Autoimmune Reaction and the Management of Recurrent Bioprosthetic Pulmonary Valve Dysfunction.

BACKGROUND: Bioprosthetic valve failure is a significant and debilitating sequela in cardiac patients caused primarily by structural changes in valve leaflets due to endocarditis, calcification, and thrombosis. CASE SUMMARY: We describe a rare case of bioprosthetic pulmonary valve failure due to an allergic reaction to molybdenum within the valve cage. We describe the clinical presentation, clinical methods leading to the diagnosis, and treatment progression until we could resolve symptoms and a significant return of valvular competency. DISCUSSION: After excluding infectious etiologies as a…

Intramolecular electron transfer in sulfite-oxidizing enzymes: probing the role of aromatic amino acids.

Sulfite oxidase (SO) is a molybdoheme enzyme that is important in sulfur catabolism, and mutations in the active site region are known to cause SO deficiency disorder in humans. This investigation probes the effects that mutating aromatic residues (Y273, W338, and H337) in the molybdenum-containing domain of human SO have on both the intramolecular electron transfer (IET) rate between the molybdenum and iron centers using laser flash photolysis and on catalytic turnover via steady-state kinetic analysis. The W338 and H337 mutants show large decreases in their IET rate constants (k (ET)) relati…

Kinetic, structural, and EPR studies reveal that aldehyde oxidoreductase from Desulfovibrio gigas does not need a sulfido ligand for catalysis and give evidence for a direct Mo-C interaction in a biological system.

Aldehyde oxidoreductase from Desulfovibrio gigas (DgAOR) is a member of the xanthine oxidase (XO) family of mononuclear Mo-enzymes that catalyzes the oxidation of aldehydes to carboxylic acids. The molybdenum site in the enzymes of the XO family shows a distorted square pyramidal geometry in which two ligands, a hydroxyl/water molecule (the catalytic labile site) and a sulfido ligand, have been shown to be essential for catalysis. We report here steady-state kinetic studies of DgAOR with the inhibitors cyanide, ethylene glycol, glycerol, and arsenite, together with crystallographic and EPR stu…

Heavy metals in seaducks and mussels from misty fjords national monument in Southeast Alaska.

Quartz Hill, in Misty Fjords National Monument near Ketchikan, Alaska, is the site of a proposed molybdenum-producing mine. To provide baseline data for use in post-development comparisons, we analyzed tissues of Barrow's goldeneyes (Bucephala islandica), common mergansers (Mergus merganser), and blue mussels (Mytilus edulis) for seven heavy metals that could potentially be released into the environment as a result of mining operations. Specimens were collected in 1980, 1981, and 1982 from two fjords likely to be used for discharge of tailings from the proposed mine and from two control fjords…

Tetrathiomolybdate inhibits copper trafficking proteins through metal cluster formation.

Tetrathiomolybdate (TM) is an orally active agent for treatment of disorders of copper metabolism. Here we describe how TM inhibits proteins that regulate copper physiology. Crystallographic results reveal that the surprising stability of the drug complex with the metallochaperone Atx1 arises from formation of a sulfur-bridged copper-molybdenum cluster reminiscent of those found in molybdenum and iron sulfur proteins. Spectroscopic studies indicate that this cluster is stable in solution and corresponds to physiological clusters isolated from TM-treated Wilson's disease animal models. Finally,…

Simultaneous exfoliation and functionalization of MoS2 with tetrapyridyl porphyrin.

Molybdenum disulfide (MoS2) attracts the attention of the scientific community due to its thickness dependent properties. To fully exploit these features, it is necessary to produce the material in mono or few-layers on a large scale. Several methodologies have been developed for this purpose, the most promising one being liquid phase exfoliation (LPE). LPE allows obtaining good quality exfoliated MoS2 in a simple and scalable manner. Herein we report the simultaneous exfoliation and functionalization of MoS2 in chloroform using a specific porphyrin, namely tetrapyridyl porphyrin. We have corr…

Stabilization of the Dinitrogen Analogue, Phosphorus Nitride.

