Hippocampal ensemble dynamics and memory performance are modulated by respiration during encoding.

During offline brain states, such as sleep and memory consolidation, respiration coordinates hippocampal activity. However, the role of breathing during online memory traces remains unclear. Here, we show that respiration can be recruited during online memory encoding. Optogenetic manipulation was used to control activation of the primary inspiratory rhythm generator PreBötzinger complex (PreBötC) in transgenic mice. When intermittent PreBötC-induced apnea covered the object exploration time during encoding, novel object detection was impaired. Moreover, the mice did not exhibit freezing behav…

Anterograde amnesia linked to benzodiazepines.

Benzodiazepines, shown to affect memory, can produce anterograde amnesia (i.e., a loss of memory for events occurring forward in time). Following the ingestion of a benzodiazepine, short-term memory is not affected, but long-term memory is impaired. The memory loss may occur because events are not transferred from short-term memory to long-term memory and thus not consolidated into memory storage. Information stored prior to the ingestion of a benzodiazepine is not affected. Memory impairment is more likely in benzodiazepines that have a high benzodiazepine-receptor affinity, that accumulate i

Morning blue light treatment improves sleep complaints, symptom severity, and retention of fear extinction memory in post-traumatic stress disorder.

Disrupted sleep is a major feature in numerous clinical disorders and is related to decrements in affective memory processing. The prevalence of sleep disruption in post-traumatic stress disorder (PTSD) is suggested to be a key feature that exacerbates the impaired ability to recall extinction memories during experimental fear conditioning. We hypothesized that an intervention employing blue-wavelength light therapy (BLT) to regulate sleep and stabilize circadian rhythms in patients with PTSD (i.e., via regulated morning exposure) would be associated with PTSD symptom improvement, decreased sl

Muscarinic receptor activation promotes destabilization and updating of object location memories in mice.

The storage of long-term memories is a dynamic process. Reminder cues can destabilize previously consolidated memories, rendering them labile and modifiable. However, memories that are strongly encoded or relatively remote at the time of reactivation can resist destabilization only being rendered labile under conditions that favour memory updating. Using the object location recognition task, here we show in male C57BL/6 mice that novelty-induced destabilization of strongly-encoded memories requires muscarinic acetylcholine receptor (mAChR) activation. Furthermore, we use the objects-in-updated

Tranquillising memories: a review of the effects of benzodiazepines on human memory.

Studies from 1973 to 1985 of the effects of benzodiazepines on memory are summarised and reviewed. Anterograde amnesia appears a common effect of all benzodiazepines although its onset and duration vary with the particular benzodiazepine, its dose and route of administration. Memory impairments increase with task difficulty. There is some evidence that partial tolerance to amnesic effects develops with repeated doses of diazepam, but research with other benzodiazepines is inconclusive. Amnesia is in part a by-product of the sedative action of benzodiazepines, although these drugs may also have

Long gradient of retrograde amnesia in mice: continuity with the findings in humans.

Mice were given a single training trial and then received either sham treatment or electroconvulsive shock (ECS; four treatments at hourly intervals) at 1 of 7 times (1-70 days) after training. Retention was always tested 2 weeks after treatment. Control animals exhibited gradual forgetting with increasing retention intervals. Mice given ECS exhibited severe retrograde amnesia, which diminished as the interval between training and ECS increased from 1 to 21 days. ECS given 21-70 days after training had no effect on memory. The finding of long, temporally graded retrograde amnesia in mice estab

Enhancing Consolidation of a New Temporal Motor Skill by Cerebellar Noninvasive Stimulation.

Cerebellar transcranial direct current stimulation (tDCS) has the potential to modulate cerebellar outputs and visuomotor adaptation. The cerebellum plays a pivotal role in the acquisition and control of skilled hand movements, especially its temporal aspects. We applied cerebellar anodal tDCS concurrently with training of a synchronization-continuation motor task. We hypothesized that anodal cerebellar tDCS will enhance motor skill acquisition. Cerebellar tDCS was applied to the right cerebellum in 31 healthy subjects in a double-blind, sham-controlled, parallel design. During synchronization

Memory and self-neuroscientific landscapes.

Relations between memory and the self are framed from a number of perspectives-developmental aspects, forms of memory, interrelations between memory and the brain, and interactions between the environment and memory. The self is seen as dividable into more rudimentary and more advanced aspects. Special emphasis is laid on memory systems and within them on episodic autobiographical memory which is seen as a pure human form of memory that is dependent on a proper ontogenetic development and shaped by the social environment, including culture. Self and episodic autobiographical memory are seen as

Epigenetic memory: the Lamarckian brain.

