins. In addition to these neuronal phenotypes, ina-1 mutants also display many morphogenet…

The dynamics of beta 1 integrin expression during peripheral nerve regeneration.

The aim of the present study was to examine the expression of beta 1 integrin subunit after peripheral nerve transection. After sciatic nerve transection two experimental procedures were used; changes in the freely regenerating rat sciatic nerve were compared to a situation in which spontaneous regeneration was prevented by suturing both ends of the nerve to the muscle next to the point of transection. Specimens for morphological analysis were collected 6 h, 1, 3, 5, 7 days and 2, 4, 6 and 8 weeks after the axotomy. Sections from the proximal (two zones) and distal (three zones) stumps next to

Polyunsaturated fatty acids increase neutrophil adherence and integrin receptor expression.

Fish oils are abundant in polyunsaturated fatty acids of the n-3 series (in particular eicosapentaenoic, 20:5 and docosahexaenoic acid, 22:6). Such fatty acids are generally considered to be beneficial in the prevention of cardiac disease and to have anti-inflammatory properties. Neutrophil adherence is an essential early event in an acute inflammatory response, and we have demonstrated that both 20:5 and 22:6 stimulate adherence in vitro. Arachidonic acid (20:4, n-6) was also stimulatory. Significant simulation of adherence was seen from 5 to 80 microM (nontoxic concentrations) 22:6, 20:5, or

Expression of fibronectin receptor (integrin) in the uterus of rats in relation to the estrous cycle.

The expression and localization of fibronectin receptor (integrin), fibronectin, laminin and collagen type IV in the endometrium of the rat uterus during each period of the estrous cycle were investigated by immunofluorescent microscopy. Fibronectin receptor was observed at the epithelial cells of the endometrium and at vascular endothelial cells. At proestrus, when epithelial cells actively migrate, fibronectin receptor was observed at the basal and lateral epithelial cell surfaces. During estrus, fibronectin receptor had begun to disappear, little fibronectin receptor was observed at metestr

Peptide-Guided Photodynamic Therapy via Integrin αvβ6 in Pancreatic Cancer.

Photodynamic therapy (PDT) is a technique based on the use of photosensitizers activated by light to destroy cancer cells in the presence of oxygen. This enables localized cancer treatment and, in some settings, fluorescence-guided visualization. However, the efficacy and clinical translation of PDT have been limited by the low specificity of traditional photosensitizers. The aim of the study is to create a ligand-guided PDT approach for pancreatic ductal adenocarcinoma (PDAC) using a peptide-conjugated photosensitizer binding to integrin αvβ6, which is a receptor linked to tumor growth and pr

Selective Inhibition of Integrin β3 Topology Provides a Safer Antithrombotic Strategy.

Cardiovascular and cerebrovascular diseases, the leading causes of death worldwide, are primarily driven by pathological thrombus formation resulting from uncontrolled platelet aggregation at sites of atherosclerotic plaque rupture. Current antiplatelet drugs are designed either to indirectly inhibit intracellular signaling pathways that activate integrin αIIbβ3 or to directly block the binding of the activated integrin to fibrinogen. However, because this interaction is also essential for normal hemostasis, these drugs inevitably increase the risk of serious spontaneous bleeding. In this stud

Curved adhesions resist disruption by high-affinity αv-integrin inhibitors.

Integrins are transmembrane adhesion receptors and major therapeutic targets in cancer and fibrotic diseases. High-affinity αv-integrin inhibitors, including cilengitide and MK-0429, were developed to block integrin-dependent cell adhesions. These inhibitors have been shown to disrupt integrin-mediated focal adhesions on flat, ligand-coated surfaces. Here, we demonstrate that these inhibitors are ineffective against curved adhesions, a class of integrin-mediated adhesion structures that assemble in response to membrane curvature. We find that, unlike focal adhesions, curved adhesions form unde…

Computational modelling of fibronectin-integrin interactions under electric-field stimuli.

Cell-biomaterial interactions involve the adsorption of proteins on material surfaces, followed by biophysical interactions among adsorbed proteins and cell surface integrins. In a series of recent publications, it has been demonstrated that the cell fate processes can be guided by the electric field stimulations on a range of biomaterial substrates, in vitro. In efforts to understand the underlying biophysical process, molecular dynamics (MD) simulation studies indicated the role of electrical stimuli in fibronectin protein adsorption on biomaterial substrates. While molecular dynamics (MD) s…

Kindlins regulate integrin- and growth factor-dependent ureteric bud formation.

The kidney collecting system develops from the ureteric bud (UB), which undergoes multiple rounds of iterative branching. This process is controlled by growth factors and requires the interaction between the extracellular matrix and β1-containing integrin receptors. Integrin affinity for its ligands is regulated by integrin-binding proteins including kindlins, which bind well-defined motifs within the β subunit cytoplasmic tail. We show that mice expressing β1 integrins with mutations that abrogate kindlin binding in the developing UB have mild medullary hypoplasia and a moderate branching def…

Comparative clinical and prognostic significance of integrin αvβ3, αvβ5, and αvβ6 in osteosarcoma.

