The aim of this work was to investigate the dynamics of the quantity of LINE in the DNA composition of blood plasma and extrachromosomal DNA of liver cells under conditions of either immobilization or exposure to ionizing radiation or a combination of both factors. It has been established that post-stress changes in the quantity of genetic elements may be interpreted as an activation of the mechanisms of retroposition and intercellular transfer of genetic elements that ensure the formation of adaptive responses at the systemic level. The formation of adaptive responses is expressed through the

Selection of a phage-stable strain of a new species of the rifamycin-producing organism was carried out. The phage-stable mutants were selected with respect to the virulent phage 2739 isolated from a lysogenic culture of the rifamycin-producing organism. Spontaneous phage-stable mutants formed at a rate of 0.8 per cent. Most of them belonged to the morphological colony type with a decreased activity level. No shifts in variation with respect to the property of the antibiotic production were noted under the action of phage 2739. 62 per cent of the phage-stable variants isolated from the seconda

The distribution of X-ray induced aberrations between trypsin-banded regions within human chromosomes has been studied in over a thousand cells from normal and abnormal individuals. An analysis of this data has been performed for each type of aberration separately, taking into account the relative amount of material at risk in reach region. A number of significant deviations from expectation were detected but many of these could be explained in terms of the failure of detection of some aberrations.

For radiation protection purposes, noncancer effects with a threshold-type dose-response relationship have been classified as tissue reactions (formerly called nonstochastic or deterministic effects), and equivalent dose limits aim to prevent occurrence of such tissue reactions. Accumulating evidence demonstrates increased risks for several late occurring noncancer effects at doses and dose rates much lower than previously considered. In 2011, the International Commission on Radiological Protection (ICRP) issued a statement on tissue reactions to recommend a threshold of 0.5 Gy to the lens of

Specialized transducing phages for gluconate 6-phosphate dehydrogenase (gnd), a constitutive enzyme in Escherichia coli, have been isolated using a method previously described for other genes. The gnd-his region, carried on an F' episome, was first transposed to tonB. Rare phages carrying gnd were selected, by transduction, from phi80 lysogens of these strains; one phage also carried his (phi80gndhis). From the transductants, high-frequency transducing lysates were obtained; low multiplicity of infection then yielded defective lysogens. tonB deletion analysis of the phi80dgndhis lysogen shows

Chronic radiation dermatitis is a late side effect of skin irradiation, which may deteriorate patients' quality of life. There is a lack of precise data about its incidence; however, several risk factors may predispose to the development of this condition. It includes radiotherapy dose, fractionation, technique, concurrent systemic therapy, comorbidities, and personal and genetic factors. Chronic radiation dermatitis is mostly caused by the imbalance of proinflammatory and profibrotic cytokines. Clinical manifestation includes changes in skin appearance, wounds, ulcerations, necrosis, fibrosis

The gastrointestinal tract is a critical target of acute radiation exposure. Severe radiation exposure can deplete epithelial stem and progenitor cells, compromise barrier integrity, alter host-microbe interactions, and drive fluid loss, inflammation, and systemic decline. Mammalian models remain essential for clinical translation, but their cost and complexity constrain sample sizes, statistical power, large-scale mechanistic discovery, and countermeasure screening. The adult Drosophila melanogaster midgut provides a complementary in vivo platform containing intestinal stem cells, absorptive

Variability in radiation-related health risk and genetic susceptibility to radiation effects within a population is a key issue for radiation protection. Besides differences in the health and biological effects of the same radiation dose, individual variability may also affect dose distribution and its consequences for the same exposure. As exposure to radon progeny affects a large population and has a well-established dose-effect relationship, investigating individual variability upon radon exposure may be particularly important. Using the Stochastic Lung Model combined with mucociliary clear

BACKGROUND: High-dose radiation altering the genetic material in patients' bone marrow cells can lead to hematopoietic radiation syndrome. Accordingly, the presence of radiation protections agents is critical to preventing these adverse effects.
OBJECTIVE: This study aimed to evaluate the radioprotection of the exclusive or combination effect of resveratrol and crocin extracts at various concentrations on irradiated human lymphocytes.
MATERIAL AND METHODS: In this experimental study, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method was used to evaluate the cell v

