Inhibition of Phosphodiesterase 10A by MP-10 Rescues Behavioral Deficits and Normalizes Microglial Morphology and Synaptic Pruning in A Mouse Model of FOXP1 Syndrome.

FOXP1 syndrome caused by FOXP1 haploinsufficiency is characterized by intellectual disability, speech, and language impairment, autistic features, and neuropsychiatric abnormalities such as anxiety and hyperactivity. Behavioral changes in patients are mirrored in Foxp1± mice. It is shown that decreased Foxp1 in the Foxp1± striatum results in a significant decrease in phosphodiesterase 10a (Pde10a). Predominantly expressed in medium spiny neurons (MSNs), Pde10a modulates basal ganglia circuitry. Furthermore, the Foxp1± striatum exhibits microglial activation, reduced synaptic pruning, and dysre

Rapid iPSC-derived neuromuscular junction model uncovers motor neuron dominance in amyotrophic lateral sclerosis cytopathy.

The neuromuscular junction (NMJ) is essential for transmitting signals from motor neurons (MNs) to skeletal muscles (SKMs), and its dysfunction can lead to severe motor disorders. However, our understanding of the NMJ is limited by the absence of accurate human models. Although human induced pluripotent stem cell (iPSC)-derived models have advanced NMJ research, their application is constrained by challenges such as limited differentiation efficiency, lengthy generation times, and cryopreservation difficulties. To overcome these limitations, we developed a rapid human NMJ model using cryoprese

Mechanisms underlying seizures and hypothermia during busulphan administration.

Busulphan (Bu) is used as a part of the conditioning regimen prior to HSCT. Neurotoxicity is one of Bu major adverse-effects. We investigated the kinetics of busulphan and its metabolites (tetrahydothiophene, tetrahydrothiophene-1-oxide, sulfolane, 3-OH-sulfolane) in patients and mice as well as the mechanisms underlying CNS-toxicity in mice. Busulphan metabolites were detectable in plasma and urine up to 72-h after the last Bu-dose. Sulfolane levels were high and reached maximum concentration at the time-point reported for the convulsions' occurrence. Mice were treated with either busulphan o

The Cutaneous Pathology of Erythromelalgia and Its Role in Establishing Critical Clues Regarding Pathogenesis.

Erythromelalgia, a rare cutaneous pain syndrome, is characterized by acral burning pain and flushing, often alleviated by cold and rest. Primary erythromelalgia is caused by gain-of-function mutations of genes encoding for sodium channels, resulting in hyperexcitability of pain signaling neurons. Autoimmunity and hematologic dyscrasias such as thrombocythemia have been implicated in secondary erythromelalgia. The pathology of this rare disease remains poorly defined. The Weill Cornell dermatopathology database was analyzed for acral biopsies procured from patients here there was a clinical con

Infantile Cerebellar-Retinal Degeneration Associated With Novel ACO2 Variants: Clinical Features and Insights From a Drosophila Model.

Infantile Cerebellar-Retinal Degeneration (ICRD) is an autosomal recessive neuro-disability associated with hypotonia, seizures, optic atrophy, and retinal degeneration. Recessive variants of the mitochondrial aconitase gene (ACO2) are a known cause of ICRD. Here, we present a paediatric male patient with ICRD, where whole genome sequencing of the family trio revealed segregating heterozygous variants of unknown significance in ACO2. At 4 months, he displayed generalised hypotonia, and by 6 years, visual electrophysiology indicated bilateral optic atrophy. Magnetic Resonance Imaging (MRI) at a

Dynamic imbalance of autophagy: A molecular link between age-related hearing loss and cognitive dysfunction.

Age-related hearing loss (ARHL) and cognitive dysfunction are established comorbidities, yet their molecular links remain unclear. This study employed a D-galactose-induced ARHL guinea pig model, along with multimodal analysis, to elucidate the bridging role of autophagy. ARHL animals exhibited significantly elevated hearing thresholds (>65 dB SPL) and spatial memory deficits (Morris Water Maze escape latency increased 0.77-fold). PET/CT revealed a 28.57 % reduced hippocampal glucose metabolism (SUV), negatively correlating with cognitive impairment. Molecular analysis revealed that the autoph

Age-related loss of Notch3 underlies brain vascular contractility deficiencies, glymphatic dysfunction, and neurodegeneration in mice.

