Two differing salmon GnRH precursor mRNAs are co-expressed in the brain of sockeye salmon.

The localization of two salmon-type gonadotropin-releasing hormone (sGnRH) precursors, pro-sGnRH-I (short type) and pro-sGnRH-II (long type), was investigated by using in situ hybridization techniques in the brain of the landlocked sockeye salmon, Oncorhynchus nerka. We used 30-mer oligonucleotide probes complementary to pro-sGnRH-I and pro-sGnRH-II cDNA. No significant differences were observed in the localization of sGnRH neurons expressing pro-sGnRH-I and pro-sGnRH-II mRNAs; both were expressed in the olfactory nerve, the olfactory bulbs, the regions between the olfactory bulb and telenceph…

Piscirickettsiosis in Farmed Turbot, Scophthalmus maximus.

Piscirickettsia salmonis is a well-established intracellular pathogen of farmed Atlantic salmon around the world. The bacterium has also been detected from a number of other hosts including lumpfish (Cyclopterus lumpus), seabass (Dicentrarchus labrax) and rainbow trout (Oncorhynchus mykiss). In February 2025, increased mortality was observed in a population of turbot, Scophthalmus maximus, farmed in Norway. Macroscopic signs of disease included stomach mucosa bleedings. Piscirickettsiosis was diagnosed based on histopathological findings including rickettsia-like inclusions immunostaining posi…

Rapid Detection of Sublethal Toxicity Using Locomotor Activity of Rainbow Trout Juveniles.

This study examined changes in locomotor activity of the rainbow trout (Oncorhynchus mykiss) juveniles exposed to sublethal concentrations of hexavalent chromium (Cr6+) (as a single pollutant) and landfill leachate (as an complex mixture of mainly organic compounds, including trace amounts of metals). Fish were first examined for baseline behavior patterns for 3 h to determine the appropriate duration of acclimation and the control level. The average velocity of fish was found to be the most informative among other endpoints throughout the 3 h long acclimation. Under the effect of both test su…

Triploidy in rainbow trout determined by computer-assisted analysis.

This study was designed to assess the use of a computer-assisted system based on erythrocyte measurements as a possible alternative to flow cytometry for identifying triploid rainbow trout (Oncorhynchus mykiss). Blood smears were prepared from 26 triploid and 26 diploid specimens, as determined by flow cytometry after staining blood cells with propidium iodide. The cell and nucleus lengths of 10 erythrocytes were determined in each fish. This was followed by discriminatory analysis to distinguish between diploids and triploids based on their score profiles. Triploid trout showed significantly …

Immune cell-resolved transcriptomics provides insights into the basis for variations of fish genetic resistance to viral disease.

BACKGROUND: The genetic basis of host resistance to viral infections is generally shaped by complex interactions between host genetic variations affecting antiviral immunity and the rapid evolutionary adaptability of viruses. In this study, we investigated two isogenic rainbow trout lines exhibiting extreme resistance or susceptibility to the rhabdovirus VHSV. We compared transcriptomes of the pronephros - a major lymphoid organ in fish - at steady state and following VHSV infection. By integrating bulk tissue RNA-seq with single-cell RNA-seq, we mapped the divergent transcriptomic responses o

Non-microcystin extracellular metabolites of Microcystis aeruginosa impair viability and reproductive gene expression in rainbow trout cell lines.

Microcystis aeruginosa is a ubiquitous freshwater cyanobacterium best known for producing hepatotoxic microcystins; however, this common bloom-forming species also produces myriad biologically active and potentially deleterious other metabolites. Our understanding of the effects of these non-microcystin metabolites on fish is limited. In this study, we evaluated cytotoxicity of extracellular metabolites harvested from both microcystin-producing (MC+) and non-producing (MC-) strains of M. aeruginosa on rainbow trout (Oncorhynchus mykiss) cell lines derived from tissues of the brain, pituitary, …

The Cytotoxicity, DNA Fragmentation, and Decreasing Velocity Induced By Chromium(III) Oxide on Rainbow Trout Spermatozoa.

The present study aimed to determine the cytotoxicity of chromium(III) oxide micro particles (Cr2O3-Ps) in rainbow trout (Oncorhynchus mykiss) spermatozoa. Firstly, Cr2O3-Ps were synthesized and structurally characterized the surface, morphological for particle size and thermal properties. In addition, its structural and elemental purity was determined using energy-dispersive X-ray (EDX) spectrum and elemental maps. Structural purity, thermal properties, and stability of Cr2O3-Ps were also examined in detail by performing thermal analysis techniques. The cytotoxicity of Cr2O3-Ps was measured b…

Effect of copper nanoparticles and copper ions on the architecture of rainbow trout olfactory mucosa.

Olfactory epithelial cells are in direct contact with myriad environmental contaminants which may consequently disrupt their structure and function. Copper ions (Cu2+) and copper nanoparticles (CuNPs) are two types of olfactory neurotoxicants. However, their effects on the structure of olfactory epithelium are largely uninvestigated. The density of olfactory goblet cells in CuNP- and Cu2+ - exposed rainbow trout was assessed using light microscopy throughout time. In both copper (Cu) treatments, the number of goblet cells increased initially over the 24 h exposure and then recovered to normal

The impact of Tetracapsuloides bryosalmonae and Myxobolus cerebralis co-infections on pathology in rainbow trout.

