Ultrastructural studies of the mechanism of changes developing in pemphigoid using peroxidase-labeled antibodies.

The application of peroxidase-labeled antiserum to human gamma globulin allowed the immunological reaction in vivo to be localized in the basement membrane, particularly in its part termed the lucid zone. The bulla is formed between the basal cell membrane and the dark lamina of the basement membrane.

The fine structural localization of peroxidase activity in goblet cells of the conjunctival epithelium of rats.

Cytochemical investigation of peroxidase was made on the conjunctival goblet cells, using the diaminobenzidine method. Many goblet cells in the conjunctiva demonstrated this reaction. The reaction was present in secretory granules as well as in the perinuclear cisterna, the rough endoplasmic reticulum, several Golgi saccules and adjacent small smooth-surfaced vesicles, and condensing vacuoles. Each secretory granule within a goblet cell showed a different degree of reaction, hence it is concluded that some goblet cells probably produce both mucous substance and peroxidase in one cell. The mode…

Glutathione Peroxidase-1 Knockout Facilitates Memory Impairment Induced by β-Amyloid (1-42) in Mice via Inhibition of PKC βII-Mediated ERK Signaling; Application with Glutathione Peroxidase-1 Gene-Encoded Adenovirus Vector.

A growing body evidence suggests that selenium (Se) deficiency is associated with an increased risk of developing Alzheimer's disease (AD). Se-dependent glutathione peroxidase-1 (GPx-1) of a major antioxidant enzyme, and the most abundant isoform of GPx in the brain. In the present study, we investigated whether GPx-1 is protective against memory impairments induced by beta-amyloid (Aβ) (1-42) in mice. As the alteration of protein kinase C (PKC)-mediated ERK activation was recognized in the early stage of AD, we examined whether the GPx-1 gene modulates Aβ (1-42)-induced changes in PKC and ERK…

Oximes, peroxidases, and oxidative nitric oxide production: a new framework for plant NO metabolism?

Nitric oxide (NO) is a small, gaseous reactive species that plays key roles in plant development and responses to environmental stimuli. In mammals, NO is produced via both nitrate/nitrite reduction and the oxidation of l-arginine by nitric oxide synthase (NOS). In plants, while the reductive pathway was identified early on, a NOS-like equivalent remained elusive for decades. In this opinion article, we describe recent discoveries that suggest a modular oxidative pathway, in which peroxidases may play a key role. Together with earlier observations, this model supports a broader conceptual fram…

Sustainable biodegradation of phenol using peroxidases extracted from local agricultural wastes.

The present study focused on the enhanced biodegradation of phenol as a toxic compound using peroxidases extracted from agricultural wastes (skin peels of luffa and jicama plants) mediated bioprocess under H2O2-infusion. The extracted crude luffa peroxidase (LP) and jicama peroxidase (JP) showed enzyme activities of 1.4 ± 0.2 U/mL and 1.6 ± 0.1 U/mL, respectively. LP achieved a phenol removal efficiency exceeding 95% at H₂O₂ concentrations between 6-8 mM and pH 7, whereas JP achieved 94.5% removal at pH 7, with minimal impact from variations in H₂O₂ concentrations. Maximum removal efficiencies…

Regulation of Glutathione Peroxidases (GPxs) in Breast Cancer: From Mechanisms to Targeted Therapeutics.

Breast cancers (BCs) are the most widespread cancer type among females and constitute the second leading cause of cancer fatality. Although there have been some advances in recent years, BC mortality rates remain high, a grim fact that underlines the need for novel therapeutic strategies. A number of recent studies have revealed that reactive oxygen species (ROS) are associated with the molecular mechanisms underlying BC manifestation and pathogenesis. It is important to note that glutathione peroxidases (GPxs) can regulate ROS. GPxs are an enzyme family involved in protecting the cells agains…

The effects of vitamin D and gene polymorphisms on susceptibility to thyroid peroxidase antibody positivity.

