Pancreatic acinar cell pyroptosis played a key role in acute pancreatitis (AP), while the underlying molecular mechanisms were unclear. In this study, hyperlipidemic acute pancreatitis (HLAP) model mice and primary acinar cells were employed for the in vivo and in vitro experiments, respectively. Western blot was employed to assess Gasdermin D-N-terminal pore-forming domain (GSDMD-N) oligomerization; Flow cytometry was employed to assess cell pyroptosis; Immunofluorescence was performed to assess GSDMD-N location in the cytoplasm; HE and TUNEL staining were employed to assess the pathological

BACKGROUND: Obesity-related hypertension (OH) is driven by endothelial dysfunction, chronic inflammation, and oxidative stress, yet effective targeted therapies are lacking. This study aims to investigate the mechanisms by which brown adipose tissue-derived exosomes (BAT-Exos) improve vascular endothelial function in OH via HuR protein delivery.
METHODS: Rat models with OH and palmitic acid-treated endothelial cell injury models were established. BAT-Exos were compared with white adipose tissue-derived exosomes (WAT-Exos) for therapeutic efficacy. In vitro, primary endothelial cells isolated

OBJECTIVE: To identify the role of triglyceride-rich lipoproteins (TGRLs) and apoE, a major apolipoprotein in TGRLs, in adipose tissue inflammation with high-fat diet (HFD)-induced obesity.
METHODS: Male apoE(-/-) and C57BL/6J wild-type (WT) mice fed HFD for 12 weeks were assessed for metabolic and inflammatory parameters. ApoE(-/-) and WT mice were orally gavaged with [(3)H]palmitic acid to examine the role of apoE in fat delivery to adipose tissue. VLDL from obese apoE(-/-) mice were intravenously injected into lean WT or apoE(-/-) mice to test potential contribution of TGRLs-derived fat de

Diabetic cardiomyopathy is characterized by interstitial fibrosis and cardiomyocyte hypertrophy and apoptosis. Also known as CCN2, connective tissue growth factor (CTGF) is implicated in the fibrosis; however, whether it contributes to cardiomyocytes changes and adverse effects of high glucose and lipids on these cells remains unknown. Hearts from streptozotocin-induced diabetic rats had elevated CTGF and changes of pathological myocardial hypertrophy, fibrosis, and cardiomyocyte apoptosis. Rat H9c2 cardiomyocytes were then treated with recombinant human (rh)CTGF, high glucose, or the saturate

Acne vulgaris, an epidemic inflammatory skin disease of adolescence, is closely related to Western diet. Three major food classes that promote acne are: 1) hyperglycemic carbohydrates, 2) milk and dairy products, 3) saturated fats including trans-fats and deficient ω-3 polyunsaturated fatty acids (PUFAs). Diet-induced insulin/insulin-like growth factor (IGF-1)-signaling is superimposed on elevated IGF-1 levels during puberty, thereby unmasking the impact of aberrant nutrigenomics on sebaceous gland homeostasis. Western diet provides abundant branched-chain amino acids (BCAAs), glutamine, and p

CONTEXT: Little is known about the link between the endocannabinoid (EC) system and the in vivo metabolic function of white adipose tissue (WAT).
OBJECTIVE: We aimed to evaluate whether ECs are linked to postprandial fatty acid metabolism and WAT metabolic function.
METHODS: Men and women, with (IGT, n = 20) or without impaired glucose tolerance (NGT, n = 20) underwent meal testing with oral and intravenous stable isotope palmitate tracers and positron emission tomography with intravenous [11C]-palmitate and oral [18F]-fluoro-thia-heptadecanoic acid to determine systemic and organ-specific d

BACKGROUND: Previous studies suggested that fibrillar human IAPP (hIAPP) is more likely to deposit in β-cells, resulting in β-cell injury. However, the changes in the conformation of hIAPP in lipid environment and the mechanism involved in β-cell damage are unclear.
METHODS: Synthetic hIAPP was incubated with five types of free fatty acids and phospholipids 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) and 1-palmitoyl-2-oleoyl-sn-glycero-3-phospho-l-serine (POPS), which constitute the cell membrane. Thioflavin-T fluorescence assay was conducted to analyze the degree of hIAPP fibrosi

Between 1 and 7 days of life, the newborn rabbit heart shifts from predominantly using carbohydrates to predominantly using fatty acids as an energy substrate. We therefore used isolated working hearts from 1- or 7-day-old rabbits to determine the effects of fatty acids on myocardial glucose use and the ability of hearts to recover following various periods of transient no-flow ischemia. One-day-old hearts were perfused via the inferior vena cava and ejected buffer through the cannulated aorta and pulmonary artery. Seven-day-old hearts were perfused via the left atrium and ejected buffer throu

The effect of vitamin A supplementation on stearoyl-CoA desaturase gene 1 expression in mouse liver was characterized. Normal BALB/c mice were fed 0.01% and 0.1% retinol palmitate as components of nonpurified diets. This treatment resulted in a 3-fold and a 7-fold induction of SCD1 mRNA levels, respectively, as determined by RNase protection analysis. Vitamin A-deficient animals were also fed diets containing 0.01% and 0.1% retinol palmitate, resulting in a similar pattern of SCD1 mRNA induction. Fatty acid synthase and beta-actin mRNA levels did not respond consistently or significantly to re

