Immunometabolic determinants of long-term response in leukemia patients receiving CD19 CAR T cell therapy.

Although most patients with relapsed/refractory B-cell acute lymphoblastic leukemia (B-ALL) receiving CD19-targeted chimeric antigen receptor (CAR) T cell therapy achieve remission, loss of CAR T cell functionality and subsequent relapse remains an unmet therapeutic need. Herein, we apply an integrative approach to study the immunometabolism of pre- and post-infusion CD19-CAR T cells of patients with relapsed/refractory B-ALL. Pre-infusion CAR T cells of long-term responders (LTR) have increased oxidative phosphorylation, fatty acid oxidation, and pentose phosphate pathway activities, higher m

Polyploid cancer cells surviving cisplatin reallocate central carbon sources to fuel antioxidant metabolism for survival.

Therapy resistance is the leading cause of cancer-related deaths. Polyploid cancer cells mediate resistance through adaptive cell states transitions that promote survival and tumor recurrence. Here, we investigate metabolic differences between cisplatin-surviving polyploid cells and parental cancer cells using integrated fluxomics. Transcriptomic and proteomic profiling and extracellular flux analyses revealed that surviving cells upregulate glycolysis and gluconeogenesis while reducing oxidative phosphorylation, indicating a shift in central carbon metabolism. Isotope tracing and metabolic mo

Transcriptional profiling of Saccharomyces cerevisiae T2 cells upon exposure to hardwood spent sulphite liquor: comparison to acetic acid, furfural and hydroxymethylfurfural.

Global gene expression was analyzed in Saccharomyces cerevisiae T2 cells grown in the presence of hardwood spent sulphite liquor (HW SSL) and each of the three main inhibitors in HW SSL, acetic acid, hydroxymethyfurfural (HMF) and furfural, using a S. cerevisiae DNA oligonucleotide microarray. The objective was to compare the gene expression profiles of T2 cells in response to the individual inhibitors against that elicited in response to HW SSL. Acetic acid mainly affected the expression of genes related to the uptake systems of the yeast as well as energy generation and metabolism. Furfural

Could thiamine pyrophosphate be a regulator of the nitric oxide synthesis in the endothelial cell of diabetic patients?

Thiamine (Vitamin B1) is considered an essential micronutrient for humans; its deficient intake brings about the Wernicke-Korsakoff syndrome (encephalopathy and psychosis) or beriberi (a neurological and cardiovascular disease). Once thiamine enters the cells it is phosphorylated by thiamine pyrophosphokinase (TPPK), and converted into the coenzyme thiamine pyrophosphate (TPP), the active form of thiamine. TPP is a relevant cofactor for transketolase (TK), α-ketoglutarate dehydrogenase (αKDH), and pyruvate dehydrogenase (PDH), all these enzymes are fundamental for glucose metabolism. Diabetes

Inhibition of hexose monophosphate shunt in young erythrocytes by pyrimidine nucleotides in hereditary pyrimidine 5' nucleotidase deficiency.

Recent reports have suggested that haemolytic anaemia in pyrimidine 5' nucleotidase (P5'N) deficiency might be due to impaired erythrocyte hexose monophosphate shunt (HMS). To investigate the relationship between pyrimidine accumulation, HMS impairment and shortened red-cell survival, we tested glucose 6-phosphate dehydrogenase (G-6PD), HMS, P5'N activities and the UV spectrum in whole red cells and in red cells of different age from 2 P5'N-deficient patients with different degrees of haemolytic anaemia. In whole red cells we found a reduction of both G-6PD and stimulated HMS activity in the p

The activation of gold complexes by cyanide produced by polymorphonuclear leukocytes--I. The effects of aurocyanide on the oxidative burst of polymorphonuclear leukocytes.

It has been suggested that the antiarthritic gold complex, aurothiomalate (Autm), is activated by its conversion to aurocyanide by polymorphonuclear leukocytes (PMN) which generate cyanide from thiocyanate. In an examination of this hypothesis, a study has been conducted on the effects of aurocyanide on the oxidative burst of polymorphonuclear leukocytes (PMN) and monocytes activated by phorbol myristate acetate (PMA). Aurocyanide produced delayed inhibition of the oxidative burst as shown by its effect on both lucigenin and luminol-dependent chemiluminescence and on the production of superoxi

Abnormalities of neutrophil phagocytosis, intracellular killing and metabolic activity in alcoholic cirrhosis and hepatitis.

