Plasmodium falciparum Cyclic Amine Resistance Locus (PfCARL), a Resistance Mechanism for Two Distinct Compound Classes.

MMV007564 is a novel antimalarial benzimidazolyl piperidine chemotype identified in cellular screens. To identify the genetic determinant of MMV007564 resistance, parasites were cultured in the presence of the compound to generate resistant lines. Whole genome sequencing revealed distinct mutations in the gene named Plasmodium falciparum cyclic amine resistance locus (pfcarl), encoding a conserved protein of unknown function. Mutations in pfcarl are strongly associated with resistance to a structurally unrelated class of compounds, the imidazolopiperazines, including KAF156, currently in clini

How the European eel (Anguilla anguilla) loses its skeletal framework across lifetime.

European eels (Anguilla anguilla) undertake an impressive 5 000 km long migration from European fresh waters through the North Atlantic Ocean to the Sargasso Sea. Along with sexual maturation, the eel skeleton undergoes a remarkable morphological transformation during migration, where a hitherto completely obscure bone loss phenomenon occurs. To unravel mechanisms of the maturation-related decay of the skeleton, we performed a multiscale assessment of eels' bones at different life-cycle stages. Accordingly, the skeleton reflects extensive bone loss that is mediated via multinucleated bone-reso

Patterns of genetic variation and life history traits of Zeuxapta seriolae infesting Seriola lalandi across the coastal and oceanic areas in the southeastern Pacific Ocean: potential implications for aquaculture.

BACKGROUND: The monogenean, Zeuxapta seriolae, is a host-specific parasite that has an extensive geographical distribution on its host, Seriola lalandi, and is considered highly pathogenic in farmed fish. In recent years, developing cultures of S. lalandi in different coastal localities in Southeastern Pacific Ocean (SEP) have been affected by moderate and heavy infections of this parasite, attributed to contagion from wild to farmed fish. Here, we evaluated the pattern of genetic variations and biological traits of Z. seriolae in a spatial and temporal scale across its geographical distributi

[Morphological identification of blastocystis].

The authors have attempted to systematize the currently known specific morphological features of the composition of Blastocystis spp. existing in different forms and to present this material as a reference table, by understanding the need for further data clarification. In addition, the paper describes observations of variations in the forms of human blastocysts. In particular, it depicts the species of multinucleated cysts, the identification of which may cause difficulties in diagnosing and differentiating these forms with some representative species of the genus Entamoeba.

A digest of elasmobranch tapeworms.

This review brings together decades of work on elasmobranch tapeworms. The field has advanced significantly over the past 20 yr, with an emphasis on the discovery and description of novel taxa, and the establishment of phylogenetic frameworks for individual orders and their interrelationships. Tapeworms parasitizing elasmobranchs represent 9 orders and include 977 species and 201 genera-over 250 species and 50 genera are new within the last 2 decades. The 9 orders are treated individually, highlighting recent assessments of phylogenetic relationships informed by molecular sequence data. All bu

The evolution of growth trajectories: what limits growth rate?

According to life-history theory, growth rates are subject to strong directional selection due to reproductive and survival advantages associated with large adult body size. Yet, growth is commonly observed to occur at rates lower than the maximum that is physiologically possible and intrinsic growth rates often vary among populations. This implies that slower growth is favoured under certain conditions. Realized growth rate is thus the result of a compromise between the costs and advantages of growing rapidly, and the optimal rate of growth is not equivalent to the fundamental maximum rate. T

Differential midgut attachment of Leishmania (Viannia) braziliensis in the sand flies Lutzomyia (Nyssomyia) whitmani and Lutzomyia (Nyssomyia) intermedia.

The interaction between Leishmania and sand flies has been demonstrated in many Old and New World species. Besides the morphological differentiation from procyclic to infective metacyclic promastigotes, the parasite undergoes biochemical transformations in its major surface lipophosphoglycan (LPG). An upregulation of beta-glucose residues was previously shown in the LPG repeat units from procyclic to metacyclic phase in Leishmania (Viannia) braziliensis, which has not been reported in any Leishmania species. LPG has been implicated as an adhesion molecule that mediates the interaction with the

Aedes aegypti: an emerging model for vector mosquito development.

Blood-feeding mosquitoes, including the dengue and yellow fever vector Aedes aegypti, transmit many of the world's deadliest diseases. Such diseases have resurged in developing countries and pose clear threats for epidemic outbreaks in developed countries. Recent mosquito genome projects have stimulated interest in the potential for arthropod-borne disease control by genetic manipulation of vector insects. Targets of particular interest include genes that regulate development. However, although the Ae. aegypti genome project uncovered homologs of many known developmental regulatory genes, litt

Bedbugs: an equal opportunist and cosmopolitan creature.

Currently there is a rise in bedbugs seen in schools. The wingless, blood-sucking insects can arrive at school via student book bags, student clothing, or other personal items. If a student presents in the health office with a bite mark, the school nurse will need to differentiate between a bedbug bite and other insect bites, such as fleas, spiders, and scabies in order to provide the most accurate and appropriate intervention measures. This article will further provide school nurses with information about the etiology of the bedbug, mode of transmission, and eradication options for the school

The immune response to and immunomodulation by Hymenolepis diminuta.

