Animal cell mutants represent two complementation groups of peroxisome-defective Zellweger syndrome.

Generalized peroxisome-deficient disorders including cerebro-hepato-renal Zellweger syndrome, neonatal adrenoleukodystrophy, and infantile Refsum disease are autosomal recessive diseases, where catalase-containing particles (peroxisomes) are morphologically absent. We previously isolated two Chinese hamster ovary (CHO) cell mutants (Z24 and Z65) that resemble the fibroblasts from patients with such diseases, in their defective peroxisome assembly (Tsukamoto, T., S. Yokota, and Y. Fujiki. 1990. J. Cell Biol. 110:651-660). Here we report isolation by the P9OH/UV method of a peroxisome-deficient …

Arachidonic acid metabolism in fibroblasts from patients with peroxisomal diseases: response to interleukin 1.

Prostaglandin E2 synthesis and eicosanoid biosynthetic enzyme activities (arachidonyl CoA synthetase, cyclooxygenase and phospholipase A2) were measured in dermal fibroblasts from patients with metabolic disorders of peroxisomal origin and compared to those from normal subjects and patients with other metabolic disorders of lipid metabolism. Basal- as well as interleukin 1-stimulated prostaglandin E2 syntheses were higher in fibroblasts from patients with X-linked adrenoleukodystrophy, the Zellweger cerebrohepatorenal syndrome and rhizomelic chondrodysplasia punctata than in normals. Basal cyc

Peroxisomal L-pipecolic acid oxidation is deficient in liver from Zellweger syndrome patients.

L-Pipecolic acid, a cyclic imino acid produced during the degradation of lysine, accumulates in body fluids of infants with the generalized peroxisomal disorders, including Zellweger syndrome, neonatal adrenoleukodystrophy, and infantile Refsum disease. Peroxisome-enriched fractions from normal human liver oxidized L-[3H]pipecolic acid to alpha-[3H]aminoadipic acid (AAA). When human liver organelles were separated on a Percoll gradient, L-[3H]pipecolic acid oxidation activity (as measured by [3H]AAA formation) most closely segregated with the peroxisomal marker, catalase, and was not associate

Infantile Refsum disease: an inherited peroxisomal disorder. Comparison with Zellweger syndrome and neonatal adrenoleukodystrophy.

Three patients affected by infantile Refsum disease are described with mental retardation, minor facial dysmorphia, chorioretinopathy, sensorineural hearing deficit, hepatomegaly, failure to thrive and hypocholesterolaemia. Initially, only an accumulation of phytanic acid was thought to be present. More recent findings showed a biochemical profile very similar to that found in classical Zellweger syndrome or neonatal adrenoleukodystrophy. Morphologically typical peroxisomes were absent in the liver. All three disorders are associated with multiple peroxisomal dysfunction. Because of these simi…

Lignoceric acid is oxidized in the peroxisome: implications for the Zellweger cerebro-hepato-renal syndrome and adrenoleukodystrophy.

The deficient oxidation and accumulation of very-long-chain fatty acids in the Zellweger cerebro-hepato-renal syndrome (CHRS) and X chromosome-linked adrenoleukodystrophy (ALD), coupled with the observation that peroxisomes are lacking in CHRS, prompted us to investigate the subcellular localization of the catabolism of lignoceric acid (C24:0). Peroxisomal and mitochondrial-rich fractions were separated from rat liver crude mitochondria by sucrose density gradient centrifugation. Enzyme activity for the oxidation of [1-14C]palmitic acid to water-soluble acetate was 2- to 3-fold higher in the m…

Hilpda gene expression in steatotic liver is regulated by peroxisome proliferator-activated receptor γ.

Hypoxia-inducible lipid droplet-associated protein (HILPDA) promotes hepatic lipid accumulation. Hilpda gene expression is directly regulated by hypoxia-inducible factors (HIF1A and HIF1B) and the nuclear receptor peroxisome proliferator-activated receptor alpha (PPARα). However, whether the expression of Hilpda is regulated by peroxisome proliferator-activated receptor gamma (PPARγ) in fatty liver remains unclear. In this study, we investigated the role of PPARγ in regulating Hilpda expression in fatty liver. The analysis of publicly available microarray datasets revealed that Hilpda expressi

Blocking MOXD1-derived ACOX1 peroxisome trafficking suppresses metabolic dysfunction-associated steatohepatitis.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) has become a major global health burden yet effective pharmacological tr … MOXD1 directly interacted with the ACOX1-PEX5 translocation complex, promoting ACOX1 trafficking to peroxisomes to block lipolysis, lipophagy. We further identified four key MOXD1 residues required for ACOX1 binding and resultant pro-MASH capacity. Importantly, based on the AI model and interacting details of MOXD1-ACOX1, we identified a small molecule rM15 t …

Targeted knockout of a host peroxisomal peptidase confers field resistance to maize lethal necrosis.