BACKGROUND: Myxozoan parasites pose emerging health issues for wild and farmed salmonid fish. Rainbow trout (Oncorhynchus mykiss) is a particularly susceptible species to Tetracapsuloides bryosalmonae (Malacosporea), the etiological agent of Proliferative Kidney Disease (PKD), and to Myxobolus cerebralis (Myxosporea), the etiological agent of Whirling Disease (WD). The objective of this study was to investigate the impact of myxozoan co-infections on the pathogenesis of PKD and WD in the rainbow trout. METHODS: Two groups of rainbow trout (96 fish each) were primarily infected with T. bryosal…

Histopathologic Changes in Disease-Resistant-Line and Disease-Susceptible-Line Juvenile Rainbow Trout Experimentally Infected with Flavobacterium psychrophilum.

Abstract A challenge to selectively breeding fish populations for improved disease resistance in aquaculture is an incomplete understanding of how artificial selection affects innate immunity at the host-pathogen level. The objective of this study was to determine whether Rainbow Trout Oncorhynchus mykiss bred for differential susceptibility to bacterial cold-water disease exhibited altered tissue damage and cellular inflammatory response following experimental challenge with Flavobacterium psychrophilum. Fish from disease-resistant (ARS-Fp-R) and disease-susceptible (ARS-Fp-S) lines were expe…

Behavioral responses by migratory chum salmon to amino acids in natal stream water.

We propose that amino acids in natal stream water have Important roles in Pacific salmon homing. This study hypothesized that amino acids found in natal stream water have a role in the ability of mature male chum salmon (Oncorhynchus keta) to home to the Osaru River (OR), Hokkaido, Japan. Behavioral experiments were conducted in a two-choice test tank using various combinations of control water (natural Toya Lake water; NLW and three artificial stream waters using amino acids: 1) artificial OR water (AOR); 2) AOR without L-glutamic acid, the major amino acid in OR water (AOR-E); and 3) artific…

Aquatic ecological risks due to cyanide releases from biomass burning.

Aquatic toxicity due to the creation and mobilization of chemical constituents by fire has been little studied, despite reports of post-fire fish kills attributed to unspecified pyrogenic toxicants. We examined releases of cyanides from biomass burning and their effect on surface runoff water. In laboratory test burns, available cyanide concentrations in leachate from residual ash were much higher than in leachate from partially burned and unburned fuel and were similar to or higher than the 96-h median lethal concentration (LC50) for rainbow trout (45 microg/l). Free cyanide concentrations in

Comparative palatability and efficacy of two praziquantel isomers in rainbow trout infected by eyefluke.

Praziquantel is a widely used anthelmintic in both human and veterinary medicine, but low palatability of the commonly used compound, which is a mixture (M) of two isomers (D and L forms), may result in rejection of oral treatments. Purification of the D-isomer of praziquantel allows a comparative evaluation of both palatability and efficacy of the isomers. We showed that rainbow trout (Oncorhynchus mykiss) preferred feed containing D-PZQ feed (60% acceptance) compared to M-PZQ feed (20% acceptance) (inclusion of 400 g PZQ/kg feed). Subsequent efficacy studies using eyefluke Diplostomum pseudo…

Enantioselective toxicokinetics drive the differential toxicity of chiral 6PPD-quinone in rainbow trout.

The tire-derived contaminant N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPD-Q) is widespread in aquatic ecosystems and poses significant ecological risk. As a chiral compound, its enantiomers may exhibit differential toxicity, yet the underlying mechanisms remain limited. Here, we found that (+)-6PPD-Q was 2.4-fold more toxic to rainbow trout than (-)-6PPD-Q (p < 0.05). In addition, in vitro metabolism assays using liver S9 fraction showed that (+)-6PPD-Q was metabolized 2.5-fold faster than (-)-6PPD-Q. Semi-quantitative nontarget analysis revealed that hydroxylated metabolit

Circulating microRNAs reveal egg-brain crosstalk and a brain-predominant microRNA linked to the onset of the next reproductive cycle in iteroparous salmonids.

BACKGROUND: Mechanisms regulating the transition between two consecutive reproductive cycles are complex and remain poorly understood, mostly because they involve a dialog between the ovary and the central nervous system that is difficult to disentangle. RESULTS: In rainbow trout (Oncorhynchus mykiss), an iteroparous species spawning every year, removal of the eggs from the body cavity was used as a switch to trigger the onset of the next reproductive cycle. Changes in circulating miRNAs (c-miRNAs) levels in blood plasma and ovarian fluid were then monitored over time. Upon removal of the egg…

Neuroanatomical profiling of the rainbow trout brain parenchyma and meninges reveals specialized immune niches and region-specific hubs for bacterial immune surveillance.

Several studies have described immune responses in the teleost brain and meninges during infection, however, fundamental studies that systematically dissect how different regions of the brain maintain immune homeostasis in teleosts are missing. Here we present an in-depth investigation of the immune status of the brain parenchyma and meninges of juvenile rainbow trout (Oncorhynchus mykiss) at the steady state. We dissected four parenchymal brain regions including olfactory bulbs (OB), telencephalon (Tel), optic tectum (OT) and cerebellum (Cer) and its corresponding dorsal meninges. Gene expres…

Stunning techniques and climatic influence in rainbow trout: Sensibility state, welfare and recovery ability.

This study evaluated the effects of climatic conditions and three stunning techniques on the stress response, sensibility state, and recovery ability of rainbow trout (Oncorhynchus mykiss). Stunning methods included ice-water immersion (ICE) and two electric shock treatments combined with ice-water immersion (E200: 200 mA for 2 s; E400: 400 mA for 0.5 s followed by 200 mA for 1.5 s). Rainbow trout were exposed to these methods in winter and summer to assess the impact of seasonality. Under winter conditions, fish in the ICE group retained sensibility, whereas electrically stunned fish lost sen…
