Techniques to Study Common Root Responses to Beneficial Microbes and Iron Deficiency.

Iron (Fe) plays a central role in the vital processes of a plant. The Fe status of a plant influences growth and immunity, but it also dictates interactions of roots with soil microbiota through the production of Fe mobilizing, antimicrobial fluorescent phenolic compounds called coumarins. To adapt to low Fe availability in the soil, plants deploy an efficient Fe deficiency response. Interestingly, this Fe deficiency response is hijacked by root-colonizing microbes in the root microbiome to establish a mutually beneficial relationship. In this chapter, we describe how we cultivate plants and m

Plant responses to UV radiation and links to pathogen resistance.

Increased incident ultraviolet (UV) radiation due to ozone depletion has heightened interest in plant responses to UV because solar UV wavelengths can reduce plant genome stability, growth, and productivity. These detrimental effects result from damage to cell components including nucleic acids, proteins, and membrane lipids. As obligate phototrophs, plants must counter the onslaught of cellular damage due to prolonged exposure to sunlight. They do so by attenuating the UV dose received through accumulation of UV-absorbing secondary metabolites, neutralizing reactive oxygen species produced by

Cotton Recruits Soil-Derived Delftia tsuruhatensis to Suppress Aphid Detoxification Via Salicylic Acid-Mediated Defense.

The synergistic defense between microorganisms and plants holds significant ecological importance in resisting pest attacks. Herbivorous insects pose a significant threat to global agriculture. While plants deploy defense hormones like jasmonic acid (JA) and salicylic acid (SA), the role of beneficial microbes in enhancing these defenses remains underexplored. This study unveils a novel tripartite interaction in which cotton actively mobilizes the soil-derived bacterium Delftia tsuruhatensis to roots and leaves upon aphid attack, reshaping its microbiome for defense. Following aphid infestatio

Cell cycle reprogramming in plant symbiotic and pathogenic interactions.

Intracellular plant-microbe interactions rely on host-derived interface membranes: as sites for reciprocal nutrient and signal exchange duri … thogens can reprogram host cell cycle pathways to establish metabolically favorable niches, whereas plant immunity can engage cell cycle checkpoints to restrict resource allocation and reinforce physical barriers. In arbuscular mycorrhizal symbiosis, localized endoreduplication in host cells could function as a "metabolic amplification program" to boost biosynth …

Assessing the effects of a novel biostimulant to enhance leafminer resistance and plant growth on common bean.

The leafminer Liriomyza trifolii is one of the major insects that affect Phaseolus vulgaris production worldwide. Novel and safe biobased stimulator compound (BSTC) with micronutrient-amino acid chelated compounds was developed from natural compounds and was used for foliar spray of P. vulgaris. Treated plants showed significantly increased in quality and productivity as well as significant reduction in leafminer infestation by close the tunnel end resulting in larvae suffocation and death. BSTC contains chemical composition that has important function in inducing immunity and resistance again

Target of rapamycin signaling orchestrates growth-defense trade-offs in plants.

Plant defense to microbial pathogens is often accompanied by significant growth inhibition. How plants merge immune system function with normal growth and development is still poorly understood. Here, we investigated the role of target of rapamycin (TOR), an evolutionary conserved serine/threonine kinase, in the plant defense response. We used rice as a model system and applied a combination of chemical, genetic, genomic and cell-based analyses. We demonstrate that ectopic expression of TOR and Raptor (regulatory-associated protein of mTOR), a protein previously demonstrated to interact with T

Pathogen-mediated priming induces intergenerational immunity against spot blotch in wheat.

Spot blotch, caused by Bipolaris sorokiniana, is one of the most destructive diseases of wheat (Triticum aestivum), particularly in warm and humid regions where yield losses may exceed 30-50%. Heavy dependence on fungicides is neither sustainable nor environmentally safe, creating an urgent need for innovative strategies to enhance crop resilience. Defense priming, a phenomenon in which prior exposure to a mild stimulus enables plants to mount faster and stronger defenses upon subsequent stress offers a promising alternative. Pathogens or pathogen-derived molecules such as PAMPs and effectors

Versatile effectors of phytopathogenic fungi target host immunity.

Phytopathogenic fungi secrete a large arsenal of effector molecules, including proteinaceous effectors, small RNAs, phytohormones and derivatives thereof. The pathogenicity of fungal pathogens is primarily determined by these effectors that are secreted into host cells to undermine innate immunity, as well as to facilitate the acquisition of nutrients for their in planta growth and proliferation. After conventional and non-conventional secretion, fungal effectors are translocated into different subcellular compartments of the host cells to interfere with various biological processes. Lipid droplets confer SA-dependent antiviral defense without growth penalty.

