A Common Eurasian Fish Tapeworm, Caryophyllaeides fennica (Cestoda), in Western North America: Further Evidence of 'Amphi-Pacific' Vicariance in Freshwater Fish Parasites.

The freshwater fish faunas of the Palaearctic and Nearctic regions show similarities but also notable differences, resulting in diverse distributions of their parasites. Relatively few parasite species occur in both regions and fewer still have been examined using molecular data. We report a rare example of 'amphi-Pacific' distribution, involving the common Palaearctic parasite, the caryophyllidean cestode Caryophyllaeides fennica (Schneider, 1902), in the chiselmouth Acrocheilus alutaceus Agassiz and Pickering, 1855, an endemic cyprinid in northwestern Nearctic, Oregon. Pinworm diversity in free-ranging howler monkeys (Alouatta spp.) in Mexico: Morphological and molecular evidence for two new Trypanoxyuris species (Nematoda: Oxyuridae).

Two new species of Trypanoxyuris are described from the intestine of free-ranging howler monkeys in Mexico, Trypanoxyuris multilabiatus n. sp. from the mantled howler Alouatta palliata, and Trypanoxyuris pigrae n. sp. from the black howler Alouatta pigra. An integrative taxonomic approach is followed, where conspicuous morphological traits and phylogenetic trees based on DNA sequences are used to test the validity of the two new species. The mitochondrial cytochrome oxidase subunit 1 gene, and the nuclear ribosomal 18S and 28S rRNA genes were used for evolutionary analyses, with the concatenat

The complete mitochondrial genome of rabbit pinworm Passalurus ambiguus: genome characterization and phylogenetic analysis.

Passalurus ambiguus (Nematda: Oxyuridae) is a common pinworm which parasitizes in the caecum and colon of rabbits. Despite its significance as a pathogen, the epidemiology, genetics, systematics, and biology of this pinworm remain poorly understood. In the present study, we sequenced the complete mitochondrial (mt) genome of P. ambiguus. The circular mt genome is 14,023 bp in size and encodes of 36 genes, including 12 protein-coding, two ribosomal RNA, and 22 transfer RNA genes. The mt gene order of P. ambiguus is the same as that of Wellcomia siamensis, but distinct from that of Enterobius ve

Up-regulation of extracellular copper/zinc superoxide dismutase mRNA after transmission of the filarial parasite Acanthocheilonema viteae in the vertebrate host Meriones unguiculatus.

The gene encoding the cytoplasmic copper/zinc superoxide dismutase (AVSOD1) from the filarial parasite Acanthocheilonema viteae was isolated from a genomic DNA library using a degenerate oligonucleotide probe. Additionally, cDNAs of the AVSOD1 and the secreted extracellular SOD (AVSOD2) were both cloned by RT-PCR, and the AVSOD2 was expressed at high levels in E. coli. The amino acid sequence of the AVSOD1 is 89.5 and 87.5% identical to that of the corresponding enzymes of Brugia pahangi and Onchocerca volvulus, respectively. In contrast, the AVSOD2 shows a lower degree of identity to the othe

A novel RFLP-PCR method for the rapid diagnosis of Echinococcus multilocularis and different Echinococcus granulosus sensu lato species from tissue samples.

Both alveolar echinococcosis and cystic echinococcosis are zoonotic diseases affecting thousands of humans in Europe annually. Five Echinococcus species are known to be endemic in Europe, but with varying relative importance in humans and animals. In order to identify these Echinococcus species from tissue samples, an RFLP-PCR method was developed and validated in accordance with the French quality standards for molecular diagnostics (NF U47-600-2). Amplification by PCR of about 846 bp from the 12S ribosomal RNA gene was followed by a single digestion step with three restriction enzymes (AccI,

Integrative Taxonomy of Didymozoids Parasitizing Thunnus obesus (Scombridae) from Southwest Atlantic Ocean: A New Genus and Species.

The global fauna of Didymozoidae infecting fishes is very diverse and includes 270 species. Integrative taxonomic studies in this group are rare, and genetic data are lacking for the molecular identification of species. In this study, a new genus and species, Platodidymocystis yamagutii n. gen., n. sp., is described based on morphological and genetic analyses. The new genus is allocated to Didymozoinae but differs from all other genera of this subfamily, mainly by the morphology of the testes and ovary. The genetic analysis and molecular phylogeny using 28S rDNA and ITS2 markers showed P. yama

Genetic Variation Within and Between G1 and G3 Genotypes of Echinococcus granulosus sensu stricto from Chile Revealed by Partial DNA Sequencing of rrnS Gene Marker.

