A case of ALL in a 2 1/2-year-old boy with fatal outcome is presented. Cytogenetic analysis revealed a hypodiploid karyotype: 45,X-Y,-2,+der (2)t(Y;2),-12,i(17q),+mar. Some metaphases represented a sideline with 44 chromosomes and monosomy 8 was a consistent anomaly. These findings are rather uncommon in ALL. Hypodiploidy and the translocation, however, indicated poor prognosis in this case.

A lymphocyte chromosome analysis was done in 85 cases of trisomy 21 Down syndrome, the objective being the identification of the frequency of mosaicism. In each case an attempt was made to analyse at least 100 metaphases. Sixteen cases of mixoploidy were found (18.8%) with two or more cellular lines. These findings are discussed in relation with the frequency of the association between Down's syndrome and other aneuploidies, the frequency of the diploid/trisomy 21 mosaicism, and the possible origin of the latter.

Autologous bone marrow transplantation is now a major tool in the treatment of human leukemias and lymphomas. Evaluation of residual disease by standard cytological methods is difficult. Cytogenetics provide clonal markers which are specific features of leukemic cells. Detection of minimal disease by chromosomal methods is possible in acute leukemias, it requires karyotyping of a few hundred metaphases from a short term culture. A preselection of the material to be examined will improve the degree of sensitivity of the method.

The high risk for malignancy for carriers of several congenital chromosomal abnormalities is discussed. We report herein a patient with Klinefelter's syndrome associated with a high grade malignant non-Hodgkin lymphoma. Chromosome analysis of PHA-stimulated lymphocytes showed a mosaic of 46,XXY (90%) and normal 46,XY (10%) metaphases. To our knowledge, this is the third reported case of a malignant non-Hodgkin lymphoma in a patient with Klinefelter's syndrome in the literature.

Cytogenetic investigations performed on lymphocytes from a 29-year-old woman with no severe anomalies, allowed us to recognize a mild form of Roberts syndrome. The proposita's metaphases showed a consistent centromere splitting, especially affecting chromosomes 16, 19, 21, and 22. This centromere separation sequence seems to be unique to Roberts syndrome cells. The experiments also showed that no diffusible factor, involved in the mechanism of sister chromatid pairing-disjunction, exists.

We report here on two mosaic patients with both an idic(Yp) and a microchromosome. FISH with the DYZ3 alphoid repeat demonstrated that the isodicentrics effectively exhibited two alphoid clusters whereas the small markers had a Y-centromere. The frequencies of dicentric chromosomes and of partial premature chromosome condensation (PCC) in definitely first, second, or third mitoses after X-irradiation of human leukocytes, as determined with a bromodeoxyuridine (BUdR)/ Giemsa labeling technique, were investigated. The results show that practically no PCCs occur in first mitoses after irradiation and culture start in contrast to second or third ones. The frequencies of dicentrics from first to second, as well as from second to third mitoses after irradiation decline by about 50% for each division step.