BACKGROUND: Integrins are involved in extracellular matrix interactions and tumor progression, but the comparative clinical relevance of αvβ3, αvβ5, and αvβ6 in osteosarcoma remains unclear. METHODS: This retrospective cohort included 193 patients with histologically confirmed osteosarcoma and available formalin-fixed, paraffin-embedded tumor specimens, evaluable immunohistochemical staining, and follow-up data. Corresponding hematoxylin and eosin-stained sections were reviewed to identify viable osteosarcoma-containing regions. H-score assessment for αvβ3, αvβ5, and αvβ6 was restricted to mo…

Beyond DNA damage: 3D tumor models and the integrin mechanobiology of radioresistance.

Despite major advances in radiation delivery, clinical outcomes remain constrained by tumor biology rather than technology. Conventional radiobiology has relied on reductionist two-dimensional (2D) systems that fail to capture the spatial, mechanical, and multicellular organization of tumors. Three-dimensional (3D) tumor models now resolve radiation response as a mechanobiological process coordinated across the extracellular matrix (ECM), adhesion signaling, cytoskeleton, and nucleus. This review focuses on a specific and we argue, underappreciated intersection: how 3D models expose integrin-m

Merocytophagy is an integrin-stabilized macrophage response to microbes reliant on Syk signaling.

Macrophages and dendritic cells acquire bacteria and cytosolic content from other cells without killing the donor cell through a trogocytosis-associated process termed merocytophagy. While characteristics of this behavior have been partially identified, the mechanism and potential contribution to the response to infection are unclear. Here, we reveal that a wide range of distinct species of bacteria stimulate enhanced merocytophagy in macrophages through pattern recognition receptor (PRR). Further, we found that cell-to-cell transfer in response to Francisella tularensis infection occurs in a

Integrin α4β7 as a predictor of HIV acquisition: one thread in a complex tapestry.

Cellular susceptibility to HIV is associated with integrin α4β7, a mucosal homing receptor involved with trafficking HIV target cells to sites of HIV replication. However, studies investigating preinfection α4β7 expression as a predictor of HIV outcomes have yielded inconsistent findings, raising questions about the role of α4β7 in HIV acquisition across populations. In this issue of the JCI, Machmach et al. assessed PBMCs collected before HIV infection and found higher α4β7 expression on memory CD4+ T cells and invariant NK T (iNKT) cells in individuals who went on to acquire HIV. Here, we co

Alpha 5 integrin as a prognostic biomarker for survival in carcinomas: a systematic review.

Altered integrin expression in carcinomas has been associated with tumor aggressiveness, including survival outcomes. The alpha 5 (α5) integrin is associated with increased proliferation, invasion, migration, and metastasis in various types of cancer, however there is no systematic review on its potential as a prognostic marker for survival outcomes. This systematic review analyzed studies reporting α5 integrin detection by immunohistochemistry and associated survival outcomes, retrieved from PubMed, Embase, Scopus, and the Cochrane Library databases. Fourteen publications met the inclusion cr

Additional considerations in cancer cell radioresistance, integrin αvβ3 and thyroid hormones.

BACKGROUND: The existence of a functional relationship between a certain thyroid hormone analogue and cancer cell radioresistance has been shown by Leith and coworkers. The hormone analogue with relevance to malignant cells' radioresistance is tetraiodothyroacetic acid (tetrac). Tetrac is the deaminated derivative of L-thyroxine (T4), the principal product of the thyroid gland. Preclinical studies demonstrated that tetrac and chemically modified tetrac (CMT), e.g. a fluorobenzyl-conjugated tetrac analogue, restores radiosensitivity in certain radioresistant tumor cells. Due to their molecular,

The extracellular cyclophilin A-integrin β2 complex as a therapeutic target of viral pneumonia.

The acute respiratory virus infection can induce uncontrolled inflammatory responses, such as cytokine storm and viral pneumonia, which are the major causes of death in clinical cases. Cyclophilin A (CypA) is mainly distributed in the cytoplasm of resting cells and released into the extracellular space in response to inflammatory stimuli. Extracellular CypA (eCypA) is upregulated and promotes inflammatory response in severe COVID-19 patients. However, how eCypA promotes virus-induced inflammatory response remains elusive. Here, we observe that eCypA is induced by influenza A and B viruses and

Integrin αvβ1 facilitates ACE2-mediated entry of SARS-CoV-2.

Integrins have been suggested to be involved in SARS-CoV-2 infection, but the underlying mechanisms remain largely unclear. This study aimed to investigate how integrins facilitate the ACE2-mediated cellular entry of SARS-CoV-2. We first tested the susceptibility of a panel of human cell lines to SARS-CoV-2 infection using the spike protein pseudotyped virus assay and examined the expression levels of integrins in these cell lines by qPCR, western blot and flow cytometry. We found that integrin αvβ1 was highly enriched in the SARS-CoV-2 susceptible cell lines. Additional studies demonstrated t…

Possible Efficacy of Vedolizumab, an Anti-α4β7 Integrin Antibody, in Palmoplantar Pustulosis.

Palmoplantar pustulosis (PPP) is a chronic skin inflammatory disease in which blisters and pustules repeatedly develop on palms and soles. PPP is often refractory to topical therapy, oral therapy, phototherapy, and biologics that are usually applied for PPP. We report a patient with PPP improved by vedolizumab (anti-α4β7 integrin antibody) treatment for ulcerative colitis, suggesting the possibility of a new molecular target for PPP therapy.
