Multidrug-resistant Proteus mirabilis isolates carrying blaOXA-1 and blaNDM-1 from wildlife in China: increasing public health risk.

The emergence of multidrug resistance (MDR) in Proteus mirabilis clinical isolates is a growing public health concern and has serious implications for wildlife. What is the role of wildlife has been become one of the hot issues in disseminating antimicrobial resistance. Here, 54 P. mirabilis isolates from 12 different species were identified. Among them, 25 isolates were determined to be MDR by profile of antimicrobial susceptibility; 10 MDR P. mirabilis isolates were subjected to comparative genomic analysis by whole genome sequencing. Comprehensive analysis showed that chromosome of P. mirab…

Antimicrobial profiling and molecular characterization of antibiotic resistant genes of Proteus vulgaris isolated from tertiary care hospital, Islamabad, Pakistan.

Urinary tract infections (UTIs) are among the most common bacterial infections acquired from hospitals and community. Pseudomonas and Proteus species are the common cause of these UTIs. Generally, UTIs are self-limiting but have potential to re-occur. Extensive treatment therapy with antibiotics lead to the development of resistance in uropathogens. The development of antibiotic resistance is leading to the failure of currently available antibiotic based therapies thus making the situation worse. The objective of the present study was to access antimicrobial sensitivity and to characterize ant…

Dissemination of integrons and carbapenemase-encoding genes among multidrug resistant Proteus mirabilis isolated from urinary tract infections in Egypt.

BACKGROUND: Proteus mirabilis (P. mirabilis) is an opportunistic pathogen responsible for various community-acquired and nosocomial infections, particularly urinary tract infections (UTIs). Rising resistance to broad-spectrum antibiotics, including carbapenems, is narrowing treatment options and poses a major public health concern. This study aimed to investigate the antimicrobial susceptibility patterns of P. mirabilis isolates obtained from UTI patients. Then, to assess the molecular determinants of carbapenem-resistant isolates with a focus on the role of integrons in resistance gene dissem…

AmpC β-Lactamases in SPICE (Serratia, Pseudomonas, Indole-Positive Proteus, Citrobacter, Enterobacter) Organisms: A Rising Threat to Antimicrobial Therapy.

The global rise of antimicrobial resistance (AMR) among Gram-negative bacteria is a major public health threat. AmpC β-lactamases, particularly in SPICE (Serratia, Pseudomonas, indole-positive Proteus, Citrobacter, Enterobacter) organisms, contribute significantly to multidrug resistance by hydrolyzing cephamycins and other β-lactam antibiotics. Unlike extended-spectrum β-lactamases (ESBLs), AmpC β-lactamases are generally not inhibited by classical β-lactamase inhibitors such as clavulanic acid, sulbactam, and tazobactam. However, newer non-β-lactam β-lactamase inhibitors, including avibactam…

Prevalence and characterization of IncQ1α-mediated multi-drug resistance in Proteus mirabilis Isolated from pigs in Kunming, Yunnan, China.

BACKGROUND: Proteus mirabilis is a conditionally pathogenic bacterium that is inherently resistant to polymyxin and tigecycline, largely due to antibiotic resistance genes (ARGs). These ARGs can be horizontally transferred to other bacteria, raising concerns about the Inc plasmid-mediated ARG transmission from Proteus mirabilis, which poses a serious public health threat. This study aims to investigate the presence of Inc plasmid types in pig-derived Proteus mirabilis in Kunming, Yunnan, China. METHODS: Fecal samples were collected from pig farms across six districts of Kunming (Luquan, Jinni…

Catalytic and structural properties of IRT-21 beta-lactamase (TEM-77) from a co-amoxiclav-resistant Proteus mirabilis isolate.

Proteus mirabilis strain MAG1, a clinical isolate that is resistant to broad-spectrum penicillins and co-amoxiclav, produces inhibitor-resistant TEM (IRT)-21, a novel mutant of TEM beta-lactamase. This enzyme has a pI of 5.2 and is derived from the bla(TEM-1a) gene ancestor. It contains two major amino acid substitutions specific for co-amoxiclav resistance (Leu-69 for Met and Ser-244 for Arg) that have never been found together previously. The dramatic loss of sensitivity to clavulanic acid, the enhancement of K(m) for all beta-lactams and markedly for ticarcillin, and the decrease in the cat…

Natural History of Patients Eligible for PROTEUS Diagnosed with Prostate Cancer and Treated with Radical Prostatectomy Without Perioperative Systemic Treatment.

The ongoing PROTEUS study aims to evaluate the effectiveness of perioperative Apalutamide combined with androgen-deprivation therapy in improving oncological outcomes for a specifically defined high-risk prostate cancer (PCa) group who were treated with radical prostatectomy (RP). Interestingly, the PROTEUS inclusion criteria did not rely on established risk categories such as the European Association of Urology (EAU) or the National Comprehensive Cancer Network (NCCN) high-risk classification. Consequently, little is known about the natural course of patients eligible for PROTEUS who were dia…

Co-infection with Proteus mirabilis and extraintestinal pathogenic Escherichia coli aggravated liver injury in chickens by disrupting the jejunal barrier.

Avian pathogenic Escherichia coli (APEC), belong to extraintestinal pathogenic E. coli (ExPEC), causes colibacillosis in poultry, characterized by fibrinous inflammation of internal organs, high morbidity and mortality, economic losses, and food safety concerns. Proteus mirabilis is frequently isolated alongside APEC from diseased chickens, yet its role in colibacillosis remains unclear. As the intestinal lumen serves as a reservoir for both E. coli and P. mirabilis, it is possible that APEC triggers colibacillosis from the intestines, though this pathway has not been extensively studied. The …

GlpC gene is responsible for biofilm formation and defense against phagocytes and imparts tolerance to pH and organic solvents in Proteus vulgaris.

Biofilm-forming bacteria are highly resistant to antibiotics, host immune defenses, and other external conditions. The formation of biofilms plays a key role in colonization and infection. To explore the mechanism of biofilm formation, mutant strains of Proteus vulgaris XC 2 were generated by Tn5 random transposon insertion. Only one biofilm defective bacterial species was identified from among 500 mutants. Inactivation of the glpC gene coding an anaerobic glycerol-3-phosphate dehydrogenase subunit C was identified by sequence analysis of the biofilm defective strain. Differences were detected…
