Behavioral and molecular exploration of the AR-CMT2A mouse model Lmna (R298C/R298C).

In 2002, we identified LMNA as the first gene responsible for an autosomal recessive axonal form of Charcot-Marie-Tooth disease, AR-CMT2A. All patients were found to be homozygous for the same mutation in the LMNA gene, p.Arg298Cys. In order to investigate the physiopathological mechanisms underlying AR-CMT2A, we have generated a knock-in mouse model for the Lmna p.Arg298Cys mutation. We have explored these mice through an exhaustive series of behavioral tests and histopathological analyses, but were not able to find any peripheral nerve phenotype, even at 18 months of age. Interestingly at th

Molecular etiology of atherogenesis--in vitro induction of lipidosis in macrophages with a new LDL model.

BACKGROUND: Atherosclerosis starts by lipid accumulation in the arterial intima and progresses into a chronic vascular inflammatory disease. A major atherogenic process is the formation of lipid-loaded macrophages in which a breakdown of the endolysomal pathway results in irreversible accumulation of cargo in the late endocytic compartments with a phenotype similar to several forms of lipidosis. Macrophages exposed to oxidized LDL exihibit this phenomenon in vitro and manifest an impaired degradation of internalized lipids and enhanced inflammatory stimulation. Identification of the specific c

Interaction of CTX-M-15 enzyme with cefotaxime: a molecular modelling and docking study.

Extended-spectrum β-lactamases (ESBLs) are the bacterial enzymes that make them resistant to advanced-generation cephalosporins. CTXM enzymes (the most prevalent ESBL-type) target cefotaxime. Aims of the study were: Modelling of CTX-M enzyme from bla(CTX-M) sequences of clinical Escherichia coli isolatesDocking of cefotaxime with modelled CTX-M enzymes to identify amino acid residues crucial to their interaction To hypothesize a possible relationship between 'interaction energy of the docked enzyme-antibiotic complex' and 'minimum inhibitory concentration (MIC) of the antibiotic against the ba

Molecular and histological changes following central retinal artery occlusion in a mouse model.

The aim of this study was to characterize the molecular and histological changes that occur in the retina following central retinal artery occlusion (CRAO) in a mouse model. CRAO was induced in 60 mice by laser photoactivation of intravenously injected rose bengal. Mice were sacrificed at 3, 6, 12, and 24h and 7 and 21 days after CRAO induction for molecular analysis (5-13 mice/time point) and histological and apoptosis studies (3-4 mice/time point). Fundus examination and fluorescein angiography were also performed at various points. Retinal mRNA was analyzed for expression of T-cell antigen

Structure based drug design for HIV protease: from molecular modeling to cheminformatics.

Significant progress over the past decade in virtual representations of molecules and their physicochemical properties has produced new drugs from virtual screening of the structures of single protein molecules by conventional modeling methods. The development of clinical antiviral drugs from structural data for HIV protease has been a major success in structure based drug design. Techniques for virtual screening involve the ranking of the affinity of potential ligands for the target site on a protein. Two main alternatives have been developed: modeling of the target protein with a series of r

The molecular pathophysiology of bacterially induced preterm labor: insights from the murine model.

Premature delivery, the most important problem in obstetrics in developed countries, continues to vex clinicians and researchers. Despite decades of investigation, the pathophysiology of premature labor is incompletely understood, and therapies or preventive strategies tailored to each of the many potential causes do not exist. The present review addresses one cause of prematurity, namely, intrauterine bacterial infection. Given the vastness of the literature for even this single etiology, we focus on the mouse as a model organism from which much can be learned about mammalian parturition. The

Notochord anomalies in the adriamycin rat model: A morphologic and molecular basis for the VACTERL association.

BACKGROUND/PURPOSE: The Adriamycin rat model (ARM) is a reliable model of the VACTERL association. The notochord is structurally abnormal in the region of the foregut, midgut, and hindgut in the ARM. The authors hypothesised that notochord anomalies allow ectopic expression of molecular signals in the developing embryo and thus lead to VACTERL malformations. This study was designed to investigate this hypothesis. METHODS: Adriamycin (1.75 mg/kg) was administered intraperitoneally to pregnant rats on days 7, 8, and 9 of gestation. Control animals were given saline. Embryos were recovered on ge

Intragastric ethanol infusion model for cellular and molecular studies of alcoholic liver disease.

