AAV mediated GDNF secretion from retinal glia slows down retinal degeneration in a rat model of retinitis pigmentosa.

Mutations in over 80 identified genes can induce apoptosis in photoreceptors, resulting in blindness with a prevalence of 1 in 3,000 individuals. This broad genetic heterogeneity of disease impacting a wide range of photoreceptor functions renders the design of gene-specific therapies for photoreceptor degeneration impractical and necessitates the development of mutation-independent treatments to slow photoreceptor cell death. One promising strategy for photoreceptor neuroprotection is neurotrophin secretion from Müller cells, the primary retinal glia. Müller glia are excellent targets for sec

Active DNA demethylation is upstream of rod-photoreceptor fate determination and required for retinal development.

Retinal cell fate specification from multipotent retinal progenitors is governed by dynamic changes in chromatin structure and gene expression. Methylation at cytosines in DNA (5mC) is actively regulated for proper control of gene expression and chromatin architecture. Numerous genes display active DNA demethylation across retinal development; a process that requires oxidation of 5mC to 5-hydroxymethylcytosine (5hmC) and is controlled by the ten-eleven translocation methylcytosine dioxygenase (TET) enzymes. Using an allelic series of conditional TET enzyme mutants, we determine that DNA demeth

Olfactory ensheathing cell-derived extracellular vesicles protect the degenerated retinal pigment epithelium by delivering phagocytosis-related proteins.

Aim: Olfactory ensheathing cells (OECs) are commonly utilized in cell-based therapies for neurodegenerative diseases and central nervous system injuries, due to their unique properties of suppressing gliosis and providing neuroprotection. Small extracellular vesicles (sEVs) have emerged as promising therapeutic agents. This study aimed to investigate OEC-sEVs' effect on retinal degeneration. Methods: Proteomics analysis identified differentially expressed proteins between OEC-sEVs and OECs. We conducted subretinal injection of OEC-sEVs and assessed their protective mechanisms in degenerative r

Opsin synthesis in the C57BL/6-mivit/mivit mouse model of retinal degeneration.

The capacity of photoreceptor cells to synthesize opsin was evaluated in a newly-described mouse model of retinal degeneration, the C57BL/6-mivit/mivit. The mivit/mivit mouse loses photoreceptor cells at a rate of about one row per month beginning at 8 weeks, ROS are severely disrupted at 4 months, RPE is unevenly pigmented. Retinas of affected and control mice ages 4, 6, 8, 12, 16, 20, 24, 28, 32 and 52 weeks were incubated for 2 hours in medium containing [3H] leucine. Homogenates of retina samples were subjected to SDS-PAGE using disc gels. The gels were sliced and counted by scintillation.…

Hyperpolarization-activated cyclic nucleotide-gated channels preserve excitability in ON-bipolar cells after degeneration.

Retinal degeneration leads to extensive remodeling of inner retinal circuits following photoreceptor loss, but the intrinsic membrane properties of surviving bipolar cells remain incompletely understood. Here, we investigated the expression and functional contribution of hyperpolarization-activated cyclic nucleotide-gated (HCN) channels in ON-bipolar cells using the rd10 mouse model of retinitis pigmentosa. High-resolution fluorescence in situ hybridization showed no significant differences in Hcn1, Hcn2, and Hcn4 transcript levels between wild-type (WT) and rd10 bipolar cells. Whole-cell patc

The effects of laser photocoagulation in the retinal capillaries.

Using ruby and argon lasers, we produced multiple lesions on normal rhesus monkey retinas. The clinically used energy levels of both lasers … cially affect laser treatment of diabetic retinopathy. The retinal tissue appears to have excessive photoreceptor cells and the horizontal connection of other cells seems to compensate for visual function after loss of many photoreceptor cells. We demonstrated severe damage in the ganglion cells by argon laser when it was aimed at the thick blood films in the vitreou …

Bright environmental light accelerates rhodopsin depletion in retinoid-deprived rats.

PURPOSE: Dietary deficiency in the retinoid precursors of the visual pigment chromophore 11-cis retinal eventually results in selective degeneration of the photoreceptor cells of the vertebrate retina. An early effect of retinoid deficiency is depletion of chromophore from the photoreceptor outer segments. Experiments were conducted to determine whether the rate of chromophore depletion was affected by the intensity of environmental light. METHODS: Rats were maintained on diets either containing or lacking retinoid precursors of 11-cis retinal for up to 30 weeks. Animals in both dietary group…

Tracing the progression of retinitis pigmentosa via photoreceptor interactions.

Retinitis pigmentosa (RP) is a group of inherited degenerative eye diseases characterized by mutations in the genetic structure of the photoreceptors that leads to the premature death of both rod and cone photoreceptors. Defects in particular genes encoding proteins that are involved in either the photoreceptor structure, phototransduction cascades, or visual cycle are expressed in the rods but ultimately affect both types of cells. RP is "typically" manifested by a steady death of rods followed by a period of stability in which cones survive initially and then inevitably die too. In some RP c

Decreased photoreceptor count in human eyes with secondary angle-closure glaucoma.

Glaucoma has been known to be associated with a loss of retinal ganglion cells and their axons throughout the fundus and a decreased count of photoreceptors and retinal pigment epithelial (RPE) cells in the parapapillary region. This study investigated whether glaucomatous changes of the deep retinal layer occur outside the parapapillary region. The nuclei of the retinal photoreceptors and RPE cells were counted in histologic slides of 23 eyes with painful secondary angle-closure glaucoma resulting from perforating corneal injuries. Fourteen eyes with malignant choroidal melanoma not involving

Structure and function of the retinal pigment epithelium.