Vascular aging affects multiple organ systems, including the brain, where it can lead to vascular dementia. However, a concrete understanding of how aging specifically affects the brain vasculature, along with molecular readouts, remains vastly incomplete. Here, we demonstrate that aging is associated with a marked decline in Notch3 signaling in both murine and human brain vessels. To clarify the consequences of Notch3 loss in the brain vasculature, we used single-cell transcriptomics and found that Notch3 inactivation alters regulation of calcium and contractile function and promotes a notabl

An fMRI study of action observation and action execution in childhood.

Although many studies have examined the location and function of the mirror neuron system (MNS) in human adults, we know relatively little about its development. The current study fills this gap by using fMRI to examine for the first time the development of the brain regions implicated in action execution, action observation, and their overlap. We examined age-related differences in brain activation by contrasting a group of children (n = 21) and adults (n = 18). Surfaced-based analyses of action execution and action observation revealed that brain activity for action observation and execution

Oculo-visual abnormalities in Parkinson's disease: Possible value as biomarkers.

Parkinson's disease (PD) is one of the most common neurodegenerative disorders and the second leading cause of dementia worldwide. With an aging population, the prevalence of the disease has dramatically increased. Clinical management has advanced through recent developments in dopaminergic imaging and genetic risk profiling. However, early and accurate diagnosis of the disorder remains a challenge, largely because of the lack of noninvasive and inexpensive reliable diagnostic tests. Besides the well-studied cerebral neurodegeneration that underlies the cardinal symptoms of PD (ie, bradykinesi

[Biological significance of Broca's region].

Since P. Broca (France) reported in 1960's the case with disturbance of speech caused by the left infero-posterior frontal lesion, this region was considered to be a kineticism speech center (Broca's area: BA). Linguistic functions have been clarified afterwards by many studies and case reports. Doubt whether the former BA where Homo Sapiens got a linguistic functions would take any kind of function is left as expected. We have faced an original function of BA based on results provided accidentally in our studies related to sensori-motor linkage mechanisms. Then, we have clarified brain functi

Pilot study: Computer-based virtual anatomical interactivity for rehabilitation of individuals with chronic acquired brain injury.

Deficiencies in upper-limb motor function and executive functioning can compromise an affected individual's ability to complete everyday activities. Impaired motor and executive functioning therefore pose a risk to increasing numbers of veterans who have been diagnosed with acquired brain injury. This article reports on changes in upper-limb motor function and executive functioning of 12 adult participants with chronic acquired brain injury using a novel, computer-based, motor and cognitive rehabilitation program called PreMotor Exercise Games (PEGs). Manual muscle, goniometric range of motion

Synergistic Effects of Constraint-Induced Movement Therapy and Mirror Therapy on Neuroplasticity and Functional Recovery in Rats With Hemiplegic Cerebral Palsy.

IMPORTANCE: The mechanisms underlying the combined effects of constraint-induced movement therapy (CIMT) and mirror therapy (MT) on neuroplasticity in hemiplegic cerebral palsy (HCP) remain unclear. OBJECTIVE: This study investigated whether combined CIMT and MT improves functional recovery more than either therapy alone and examined accompanying motor cortical neuroplastic changes. DESIGN: HCP rats were randomly assigned to HCP, HCP+CIMT, HCP+MT, and HCP+CIMT+MT groups (n = 13 per group), with a separate control group. SETTING: The study was conducted at the Experimental Animal Center, Gua

Surface-based information mapping reveals crossmodal vision-action representations in human parietal and occipitotemporal cortex.