These studies were performed to follow a spectrum of relevant parameters in male Syrian golden hamsters fed either a vitamin A-deficient diet or the same diet supplemented with retinoic acid at 3 micrograms/g diet. Body weight and life span were not affected by the vitamin A-deficient diet until after 6-7 wk. Squamous metaplastic lesions of the Formalin-fixed tracheas were not generally observed in the hamsters fed the deficient diet until 6-7 wk, at which time blood retinol and liver retinyl palmitate levels had also decreased. Blood glucose levels remained normal (90 mg/dl) until about 7 wk

Stimulatory effects of saturated fatty acids consisting of 4 (butyrate), 8 (octanoate), 12 (laurate) and 16 (palmitate) carbon atoms, as well as acetylcholine on pancreatic amylase release were assessed in tissue segments isolated from sheep, rats, hamsters, field voles and mice. The amount of amylase release induced by the fatty acids (1 mumol.l-1 to 10 mml.l-1) and by acetylcholine (10 nmol.l-1 to 100 mumol.l-1) increased in a concentration-dependent manner, and the maximum response in response to the fatty acids was obtained at the maximal dose used. The maximum increase in amylase release

BACKGROUND: Endothelial dysfunction and monocyte adhesion driven by free fatty acids (FFAs) are critical early events in atherosclerosis. Palmitate, a long-chain saturated free fatty acid, upregulated GPR84 expression. Although GPR84 is canonically activated by medium-chain FFAs, its induction under lipotoxic stress suggests a compensatory, context-dependent role. Its role in endothelial inflammation is poorly defined.
OBJECTIVES: To investigate the functional role of GPR84 activation in FFA-induced endothelial inflammation and elucidate the underlying mechanisms.
METHODS: Human aortic endot

This study optimized ultrasonic-assisted ethanol extraction of dried Litsea cubeba fruits using response surface methodology, identified chemical components by GC‑MS, and explored the antibacterial mechanism against Staphylococcus aureus. The optimal conditions were 60 °C, 60 min, and solid‑liquid ratio 1:25 g/mL, with an antibacterial rate of 83.42%. GC‑MS detected 20 compounds, dominated by lauric acid, oleic acid, and palmitic acid. The extract showed concentration‑dependent bactericidal activity with MIC 0.16 mg/mL and MBC 0.31 mg/mL. It disrupted bacterial cell wall and membrane integrity

Sarcosphaera crassa is an edible but deadly mushroom that is poisonous in its raw form. In this study, fatty acids profiles, total phenolic, along with in vitro antioxidant and anticholinesterase activities of baked and raw forms of S. crassa were reported for the first time. Linoleic acid (53.6%), oleic acid (24.2%), and palmitic acid (11.5%) were present in the highest amount in the baked sample, while oleic acid (50.5%), palmitic acid (22.4%) and linoleic acid (16.6%) were found in the raw petroleum ether extract (in phenolic equivalents, PEs). PE extracts of raw and baked samples exhibited

Palm oil is the world's most commonly used vegetable oil and extracted both from fruit and seed of palm tree. However, its high saturated fatty acid content raised controversies over consumption of the oil. Few scientific findings suggest it as a risk factor for cardiovascular disease and increased consumer's awareness over healthy diet raised claim over it. So that, this article aimed to review literatures on palm oil extraction process and its positive and negative health consequences and besides suggest strategies for healthy diet. Literature search of relevant articles was conducted by usi

Alveolar Type II cells were isolated from control and vitamin A deficient rats and allowed to form a monolayer in plastic dishes for 16-18 hours. The vitamin A content (retinol plus retinyl palmitate) of deficient cells was 50-75% less than in control cells on a per mg protein basis. Isolated Type II cells took up [3H]-retinol, synthesized [3H]-retinyl palmitate, and after 4 hours, 24% of the radioactivity in the Type II cells was [3H]-retinoic acid. Deficiency did not appear to alter retinoic acid synthesis. Phosphatidylcholine (PC) and disaturated phosphatidylcholine (DSPC) synthesis, were s

Non-alcoholic fatty liver disease (NAFLD) is characterized by the accumulation of lipids within hepatocytes, which compromises liver functionality following mitochondrial dysfunction and increased production of reactive oxygen species (ROS). Lipoic acid is one of the prosthetic groups of the pyruvate dehydrogenase complex also known for its ability to confer protection from oxidative damage because of its antioxidant properties. In this study, we aimed to investigate the effects of lipoic acid on lipotoxicity and mitochondrial dynamics in an in vitro model of liver steatosis. HepG2 cells were

Vitamin C (ascorbic acid) is a non-enzymatic antioxidant important in protecting the lung against oxidative damage and is decreased in lung lining fluid of horses with airway inflammation. To examine possible therapeutic regimens in a species with ascorbate-synthesising capacity, we studied the effects of oral supplementation of two forms of ascorbic acid, (each equivalent to 20 mg ascorbic acid per kg body weight) on the pulmonary and systemic antioxidant status of six healthy ponies in a 3 x 3 Latin square design. Two weeks supplementation with ascorbyl palmitate significantly increased mean