Neutrophil functions of phagocytosis and intracellular killing of bacteria were examined in 40 patients with alcoholic cirrhosis of whom 18 had a superimposed acute alcoholic hepatitis. In 65% of these, defective neutrophil phagocytosis was demonstrable, and in 62.5% there was a defect of intracellular killing of either Staphylococcus aureus or Escherichia coli. Studies of the patients' serum failed to reveal inhibitors of neutrophil function. Additional assays of superoxide (O2-) and hydrogen peroxide production, hexose monophosphate shunt activity, degranulation and cellular levels of granul

Neutropenia and impaired neutrophil function in glycogenosis type Ib.

The impairment of different neutrophil functions has recently been reported in some patients with glycogenosis Ib and neutropenia. However, no satisfactory explanation for these findings has so far been supplied. In order to investigate this problem, we have studied neutrophil functions (random locomotion and chemotaxis, O-2 release, [1-14C]glucose oxidation and cellular cytotoxicity) in two further patients with glycogenosis Ib and neutropenia. The results show that neutrophil dysfunctions related to the involvement of both hexose monophosphate shunt and anaerobic glycolysis were variable. Th

Reprogramming a Hgt-family Kluyveromyces marxianus sugar transporter by site-directed mutagenesis to enable co-consumption of glucose and xylose.

Efficient utilization of lignocellulosic hydrolysates in yeast-based biorefineries requires simultaneous consumption of glucose and xylose, which is often limited by preferential glucose uptake. In Kluyveromyces marxianus, we kinetically characterized two native xylose transporters, KMAR_10 531 and KMAR_60 179, identifying medium- and low-affinity xylose transporters, respectively. KMAR_10 531 also mediated high-affinity glucose uptake (Km 0.28 ± 0.1 mM), limiting xylose utilization in mixed-sugar media. Guided by structural modelling, we engineered the KMAR_10 531 N325V variant, which reduced

Simultaneous optimization of lignocellulosic sugar catabolism via systematic laboratory evolution under complex selection pressure.

Efficient co-utilization of hexose and pentose sugars from lignocellulose is essential for microbial bioconversion, yet engineered catabolic pathways can be unstable or suboptimal in complex resource environments. Here, we use a Pseudomonas putida strain engineered to catabolize xylose and arabinose to examine how resource abundance, temporal availability, and subculturing shape evolutionary outcomes. Using an automated adaptive laboratory evolution (ALE) platform, we evolve the strain under simple single-substrate and complex multi-substrate selection pressures. These environments drive diver

Regulation of Ansamitocin P-3 Biosynthesis by the Response Regulator CrsB in Actinosynnema pretiosum.

Ansamitocin P-3 (AP-3) is an ansamycin antibiotic produced by Actinosynnema pretiosum with high clinical and commercial value. Its biosynthesis is tightly controlled, yet the underlying transcriptional network is not fully understood. Here, we identified the two-component system CrsAB and characterized the response regulator CrsB. Deletion of crsB resulted in a 57% reduction in AP-3 yield without impairing morphological development or biomass accumulation. RNA‑Seq showed that CrsB acts as a global regulator, influencing energy metabolism, cell integrity, and biosynthetic capacity. At the ferme

Elevated ozone reprograms secondary metabolism in Abelmoschus moschatus pods, enhancing antidiabetic and antiobesity bioactivities.

Critical insight: Elevated ozone (EO3) induced selective upregulation of TCA cycle, phenylpropanoid?flavonoid pathways and downregulation of lignans and specific lipids. This coordinated carbon?nitrogen reallocation enhances quercetin-based ROS scavenging and respiratory energy supply, strengthening ozone stress tolerance. This study examines metabolic changes in A. moschatus pods under elevated ozone (EO3) stress by using open-top chambers (OTC). The metabolomic analysis revealed the carbon and nitrogen distribution, with reduced glycolysis and TCA cycle activity, while the pentose phosphate

Metabolic engineering of Pichia pastoris for sustainable production of 1,8-cineole from methanol.

