Metformin's disruption of gluconeogenesis in type 2 diabetes impairments of the cori and alanine cycles as hidden drivers of metabolic waste accumulation, NF-κBHIF-1α-mediated low-grade inflammation, and multisystem dysfunction.

Gluconeogenesis, a dual-purpose pathway in type 2 diabetes mellitus (T2DM), not only synthesizes glucose but also clears metabolic waste via the Cori cycle (lactate recycling through LDH, PC, PEPCK) and Alanine cycle (nitrogen disposal via ALT, GDH, urea cycle), preventing acidosis, ROS accumulation, and ammonia toxicity. Metformin, the cornerstone T2DM therapy, inhibits gluconeogenesis by targeting mitochondrial complex I, elevating AMP/ATP ratios, and activating AMPK-PKCι/λ signaling to repress CREB-CRTC2-driven PEPCK/G6Pase expression, disrupting these cycles. This leads to lactate, pyruvat

Wet-dry cycles control the emissions and sources of greenhouse gases in agricultural soil: An incubation study.

This study examines the impact of wet-drying cycles and nitrogen (N) fertilization on soil greenhouse gas fluxes, specifically nitrous oxide (N2O) and carbon dioxide (CO2). Nine treatments were tested, combining three soil moisture regimes (55% constant, 55%-30% cycle, and 80%-55% cycle) with three N addition rates (0, 100, and 150 kg N ha-1) using 15N-labeled urea. Soil samples from a potato (Solanum tuberosum) field in Lethbridge, Alberta, were incubated for 28 days under controlled conditions. Wet-drying cycles involved initially wetting the soil to the upper threshold (55% or 80% WFPS) and

Effect of biochar combined with a biotrickling filter on deodorization, nitrogen retention, and microbial community succession during chicken manure composting.

The high-nitrogen content and dense structure of poultry manure compost cause volatilization of N to ammonia (NH3). This study evaluated the combined application of biochar and biotrickling filtration (BTF) to remove of odor in chicken manure mixed straw compost (w/w, 2.5:1). Adding of 10% biochar reduced NH3, hydrogen sulfide (H2S), and total volatile organic compounds (TVOCs) contents by 20.04%, 16.18%, and 17.55% respectively, and decreased the N loss rate by 8.27%, compared with those observed in control. The organic matter content decreased by 28.11% and germination index reached 97.36% i

Trichorrhexis nodosa secondary to argininosuccinicaciduria.

Argininosuccinicaciduria is a rare metabolic disorder of the urea cycle associated with the inability to excrete nitrogenous waste in the form of urea. Along with low serum arginine, hepatomegaly, and mental retardation, congenital trichorrhexis nodosa is a distinguishing feature of the disorder. We present a 3.5-year-old girl diagnosed with argininosuccinicaciduria who presented to the dermatology clinic with hair thinning and loss since birth. Microscopic evaluation revealed nodular swellings on the hair shafts and frayed cortical fibers consistent with the diagnosis of trichorrhexis nodosa

Identification of novel mutations in the human ornithine transcarbamylase (OTC) gene of Korean patients with OTC deficiency and transient expression of the mutant proteins in vitro.

The urea cycle plays key roles to prevent the accumulation of toxic nitrogenous compound and synthesize arginine de novo. Ornithine transcarbamylase (OTC) deficiency is the most common inborn error of urea cycle, which is inherited in an X-linked manner. This study was undertaken to characterize molecular defects in Korean patients with OTC deficiency. With direct sequence analysis of OTC gene of 26 unrelated Korean patients with OTC deficiency, 23 different mutations were identified. Among these mutations, eleven were novel mutations. The novel mutations were p.Leu9X, p.Arg26Pro, p.Gly100Arg,

Sulfur disproportionation in engineered environmental systems: A critical review of mechanisms, applications, and emerging perspectives.

Sulfur disproportionation (SDP), a microbial metabolism that couples the oxidation and reduction of intermediate-valence sulfur compounds, is increasingly recognized as a key process in both natural sulfur cycling and engineered environmental systems. Despite growing interest, our understanding of SDP remains incomplete, and its practical significance has often been overlooked. This critical review integrates recent advances in ecophysiology, molecular mechanisms, and reactor-scale applications to establish a unified framework and provide practical guiding principles for SDP. We first summariz

Cation-regulated nitrate electroreduction to ammonia on Fe-deposited carbon rod electrodes with correction of spontaneous contribution.

