The neuroendocrine control of gonadotrophin release in the Japanese quail. III. The role of the tuberal and anterior hypothalamus in the control of ovarian development and ovulation.

Reproduction in the Japanese quail (Coturnix coturnix japonica), as in many other species, is controlled by the length of the daily photoperiod. The present experiments have investigated the neuroendocrine regulation of both the photoperiodically induced 'tonic' gonadotrophin secretion, which causes the development and maintenance of the ovary, and the 'cyclic' surge release of luteinizing hormone (LH), which stimulates ovulation. Areas within the anterior and tuberal hypothalamus were destroyed by means of electrolytic lesions while a small knife was used to isolate, to various degrees, the t…

Sympathetic activation links light exposure and rest-activity rhythm to liver injury and bile acid dysregulation.

Prolonged light exposure and rest-activity alteration are increasingly prevalent in modern society, yet their health consequence and the underlying mechanism remain poorly characterized. The liver is an organ central to metabolic and circadian regulation. We hypothesized that extended wakefulness, which was modeled as long photoperiod in humans and short photoperiod in nocturnal mice, would affect liver integrity and function. Actigraphic watch, wheel running, morphological, biochemical, transcriptomic, and chromatographic techniques were employed to examine the impact of light exposure altera…

Gestational exposure to continuous light impairs the development of the female reproductive system in adult Wistar rat offspring.

INTRODUCTION: It has been suggested that maternal exposure to constant light during the gestational period could be considered as a chronic stressor, impairing offspring development by interfering in neuroendocrine and behavior responses. OBJECTIVE: This study aimed to evaluate whether maternal exposure to continuous light during pregnancy affects the adult reproductive system in the female offspring. MATERIALS AND METHODS: Pregnant Wistar rats were allocated into light-dark (LD) group, exposed to light and dark photoperiod during gestation, and the light-light (LL) group, exposed to a photo…

Urinary 6-sulphatoxymelatonin excretion reflects pineal melatonin secretion in the Djungarian hamster (Phodopus sungorus).

To monitor pineal function in the Djungarian hamster (Phodopus sungorus), we measured the urinary excretion of the melatonin metabolite 6-sulphatoxymelatonin (aMT6s) at 3-hr intervals by radioimmunoassay. Hamsters maintained in either long photoperiod (LP, LD 16:8) or short photoperiod (SP, LD 8:16) showed marked daily rhythms in aMT6s excretion, with elevated levels during the dark phase. In both photoperiods, we found large interindividual differences, mainly in the amplitude of the signal. However, the amplitude as well as the duration of nocturnal aMT6s excretion was higher in SP than in L…

Plasma prolactin and luteinizing hormone during termination and onset of photorefractoriness in intact and pinealectomized European starlings (Sturnus vulgaris).

Plasma levels of prolactin and luteinizing hormone have been examined in intact and pinealectomized European starling (Sturnus vulgaris) during termination and onset of photorefractoriness. Photorefractory starlings exposed to long (18L:6D) daily photoperiods for 10 weeks were pinealectomized. One week after pinealectomy these birds, along with the intact and sham-operated birds, were transferred to short (8L:16D) daily photoperiods. After 10 weeks on short day lengths the daily photoperiod was switched to long (18L:6D) and the birds remained on this regime for a further 10 weeks. Plasma prola…

Signaling pathways to and from the hypophysial pars tuberalis, an important center for the control of seasonal rhythms.

Seasonal (circannual) rhythms play an important role for the control of body functions (reproduction, metabolism, immune responses) in nearly all living organisms. Also humans are affected by the seasons with regard to immune responses and mental functions, the seasonal affective disorder being one of the most prominent examples. The hypophysial pars tuberalis (PT), an important interface between the hypophysial pars distalis and neuroendocrine centers in the brain, plays an essential role in the regulation of seasonal functions and may even be the seat of the circannual clock. Photoperiodic s…

Vitamin D and serotonin in winter.

