A suppression-modification gene drive for malaria control targeting the ultra-conserved RNA gene mir-184.

Gene drive technology presents a promising approach to controlling malaria vector populations. Suppression drives are intended to disrupt essential mosquito genes whereas modification drives aim to reduce the individual vectorial capacity of mosquitoes. Here we present a highly efficient homing gene drive in the African malaria vector Anopheles gambiae that targets the microRNA gene mir-184 and combines suppression with modification. Homozygous gene drive (miR-184D) individuals incur significant fitness costs, including high mortality following a blood meal, that curtail their propensity for m

Insecticide resistance and the role of target-site insensitivity mutations among malaria vectors in China: A systematic review and meta-analysis.

BACKGROUND: Malaria, which is transmitted by Anopheles mosquitoes (Diptera: Culicidae), remains a critical global public health problem. Vector control interventions, particularly insecticide-based strategies, are pivotal for malaria control and elimination, as the efficacy of these interventions is heavily dependent on the high susceptibility of Anopheles mosquitoes to insecticides. However, insecticide resistance in mosquito vectors poses a considerable threat to the sustainability of these control efforts. Notably, no synthesis data on insecticide resistance have been reported in China in r

A costing analysis of an intermittent biolarviciding intervention with limited epidemiological effect in Tanga Region, Tanzania.

BACKGROUND: Malaria vector control strategies in sub-Saharan Africa have largely relied on the use of Insecticide Treated-mosquito Nets and Indoor Residual Spraying. However, after substantial reductions in malaria burden associated with the scale-up of these interventions, little progress has been achieved over the last decade. With the increase in insecticide resistance and outdoor biting of mosquitoes, larviciding offers the possibility to be an additional malaria control tool. This study is part of a comprehensive evaluation and aimed to assess prospectively the financial cost of a large-s

Human and vector behavioural determinants of malaria transmission in the Nandi highlands, western Kenya.

BACKGROUND: Malaria transmission, characterized by spatial and temporal heterogeneity and complex vector behaviours, persists in Kenya's highlands despite widespread use of long-lasting insecticidal nets (LLINs). The role of human activity in exposure risk remains underexplored. Identifying vulnerable times and locations is crucial for designing and optimizing targeted control strategies that address the intricate interplay between human activity and local vector behaviour that results in transmission. This study examined human-mosquito interactions in three different ecological settings in Na

Modeling the Dynamics of Dengue Transmission with Awareness and Optimal Control Analysis.

Dengue fever is a vector-borne disease which is transmitted by the bites of mosquitoes infected with dengue viruses. This disease is spread around the world and still as a global health problem. In this work, we formulate the dengue model by considering the population of aware humans. The real data of dengue infection in East Java, Indonesia is employed to estimate the parameters of the dengue model. The estimation of parameters is done by using monthly cumulative data on humans infected dengue that recorded at East Java Health Office on 2018-2020. We then analyze the stability of the equilibr

An AI-based gravitrap surveillance for spatial interaction analysis in predicting aedes risk.

BACKGROUND: Dengue fever is transmitted to humans through bites of Aedes mosquito vectors. Therefore, controlling the Aedes population can decrease the incidence and block transmission of dengue fever and other diseases transmitted by these mosquito species. In many countries, gravitraps are used to monitor mosquito vector densities, but this approach usually underestimates the population of Aedes mosquitoes. Moreover, literature on the spatio-temporal dynamics of Aedes populations in a single city is limited. For example, in Kaohsiung of Taiwan, population densities vary substantially between

Ecological dynamics of field Aedes albopictus populations under Wolbachia-mediated suppression.

BACKGROUND: The incompatible insect technique (IIT), based on Wolbachia-induced conditional sterility, has proven highly effective in suppressing mosquito populations for dengue control. However, concerns that accidental release of infected females could drive population replacement have prompted integration of IIT with irradiation or advanced sex-separation technologies. Moreover, the broader ecological consequences of IIT-based suppression remain insufficiently understood. Here, we investigated whether standalone IIT, leveraging Wolbachia-associated fitness costs under real-world conditions,

"Basically a PR exercise…": a qualitative study of expert perspectives on 'performative versus evidence-informed' dengue vector control in the Asia-Pacific region.

BACKGROUND: Dengue cases are increasing despite continued efforts to control its vectors, with the Asia-Pacific region projected to bear a substantial proportion of the global burden. However, literature on dengue vector control methods and evidence use in national control programmes in the region is limited. We thus aimed to explore expert perspectives on current methods and how to improve dengue vector control programmes across the region. METHODS: We conducted remote semi-structured interviews with a purposive sample of 23 dengue vector control experts with experience in the Asia-Pacific r

High-resolution mapping of urban Aedes aegypti immature abundance through breeding site detection based on satellite and street view imagery.

Identification of Aedes aegypti breeding hotspots is essential for the implementation of targeted vector control strategies and thus the prevention of several mosquito-borne diseases worldwide. Training computer vision models on satellite and street view imagery in the municipality of Rio de Janeiro, we analyzed the correlation between the density of common breeding grounds and Aedes aegypti infestation measured by ovitraps on a monthly basis between 2019 and 2022. Our findings emphasized the significance (p ≤ 0.05) of micro-habitat proxies generated through object detection, allowing to expla

Insecticide tolerance of the malaria vector Anopheles gambiae following larval exposure to microplastics and insecticide.