Electrochemical nitrate reduction to ammonia (NO3RR) offers a promising route for nitrogen-resource recovery under mild conditions. However, for metal-based nitrate electrolysis systems, the detected ammonia may not originate solely from potential-driven electrocatalysis because the metal electrode can also react with nitrate under open-circuit conditions, leading to overestimation of the apparent ammonia yield and faradaic efficiency (FE). Herein, an Fe-deposited carbon rod electrode (Fe/CR) was used as a model system to examine electrolyte/cation effects and quantify the spontaneous contribu

Identification of elevated urea as a severe, ubiquitous metabolic defect in the brain of patients with Huntington's disease.

Huntington's disease (HD) is a neurodegenerative disorder wherein the aetiological defect is a mutation in the Huntington's gene (HTT), which alters the structure of the huntingtin protein through the lengthening of a polyglutamine tract and initiates a cascade that ultimately leads to dementia and premature death. However, neurodegeneration typically manifests in HD only in middle age, and processes linking the causative mutation to brain disease are poorly understood. Here, our objective was to elucidate further the processes that cause neurodegeneration in HD, by measuring levels of metabol

Carbon Dioxide Fixation Cascade for the Production of Single-Cell Protein by Clostridium ljungdahlii and Yarrowia lipolytica.

Clostridium species and yeast single-cell protein (SCP), produced via industrial fermentation, offer a sustainable, high-protein alternative to conventional sources while advancing circular economy goals through CO2 valorization. Here, we developed an integrated two-stage bioprocess for SCP production from CO2. First, Clostridium ljungdahlii converted CO2 and H2 into SCP and acetate via gas fermentation. Optimizing nitrogen and sulfur feeding based on acetate synthesis rate increased acetate titer from 6 to 53.2 g/L and biomass from 1.1 to 2.8 (OD600). The fermentation broth was then directly

Cytotoxicity of nitroxyl (HNO/NO-) against normal and cancer human cells.

Nitroxyl (HNO/NO(-)) is a molecule which results from one-electron reduction of nitric oxide (NO). It is considered to be a pharmacologically important particle because of its cardiovascular effects and potential anticancer activity. Due to molecule instability studies on nitroxyl biological activity require the use of donor compounds. In the present study Angeli's salt (Na2N2O3) was used as the nitroxyl donor and cytotoxicity of HNO/NO(-) against human lymphocytes and A549 and HT29 cancer cells was examined. The studies were performed under different pH conditions (pH 6.2 and 7.4) because it

Increased Soil Nitrogen Availability Suppresses Annual Soil Respiration in Mixed Temperate Forests Regardless of Acidification.

Soil respiration (Rsoil) is the second largest terrestrial carbon (C) flux, and therefore, it is imperative to understand and quantify its responses to global environmental change. Rsoil consists of two component CO2 fluxes: autotrophic respiration from the metabolic activity of roots (Ra-root) and heterotrophic respiration (Rh) derived from the metabolic activity of mycorrhizal fungi and microbial decomposition of detritus, soil organic matter, and rhizodeposits. Increased nitrogen (N) availability often reduces Rsoil in forest ecosystems, but it remains unclear which contributing fluxes gove

The impact of urbanisation on community structure, gene abundance and transcription rates of microbes in upland swamps of Eastern Australia.

The Temperate Highland Peat Swamps on Sandstone of the Sydney Basin occur in the headwaters of Sydney's drinking water catchments and are listed as endangered ecosystems, yet they have suffered habitat losses and degradation due to human impacts such as urbanisation. Despite ongoing efforts to restore and better protect upland swamps, they remain poorly understood, potentially hindering the effectiveness of management efforts. Essential to overall ecosystem function and the provision of services for human and environmental benefit are the microbial component of wetland ecosystems. In the case

Modulating plant growth-metabolism coordination for sustainable agriculture.

Enhancing global food security by increasing the productivity of green revolution varieties of cereals risks increasing the collateral environmental damage produced by inorganic nitrogen fertilizers. Improvements in the efficiency of nitrogen use of crops are therefore essential; however, they require an in-depth understanding of the co-regulatory mechanisms that integrate growth, nitrogen assimilation and carbon fixation. Here we show that the balanced opposing activities and physical interactions of the rice GROWTH-REGULATING FACTOR 4 (GRF4) transcription factor and the growth inhibitor DELL

Synthesis and Biological Activity Assessment of Caffeic Acid Azaheterocyclic Amide Derivatives.