Calcitriol accumulates in the nuclei of adrenal medullary cells, stimulating the tyrosine hydroxylase gene expression. The transcription of the CREM gene is induced by adrenergic input to the pineal gland at night. Depending on the photoperiod of the prior night length, the CREM gene is either subsensitive or supersensitive to induction. Guided by the memory of past photoperiods, the induced changes in melatonin synthesis may mediate either the inhibitory or stimulatory effects on bodily functions. Calcitriol might inhibit the binding of melatonin to the nuclear retinoid Z receptors, which wou…

Effect of ambient temperature on serotonin, norepinephrine, and epinephrine contents in the pineal-paraphyseal complex of the soft-shelled turtle (Lissemys punctata punctata).

The aim of the present investigation was to ascertain the influence of ambient temperature on serotonin, norepinephrine, and epinephrine contents in the pineal-paraphyseal complex of soft-shelled turtles. Turtles were exposed to low (10 degrees) and high (32 degrees) temperatures for 7 days in an incubator with the same photoperiod (12L:12D). Control animals were also kept in the same incubator at 25 degrees for similar duration with the same photoperiod. The findings revealed that ambient temperatures significantly altered serotonin, norepinephrine, and epinephrine content in the pineal-parap…

[Melatonin, synthetic analogs, and the sleep/wake rhythm].

INTRODUCTION: Melatonin, a widespread hormone in the animal kingdom, is produced by several organs and tissues besides the pineal gland. Whilst extrapineal melatonin behaves as a cytoprotective molecule, the pineal produces the hormone in a rhythmic manner. The discovery of melatonin in 1958, and the characterization of its synthesis somewhat later, let to the description of its photoperiodic regulation and its relationship with the biological rhythms such as the sleep/wake rhythm. DEVELOPMENT: The suprachiasmatic nuclei are the anatomical seat of the biological clock, represented by the cloc…

Influence of continuous light and darkness on the secretory pinealocytes of Heteropneustes fossilis.

In an earlier study on Heteropneustes fossilis, evidence of secretory activity in the pinealocytes had been demonstrated at the electron microscopic (EM) level and it was found to exist in two phases: a secretory phase (light cells) and a storage phase (dark cells). In the present investigation, H. fossilis was subjected to artificial photoperiods of continuous illumination and continuous darkness for a period of ten days and the effect on the secretory pinealocytes was studied at the EM level. Marked results were observed within the short period of ten days emphasizing the role of environment…

Colic, sleep inertia, melatonin and circannual rhythms.

Colic is periodic behavior occurring at the end of the day during the first 3 months of life characterized by crying. It is hypothesized tha … fficulty reconciling cues for the timing of evening sleep due to discrepancies between the expected photoperiod derived from the prenatal maternal pineal melatonin circadian rhythm and the postnatally experienced photoperiod. Data is presented showing that the incidence of colic increases with increasing latitude in support of the hypothesis that infant cryin …

Temporal Expression of c-fos and Genes Coding for Neuropeptides and Enzymes of Amino Acid and Amine Neurotransmitter Biosynthesis in Retina, Pineal and Hypothalamus of a Migratory Songbird: Evidence for Circadian Rhythm-Dependent Seasonal Responses.

This study investigated whether, in photoperiodic songbirds, the circadian pacemaker system (CPS) connects to the seasonal photoperiodic responses, by changes at transcriptional level in the level and 24-h rhythm of its constituent neurotransmitters. We used black-headed buntings (Emberiza melanocephala), which exhibit distinct seasonal states in captivity under appropriate photoperiods and hence served as a useful model system. Under short days, buntings remain in the photosensitive state (Pse) (winter phenotype: non-migratory, non-breeding). Under long days, however, buntings undergo through…

Photomanipulation of sexual maturation and breeding cycle of the steppe polecat (Mustela eversmanni) and other techniques for more rapid propagation of the species.