The intragastric alcohol infusion rat model (IAIRM) of alcoholic liver disease (ALD) has been utilized in various laboratories to study various aspects of ALD pathogenesis including oxidative stress, cytokine upregulation, hypoxic damage, apoptosis, ubiquitin-proteasome pathway and CYP2E1 induction. The basic value of the model is that it produces pathologic changes which resemble ALD including microvesicular and macrovesicular fat, megamitochondria, apoptosis, central lobular and pericellular fibrosis, portal fibrosis, bridging fibrosis, central necrosis, and mixed inflammatory infiltrate inc

Molecular basis of feline beta-glucuronidase deficiency: an animal model of mucopolysaccharidosis VII.

A family of domestic cats was found that exhibited clinical and biochemical abnormalities consistent with mucopolysaccharidosis VII, an autosomal recessive lysosomal storage disorder caused by beta-glucuronidase deficiency. beta-Glucuronidase activity was undetectable in affected cat fibroblasts and restored by retroviral gene transfer of rat beta-glucuronidase cDNA. beta-Glucuronidase mRNA was normal in affected cat testis by Northern blot analysis. Normal feline beta-glucuronidase cDNA was cloned and characterized, and amplified from affected cat fibroblasts by reverse transcription coupled

Juvenile retinoschisis: a model for molecular diagnostic testing of X-linked ophthalmic disease.

BACKGROUND AND PURPOSE: X-linked juvenile retinoschisis (RS) provides a starting point to define clinical paradigms and understand the limitations of diagnostic molecular testing. The RS phenotype is specific, but the broad severity range is clinically confusing. Molecular diagnostic testing obviates unnecessary examinations for boys at-risk and identifies carrier females who otherwise show no clinical signs. METHODS: The XLRS1 gene has 6 exons of 26-196 base-pair size. Each exon is amplified by a single polymerase chain reaction and then sequenced, starting with exons 4 through 6, which cont

The molecular basis of ornithine transcarbamylase deficiency: modelling the human enzyme and the effects of mutations.

Human ornithine transcarbamylase is a trimer with 46% amino acid sequence homology to the catalytic subunit of E coli aspartate transcarbamylase. Secondary structure predictions, distributions of hydrophilic and hydrophobic regions, and the pattern of conserved residues suggest that the three dimensional structures of the two proteins are likely to be similar. A three dimensional model of ornithine transcarbamylase was generated from the crystal structure of the catalytic subunit of E coli aspartate transcarbamylase in the holoenzyme, by aligning the sequences, building in gaps, and minimising

Interaction-constrained 3D molecular generation using a diffusion model enables structure-based pharmacophore modeling for drug design.

A key challenge in structure-based drug design is generating three-dimensional molecules while preserving essential protein-ligand interactions. We propose DiffPharma, a structure-based pharmacophore modeling framework based on a conditional diffusion model, to generate molecules that satisfy specified interaction constraints. The proposed method incorporates a semantic fusion architecture that integrates multiple interaction-specific neural networks, each designed to capture distinct molecular interactions such as hydrogen bonds and hydrophobic interactions. Experimental results demonstrate t

Validation of the antihyperglycaemic and hepatoprotective activity of the flavonoid rich fraction of Brachychiton rupestris using in vivo experimental models and molecular modelling.

Oxidative stress leads to many disorders as diabetes mellitus and liver diseases. This study evaluates the antihyperglycemic and hepatoprotective activities of Brachychiton rupestris (Malvaceae). The antihyperglycemic activity of the total methanol extract of B. rupestris leaves (BRT) and its ethyl acetate fraction (BRE) was evaluated using streptozotocin induced diabetic rats. The hepatoprotective activity was assessed using carbon-tetrachloride induced hepatotoxicity. Oral administration of 50 mg/kg b.wt (body weight) of BRT and BRE to Streptozotocin -diabetic rats caused a notable decrease

Vibrational dynamics of TODD-graphene nanosheets via molecular dynamics simulations and machine-learning surrogate models: implications for adaptive nanomechanical devices.

