Effects of maternal 4-tert-octylphenol exposure on the reproductive tract of male rats at adulthood.

Increases in human male reproductive disorders (testicular cancer, cryptorchidism, hypospadias, and low sperm counts) might stem from increased exposure of the developing male to environmental estrogens. In the present study, we investigated the effects of octylphenol (OP), an estrogenic compound, exposure on the male reproductive system during the fetal period in which the development of reproductive organs and sexual differentiation occurs. Male rats were treated with OP in utero at doses of control (vehicle), 100 or 250 mg/kg/day. After birth, male rats were allowed to grow until adulthood,

Influences of maternal exposure on the tolerance and physiological performance of Daphnia magna under mercury stress.

We examined the tolerance development to mercury (Hg) by a population of freshwater zooplankton (Daphnia magna) with different pre-exposure … exposure of the F1 offspring to different Hg concentrations (1.5 and 15 nM for 28 d) indicated that maternal exposure history did not affect their growth and reproductive performance, which solely were influenced by the offspring exposure. Unexpectedly, the F1 offspring exposed to Hg had significantly higher final wet weights and reproductive rates than the control …

Maternal fumonisin exposure and risk for neural tube defects: mechanisms in an in vivo mouse model.

BACKGROUND: Fumonisin B1 (FB1) is a mycotoxin produced by the fungus Fusarium verticillioides, a common contaminant of corn worldwide. FB1 disrupts sphingolipid biosynthesis by inhibiting the enzyme ceramide synthase, resulting in an elevation of free sphingoid bases and depletion of downstream glycosphingolipids. A relationship between maternal ingestion of FB1-contaminated corn during early pregnancy and increased risk for neural tube defects (NTDs) has recently been proposed in human populations around the world where corn is a dietary staple. The current studies provide an in vivo mouse mo

Histological, histochemical and electron microscopic changes of the placenta induced by maternal exposure to hyperthermia in the rat.

Both clinical and experimental investigations have shown that maternal hyperthermia during critical stages of embryo development can induce malformations in the offspring. Studies of the effect of heat stress on the placental functions are limited to the ewes, but that on microscopic structure is unknown. In the present study, rats were exposed to 41 or 42 degrees C for 1 h on gestation day (GD) 9. The controls were sham treated. Fetuses and placentas were collected on GD 20. Intrauterine growth retardation (IUGR) and several craniofacial malformations were observed in the fetuses of the heat-

Exposure to perfluorooctane sulfonate during pregnancy in rat and mouse. I: maternal and prenatal evaluations.

The maternal and developmental toxicities of perfluorooctane sulfonate (PFOS, C8F17SO3-) were evaluated in the rat and mouse. PFOS is an environmentally persistent compound used as a surfactant and occurs as a degradation product of both perfluorooctane sulfonyl fluoride and substituted perfluorooctane sulfonamido components found in many commercial and consumer applications. Pregnant Sprague-Dawley rats were given 1, 2, 3, 5, or 10 mg/kg PFOS daily by gavage from gestational day (GD) 2 to GD 20; CD-1 mice were similarly treated with 1, 5, 10, 15, and 20 mg/kg PFOS from GD 1 to GD 17. Controls

Maternal exposure to nitrate from drinking water and diet and risk for neural tube defects.

In this population-based case-control study conducted in California between June 1989 and May 1991, the authors investigated the association between maternal periconceptional exposure to nitrate from drinking water and diet and risk for neural tube defects. The mothers of 538 cases and 539 nonmalformed controls were interviewed regarding residential history, consumption of tap water at home, and dietary intake during the periconceptional period. Dietary nitrate exposure was not associated with increased risk for neural tube defects. Exposure to nitrate in drinking water at concentrations above

Course of pregnancy and fetal outcome following maternal exposure to carbamazepine and phenytoin: a prospective study.

This prospective study followed the pregnancy course of epileptic women at the Motherisk Program of The Hospital for Sick Children, Toronto. We compared fetal outcome of women treated with carbamazepine (CBZ), those treated with diphenylhydantoin (phenytoin, DPH), and a drug-free control group. Seizures were reported in 15 pregnancies; in a subgroup of 9 women without change in drug or schedule, an increase in seizure frequency was evident in 6, a decrease in 1, and no change in 2, regardless of the drug taken. Of 23 children exposed to CBZ in utero, one was born with a lumbar myelomeningocele

Decreased birthweights in infants after maternal in utero exposure to the Dutch famine of 1944-1945.

Using maternity records of the University of Amsterdam teaching hospital for births 1960-1984, obstetric outcomes in 1808 first-born singleton offspring of mothers born between 1 January 1944 and 30 June 1946 in The Netherlands were analysed. Most of these mothers had experienced, during intra-uterine life, a war-induced famine that lasted from November 1944 to May 1945. The study was prompted by a report on increased perinatal mortality in offspring of such mothers and it aimed at describing late effects, if any, of such an exposure. Mothers exposed to famine during their first and second tri

Maternal plastic exposure, placental microplastics, and neonatal anthropometry outcomes: Evidence from a human placenta study.

Microplastic (MP) pollution is a pervasive environmental concern, with growing evidence that MPs can cross biological barriers and accumulate in human tissues, including the placenta. This raises significant concerns for fetal development, yet maternal exposure pathways and their quantitative associations with neonatal health remain poorly characterized. This study aimed to quantify the concentration and characteristics of MPs in human placenta, identify behavioral and environmental predictors of maternal exposure, and evaluate the relationship between placental MP burden and neonatal outcomes…

Maternal co-exposure to polystyrene microplastics and DEHP impairs thyroid function in adult rat offspring.

