Immunogenic integral membrane proteins of Borrelia burgdorferi are lipoproteins.

The pathogenic spirochete Borrelia burgdorferi contains a set of integral membrane proteins which were selectively extracted into the detergent phase upon solubilization of intact B. burgdorferi with the nonionic detergent Triton X-114. Virtually all of these hydrophobic proteins were recognized by antibodies in pooled sera from patients with chronic Lyme arthritis, demonstrating that proteins partitioning into the detergent phase of Triton X-114 encompass the major B. burgdorferi immunogens. Furthermore, most of these immunogenic proteins, including the previously characterized OspA and OspB

Measurement of true salicylate concentrations in serum from patients with Reye's syndrome.

Patients with Reye's syndrome who have been given aspirin are said to maintain higher-than-anticipated salicylate concentrations in blood, for longer than expected. We explored whether this could be attributed to spurious results from nonsalicylate compounds in the Trinder reaction for salicylates. All of 63 organic acids and amines examined that form colored complexes with Trinder's reagent had detectable absorbance at 540 nm at 0.2 g/L, including some endogenous compounds known to be increased in Reye's syndrome patients and many others endogenous in humans. By subjecting deproteinized sera

High α-tocopherol dosing increases lipid metabolism by changing redox state in damaged rat gastric mucosa and liver after ethanol treatment.

Regeneration of ethanol-injured rat gastric mucosa must undergo changes in major metabolic pathways to achieve DNA replication and cell prol … the mitochondrial redox NAD/NADH ratio was much less affected. In addition, α-tocopherol increased palmitic acid utilization in the gastric mucosa, which was accompanied by the induction of 'mirror image' effects on the cell redox state, reflected in an inhibited cell gastric mucosa proliferation by the vitamin administration. In conclusion, the present study …

[Effects of various modes of therapy on the fatty acid spectrum in hyperlipoproteinemia].

Gas-chromatographic investigations of the fatty acid pattern of the fractions of serum triglyceride and serum cholesterol ester revealed characteristic changes under different therapeutic measures in altogether 120 patients with primary hyperlipoproteinaemia of the types IIa, IIb and IV. Already under the treatment with basal metabolic diet, more clearly by medication with clofibrin acid, in the hyperlipoproteinaemia type IV the decrease of the palmitic acid, the palmitoleic acid, the stearic acid and oleic acid and the increase of linoleic acid, linoleneic acid, arachidonic acid and eikopenta…

Probing the Global Cellular Responses to Lipotoxicity Caused by Saturated Fatty Acids.

Excessive levels of saturated fatty acids are toxic to cells, although the basis for this lipotoxicity remains incompletely understood. Here, we analyzed the transcriptome, lipidome, and genetic interactions of human leukemia cells exposed to palmitate. Palmitate treatment increased saturated glycerolipids, accompanied by a transcriptional stress response, including upregulation of the endoplasmic reticulum (ER) stress response. A comprehensive genome-wide short hairpin RNA (shRNA) screen identified >350 genes modulating lipotoxicity. Among previously unknown genetic modifiers of lipotoxicity,

A component-resolved in vitro skin barrier model for assessing nanoplastic retention and barrier susceptibility.

Human-relevant in vitro models are needed to assess dermal nanoplastic hazards, yet current skin penetration approaches often treat the stratum corneum as a compositionally uniform barrier. This limits mechanistic understanding of how barrier biochemistry relates to nanoplastic retention, particularly in skin states with altered lipid or protein organization. Here, we developed a component-resolved in vitro stratum corneum model to quantify interactions between 50 nm polystyrene nanoplastics and six major skin barrier constituents: ceramide, cholesterol, keratin, palmitic acid, proline, and ph…

The effect of dietary ethanol on the composition of lipids of Drosophila melanogaster larvae.

At a moderate concentration (2.5%, v/v) dietary ethanol reduced the chain length of total fatty acids (FA) and increased the desaturation of short-chain FA in Drosophila melanogaster larvae with a functional alcohol dehydrogenase (ADH). The changes in length in total FA were postulated to be due to the modulation of the termination specificity of fatty acid synthetase. Because the ethanol-stimulated reduction in the length of unsaturated FA was blocked by linoleic acid, it was thought to reflect the properties of FA 9-desaturase. Although the ethanol-stimulated reduction in chain length of uns

Nucleotide sequence of a cDNA clone encoding the entire glycoprotein from the New Jersey serotype of vesicular stomatitis virus.

The nucleotide sequence of the mRNA encoding the glycoprotein from the New Jersey serotype of vesicular stomatitis virus (VSV) was determined from a cDNA clone containing the entire coding region. The sequence of 12 5'-terminal noncoding nucleotides present in the mRNA but not in the cDNA clone was determined from a primer extended to the 5' terminus of the mRNA. The mRNA is 1,573 nucleotides long (excluding polyadenylic acid) and encodes a protein of 517 amino acids. Only six nucleotides occur between the translation termination codon and the polyadenylic acid. Short homologies between the un

Lipid-driven epigenetic exhaustion: linking palmitate metabolism to CD8+ T-cell dysfunction in cancer.

In a recent study published in Science Immunology, Tiberti and colleagues demonstrate that palmitate, a saturated fatty acid enriched in tumors, directly impairs CD8+ cytotoxic T lymphocyte function through mitochondrial and epigenetic reprogramming. Palmitate exposure reduced mitochondrial fitness, oxidative phosphorylation, and adenosine triphosphate production, resulting in defective proliferation, cytokine production, and antitumor activity. Mechanistically, mitochondrial dysfunction decreased intracellular acetyl-CoA availability, leading to reduced histone acetylation and loss of chromat

Microwave-assisted click synthesis of levoglucosan triazolyl glycoconjugates using ascorbyl palmitate sodium salt (Asp-Na) as a green Co-catalyst.

Ascorbyl palmitate sodium salt (Asp-Na) 2 has been found to be a highly effective co-catalyst in copper catalyzed azide-alkyne cycloaddition (CuAAC) reactions under microwave (MW) condition. This co-catalyst provides a more cost-efficient option compared to sodium ascorbate by reducing the stoichiometric amount of ascorbic acid required for the CuAAC reaction. With its palmitate side chain, Asp-Na 2 acts as a surfactant by improving the solubility of substrates, particularly the lipophilic compounds. Notably, it can be obtained in high purity by semi-synthesis from non-edible oils, aligning wi

The Myxobacterial Metabolite Soraphen A Inhibits HIV-1 by Reducing Virus Production and Altering Virion Composition.

Soraphen A is a myxobacterial metabolite that blocks the acetyl-coenzyme A carboxylase of the host and was previously identified as a novel HIV inhibitor. Here, we report that soraphen A acts by reducing virus production and altering the gp120 virion content, impacting entry capacity and infectivity. These effects are partially reversed by addition of palmitic acid, suggesting that inhibition of HIV envelope palmitoylation is one of the mechanisms of antiviral action.