Background: Cystic echinococcosis (CE) caused by the parasite Echinococcus granulosus sensu lato is one of the most important zoonoses worldwide, causing a great impact on human and animal health. Despite its impact, control programs have not been able to eradicate the parasite; in part, because its transmission cycles, its phylogeny, and the host-parasite relationships are still not well known. In order to deepen knowledge about Echinococcus spp. populations in the endemic areas of southern Chile, the present study was carried out. Materials and Methods: Ten isolates of CE cysts were analyzed

Morphological and molecular data on Pseudozoogonoides ugui Shimazu, 1974 (Digenea: Microphalloidea: Zoogonidae) ex Pseudaspius hakonensis (Günther, 1877) and taxonomic problems in Zoogoninae genera.

New morphological and molecular data were generated for trematodes recovered from the intestines of the fish Pseudaspius hakonensis from two locations in the south of the Russian Far East. Morphologically, these trematodes are identical to Pseudozoogonoides ugui (Microphalloidea: Zoogonidae) from Japan. According to results of phylogenetic analysis based on 28S rDNA sequence data, P. ugui was closely related to Zoogonoides viviparus, and P. subaequiporus appears as a sister taxon to these two species. Genetic distance values, calculated based on both 28S rDNA and ITS2 rDNA, between P. ugui and

Is Saccocoelioides bacilliformis Szidat, 1973 (Digenea: Haploporidae) a valid species? Genetic and morphological studies of this controversial species.

Saccocoelioides is a genus within the family Haploporidae and is distributed throughout the Americas. The recent application of molecular techniques has facilitated the reorganization of this genus and the description of new species, resulting in a current total of 28 species. In Argentina, 11 species have been identified; however, the validity of Saccocoelioides bacilliformis and Saccocoelioides octavus remains controversial. Recently, specimens of digeneans were collected from two distinct geographical locations: the Paraná River basin (Misiones province) and the Río de la Plata basin (Bueno

Function of the lesswright (lwr) gene in the growth, development, and reproduction of Schistosoma japonicum.

The lesswright (lwr) gene and its products are essential molecules in mitosis, DNA repair, and embryo formation in many eukaryotes. In this study, immunohistochemical analysis revealed that the Lwr protein was located in the internal tissues and the surface layer of the adult Schistosoma japonicum (Sj) worms. The mRNA expression levels of SjLwr at different points were evaluated by quantitative real-time RT-PCR. The expression of SjLwr peaked at 14 days and then decreased thereafter. SjLwr expression was relatively more stable in male worms than in female worms. The functions of SjLwr were exp

Accuracy of molecular biology techniques for the diagnosis of Strongyloides stercoralis infection-A systematic review and meta-analysis.

BACKGROUND: Strongyloides stercoralis infection is a neglected tropical disease which can lead to severe symptoms and even death in immunosuppressed people. Unfortunately, its diagnosis is hampered by the lack of a gold standard, as the sensitivity of traditional parasitological tests (including microscopic examination of stool samples and coproculture) is low. Hence, alternative diagnostic methods, such as molecular biology techniques (mostly polymerase chain reaction, PCR) have been implemented. However, there are discrepancies in the reported accuracy of PCR. METHODOLOGY: A systematic revi

Reliable molecular differentiation of Trichuris ovis and Trichuris discolor from sheep (Ovis orientalis aries) and roe deer (Capreolus capreolus) and morphological characterisation of their females: morphology does not work sufficiently.

The main aim of the study was to evaluate associations between morphological variability of Trichuris females from sheep and roe deer and their rDNA polymorphism in whipworm populations from the Czech Republic. The results introduced the use of new molecular markers based on the internal transcribed spacer (ITS)1-5.8S RNA-ITS2 region polymorphisms, as useful tools for the unambiguous differentiation of congeners Trichuris ovis and Trichuris discolor. These markers revealed both parasites in roe deer and in sheep; however, T. ovis females predominated in sheep while T. discolor females occurred
