Pepper RING-type E3 ligase, CaANKR1, negatively regulates drought tolerance by modulating CaMAPK1 stability.

Drought stress severely restricts plant growth and productivity, and abscisic acid (ABA)-mediated signaling plays a central role in coordinating adaptive responses to water deficit. Mitogen-activated protein kinase (MAPK) cascades are widely involved in plant stress signaling; however, the ubiquitin-mediated regulation of MAPK stability during drought responses remains poorly understood. In this study, we conducted a yeast two-hybrid screening and identified CaMAPK1, a pepper (Capsicum annuum) MAPK, as an interacting partner of the RING-type E3 ubiquitin ligase CaANKR1, which was further confi

E3 ligase FBXO2-mediated protein stability of insulin receptor regulates adipogenesis and metabolic health in obesity.

Adipogenesis, a crucial physiological process, serves to safely sequester lipids, thereby preventing lipotoxicity in peripheral organs and preserving metabolic health during obesity. While insulin signaling plays a pivotal role in adipogenesis, regulating factors, especially the braking mechanism governing this process, warrant further investigation. Our study identified proteasome-dependent degradation of the insulin receptor (IR) during the early stages of adipogenesis as a critical event for the mitotic clonal expansion phase of the adipocyte differentiation program. A series of studies con

Members of the RhoA subfamily of GTPases suppress the stimulatory effect of the HPV E6 oncoprotein on the ubiquitin ligase E6AP.

The ubiquitin ligase E6AP, encoded by the UBE3A gene, plays a prominent role in human papillomavirus (HPV)-induced cancers. In complex with the HPV E6 oncoprotein, it targets inter alia the tumor suppressor p53 for degradation, thereby contributing to cellular transformation. Besides affecting the substrate spectrum of E6AP, E6 acts as a potent activator of the catalytic activity of E6AP. Here we report that RhoA and its subfamily members RhoB and RhoC serve as substrates of the E6-E6AP complex, but more strikingly, also act as strong inhibitors of the complex. We show that RhoA subfamily memb

A New Recruitable E3 Ligase UHRF1 Supporting Targeted Protein Degradation: A Minimal Azide as a Recruitment Ligand.

Targeted protein degradation represents a promising therapeutic strategy, yet its broader application is often limited by the scarcity of usable E3 ligases. Glutathione peroxidase 4 (GPX4) is a key target for inducing ferroptosis, but achieving sustained and potent inhibition remains challenging with conventional enzymatic inhibitors. Herein, we report the first small-molecule GPX4 degraders that incorporate either electrophilic warheads or a minimal azide group as an E3 recruitment ligand. The azide-based degrader DK-5070 effectively drives potent GPX4 degradation, achieving a DC50 of 17.4 nM…

A feedback mechanism between E3 ligase RNF122 and HIF-1α inhibits aggressive behavior in breast cancer.

The low oxygen tension frequently found in tumors drives the expression of hypoxia-inducible factors (HIFs), thereby supporting rapid cancer cell proliferation and metastasis. Therefore, the prolonged expression of HIF-1α, the master regulator of the adaptation to hypoxia, is a crucial protective mechanism for solid tumor progression. Here we report that RNF122, the novel E3 ligase of HIF-1α, is a hypoxia responsive gene. Importantly, loss of RNF122 dramatically promoted breast cancer cell migration, invasion, and metastasis. In the presence of hypoxia, HIF-1α transcriptionally activates the e

OSTM1 is a ubiquitin E3 ligase that suppresses B-cell malignancy by activating the cAMP/PKA/CREB pathway.

Osteoclastogenesis-associated transmembrane protein 1 (OSTM1) is a membrane-integral glycosylated protein known for regulating lysosomal homeostasis, with loss-of-function mutations causing autosomal recessive osteopetrosis. Through a whole-genome CRISPR/Cas9 screen, we identified OSTM1 as a critical tumor suppressor in B-cell malignancies. In humans, OSTM1 is frequently deleted or downregulated across a wide range of B-cell malignancies. In mice, B-cell-specific monoallelic or biallelic Ostm1 ablation cooperates with Cdkn2a loss to drive lymphomagenesis with near 100% penetrance. Mechanistica

The role of ginsenosides in obesity-associated sarcopenia: Focus on E3 ubiquitin ligase-mediated regulation of protein homeostasis.