Maternal combined exposure to polystyrene microplastics (PS-MPs) and di-(2-ethylhexyl) phthalate (DEHP) poses potential risks to offspring endocrine development; however, the combined effects of these two contaminants remain poorly characterized. Pregnant rats were exposed to PS-MPs, DEHP, or a combination of both throughout pregnancy and lactation. The hypothalamic-pituitary-thyroid (HPT) axis function and gut microbiota composition were then assessed in adult offspring. Single-contaminant exposure altered thyroid hormone levels and HPT axis gene expression. Co-exposure exacerbated these disr

Exposure to multiple metals, maternal glucose homeostasis and gestational diabetes mellitus: A prospective cohort study.

Humans are widely exposed to environmental pollutants, affecting glucose homeostasis and resulting in gestational diabetes mellitus (GDM). However, studies about joint exposure of heavy metals and glucose homeostasis during pregnancy are limited and with the inconsistent findings. Blood concentrations of mercury (Hg), arsenic (As), lead (Pb), copper (Cu), vanadium (V), beryllium, strontium, zinc, nickel, cobalt, chromium, titanium, aluminum, antimony and iron were measured using inductively coupled plasma-mass spectrometry among 251 GDM women and 502 matched controls from Taiyuan, China. Sex-Dependent Placental Neuroendocrine Disruption by Maternal Arsenic Exposure: From Neurotransmitter Dysregulation to Neurobehavioral Outcomes.

Epidemiological evidence suggests that developmental arsenic (As) exposure increases behavioral abnormalities in offspring. However, how placental neuroendocrine function and sex-specific responses contribute to these effects remains poorly understood. Here, we demonstrate that maternal As exposure induces trophoblastic damage, vascular disruption, and placental labyrinth thinning, leading to placental dysplasia, fetal growth restriction, and sex-specific behavioral outcomes in offspring. Integrated transcriptomic and neurobehavioral analyses revealed sex-dependent alterations primarily involv

Tert-Butylhydroquinone Reversed Age-Dependent Hypothalamic Mitochondrial Dysfunction Linking Maternal Fructose Exposure to Synaptic Vulnerability.

Maternal nutrition shapes offspring brain metabolism and later-life vulnerability. Excessive fructose intake during pregnancy and lactation has been linked to hypothalamic metabolic dysregulation, yet the mitochondrial basis of this programming remains unclear. We examined female Sprague-Dawley offspring from dams fed a normal diet (ND) or high-fructose diet (HFD) during gestation and lactation at 1 and 3 months of age. Maternal HFD produced age-dependent suppression of hypothalamic electron transport chain activity, accompanied by increased oxidative protein modification, together with a shif

Perinatal exposure to perfluoroalkyl substances impairs maternal care and induces depressive-like behavior in mice.

Postpartum mental health disorders are a critical yet understudied aspect of maternal health. Exposure to environmental toxicants such as per- and poly-fluoroalkyl substances (PFAS) has been associated with adverse health outcomes, including reproductive and neurobehavioral dysfunction, whereas their specific effects on maternal behavior and mental health remain poorly characterized. This study investigated the effects of perinatal exposure to environmentally relevant concentrations of a PFAS mixture comprising 10 individual PFAS (PFHxA, PFPeA, PFHpA, PFBA, perfluorooctanoic acid [PFOA], PFOS,

Maternal cold exposure improves offspring metabolic health via a milk lithocholic acid-microbiota-Th17 axis.

Metabolic diseases are rising with a trend toward earlier onset, yet effective preventive strategies remain limited. While cold exposure improves metabolic health in adults, its role during pregnancy in shaping offspring metabolic outcomes remains unknown. Herein, we demonstrate that maternal cold exposure in early pregnancy markedly improved offspring glucose tolerance, insulin sensitivity, and hepatic lipid metabolism when challenged with a Western diet, and the benefits persisted into late adulthood. Transcriptomic and immunophenotyping analyses revealed that offspring with cold-exposed dam

Maternal per- and polyfluoroalkyl substances exposure, cord blood lipidomics and infant anthropometry: A mediation analysis.

Maternal exposure to per- and polyfluoroalkyl substances (PFAS) has been associated with offspring adiposity; however, underlying mechanisms remain unclear. In this study, we quantified 11 PFAS in maternal plasma collected between 12 and 16 gestational weeks and 104 lipid metabolites in the cord blood of 525 mother-infant pairs. Principal components of multiple PFAS compounds, extracted by principal component analysis, were employed to investigate the effect of the PFAS mixture. Infant anthropometric indicators included weight, length, waist/arm circumference, and abdominal/triceps/subscapular…

Maternal Nutritional Programming: Sex-Specific Cardiovascular and Immune Outcomes Following Perinatal High-Fat Diet Exposure.

Background: The long-term effects of a perinatal high-fat diet on the cardiovascular function of offspring are not well elucidated. We hypothesize that perinatal exposure to a high-fat diet alters adult cardiovascular and immune responses in a sex-specific manner. Methods: Male and female offspring were born to perinatal high-fat (pHFD) or control diet (pCD)-fed C57BL/6 mothers and weaned to a control diet. Cardiovascular function (baseline and response to an acute isoproterenol stress test) was quantified at 8 weeks of age, and acute blood inflammatory response to a single low dose of lipopol
