The Mitochondrial Guardian α-Amyrin Mitigates Alzheimer's Disease Pathology via Modulation of the DLK-SARM1-ULK1 Axis.

High consumption of colorful fruits and vegetables correlates with low dementia risk, but the exact molecules and the underlying biological mechanisms governing their bioactive profiles are largely unknown. Using a 10-year observational cohort study coupled with an AI-driven systems pharmacology platform, we identified a natural triterpenoid compound found in colorful fruits and vegetables, α-Amyrin (αA), as a therapeutic candidate for Alzheimer's disease (AD). The efficacy of αA in treating the symptoms of AD, such as Tau tangles, damaged mitochondria, and memory loss, was examined using cros

Integration of multi-omics and machine learning to reveal the anti-osteoporosis effects of Hederagenin by inhibiting lipid peroxidation via the GPX4/p-STAT1/PTGS2 axis.

Osteoporosis (OP) is a multifactorial disease, leading to abnormal bone remodeling, requiring targeted therapeutic interventions. This study investigated the pharmacological effects of Hederagenin (Hed), a triterpenoid saponin, in ovariectomized (OVX) mice. Hed treatment significantly ameliorated bone loss by improving trabecular microstructure and reducing osteoclast formation and activity. Integrated metabolomics and network pharmacology revealed that Hed primarily restored glycerophospholipid metabolic homeostasis and suppressed oxidative stress. Crucially, biochemical assays demonstrated t

The NRF2 inducer CDDO-2P-Im provokes a reduction in amyloid β levels in Alzheimer's disease model mice.

Alzheimer's disease (AD) is the most common aetiology of dementia. The transcription factor NF-E2-related factor 2 (NRF2) induces the expression of genes encoding phase II detoxification and antioxidant genes. NRF2 is regulated by Kelch-like ECH-associated protein 1 (KEAP1), and the KEAP1-NRF2 system is the key regulatory system involved in cytoprotection. To examine whether pharmacological induction of NRF2 expression alleviates AD phenotypes in vivo, we employed two AD mouse models, i.e. App  NL-G-F/NL-G-F (AppNLGF) and APPV717I::TAUP301L (APP/TAU) mice. As the synthetic oleanane triterpenoi

Saikosaponin inhibits Eimeria tenella infection by modifying the NF-κB pathway and regulating cytokines and the intestinal microbial community.

Eimeria tenella (E. tenella) is an intestinal parasite that not only endangers the health of broiler chickens but may also cause death in severe cases. However, the growing critical problem of drug resistance in E. tenella complicates therapy. Consequently, a more natural and safer technique for treating E. tenella is urgently warranted. Saikosaponin (SS) is a saponin component extracted from the traditional Chinese herb Chaihu that has been demonstrated to treat various diseases. However, little is known regarding the function of SS in E. tenella treatment. In the present investigation, SS lo

Saikosaponin A protects chickens against pullorum disease via modulation of cholesterol.

The worsening problem of antibiotic resistance prompts the need for alternative strategies that do not directly target bacteria. Virulent Salmonella pullorum strains can invade macrophages and lead to a systemic infection. Saikosaponin A (SSa), a bioactive saponin isolated from Radix bupleuri, has been demonstrated to exhibit anti-inflammatory, hepatoprotective, and cholesterol regulatory activity. The aim of this study was to investigate the effects of SSa on Salmonella-induced pullorum disease in chickens and clarify the possible mechanism. A S. pullorum-induced pullorum disease chicken mode

Natural products and long noncoding RNA signatures in gallbladder cancer: a review focuses on pathogenesis, diagnosis, and drug resistance.

Gallbladder cancer (GBC) is an aggressive and lethal malignancy with a poor prognosis. Long noncoding RNAs (lncRNAs) and natural products have emerged as key orchestrators of cancer pathogenesis through widespread dysregulation across GBC transcriptomes. Functional studies have revealed that lncRNAs interact with oncoproteins and tumor suppressors to control proliferation, invasion, metastasis, angiogenesis, stemness, and drug resistance. Curcumin, baicalein, oleanolic acid, shikonin, oxymatrine, arctigenin, liensinine, fangchinoline, and dioscin are a few examples of natural compounds that ha…

Cyclophosphamide-Mediated Induction of Myeloid-Derived Suppressor Cells In Vivo: Kinetics of Accumulation, Immune Profile, and Immunomodulation by Oleanane-Type Triterpenoids.

Myeloid-derived suppressor cells (MDSCs) are immature myeloid cells that strongly suppress immunity and expand during tumor progression. Various antitumor chemotherapy agents can induce MDSC accumulation, reducing treatment effectiveness. We investigated the impact of the CHOP regimen and its components (cyclophosphamide (CTX), doxorubicin, vincristine, and prednisolone) on the dynamics of MDSC accumulation and the associated changes in immune cell profiles in the peripheral blood and spleen of healthy and lymphosarcoma RLS40-bearing CBA mice. CHOP induced significant thymic atrophy and spleno