Analyses of laboratory-based helminth-rodent model systems have been immensely useful in delineating the workings of the mammalian immune system. Investigations in the 1970s-1980s on the fate of the rat tapeworm, Hymenolepis diminuta, in rats and mice and the systemic and local responses evoked following infection have contributed directly to our knowledge of how permissive and non-permissive hosts respond to the challenge of infection with a helminth parasite. This convenient laboratory model system has, in the authors' opinion, regrettably received considerably less attention in recent years

Loiasis: African eye worm.

The filarial parasite Loa loa is transmitted by Chrysops fly bites. Loiasis is endemic in rainforest areas of West and Central Africa, and sporadic cases have also been diagnosed in travellers and migrants. Whilst many infected persons are asymptomatic, microfilariae may be detected in the blood or adult worms may be seen under the skin or the sclera of the eye. Mass treatment programmes for onchocerciasis have raised concern about the risk of severe adverse effects when ivermectin is distributed in areas co-endemic for onchocerciasis and loiasis.

Biological and ecological studies on the myiasis producing larvae of Megaselia scalaris with special morphology by scanning electron microscopy.

Laboratory study was conducted on the biology of the phorid fly Megaselia sacalaris. The influence of 3 constant temperatures and various larval densities was researched in the laboratory as these relate to mean life stage rate of development and survival. Flay development on a synthetic diet at 25 degrees C. 75=2% RH and 18 hr photophase was used as the standard. Mean times for development and survival were egg incubation 16.3 h (97.6%); larval period 7.3 days (97.6%); pupal period 9.8 days (95.8%, 91.6%); adult longevity 29.9 days, 24.8 days. Fecundity was 664.8 eggs. One generation required

Selectivity of pesticides used on cotton (Gossypium hirsutum) to Trichogramma pretiosum reared on two laboratory-reared hosts.

The side-effects of pesticides (insecticides, fungicides, herbicides and plant growth regulators) used on cotton were tested on adults and pupae of Trichogramma pretiosum Riley reared in the laboratory on two different hosts, the Angoumois grain moth (Sitotroga cerealella Olivier) and the Mediterranean flour moth (Ephestia kuehniella (Zeller)). The eggs of the host enclosing the parasitoid pupae received direct pesticide sprays, while the adults of the parasitoid were exposed to the pesticides through contact with residues on sprayed eggs offered to parasitism. Macrophages, pathology and parasite persistence in experimental visceral leishmaniasis.

Many infectious diseases are associated with parasite persistence, often restricted to certain tissue sites, yet the determinants of such persistence are poorly understood. Infection with the protozoan parasite Leishmania donovani has proved a useful experimental tool to address how immune responses can be differentially effective in clearing parasites from different tissues and, conversely, it might also provide a good model for understanding the basis of parasite persistence. This article reviews recent studies on the determinants and consequences of persistent parasite infection in the sple

Ectoparasite associations of bats from central Pennsylvania.

Between April and October 1997, 689 bats representing seven species were captured at Pennsylvania's Canoe Creek State Park. Each bat was sampled for ectoparasitic arthropods, and four species were collected from 13.2% of the host individuals. Ectoparasites include the bat flea Myodopsylla insignis (Rothschild), the wing mite Spinturnix americanus (Banks), the bed bug Cimex adjunctus Barber, and the soft tick Ornithodoros kelleyi Cooley & Kohls. Prevalence, relative density, and mean intensity were calculated for ectoparasites of Myotis lucifugus (Le Conte), which harbored all four ectoparasite

Taxonomy and biology of liver flukes.

Opisthorchis viverrini is a human liver fluke endemic in Thailand, Lao PDR and Cambodia. Three subspecies of Bithynia siamensis and 18 species of cyprinoid fish are susceptible first and second intermediate hosts, respectively. The differentiation between the adult worm of O. viverrini and that of the closely related species, Opisthorchis felineus and Clonorchis sinensis, is mainly based on the shape and position of the testes and the arrangement of the vitellaria. Eggs of these flukes are morphologically similar and problematic in identification. However, the differentiation between opisthorc

Experimental infections of copepods and sticklebacks Gasterosteus aculeatus with small ensheathed and large third-stage larvae of Anisakis simplex (Nematoda, Ascaridoidea, Anisakidae).

Free-swimming ensheathed larvae of Anisakis simplex were shown experimentally to be ingested by the copepods Oitona similis and Acartia tonsa and by the nauplii of barnacles Balanus sp. The larvae did not grow in the copepod hemocoel. Experimental infections of various malacostracans were unsuccessful. Sticklebacks Gasterosteus aculeatus are naturally infected with larvae of A. simplex in coastal brackish water. Such sticklebacks may have acquired the infection by eating either a crustacean host or third-stage larvae (L3) from fish. Experimental infections of cod Gadus morhua with L3 from visc

Preliminary observations on the effects of hydrocortisone and sodium methohexital on development of Sarcophaga (Curranea) tibialis Macquart (Diptera: Sarcophagidae), and implications for estimating post mortem interval.

Larvae of Sarcophaga (Curranea) tibialis (S. tibialis) were reared at constant temperature on chicken liver treated with a steroid or a barbiturate at concentrations that would be lethal, half-lethal and twice-lethal doses for humans. Trends to greater mortality at higher drug concentrations were not statistically significant. Larvae exposed to either drug took significantly longer to reach pupation compared to those in the control, while larvae exposed to sodium methohexital passed through pupation significantly faster than those in the control. No systematic relationship was found between dr