Many lines of evidence point to a tight linkage between the perceptual and motoric representations of actions. Numerous demonstrations show how the visual perception of an action engages compatible activity in the observer's motor system. This is seen for both intransitive actions (e.g., in the case of unconscious postural imitation) and transitive actions (e.g., grasping an object). Although the discovery of "mirror neurons" in macaques has inspired explanations of these processes in human action behaviors, the evidence for areas in the human brain that similarly form a crossmodal visual/moto…

Task-Parametrized dynamics: Representation of time and decisions in recurrent neural networks.

How do recurrent neural networks (RNNs) internally represent elapsed time to initiate responses after learned delays? To address this question, we trained RNNs on delayed decision-making tasks with progressively increasing temporal demands, including binary decisions, context-dependent decisions, and perceptual integration. We analyzed trained networks using connectivity statistics, eigenvalue spectra, readout alignment, and low-dimensional population trajectories. Across tasks, networks converged to qualitatively distinct but behaviourally comparable dynamical solutions, including oscillatory

Enteric and Sympathetic Nervous System Pathways Mediate Early Life Stress Effects on Gut Motility and Pain: Mechanistic Findings With Human Correlation.

BACKGROUND & AIMS: Adverse experiences during early life can disrupt gut-brain axis development, but the mechanisms linking early life stress (ELS) to long-term gastrointestinal dysmotility and pain remain unclear. METHODS: We used a maternal separation (MS) mouse model of ELS and assessed visceral pain sensitivity, intestinal motility, and enteric nervous system (ENS) composition. Two large human pediatric population cohorts were also analyzed for associations between ELS and disorders of gut-brain interaction (DGBI) risk. RESULTS: MS in mice led to visceral hypersensitivity, sex-specific m

Disruptions in speech, language and social communication in lysosomal storage diseases.

Communication enables social connection, autonomy and identity, making it a fundamental domain of human functioning. In lysosomal storage diseases (LSDs), progressive neurological dysfunction can severely disrupt speech, language and social communication, resulting in profound isolation and reduced quality of life. LSDs are a heterogeneous group of more than 70 inherited metabolic disorders caused by lysosomal dysfunction and substrate accumulation, leading to severe neurological consequences and multisystem pathology. LSDs are classified according to the biochemical nature of the accumulating

Integration of a Gripper-Equipped Humanoid Social Robot for EEG-Monitored Action Observation Paradigms.

Action Observation (AO) therapy leverages the mirror neuron system (MNS) and may support motor recovery in neurorehabilitation. In this study, we integrated Flo v2, a humanoid robot equipped with grippers and object detection system, into an AO therapy paradigm with electroencephalography (EEG) monitoring. Flo v2's enhanced design enables the execution of upper-limb actions, either transitive (involving object interaction such as grasping a cup) or intransitive (gesture-based without object manipulation such as waving). The robot control is synchronized with EEG recording to facilitate the inv

Relative contribution of gonads and sex chromosomes to sex differences in cell-type gene expression in the mouse medial septum and sex-biased disease risk.

BACKGROUND: While sex differences in the brain have traditionally been attributed to gonadal hormones, emerging evidence points to regulation by sex chromosomes. This study aims to differentiate the influence of gonads versus sex chromosomes on cellular gene expression in the mouse medial septum (MS), a critically understudied brain region. METHODS: Using single nucleus RNA-sequencing and the Sex Chromosome Trisomy mouse model, we (1) quantified sex differences in cellular gene expression and (2) isolated sex-biasing effects by identifying perturbed cell types, differentially expressed genes,

Cortical organoids reveal human-specific roles of METTL5 in neurodevelopment via regulation of CHCHD2.

METTL5 is a conserved methyltransferase responsible for catalyzing N6-methyladenosine (m 6 A) modification on 18S ribosomal RNA. Human patients with mutations in METTL5 show severe microcephaly and intellectual disability, which has not been fully recapitulated in animal models. Given its emerging role in neurodevelopment, we sought to investigate METTL5 function in a human-specific context using cortical forebrain organoids derived from human induced pluripotent stem cells. We generated METTL5 knockout organoids and observed a marked delay in neural stem cell proliferation and the timing of n