The N2 analogue phosphorus nitride (PN) was the first phosphorus-containing compound to be detected in the interstellar medium; however, this thermodynamically unstable compound has a fleeting existence on Earth. Here, we show that reductive coupling of iron(IV) nitride and molybdenum(VI) phosphide complexes assembles PN as a bridging ligand in a structurally characterized bimetallic complex. Reaction with C≡N t Bu releases the mononuclear complex [(N3N)Mo-PN]-, N3N = [(Me3SiNCH2CH2)3N]3-), which undergoes light-induced linkage isomerization to provide [(N3N)Mo-NP]-, as revealed by photocrysta…

Atomic-layer-deposited ultrafine MoS2 nanocrystals on cobalt foam for efficient and stable electrochemical oxygen evolution.

Ultrafine molybdenum sulfide (MoS2) nanocrystals are grown on a porous cobalt (Co) foam current collector by atomic layer deposition (ALD) using molybdenum hexacarbonyl and hydrogen sulfide as precursors. When used to catalyze the oxygen evolution reaction (OER), the optimal Co@MoS2 electrode, even with a MoS2 loading as small as 0.06 mg cm-2, exhibits a large cathodic shift of ca. 200 mV in the onset potential (the potential at which the current density is 5 mA cm-2), a low overpotential of only 270 mV to attain an anodic current density of 10 mA cm-2, much smaller charge transfer resistance …

Substrate specificity and evolutionary implications of a NifDK enzyme carrying NifB-co at its active site.

The in vitro reconstitution of molybdenum nitrogenase was manipulated to generate a chimeric enzyme in which the active site iron-molybdenum cofactor (FeMo-co) is replaced by NifB-co. The NifDK/NifB-co enzyme was unable to reduce N(2) to NH(3), while exhibiting residual C(2)H(4) and considerable H(2) production activities. Production of H(2) by NifDK/NifB-co was stimulated by N(2) and was dependent on NifH and ATP hydrolysis. Thus, NifDK/NifB-co is a useful tool to gain insights into the catalytic mechanism of nitrogenase. Furthermore, phylogenetic analysis of D and K homologs indicates that s…

Effect of Platinum Content on Properties of CNT-Supported Pt-Mo Catalyst for Ethanol Electrooxidation Reaction.

The CNT-supported nanodispersed Pt-Mo catalysts for the ethanol electrooxidation reaction in the alkaline solution are synthesized and their characteristics are studied. Based on the XPS studies in a wide range of platinum content (10-40 wt %), it is found that in the composition of the catalysts, platinum is predominantly in the metallic state, and molybdenum is in the hexavalent form, probably in the form of MoO3 oxide. According to the XRD and electrochemical studies, the Pt/CNT and PtMo/CNT catalysts with equal platinum contents (~20 wt %) are characterized by similar platinum crystallite …

Ion-pair chromatography coupled to inductively coupled plasma-mass spectrometry (IPC-ICP-MS) as a method for thiomolybdate speciation in natural waters.

Molybdenum precipitates preferentially under reducing conditions; therefore, its occurrence in sediment records is used as an indicator of paleoredox conditions. Although thiomolybdates (MoO4-xSx(2-) with x = 1-4) supposedly are necessary intermediates in the process of molybdenum precipitation under anoxic conditions, there is no information about their abundance in natural environments, because of a lack of element-specific methods with sufficiently low detection limits. Here, we optimized ion-pair chromatographic separation for coupling to an inductively coupled plasma-mass spectrometry det…

Cu2MoS4 nanozymes reduced bacterial defenses through a dual mechanism to amplify cuproptosis-like death and accelerate the healing of burn-infected wounds.

Burn injuries disrupt the skin barrier function, significantly predisposing patients to bacterial invasion. With the widespread use of antibiotics, bacteria have evolved various resistance mechanisms, significantly weakening the efficacy of traditional antibiotics. The application of copper-based nanozymes is still limited by issues such as concentration dependence and bacterial defense. Here, hollow-structured copper-molybdenum bimetallic Cu2MoS4 nanozymes were successfully synthesized using a simple hydrothermal method. In vitro antibacterial experiments showed that Cu2MoS4 nanozymes have mu…