CONTEXT: TODD-Graphene (TODD-G), a porous two-dimensional carbon allotrope composed of interconnected 3-8-10-12 carbon rings, has recently emerged as a promising candidate for nanoelectromechanical and adaptive nanodevice applications due to its metallic conductivity, mechanical compliance, and topologically engineered phonon structure. In this work, the vibrational dynamics of TODD-G nanosheets were systematically investigated to establish structure-property relationships governing resonant behavior under varying geometric, thermal, mechanical, and defect conditions. Fundamental frequencies w

Recent advances in small molecules for amyloid fibril inhibition: chemical strategies and molecular mechanistic insights from lysozyme and insulin models.

Protein misfolding and aggregation are central pathological processes underlying amyloidosis, a group of disorders characterized by the deposition of insoluble amyloid fibrils. These toxic aggregates form due to conformational destabilization and exposure of hydrophobic protein regions, leading to aberrant interactions that disrupt normal cellular functions. Amyloid fibrillation is a key pathological event in both neurodegenerative and systemic disorders. In metabolic and systemic amyloidosis, such as lysozyme derived and insulin derived amyloidosis, peripheral fibril deposition leads to progr

Assessment of Bedside-Adaptable Models to Predict Molecular Sepsis Subtypes in a Resource-Limited Setting: A Multicenter Analysis from Uganda.

OBJECTIVE: Biologically defined sepsis subtypes have been identified in low- and middle-income countries (LMICs), but limited access to molecular diagnostics challenges broader evaluation and implementation in resource-limited settings. We assessed whether models including bedside clinical and rapid microbiologic data could accurately stratify Ugandan adults with sepsis by molecular subtype. DESIGN: Secondary analysis of two prospective observational sepsis cohorts, testing bedside-adaptable classifier models against transcriptomic and proteomic subtype assignments. SETTING: Entebbe Regional

Molecular and topographic mapping of antipsychotic effects: a meta-analysis of postsynaptic density proteins in animal models with translational implications.

BACKGROUND: While antipsychotics primarily target dopamine D2 receptor, the putative synaptic mechanisms underlying their therapeutic effects remain unclear. Postsynaptic density (PSD) at glutamatergic synapses represents a dynamic protein network involved in synaptic plasticity and neurotransmission, whose dysfunction has been implicated in the pathophysiology of schizophrenia and bipolar disorder. This study aims to explore, for the first time, with a quantitative meta-analytical approach, how antipsychotic treatments affect PSD molecules across various brain regions in preclinical settings,

Antisense oligonucleotides targeting valosin-containing protein ameliorate muscle pathology and molecular defects in cell and mouse models of multisystem proteinopathy.

BACKGROUND: Valosin-containing protein (VCP) related disease, also known as multisystem proteinopathy 1 (MSP1), is an autosomal dominant disease caused by gain-of-function pathogenic variants of the VCP gene. The disease presents with variable combinations of inclusion body myopathy, early-onset Paget's disease of bone, frontotemporal dementia and may also overlap with familial amyotrophic lateral sclerosis. There is currently no treatment for this progressive disease associated with early demise resulting from proximal limb girdle and respiratory muscle weakness. We hypothesise that regulatin

[Molecular Characterization of Cutaneous Leishmaniasis and Canine Leishmaniasis Strains Isolated from Northern Cyprus and Their Clinical Reflections in Mice Models].

Leishmaniasis is a parasitic infection caused by Leishmania species in tropical and subtropical regions. Cyprus, located in the Eastern Mediterranean basin, is considered endemic for leishmaniasis. In our study, we aimed to investigate the origins of two Leishmania strains isolated from Turkish Republic of Northern Cyprus by sequencing and to establish in vivo animal models by examining the clinical reflections of these strains in mice. A biopsy material was taken from a seven-year-old girl with a lupoid lesion on her forehead and amastigotes were detected in the smears. Following culture of t