Maize lethal necrosis (MLN) is a severe disease caused by the combined infection of maize chlorotic mottle virus (MCMV) and a potyvirus, mos … cause of susceptibility. Confocal microscopy confirmed the localization of the MLNS1 protein within peroxisomes. Targeted knockout of the Mlns1 gene in the susceptible elite line CML536 from SSA conferred resistance comparable to KS23-6 in field trials conducted in Naivasha, Kenya. This knockout specifically blocked MCMV accumulation without affecting SCMV. T …

Peroxisomal catalase and plasmalogen biosynthesis protect from oxidative stress in Barth syndrome cardiomyopathy.

Barth Syndrome (BTHS) is an inherited mitochondrial cardiomyopathy caused by variants in the gene encoding TAFAZZIN (Taz), a transacylase ca … tochondrial FAO and to support GSH production. Here, we interrogated whether adaptive mechanisms in peroxisomes, which are closely associated with mitochondria and harbor antioxidative enzymes, can also compensate for the mitochondrial defects. We identified alterations in the peroxisomal biogenesis factors PEX14 and PEX19, indicating changes in the peroxisom …

Pharmacological Targeting of Peroxisome
Proliferator-activated Receptors for Prevention
of Radiation-induced Cognitive Decline.

BACKGROUND: Radiation-induced cognitive decline (RICD) is a common and debilitating late effect of cranial radiotherapy, particularly in long-term survivors of brain tumours and paediatric cancers. Despite advances in conformal radiation delivery and supportive care, progressive impairments in memory, attention and executive function continue to limit quality of life. Increasing evidence indicates that RICD is driven not by acute neuronal loss but by persistent neuroinflammation, oxidative stress, vascular dysfunction and metabolic failure within cognitive circuits. These converging processes

Emerging role of peroxisome proliferator-activated receptor agonists in the treatment of cholestatic liver disease.

PURPOSE OF REVIEW: Primary biliary cholangitis (PBC) and primary sclerosing cholangitis (PSC) are rare, chronic cholestatic diseases associated with significant morbidity. While previously approved therapies for PBC, including ursodeoxycholic acid (UDCA) and obeticholic acid (OCA) have substantially altered the natural course of the disease and improved patient survival, they have several limitations including an incomplete therapeutic response, patient intolerance and a lack of symptom relief. RECENT FINDINGS: Peroxisome proliferator-activated receptor (PPAR) agonists have emerged as promisi

Peroxisomal Sulfite Oxidase (SOX), an alternative source of NO in higher plants which is upregulated by H2S.

Nitric oxide (•NO) is a free radical that is endogenously produced in plant cells, though its enzymatic synthesis remains a subject of ongoing debate. Plant peroxisomes, subcellular compartments with active nitro-oxidative metabolism, play a role in various metabolic pathways. Sulfite oxidase (SOX), a peroxisomal enzyme requiring the molybdenum cofactor (MoCo), catalyzes the oxidation of sulfite (SO32-) to sulfate (SO42-), along with the concomitant production of H2O2. Using reconstituted recombinant SOX from pepper (Capsicum annuum L.) fruit, it was shown that this enzyme has the capacity to …

Endogenous NO fluctuations in Arabidopsis leaves influence peroxisomal activities and ROS, NADPH, and H2S metabolism.

Nitric oxide (NO) is a gasotransmitter that exerts signaling functions in plants. Using 30-day-old Arabidopsis thaliana plants wild type and transgenic lines with different NO content (Atnoa1 and Atnox1/cue1), the biochemical analysis of key components in the metabolism of ROS, NADPH, NO, and H2S in leaves indicates that the imbalance of endogenous cellular NO triggered differential changes in many of the analyzed biochemical parameters including the protein profile of S-glutathionylation, S-nitrosation, tyrosine and tryptophan nitration. It was remarkable the differences observed in the antio

Drosophila Models Uncover Substrate Channeling Effects on Phospholipids and Sphingolipids in Peroxisomal Biogenesis Disorders.

Peroxisomal Biogenesis Disorders Zellweger Spectrum (PBD-ZSD) disorders are a group of autosomal recessive defects in peroxisome formation that produce a multi-systemic disease presenting at birth or in childhood. Well documented clinical biomarkers such as elevated very long chain fatty acids (VLCFA) are key biochemical diagnostic findings in these conditions. Additional, secondary biochemical alterations such as elevated very long chain lysophosphatidylcholines are allowing newborn screening for peroxisomal disease. In addition, a more widespread impact on metabolism and lipids is increasing