Microplastic (MP) pollution poses a global threat to urban and rural environments and can have negative effects on a range of organisms. Mosquito larvae often breed in water contaminated with MPs, and given their important role as disease vectors, understanding the effects of larval exposure to MPs is critical for understanding the potential impact on their life history traits and subsequent methods for their control. Here, we have exposed first instar larvae of Anopheles gambiae s.s. to environmentally realistic concentrations of PET microplastics (1.0-7.5 μm) and a sub-lethal dose of insecti

Implementation of malaria vector surveillance and insecticide resistance monitoring interventions in Nigeria.

Malaria vector surveillance is required to determine disease transmission dynamics, vector insecticide susceptibility status, suitable control strategies and impact of control interventions. However, capacity and resources for vector surveillance and insecticide resistance monitoring is often inadequate in most countries at risk of vector-borne diseases. Collaborations and linkages between malaria control policy makers and existing research institutions generating vector surveillance research data are often weak, thereby hindering the availability of data for decision-making. A national vector

Histological validation of in-vivo larvicidal efficacy of marine Streptomyces sp. RD06 secondary metabolites against filariasis causing Culex quinquefasciatus and statistical media optimization for larvicidal derivatives production.

Mosquito-borne disease pandemics, such as the Zika virus and chikungunya, have escalated cognizance of how critical it is to implement proficient mosquito vector control measures. The prevention of Culicidae is becoming more difficult these days because of the expeditious imminence of synthetic pesticide resistance and the universal expansion of tremendously invasive mosquito vectors. The present study highlights the insecticidal and larvicidal efficacy of the prospective novel actinobacterium derived from the marine Streptomyces sp. RD06 secondary metabolites against Culex quinquefasciatus mo

AnophelesModel: An R package to interface mosquito bionomics, human exposure and intervention effects with models of malaria intervention impact.

In recent decades, field and semi-field studies of malaria transmission have gathered geographic-specific information about mosquito ecology, behaviour and their sensitivity to interventions. Mathematical models of malaria transmission can incorporate such data to infer the likely impact of vector control interventions and hence guide malaria control strategies in various geographies. To facilitate this process and make model predictions of intervention impact available for different geographical regions, we developed AnophelesModel. AnophelesModel is an online, open-access R package that quan

Assessing the direct and spillover protective effectiveness of Wolbachia-mediated introgression to combat dengue.

BACKGROUND: Dengue remains a global health challenge with limited treatment options, highlighting the need for effective vector control strategies. The introduction of Wolbachia pipientis into Aedes aegypti populations has shown success in reducing dengue transmission across global field trials. However, the spillover effectiveness of the technology on untreated areas is not well-known. This study estimates the spillover protective effectiveness (PE) of Wolbachia-mediated introgression on dengue. METHODS: We used the synthetic control method (SCM) under assumption of partial interference to e

Wind-assisted high-altitude dispersal of mosquitoes and other insects in East Africa.

Knowledge of insect dispersal is relevant to the control of agricultural pests, vector-borne transmission of human and veterinary pathogens, and insect biodiversity. Previous studies in a malaria endemic area of the Sahel region in West Africa revealed high-altitude, long-distance migration of insects and various mosquito species. The objective of the current study was to assess whether similar behavior is exhibited by mosquitoes and other insects around the Lake Victoria basin region of Kenya in East Africa. Insects were sampled monthly from dusk to dawn over 1 year using sticky nets suspende

Impact of different mosquito collection methods on indicators of Anopheles malaria vectors in Uganda.

BACKGROUND: Methods used to sample mosquitoes are important to consider when estimating entomologic metrics. Human landing catches (HLCs) are considered the gold standard for collecting malaria vectors. However, HLCs are labour intensive, can expose collectors to transmission risk, and are difficult to implement at scale. This study compared alternative methods to HLCs for collecting Anopheles mosquitoes in eastern Uganda. METHODS: Between June and November 2021, mosquitoes were collected from randomly selected households in three parishes in Tororo and Busia districts. Mosquitoes were collec

Association between densities of adult and immature stages of Aedes aegypti mosquitoes in space and time: implications for vector surveillance.

BACKGROUND: Mosquito control is currently the main tool available to contain the spread of several arboviruses in Brazil. We have evaluated the association between entomological surveys of female adult Aedes aegypti and the Breteau index (BI) in space and time in a hyperendemic area, and compared the human resources costs required to measure each of these indicators. METHODS: Entomological surveys were conducted between 2016 and 2019 in Vila Toninho, a neighborhood in the city of São José do Rio Preto, Brazil. Monthly records of collected mosquito specimens were made and then grouped by seaso…

Estimating the potential impact of Attractive Targeted Sugar Baits (ATSBs) as a new vector control tool for Plasmodium falciparum malaria.

BACKGROUND: Attractive targeted sugar baits (ATSBs) are a promising new tool for malaria control as they can target outdoor-feeding mosquito populations, in contrast to current vector control tools which predominantly target indoor-feeding mosquitoes. METHODS: It was sought to estimate the potential impact of these new tools on Plasmodium falciparum malaria prevalence in African settings by combining data from a recent entomological field trial of ATSBs undertaken in Mali with mathematical models of malaria transmission. The key parameter determining impact on the mosquito population is the e