Recent data support the view that epigenetic processes play a role in memory consolidation and help to transmit acquired memories even across generations in a Lamarckian manner. Drugs that target the epigenetic machinery were found to enhance memory function in rodents and ameliorate disease phenotypes in models for brain diseases such as Alzheimer's disease, Chorea Huntington, Depression or Schizophrenia. In this review, I will give an overview on the current knowledge of epigenetic processes in memory function and brain disease with a focus on Morbus Alzheimer as the most common neurodegener…

Drug development targeting the glycogen synthase kinase-3beta (GSK-3beta)-mediated signal transduction pathway: role of GSK-3beta in adult brain.

Glycogen synthase kinase-3beta (GSK-3beta) is a constitutively active kinase. Since its activation results in neurofibrillary tangle (NFT) d … amnesia. Further analysis indicated that GSK+/- mice had impaired memory reconsolidation but normal memory consolidation. Therefore, we concluded that GSK-3beta activation is required for memory reconsolidation in the adult brain.

Memory loss caused by beta-amyloid protein is rescued by a beta(3)-adrenoceptor agonist.

Accumulation of the neurotoxic beta-amyloid protein (Abeta) in the brain is a key step in the pathogenesis of Alzheimer's disease (AD). Although transgenic mouse models of AD have been developed, there is a clear need for a validated animal model of Abeta-induced amnesia which can be used for toxicity testing and drug development. Intracranial injections of Abeta(1-42) impaired memory in a single trial discriminative avoidance learning task in chicks. Memory inhibition was closely associated with the state of aggregation of the Abeta peptide, and a scrambled-sequence of Abeta(1-42) peptide fai

[Memory disorders in Parkinson's disease].

52 patients with Parkinson's diseases (PD), 11 patients with suspected Alzheimer's disease (AD) and 20 healthy individuals were observed. The age of the examined persons was 55-74 years. Both quantitative evaluation of the neurologic symptoms and estimation of high psychic functions were performed by means of A.R.Luria's method. Memory tests were also used. It was found that PD was practically always accompanied by memory impairments which were out of the normal age limits. The main mechanism of memory impairment in PD without dementia included insufficiency of independent organization of the

Sleep restoration by optogenetic targeting of GABAergic neurons reprograms microglia and ameliorates pathological phenotypes in an Alzheimer's disease model.

BACKGROUND: Alzheimer's disease (AD) patients exhibit memory disruptions and profound sleep disturbances, including disruption of deep non-rapid eye movement (NREM) sleep. Slow-wave activity (SWA) is a major restorative feature of NREM sleep and is important for memory consolidation. METHODS: We generated a mouse model where GABAergic interneurons could be targeted in the presence of APPswe/PS1dE9 (APP) amyloidosis, APP-GAD-Cre mice. An electroencephalography (EEG) / electromyography (EMG) telemetry system was used to monitor sleep disruptions in these animals. Optogenetic stimulation of GABA…

Anterograde amnesia in triazolam overdose despite flumazenil treatment: a case report.

Anterograde amnesia, possibly accompanied by acute brain syndrome, is a potential side-effect of certain benzodiazepines, particularly triazolam. Flumazenil is a benzodiazepine antagonist that is highly effective in reversing the central nervous system effects of benzodiazepine overdose. We report a case of triazolam overdose resulting in anterograde amnesia after flumazenil administration had restored clear consciousness. The defect in memory may have been due to too little flumazenil being given or failure of memory consolidation affected by the character of triazolam during the induced luce…

Exercise effects on consolidation of speech and language training in post-stroke aphasia: a case report.

Post-stroke aphasia severely impacts communication and quality of life. Aerobic exercise enhances learning and memory in healthy adults, with evidence suggesting benefits for verbal tasks. Research exploring its effects in stroke patients with aphasia is minimal. This case study investigated the effects of combining speech and language therapy (SLT) with high-intensity aerobic exercise on speech performance in post-stroke aphasia. Over 4 weeks, two participants with post-stroke anomic aphasia engaged in daily 20-min SLT sessions focused on naming activities. Speech training was followed by 20-

Splice isoforms of the histone variant macroH2A1 differentially regulate hippocampal gene expression and memory formation.

Histone variants are critical components of neuronal chromatin that are emerging as key regulators of long-term memory formation. We previously showed that depleting the macrodomain-containing histone variant macroH2A1 (mH2A1) in the mouse hippocampus impairs long-term memory, establishing this histone as essential for memory consolidation. However, mH2A1 undergoes alternative splicing to generate two isoforms, mH2A1.1 and mH2A1.2, which differ by a single exon within the macrodomain. Though mH2A1.1 and mH2A1.2 have been reported to regulate unique molecular processes in non-neuronal cells, di…