The evolution of light-perception organs started with the co-development of a double-cell structure consisting of a light-sensitive cell and a pigmented cell. In the human eye, the retinal pigment epithelium (RPE) forms the outer blood/retina barriers and has evolved into a close partnership with photoreceptors indispensable for visual function. In this interaction the RPE performs numerous functions to support the structural integrity of the photoreceptors and light sensitivity in the visual process. These functions include the absorption of light energy focused by a lens onto the retina, the

In retinitis pigmentosa TrkC.T1-dependent vectorial Erk activity upregulates glial TNF-α, causing selective neuronal death.

In some diseases the TrkC.T1 isoform is upregulated in glia, associated with glial TNF-α production and neuronal death. What remains unknown are the activating signals in glia, and how paracrine signals may be selective for a targeted neuron while sparing other proximate neurons. We studied these questions in the retina, where Müller glia contacts photoreceptors on one side and retinal ganglion cells on the other. In a mutant Rhodopsin mouse model of retinitis pigmentosa (RP) causing progressive photoreceptor death-but sparing retinal ganglion cells-TrkC.T1 and NT-3 ligand are upregulated in M

Genome Editing to Study Ca2+ Homeostasis in Zebrafish Cone Photoreceptors.

Photoreceptors are specialized sensory neurons with unique biological features. Phototransduction is well understood due in part to the exclusive expression and function of the molecular components of this cascade. Many other processes are less well understood, but also extremely important for understanding photoreceptor function and for treating disease. One example is the role of Ca2+ in the cell body and overall compartmentalization and regulation of Ca2+ within the cell. The recent development of CRISPR/Cas9 genome editing techniques has made it possible to rapidly and cheaply alter specif

Cell death in retinitis pigmentosa: gap junctions and the 'bystander' effect.

Retinitis pigmentosa (RP) constitutes a group of genetically mediated, degenerative retinal diseases that display a broad range of phenotypes. There is appreciable heterogeneity in the pathogenetic mechanisms that underlie the various forms of RP, but a substantial percentage of the known cases arise as a consequence of mutations in rhodopsin or other rod-specific proteins. However, despite the fact that the genetic defect is expressed solely in the rod photoreceptors, otherwise healthy cone photoreceptors invariably die, resulting in severe visual impairment. In this paper, the author propose

Nomen omen: Euglena gracilis possesses a rhodopsin-based photoreceptor.

The unicellular microalga Euglena gracilis has always been considered the ideal alga to investigate photoreceptive responses and systems, and it has been the subject of hundreds of articles. Moreover, because of its detectable photoreceptor, it has been given a key role in the evolution of photoreception, from single and simple cells to complex visual system of higher organisms. In this article, we report the Raman spectra recorded in vivo on photoreceptors of E. gracilis and Bos taurus retina. The almost perfect superimposability (correlation coefficient r = 0.955) of these spectra states tha

Prph2 disease mutations lead to structural and functional defects in the RPE.

Prph2 is a photoreceptor-specific tetraspanin with an essential role in the structure and function of photoreceptor outer segments. PRPH2 mutations cause a multitude of retinal diseases characterized by the degeneration of photoreceptors as well as defects in neighboring tissues such as the RPE. While extensive research has analyzed photoreceptors, less attention has been paid to these secondary defects. Here, we use different Prph2 disease models to evaluate the damage of the RPE arising from photoreceptor defects. In Prph2 disease models, the RPE exhibits structural abnormalities and cell lo

Insights into retinal remodeling in retinal degenerative disease.

The retina is a highly organized sensory structure responsible for capturing and processing visual information. Visual computation begins at the first synapse between photoreceptors, bipolar cells, and horizontal cells, before involving amacrine and ganglion cells to generate vision. Retinal degeneration disrupts the precise neural architecture required for vision, initiating a maladaptive process known as retinal remodeling. Photoreceptor degeneration in diseases, like retinitis pigmentosa (RP) and age-related macular degeneration, induces retinal remodeling, but good evidence shows glaucoma

Luteolin delays photoreceptor degeneration in a mouse model of retinitis pigmentosa.

Luteolin is neuroprotective for retinal ganglion cells and retinal pigment epithelial cells after oxidative injury, whereby it can inhibit microglial neurotoxicity. Therefore, luteolin holds the potential to be useful for treatment of retinal diseases. The purpose of this study was to investigate whether luteolin exhibits neuroprotective effects on rod cells in rd10 mice, a slow photoreceptor-degenerative model of retinitis pigmentosa. Luteolin (100 mg/kg) intraperitoneally injected daily from postnatal day 14 (P14) to P25 significantly enhanced the visual performance and retinal light respons

The relative distribution of retinal and choroidal blood in the human retina.

In this paper a model is presented to explain the relative distribution of retinal and choroidal blood to the human retina. According to this model the retina between the inner limiting membrane and the outer limiting membrane is metabolically controlled and nourished by the retinal vasculature, through Muller cells. The retina between the outer limiting membrane and Bruch's membrane is nourished by the choroidal vasculature, through the retinal pigment epithelium. Muller cells have the ability of metabolic uptake from the interphotoreceptor space as demonstrated in pathological situations suc