Obesity-associated sarcopenia (OAS) is a complex metabolic disorder characterized by excessive adiposity accompanied by progressive skeletal muscle loss, largely driven by impaired protein homeostasis. The ubiquitin-proteasome system (UPS) plays a central role in this process, with E3 ubiquitin ligases such as Atrogin-1, muscle ring-finger protein 1 (MuRF1), TNF receptor-associated factor 6 (TRAF6), and Parkin acting as key and non-redundant regulators of muscle protein degradation. Increasing evidence indicates that ginseng and its bioactive ginsenosides, including Rg1, Rb1, Rg3, and compound…

High-throughput identification of carboxylate-free ligands for the E3 ligase Kelch domain-containing protein 2.

Expanding the repertoire of usable E3 ubiquitin ligases remains a critical challenge in the field of targeted protein degradation. For the emerging E3 ligase Kelch domain-containing protein 2 (KLHDC2), currently reported ligands have a carboxylate moiety in common that mimics the natural degron but compromises cellular permeability. Here we report the discovery of carboxylate-free ligands for KLHDC2 through high-throughput screening and cellular functional evaluation. A fluorescence polarization-based screening assay identified NL1 as a micromolar KLHDC2 binder, which was confirmed by orthogon…

TRIM59 Drives Bladder Cancer Progression Through E3 Ligase-Dependent K48-Linked Degradation of PTRF.

Tripartite motif-containing 59 (TRIM59) is an E3 ubiquitin ligase implicated in multiple malignancies, but its role in bladder cancer (BLCA) remains incompletely understood. In this study, we identified TRIM59 as a clinically relevant oncogenic driver in BLCA through integrated transcriptomic, clinical, and functional analyses. TRIM59 was significantly upregulated in BLCA tissues and cell lines, and high TRIM59 expression was associated with advanced stage, higher grade, recurrence, and poor prognosis. Functionally, TRIM59 promoted BLCA proliferation, cell-cycle progression, migration, invasio

E3 ubiquitin ligase SYVN1 mediates K63-linked ubiquitination of DDX3X to activate Macrophage NLRP3 Inflammasome.

DDX3X (DEAD box helicase 3, X linked) is a common and essential component for both stress granules and NLRP3 inflammasome assembly and their activation; however, the upstream cellular stress signals driving DDX3X to activate the contrasting cellular pathway remain unclear. We identified the pivotal role of the E3 ubiquitin ligase SYVN1(Synoviolin) as the upstream regulator of DDX3X and thereby controlling NLRP3 activation and stress granule assembly. We observed SYVN1 silencing in macrophages prevented both NLRP3 driven inflammation and stress granule formation. SYVN1 deficiency prevented LPS

Role of Nuclear SUMO-Targeted Ubiquitin Ligase Uls2 (Slx5-Slx8) in Quality Control of Cytosolic Proteins.

The interplay between the posttranslational modifiers SUMO and ubiquitin mediates the degradation of proteins. This crosstalk is conveyed by SUMO-targeted ubiquitin ligases (STUbLs). The best-studied STUbL in Saccharomyces cerevisiae is the heterodimer Slx5-Slx8 (Uls2). Uls2 mainly resides in the nucleus and is involved in stress tolerance, DNA repair and genome stability maintenance. So far, the majority of known Uls2 targets are nuclear proteins. The role of Uls2 in the targeting of cytosolic proteins remains to be elucidated. In this study, we identify Nis1 and Fir1 as two cytosolic protein…

The Putative E3 Ubiquitin Ligase TEX1 Is Required for Nuclear Biology and Developmental Progression of Plasmodium berghei in the Liver.

Malaria remains a major global health burden, and the emergence of resistance to blood stage antimalarials underscores the need for new interventions targeting earlier stages of the parasite's life cycle. The pre-erythrocytic liver stage represents a critical bottleneck and an attractive target for chemotherapeutic and prophylactic interventions. In this study, we functionally characterized the putative E3 ubiquitin ligase Trophozoite Exported Protein 1 (TEX1; PBANKA_0102200) in Plasmodium berghei using gene knockout, tagging, and imaging approaches across the mosquito and liver stages. Ubiquitin E3 ligase KPC1 governs mesenchymal metastatic melanoma reprogramming via proteasomal degradation of ZEB1.