Twenty steppe polecats were divided into 2 groups, each consisting of 4 males and 6 females, and subjected to either a natural photoperiod (controls) or alternating periods of short (8 h light/16 hr dark for 8-9 weeks) and long days (16 h light/8 h dark for 16-20 weeks). The experimental photoperiod significantly accelerated sexual maturation in both sexes, with males developing maximal testis size within 57 days and females breeding after an average of 52 days exposure to 16L/8D. Males in the experimental group completed 2 1/2 testicular cycles and participated in mating during 3 successive b…

Expression and regulation of Icer mRNA in the Syrian hamster pineal gland.

Inducible-cAMP early repressor (ICER) is a potent inhibitor of CRE (cAMP-related element)-driven gene transcription. In the rat pineal gland, it has been proposed to be part of the mechanisms involved in the shutting down of the transcription of the gene coding for arylalkylamine N-acetyltransferase (AA-NAT, the melatonin rhythm-generating enzyme). In this study, we report that ICER is expressed in the pineal gland of the photoperiodic rodent Syrian hamster although with some difference compared to the rat. In the Syrian hamster pineal, Icer mRNA levels, low at daytime, displayed a 20-fold inc…

The impact of thyroid hormone in seasonal breeding has a restricted transcriptional signature.

Thyroid hormone (TH) directs seasonal breeding through reciprocal regulation of TH deiodinase (Dio2/Dio3) gene expression in tanycytes in the ependymal zone of the medio-basal hypothalamus (MBH). Thyrotropin secretion by the pars tuberalis (PT) is a major photoperiod-dependent upstream regulator of Dio2/Dio3 gene expression. Long days enhance thyrotropin production, which increases Dio2 expression and suppresses Dio3 expression, thereby heightening TH signaling in the MBH. Short days appear to exert the converse effect. Here, we combined endocrine profiling and transcriptomics to understand ho…

Generation of melatonin rhythms.

In mammals, information about the environmental photoperiod is relayed from the retina to the suprachiasmatic nuclei (SCN) in the anterior hypothalamus and via the sympathetic nervous system to the pineal gland where it influences the secretion of melatonin. Light plays a dual role: to suppress the release of melatonin and to entrain the circadian rhythm generators in the SCN, which govern the endogenous melatonin rhythm. Under normal daily light-dark cycles melatonin secretion is confined to the dark period. In most photoperiodic species the daily pattern of secretion changes in response to c…

Transcriptome profiling in fast versus slow-growing rainbow trout across seasonal gradients.

BACKGROUND: Circannual rhythms in vertebrates can influence a wide variety of physiological processes. Some notable examples include annual reproductive cycles and for poikilotherms, seasonal changes modulating growth. Increasing water temperature elevates growth rates in fishes, but increases in photoperiod regime can have similar influences even at constant temperature. Therefore, in order to understand the dynamics of growth in fish it is important to consider the background influence of photoperiod regime on gene expression differences. This study examined the influence of a declining phot…

The effects of early environmental conditions on the reproductive and somatic development of juvenile guinea pigs (Cavia aperea f. porcellus).

Little is known about the effects of the early environment on the development of non-seasonally reproducing species like the domestic guinea pig (Cavia aperea f. porcellus). Although guinea pigs reproduce throughout the year, there is evidence for environmental sensitivity of their reproductive physiology. To investigate the sensitivity of juvenile body weight and puberty to differences in the prenatal and early postnatal environment, subjects were exposed to either of two experimental conditions mimicking seasonal variation: a long photoperiod with 25 degrees C ambient temperature ("LD/25 deg…

Melatonin secretion from goldfish pineal gland in organ culture.

Pineal glands were removed from goldfish reared under 12L-12D at 25 degrees for 2 weeks. These were incubated for 6 days under (1) normal 12L-12D (lights on 0600-1800 hr), (2) reversed 12L-12D (lights on 1800-0600 hr), (3) continuous dark, or (4) continuous light condition at 25 degrees. The incubation medium was changed at 12-hr intervals (0600-1800 and 1800-0600 hr) and secreted melatonin (MLT) was measured by RIA. Under 12L-12D or reversed 12L-12D, MLT secretion was active in the dark phase and was suppressed in the light phase of a given photoperiod. Under a continuous dark condition, a la…