The role of lipid droplets (LDs), typically regarded as storage sites for neutral lipid, in plant defense remains poorly understood. In our study with barley stripe mosaic virus (BSMV), a (+)ssRNA virus, we found that infection significantly increases LD abundance in Nicotiana benthamiana and wheat. Using split-TurboID-based proximity labeling, we identified LD-associated proteins (LDAPs) at ER-chloroplast contact sites. Increasing LD formation by overexpressing mouse FIT2 or plant NbLDAP2.1 decreased viral accumulation, whereas impairing LD formation led to higher infection. Live cell imaging

The CBP60b-14-3-3 functional module regulates immunity in arabidopsis.

Transcriptional reprogramming is a critical process in plant immune responses, in which Calmodulin-Binding Protein 60b (CBP60b), a plant-specific atypical transcription factor family, plays an essential role. However, how its transcriptional activity is regulated during immunity remains unclear. Here, we report that 14-3-3 proteins, conserved scaffolding proteins in eukaryotes, interact with CBP60b in a calcium-dependent manner and enhance its transcriptional activity on downstream genes. The 14-3-3 variant deficient in calcium binding, 14-3-3λG216A, disrupted the interaction with CBP60b and f

TurboID-Based Proximity Labeling for Exploiting Effector Interacting Proteins in the Fungal Maize Pathogen Ustilago maydis.

TurboID-based proximity labeling (PL) is an advanced method for identifying proteins in the immediate molecular environment of a protein of interest under physiological conditions. TurboID enables the rapid and covalent biotinylation of proteins in proximity to the bait proteins in living cells, capturing both stable and transient interactions with high temporal resolution. Most plant pathogens secrete effectors that modulate host immunity and facilitate infection. The effector-targeted host proteins are commonly involved in plant resistance or susceptibility. Elucidating these proteins is mea

[Chromoblastomycosis and phaeohyphomycosis, overlooked fungal diseases].

Dark-pigmented microscopic fungi are worldwide-spread soil saprophytes often found on plant remnants. In chromoblastomycosis, infectious particles of these fungi enter the human body at the site of injury and may cause chronic infection, mainly in tropical and subtropical endemic areas. Chromoblastomycosis is almost exclusively diagnosed in patients with fully functioning immunity, with typically muriform cells present in infected tissue distinguishing this condition from phaeohyphomycosis. Phaeohyphomycosis, a less specific disease caused by dark-pigmented fungi, usually makes tissue necrotiz

Pathogenesis-related proteins and peptides as promising tools for engineering plants with multiple stress tolerance.

Pathogenesis-related (PR) proteins and antimicrobial peptides (AMPs) are a group of diverse molecules that are induced by phytopathogens as well as defense related signaling molecules. They are the key components of plant innate immune system especially systemic acquired resistance (SAR), and are widely used as diagnostic molecular markers of defense signaling pathways. Although, PR proteins and peptides have been isolated much before but their biological function remains largely enigmatic despite the availability of new scientific tools. The earlier studies have demonstrated that PR genes pro

A cooperative regulatory module between TAGL2 and JMJC1 activates specific defense genes against root-knot nematodes in tomato.

Plant-parasitic nematodes (PPNs) threaten global food security. Although epigenetic modifications are crucial for plant immunity, how histone modifiers contribute to root-knot nematodes (RKNs, Meloidogyne incognita) resistance remains unclear. Here, using genetic, molecular and biochemical approaches, we investigated the epigenetic and transcriptional mechanisms underlying RKN resistance mediated by the histone demethylase (HDM) JMJC1 and the MADS-box transcription factor TAGL2 in tomato (Solanum lycopersicum). We identified JMJC1 as an RKN-induced positive defense regulator targeting H3K9me3 …

Sulfur Deprivation Modulates Salicylic Acid Responses via Nonexpressor of Pathogenesis-Related Gene 1 in Arabidopsis thaliana.

Mineral nutrients are essential for plant growth and reproduction, yet only a few studies connect the nutritional status to plant innate immunity. The backbone of plant defense response is mainly controlled by two major hormones: salicylic acid (SA) and jasmonic acid (JA). This study investigated changes in the macronutrient concentration (deficiency/excess of nitrogen, phosphorus, potassium, magnesium, and sulfur) on the expression of PR1, a well-characterized marker in the SA-pathway, and PDF1.2 and LOX2 for the JA-pathway, analyzing plants carrying the promoter of each gene fused to GUS as