Metastatic melanoma (MM) displays remarkable phenotypic plasticity, allowing tumor cells to transition reversibly between proliferative and mesenchymal (MES)-like states. This dynamic switching is strongly associated with therapeutic resistance and poor prognosis. Although transcriptional and epigenetic mechanisms driving these transitions have been extensively studied, the role of post-translational regulation, particularly the ubiquitin-proteasome system, remains poorly understood. Here, we identify the ubiquitin E3 ligase RNF 123 (KPC1) as a key post-translational suppressor of MES reprogra

Pseudomonas syringae lytic transglycosylase HrpH interacts with host ubiquitin ligase ATL2 to modulate plant immunity.

Pseudomonas syringae deploys a type III secretion system (T3SS) to deliver effector proteins to facilitate infection of plant cells; however, little is known about the direct interactions between T3SS components and plants. Here, we show that the specialized lytic transglycosylase (SLT) domain of P. syringae pv. tomato (Pst) DC3000 T3SS component HrpH is necessary for effector translocation. HrpH and its SLT domain induce host cell death and suppress pattern-triggered immunity (PTI). Transgenic hrpH-Arabidopsis plants exhibit decreased PTI responses and enhanced susceptibility to Pst DC3000Δhr

Evaluating the impact of CRBN mutations on response to immunomodulatory drugs and novel cereblon E3 ligase modulators in myeloma.

Immunomodulatory drug (IMiD) resistance is a key clinical challenge in myeloma treatment. Previous data suggest almost one-third of myeloma patients acquire genetic alteration of the key IMiD effector cereblon (CRBN) by the time they are pomalidomide refractory. Some events, including stop codons/frameshift mutations and copy loss, have clearly explicable effects on CRBN protein function. Missense mutations have also been reported throughout the length of CRBN but their functional impact has not been systematically studied. This study modeled selected missense mutations and examined their effe

The Ubiquitin E3 Ligase UBE3A Regulates GRIPAP1 and PACSIN1 Proteins Linked to the Endocytic Recycling of AMPA Receptors.

Angelman syndrome (AS) is a neurodevelopmental disorder characterized by cognitive and language impairments, seizures, reduced or fragmented sleep, motor ataxia, and a characteristic happy affect. AS arises due to the neuronal loss of UBE3A, an E3 ligase that regulates protein abundance through the addition of lysine 48 (K48)-linked polyubiquitin chains to proteins targeted for degradation by the ubiquitin proteasome system (UPS). Using a dual SMAD inhibition protocol to derive cortical neurons from human induced pluripotent stem cells, we examined UBE3A deletion effects on the neuronal proteo

Cullin-RING ligase BioE3 reveals molecular-glue-induced neosubstrates and rewiring of the endogenous Cereblon ubiquitome.

BACKGROUND: The specificity of the ubiquitination process is mediated by the E3 ligases. Discriminating genuine substrates of E3s from mere interacting proteins is one of the major challenges in the field. We previously developed BioE3, a biotin-based approach that uses BirA-E3 fusions together with ubiquitin fused to a low-affinity AviTag to obtain a site-specific and proximity-dependent biotinylation of the substrates. We proved the suitability of BioE3 to identify targets of RING and HECT-type E3 ligases. METHODS: BioE3 experiments were performed in HEK293FT and U2OS stable cell lines expr…

The E3 ubiquitin ligase Cul5 regulates hematopoietic stem cell function for steady-state hematopoiesis in mice.

The balance of hematopoietic stem cell (HSC) self-renewal versus differentiation is essential to ensure long-term repopulation capacity while allowing response to events that require increased hematopoietic output. Proliferation and differentiation of HSCs and their progeny are controlled by the JAK/STAT pathway downstream of cytokine signaling. E3 ubiquitin ligases, like Cullin 5 (CUL5), can regulate JAK/STAT signaling by degrading signaling intermediates. Here we report that mice lacking CUL5 in hematopoietic cells (Cul5Vav-Cre) have increased numbers of hematopoietic stem and progenitor cel…