BACKGROUND: The etiology of Hashimoto's thyroiditis (HT) involves genetic and environmental factors. There is a lack of clarity regarding the relationship between Vitamin D and HT. This study aimed to investigate the effect of Vitamin D and gene polymorphisms on thyroid peroxidase antibody (TPOAb) positivity. METHODS: A total of 9,966 participants were included from a survey conducted in East China from 2014 to 2016. We measured the levels of 25(OH)D, thyroid hormones and autoimmune antibodies. rs11675434, rs9277555, and rs301799 were genotyped. Based on these 3 SNPs, a weighted genetic risk …

Cleavage of a Peroxide Bond via a Dual Attack by Functional Mimics of Glutathione Peroxidase.

Nonmetal-containing peroxidase enzymes, including glutathione peroxidase (GPx), and peroxiredoxins, control cellular redox levels by catalyzing the reduction of H2O2. The remarkably higher reactivity of GPx enzyme as compared to the fully dissociated synthetic selenolate/thiolate molecule is probably due to the dual-attack on the peroxide bond (HO1-O2H) by the enzyme; The first one is a nucleophilic attack of the selenolate/thiolate moiety to O1 atom and the second attack at the O2 atom of the peroxide bond by the acidic "parked proton" from Trp or His residue present at the enzyme's active si…

NADH and NADPH peroxidases as antioxidant defense mechanisms in intestinal sulfate-reducing bacteria.

Animal and human feces typically include intestinal sulfate-reducing bacteria (SRB). Hydrogen sulfide and acetate are the end products of their dissimilatory sulfate reduction and may create a synergistic effect. Here, we report NADH and NADPH peroxidase activities from intestinal SRB Desulfomicrobium orale and Desulfovibrio piger. We sought to compare enzymatic activities under the influence of various temperature and pH regimes, as well as to carry out kinetic analyses of enzymatic reaction rates, maximum amounts of the reaction product, reaction times, maximum rates of the enzyme reactions,…

Analysis of Bioavailability and Induction of Glutathione Peroxidase by Dietary Nanoelemental, Organic and Inorganic Selenium.

Dietary organic selenium (Se) is commonly utilized to increase formation of selenoproteins, including the major antioxidant protein, glutathione peroxidase (GPx). Inorganic Se salts, such as sodium selenite, are also incorporated into selenoproteins, and there is evidence that nanoelemental Se added to the diet may also be effective. We conducted two trials, the first investigated inorganic Se (selenite), organic Se (L-selenomethionine) and nanoelemental Se, in conventional mice. Their bioavailability and effectiveness to increase GPx activity were examined. The second trial focused on determi…

Thyroid peroxidase in human endometrium and placenta: a potential target for anti-TPO antibodies.

Autoimmune thyroid disease is the most common endocrine disorder during pregnancy. Thyroid autoantibodies (TAs) have been suggested to serve a role in implantation failure and spontaneous abortion. Until now, there are no data on the potential interaction of TAs with human reproductive organs. Here, we set out for the first time to test this hypothesis by studying the expression of thyroid peroxidase (TPO) at gene and protein level in human reproductive organs. Endometrial samples were taken from normal women, and placenta tissues were collected after full-term caesarian section. Expression of…

Involvement of sperm acetylated histones and the nuclear isoform of Glutathione peroxidase 4 in fertilization.

We previously demonstrated that the nuclear form of Glutathione peroxidase 4 (nGPx4) has a peculiar distribution in sperm head, being localized to nuclear matrix and acrosome and that sperm lacking nGPx4 are more prone to decondensation in vitro. In this study we have hypothesized that sperm retained acetylated histones and nGPx4 are implicated in paternal chromatin decondensation and male pronucleus formation at fertilization. Indeed, significant higher amounts of acetylated histone H4 and acetylated histone H3 were observed by both immunofluorescence and western blotting in nGPx4-KO sperm vs…

Enzymatic etching of gold nanorods by horseradish peroxidase and application to blood glucose detection.

Gold nanorods (AuNRs) have become some of the most used nanostructures for biosensing and imaging applications due to their plasmon-related … oses. To overcome this limitation we have introduced a biocatalytic species, the enzyme horseradish peroxidase (HRP) that is able to induce a gradual oxidation of AuNRs in the presence of trace concentrations of H2O2. Interestingly, the presence of halide ions has also been found to be essential for this process. As a consequence, other enzymatic reactions, …