The brain of a human and other organisms is affected by the electromagnetic field (EMF) radiations, emanating from the cell phones and mobile towers. Prolonged exposure to EMF radiations may cause neurological changes in the brain, which in turn may bring chemical as well as morphological changes in the brain. Conventionally, the identification of EMF radiation effect on the brain is performed using cellular-level analysis. In the present work, an automatic image processing-based approach is used where geometric features extracted from the segmented brain region has been analyzed for identifyi

The current report assessed the effects of low-level proton irradiation in inbred adult male Fischer 344 and Lewis rats performing an analog of the human Psychomotor Vigilance Test (PVT), commonly utilized as an object risk assessment tool to quantify fatigue and sustained attention in laboratory, clinical, and operational settings. These strains were used to determine if genetic differences in dopaminergic function would impact radiation-induced deficits in sustained attention. Exposure to head-only proton irradiation (25 or 100 cGy) disrupted rPVT performance in a strain-specific manner, wit

PURPOSE: Recent studies have suggested relative radioresistance in glioblastoma multiforme (GM) tumors in older patients, consistent with their shorter survival. Two common molecular genetic abnormalities in GM are age related: epidermal growth factor receptor (EGFR) overexpression in older patients and p53 mutations in younger patients. We tested whether these abnormalities correlated with clinical heterogeneity in GM response to radiation treatment.
METHODS AND MATERIALS: Radiographically assessed radiation response (5-level scale) was correlated with EGFR immunoreactivity, p53 immunoreacti

Ionizing radiation is a well-known risk factor of cancer development, but the mechanism of radiation induced carcinogenesis is not clear. Chromosomal rearrangements induced by radiation most likely are one of the principal genetic alterations resulting in malignant transformation. The chimeric BCR-ABL associated with chronic myelogenous leukemia (CML) and H4-RET oncogenes associated with thyroid papillary carcinoma are the result of a translocation and inversion, respectively. In vitro studies showed these genes were induced by high-doses of X-irradiation in cell lines. Studies also show that

Renewed interest in experimental photocarcinogenesis was stimulated by the observation that skin cancers induced in mice by ultraviolet radiation are highly antigenic. Most of these cancers are rejected by the immune system on transplantation into a normal, genetically identical animal. This finding raised the question of how these tumors could survive and grow progressively in the primary host without inducing immunologic rejection. Studies addressing this question produced the unexpected finding that mice that had been exposed to ultraviolet radiation, but that had not yet developed primary

This paper is a tribute to Paul Lohman at the occasion of his retirement from the position of Professor in the Medical Faculty at the Leiden University in The Netherlands and as Director of its Department of Radiation Genetics and Chemical Mutagenesis. Paul's contributions to the science of genetic toxicology are discussed in the context of more recent insights as to how mammalian cells process DNA damage, and how this may lead to cancer and, possibly, aging. Starting with his work on the characterization of UV-induced DNA repair in cultured cells from xeroderma pigmentosum patients and the de…

OBJECTIVE: To map the gene for human cartilage intermediate layer protein (CILP) in order to assess its involvement in some familial forms of calcium pyrophosphate dihydrate (CPPD) deposition disease.
METHODS: A radiation hybrid panel was analyzed for chromosomal assignment of the CILP gene within a 1-cM limit of resolution. The location of the gene for CILP was confirmed to reside at the observed radiation hybrid locus by fluorescence in situ hybridization.
RESULTS: The human CILP gene resides at chromosome 15q21.
CONCLUSION: This map location definitively excludes mutations in the CILP ge

All forms of cancer are initiated by heritable changes in gene expression. Although point mutations have been studied extensively, much less is known about homologous recombination events, despite its role in causing sequence rearrangements that contribute to tumorigenesis. Although transgenic mice that permit detection of point mutations have provided a fundamental tool for studying point mutations in vivo, until recently, transgenic mice designed specifically to detect homologous recombination events in somatic tissues in vivo did not exist. We therefore created fluorescent yellow direct rep

The aim of the present study was to evaluate the influence of the exclusion of cells with unstable aberrations in the elaboration of dose-effect curves for translocations and their implications in biological dosimetry of past exposures. To establish dose-effect curves, peripheral blood samples were irradiated with 60Co gamma rays at ten different doses and the yield of translocations analyzed by FISH was considered in all cells and in stable cells (those without dicentrics, acentrics or rings). To discriminate transmissible translocations, the dose- effect curve for total apparently simple tra
