Comprehensive machine learning analysis of PANoptosis signatures in multiple myeloma identifies prognostic and immunotherapy biomarkers.

PANoptosis is closely associated with tumorigenesis and therapeutic response, yet its role in multiple myeloma (MM) remains unclear. This study analyzed bulk transcriptomic and clinical data from the TCGA and GEO databases to identify seven PANoptosis-related genes (PRGs) using machine learning (LASSO regression and random forest models) and univariate Cox analysis, and constructed a prognostic risk model. The model demonstrated robust predictive performance across three external validation cohorts. High-risk patients exhibited higher tumor purity, increased tumor mutational burden, and distin

A Retrospective Study of Climate Change Affecting Dengue: Evidences, Challenges and Future Directions.

Climate change is unexpected weather patterns that can create an alarming situation. Due to climate change, various sectors are affected, and one of the sectors is healthcare. As a result of climate change, the geographic range of several vector-borne human infectious diseases will expand. Currently, dengue is taking its toll, and climate change is one of the key reasons contributing to the intensification of dengue disease transmission. The most important climatic factors linked to dengue transmission are temperature, rainfall, and relative humidity. The present study carries out a systematic

Applying bioinformatics to proteomics: is machine learning the answer to biomarker discovery for PD and MSA?

Bioinformatics tools are increasingly being applied to proteomic data to facilitate the identification of biomarkers and classification of patients. In the June, 2012 issue, Ishigami et al. used principal component analysis (PCA) to extract features and support vector machine (SVM) to differentiate and classify cerebrospinal fluid (CSF) samples from two small cohorts of patients diagnosed with either Parkinson's disease (PD) or multiple system atrophy (MSA) based on differences in the patterns of peaks generated with matrix-assisted desorption/ionization time-of-flight mass spectrometry (MALDI

Development and validation of an interpretable machine learning model for pulmonary heart disease in patients with pneumoconiosis.

BACKGROUND: Pneumoconiosis patients with pulmonary heart disease (PHD) face an elevated risk of mortality. Early identification of individuals at higher risk of PHD is important for timely risk stratification and clinical management. However, few risk-screening models have been specifically developed for pneumoconiosis in primary care settings, where access to advanced diagnostic resources may be limited. Therefore, this study developed and validated a machine learning model based on routinely available data to support early risk stratification and clinical decision-making in resource-limited

Discrimination of secondary hypsarrhythmias to Zika virus congenital syndrome and west syndrome based on joint moments and entropy measurements.

Hypsarrhythmia is a specific chaotic morphology, present in the interictal period of the electroencephalogram (EEG) signal in patients with West Syndrome (WS), a severe form of childhood epilepsy and that, recently, was also identified in the examinations of patients with Zika Virus Congenital Syndrome (ZVCS). This innovative work proposes the development of a computational methodology for analysis and differentiation, based on the time-frequency domain, between the chaotic pattern of WS and ZVCS hypsarrhythmia. The EEG signal time-frequency analysis is carried out from the Continuous Wavelet

Predicting Antidepressant Treatment Response From Cortical Structure on MRI: A Mega-Analysis From the ENIGMA-MDD Working Group.

Accurately predicting individual antidepressant treatment response could expedite the lengthy trial-and-error process of finding an effective treatment for major depressive disorder (MDD). We tested and compared machine learning-based methods that predict individual-level pharmacotherapeutic treatment response using cortical morphometry from multisite longitudinal cohorts. We conducted an international analysis of pooled data from six sites of the ENIGMA-MDD consortium (n = 262 MDD patients; age = 36.5 ± 15.3 years; 154 (59%) female; mean response rate = 57%). Treatment response was defined as

Clinical Prediction of Functional Decline in Multiple Sclerosis Using Volumetry-Based Synthetic Brain Networks.

BACKGROUND: Disability progression in multiple sclerosis (MS) is increasingly recognized as a consequence of large-scale brain network disruption rather than isolated regional damage. Although diffusion tensor imaging (DTI) is the reference method for assessing structural connectivity, its limited availability restricts widespread clinical application. There is therefore a critical need for alternative approaches capable of capturing network-level alterations using routinely acquired MRI data. OBJECTIVE: This study aimed to determine whether synthetic structural connectivity matrices derived

Particulate Matter Exposure and Diabetic Kidney Dysfunction: Insights from Integrated Transcriptomic and Bioinformatics Analyses.

Exposure to ambient particulate matter (PM) has been linked to renal dysfunction, particularly in diabetic populations, but the underlying mechanisms remain unclear. We performed bidirectional Mendelian randomization to assess causal relationships between PM exposure and estimated glomerular filtration rate (eGFR), integrated transcriptomic datasets to identify PM-related genes overlapping with diabetic kidney disease (DKD) differentially expressed genes, and applied machine learning approaches to select key feature genes and construct diagnostic models. Single-cell and spatial transcriptomic

Synaptic dysfunction-related gene expression spatially correlates with widespread cortical network atrophy in temporal lobe epilepsy.

Temporal lobe epilepsy (TLE) exhibits marked lateralized gray matter alterations, yet whole-brain network vulnerability patterns, particularly those independent of seizure laterality, remain incompletely understood. Furthermore, the spatial correspondence between macroscopic network disruptions and underlying molecular architectures lacks systematic characterization, limiting mechanistic insights into epileptic network pathology. We utilized voxel-based morphometry (VBM) and surface-based morphometry (SBM) to characterize whole-brain gray matter alterations in 69 TLE patients (34 left-TLE, 35

Comparative analysis of breast cancer detection in mammograms and thermograms.

In this paper, we present a system based on feature extraction techniques for detecting abnormal patterns in digital mammograms and thermograms. A comparative study of texture-analysis methods is performed for three image groups: mammograms from the Mammographic Image Analysis Society mammographic database; digital mammograms from the local database; and thermography images of the breast. Also, we present a procedure for the automatic separation of the breast region from the mammograms. Computed features based on gray-level co-occurrence matrices are used to evaluate the effectiveness of textu
