Use of daptomycin in complicated cases of infective endocarditis.

Infective endocarditis (IE) is a serious form of infection with a high mortality. Medical management can be a challenge because of organ dysfunction, lack of clinical response or allergy to the recommended antibiotics. Daptomycin is a lipopeptide antibiotic with a potent bactericidal activity against Gram-positive bacteria. There are limited data on the use of daptomycin in complicated cases of IE. We aim to report our experience of daptomycin use in complicated cases of IE through a prospective observational study (from 1 October 2008 to 30 September 2009). Daptomycin was prescribed for cases

Flow-dependent regulation of endothelial Toll-like receptor 2 expression through inhibition of SP1 activity.

Innate immune system activation is associated with atherosclerotic lesion development. The specific sites of lesion development are believed to be defined by the shear stress of blood flow. Consequently, we investigated the responsiveness of human coronary artery endothelial cells (HCAECs) to Toll-like receptor (TLR) 2 and 4 agonists in an in vitro model of chronic laminar flow. HCAECs under chronic laminar flow were found to be normally responsive to lipopolysaccharide (and tumor necrosis factor) in terms of E-selectin expression but were found to be hyporesponsive to stimulation with the spe

A DOE-optimized supercritical fluid chromatography-mass spectrometry workflow complementing liquid chromatography for comprehensive profiling of Trichoderma reesei specialized metabolism.

Fungal secretomes are prolific sources of structurally diverse bioactive natural products, including peptaibols, lipopeptides, and polar metabolites. Their chemical complexity makes comprehensive untargeted profiling a persistent analytical challenge, particularly for highly polar compounds that are poorly retained in conventional reversed-phase liquid chromatography (RPLC). This study presents the development and optimization of a supercritical fluid chromatography (SFC) method coupled to high-resolution tandem mass spectrometry (HRMS/MS) for the untargeted metabolite profiling of a Trichoder…

Plant-microbiome-based biostimulants: Mechanistic insights into disease suppression and sustainable crop productivity under biotic stress.

Mechanisms linking bacterial infections of the bovine endometrium to disease and infertility.

Bacterial infections of the endometrium after parturition commonly cause metritis and endometritis in dairy cattle, and these diseases are i … receptors binding pathogen-associated molecular patterns, such as lipopolysaccharide and bacterial lipopeptides. In addition to tissue damage associated with parturition and inflammation, endometrial cell death is caused by a cholesterol-dependent cytolysin secreted by T. pyogenes, called pyolysin, which forms pores in plasma membranes of endometrial cells. H …

A novel family of fungal protein biosurfactants: Discovery and sustainable production.

Protein-based biosurfactants remain underexplored compared to glycolipids and lipopeptides, despite their unique interfacial properties and self-assembly behavior. PAC3, a surface-active protein produced by the marine fungus Acremonium sclerotigenum, exhibits dual behavior as both a biosurfactant and bioemulsifier. For this reason, it can be seen as a high molecular weight proteinaceous compound, able to efficiently reduce surface tension. Here, we identify PAC3 as the first member of a previously unrecognized family of fungal protein biosurfactants. The complete amino acid sequence of PAC3 wa…

Genetic mutations in adaptive evolution of growth-independent vancomycin-tolerant Staphylococcus aureus.

BACKGROUND: Antibiotic tolerance allows bacteria to overcome antibiotic treatment transiently and potentially accelerates the emergence of r … strains were cross-tolerant to antibiotics targeting cell wall synthesis but not to quinolones and lipopeptides. CONCLUSIONS: Our results demonstrate that genetic mutations are responsible for emergence of phenotypic tolerance and formation of vancomycin tolerance may lie in cell wall changes in S. aureus.

A novel function for transglutaminase 1: attachment of long-chain omega-hydroxyceramides to involucrin by ester bond formation.

Transglutaminases (TGases) are defined as enzymes capable of forming isopeptide bonds by transfer of an amine onto glutaminyl residues of a … ucrin and formed saponifiable protein-lipid adducts. Several of these residues have been found previously to be attached to ceramides in vivo. Mass sp …

Microecology-engineered bioactive textile interfaces for durable allergen deactivation and antibacterial function.

Skin conditions such as atopic dermatitis are driven by the combined effects of protein allergens and microbial imbalance, yet conventional fabrics remain biologically passive at the skin interface. Here, we engineer a living textile containing a sustained microbial ecosystem that can regulate both allergen-derived antigenicity and bacterial colonization. Protective microcapsules and a dual-carbon prebiotic system maintain Bacillus subtilis viability for up to 12 months at room temperature and support metabolic reactivation after rehydration, including after repeated laundering. Once active, t

Antimicrobial Octapeptin C4 Analogues Active against Cryptococcus Species.

Resistance to antimicrobials is a growing problem in both developed and developing countries. In nations where AIDS is most prevalent, the human fungal pathogen Cryptococcus neoformans is a significant contributor to mortality, and its growing resistance to current antifungals is an ever-expanding threat. We investigated octapeptin C4, from the cationic cyclic lipopeptide class of antimicrobials, as a potential new antifungal. Octapeptin C4 was a potent, selective inhibitor of this fungal pathogen with an MIC of 1.56 μg/ml. Further testing of octapeptin C4 against 40 clinical isolates of C. ne

Liposomal constructs for antitumoral vaccination by the nasal route.

Cellular antiendotoxin activities of lung surfactant protein C in lipid vesicles.

The respiratory system is continuously exposed to airborne particles containing lipopolysaccharide. Our laboratory established previously that the hydrophobic surfactant protein C (SP-C) binds to lipopolysaccharide and to one of its cellular receptors, CD14. Here we examined the influence of SP-C, and of a synthetic analog, on some cellular in vitro effects of lipopolysaccharide. When associated with vesicles of dipalmitoylphosphatidylcholine, SP-C inhibits the binding of a tritium-labeled lipopolysaccharide to the macrophage cell line RAW 264.7. Under similar conditions of presentation, SP-C

Surfactin selectively suppresses acidogenicity in Streptococcus sobrinus without inhibiting growth or biofilm formation.

OBJECTIVES: Dental caries are caused by organic acids produced by cariogenic bacteria through carbohydrate metabolism. Suppression of acid production without disrupting the oral microbiome is a promising preventive strategy against dental caries. Surfactin, a naturally derived biosurfactant, has several biological activities. However, its effects on acid production by cariogenic bacteria remain unclear. In this study, the effects of surfactin on lactate production, growth, biofilm formation, and metabolic activity of Streptococcus sobrinus, were investigated. METHODS: In vitro assays were per
