An evaluation of cell-mediated immunity in man is described that combines the advantages of an in vitro technique, lymphocyte transformation, with the use of contact sensitization to a primary immunogen, dinitrochlorobenzene (DNCB). The present study is an evaluation of the developmental toxicity of dinocap in three rodent species using an in vivo teratology screen. Our protocol uses postnatal viability, weight gain, and morphological and behavioral development through weaning to assess the developmental toxicity of compounds. Skin sensitization with DNCB was carried out in forty-five untreated, 106 "on treatment" and fifteen fully treated patients with pulmonary tuberculosis along with fifty-five controls. Using a quantitative method based on the sponteneous flare phenomenon, contact sensitization to DNCB was studied in 143 healthy volunteers between the ages of 20 and 80 yr. A spontaneous flare reaction occurred in 96·5% of subjects tested, regardless of age, and a correlation was demonstrated between the intensity of the primary response to DNCB and the threshold dose of DNCB required to elicit an anamnestic response.