Methanol is an ideal feedstock for biomanufacturing and can be produced in large quantities from carbon dioxide via photocatalysis or electrolytic reduction, contributing to global carbon neutrality. In this study, we described an engineered Pichia pastoris strain capable of efficiently producing the monoterpene 1,8-cineole from methanol. First, a remarkable high-level synthesis of 1,8-cineole was achieved in Pichia pastoris by introducing an exogenous 1,8-cineole synthase, strengthening the MVA pathway, and suppressing the competing pathway. Subsequently, we significantly improved the yield o

High-fat diet primes vulnerability to polyethylene terephthalate nanoplastic toxicity via the gut microbiota-metabolism axis.

This study investigated whether a high-fat diet (HFD) acts as a priming factor that increases intestinal susceptibility to polyethylene terephthalate nanoplastics (PET-NPs, 100 nm, 25 mg/kg/d). After a 12-week combined exposure in mice, HFD priming potentiated the deleterious effects of PET-NPs on intestinal structure and function. Compared to HFD alone, the combined exposure (HFD + PET-NPs) exacerbated intestinal barrier injury, as evidenced by reduced tight junction proteins (Claudin-1, Occludin, and ZO-1), increased permeability, and disrupted histoarchitecture. These mice also exhibited en

Multiomics-guided discovery of protective microbiome signatures in lupus-prone mice treated with Faecalibacterium prausnitzii.

Gut microbiome dysbiosis has been implicated in the pathogenesis of systemic lupus erythematosus (SLE). However, microbiota-targeted therapeutic strategies have been lacking. Here, we report the potential of Faecalibacterium prausnitzii (strain UT1) to ameliorate gut dysbiosis and alleviate disease progression in the B6.Sle1.Yaa male mouse model of SLE. Fecal metagenomes of patients with SLE shifted carbohydrate catabolism from dietary fibers to host glycans, coinciding with depletion of F. prausnitzii. Oral administration of UT1 partially reversed lupus-associated microbiome alterations and r

Retinopathy-associated inosine monophosphate dehydrogenase 1 mutations cause metabolic and filament defects in cones.

Dominant variants in inosine monophosphate dehydrogenase 1 (IMPDH1), a key enzyme in the de novo synthesis of purine bases, cause progressive photoreceptor death, leading to blindness. To investigate the cause of degeneration, we generated the first mutant IMPDH1 animal models and expressed mutant forms of impdh1a in zebrafish cone photoreceptors. Unlike cones expressing exogenous normal impdh1a, cones containing impdh1a with the K238E mutation degenerated. Cones expressing impdh1a with the D226N mutation did not show significant cone loss by 2 years. Steady-state and flux metabolomics in zebr

The glucose metabolism reprogramming of yak Sertoli cells under hypoxia is regulated by autophagy.

Hypoxia often has negative effects on testis development and spermatogenesis of mammals. Plateau yaks have lived in the hypoxia environment for generations, but have ensured testicular function, which is closely related to their unique hypoxia response mechanism. Glucose metabolic reprogramming is an important way for cells to respond to stressful environments, especially the metabolite lactate, which is the energy basis for the development and differentiation of germ cells. In this study, hypoxia (5% O2) effectively promoted yak Sertoli cell proliferation and decreased autophagy and apoptosis

Ribitol and ribose treatments differentially affect metabolism of muscle tissue in FKRP mutant mice.

Dystroglycanopathy is characterized by reduced or lack of matriglycan, a cellular receptor for laminin as well as other extracellular matrix proteins. Recent studies have delineated the glycan chain structure of the matriglycan and the pathway with key components identified. FKRP functions as ribitol-5-phosphate transferase with CDP-ribitol as the substrate for the extension of the glycan chain. Supplement of ribitol and ribose have been reported to increase the levels of CDP-ribitol in both cells and in muscles in vivo. Clinical trials with both ribitol and ribose have been reported for treat