Natural caffeic acid derivatives are ubiquitous plant phenolic compounds with documented antitumor, antioxidant, and anti-inflammatory bioactivities. However, their clinical application is limited by low bioactivity potency and poor stability. Therefore, rational design and synthesis of novel caffeic acid derivatives are critical to improve their druglikeness and broaden biomedical applications. Herein, fourteen amide derivatives (H1-H14) were synthesized by conjugating three 3,4-substituted caffeic acid skeletons with five nitrogen-containing heterocycles. Bioactivity evaluation revealed that

Metabolic profiling of three Brachypodium species reveals different adaptive strategies to ammonium stress.

Nitrogen (N) use efficiency (NUE) in crops is a critical challenge, as only 40 % of applied nitrogen is typically recovered at harvest. Ammonium-based nutrition is proposed as a strategy to increase NUE in agrosystems. However, crops display better performance when growing with a nitrate-based nutrition. To advance in the current understanding of the interspecific variability to ammonium nutrition, we investigated the performance of the three annual species of Brachypodium genus: B. distachyon, B. stacei, and B. hybridum, focusing on their differential responses to ammonium and nitrate nutriti

The regulon of Brucella abortus two-component system BvrR/BvrS reveals the coordination of metabolic pathways required for intracellular life.

Brucella abortus is a facultative intracellular pathogen causing a severe zoonotic disease worldwide. The two-component regulatory system (TCS) BvrR/BvrS of B. abortus is conserved in members of the Alphaproteobacteria class. It is related to the expression of genes required for host interaction and intracellular survival. Here we report that bvrR and bvrS are part of an operon composed of 16 genes encoding functions related to nitrogen metabolism, DNA repair and recombination, cell cycle arrest, and stress response. Synteny of this genomic region within close Alphaproteobacteria members sugge

Metabolic regulation of Escherichia coli and its phoB and phoR genes knockout mutants under phosphate and nitrogen limitations as well as at acidic condition.

BACKGROUND: The phosphorus compounds serve as major building blocks of many biomolecules, and have important roles in signal transduction. The phosphate is involved in many biochemical reactions by the transfer of phosphoryl groups. All living cells sophisticatedly regulate the phosphate uptake, and survive even under phosphate-limiting condition, and thus phosphate metabolism is closely related to the diverse metabolism including energy and central carbon metabolism. In particular, phosphorylation may play important roles in the metabolic regulation at acidic condition and nitrogen limiting c

Nitrate modulates high CO2 effects on carbon partitioning in Gracilariopsis lemaneiformis: Trade-offs between particulate and dissolved organic carbon sequestration.

Intensive studies have explored the feasibility of using high CO2 concentrations to culture microalgae for carbon capture. Compared to microalgae, macroalgae are easier to be harvested, thereby reducing culture cost. However, little is known regarding the possibility of using macroalgae to fix and sequester high levels of CO2. In this study, Gracilariopsis lemaneiformis was cultured under low (8 μM) and high (200 μM) nitrate concentrations to clarify the physiological and molecular responses of carbon fixation and sequestration to a 10 % high CO2 concentration (HC). HC, inducing low pH, inhibi

Nitrate sources and its formation in precipitation during typhoons (In-fa and Chanthu) in multiple cities, East China.

A clear understanding of the factors governing dual isotopes (δ15N-NO3- and δ18O-NO3-) in typhoons is essential for understanding their NO3- sources and its formation mechanisms. In this study, sequential precipitation samples during typhoons, including In-fa and Chanthu, were collected from Ningbo, Hangzhou and Huzhou. The chemical compositions, nitrogen and oxygen isotopes of NO3- and oxygen isotopes of H2O (δ18O-H2O) were measured. The results showed that the δ15N-NO3- and δ18O-NO3- values ranged from -6.3‰ to 6.0‰, and 38.0‰ to 66.5‰, respectively. The lower δ18O-NO3- values (less than 52‰

Environmental evaluation of eicosapentaenoic acid production by Phaeodactylum tricornutum.

Polyunsaturated fatty acids (PUFAs) play an important role in human health. Due to the increased market demand, the production of PUFAs from potential alternative sources such as microalgae is receiving increased interest. The aim of this study was to perform a life cycle assessment (LCA) of the biotechnological production of eicosapentaenoic acid (EPA) from the marine diatom Phaeodactylum tricornutum, followed by the identification of avenues to improve its environmental profile. The LCA tackles two production schemes of P. tricornutum PUFAs with an EPA content of 36%: lab and pilot scales. T
