Structural and biochemical insights into a hyperthermostable urate oxidase from Thermobispora bispora for hyperuricemia and gout therapy.

Microbial urate oxidase has emerged as a potential source of therapeutic properties for hyperuricemia in arthritic gout and renal disease. The thermostability and long-term thermal tolerance of the enzyme need to be established to prolong its therapeutic effects. Here, we present the biochemical and structural aspects of a hyperthermostable urate oxidase (TbUox) from the thermophilic microorganism Thermobispora bispora. Enzymatic characterization of TbUox revealed that it was active over a wide range of temperatures, from 30 to 70 °C, with optimal activity at 65 °C and pH 8.0, which suggests i

Carboxymethyl cellulose-ferulic acid nanocomposite enhances salinity tolerance and phytochemical accumulation in Dracocephalum moldavica via coordinated redox and osmotic regulation.

BACKGROUND: Salinity is a significant constraint on medicinal and aromatic plants that limits their growth and photosynthetic activity, increases oxidative stress, and disrupts water balance. RESULTS: The present paper prepared a carboxymethyl cellulose-ferulic acid nanocomposite (FA-NC), which is a bio-based nano-formulation to enhance the performance of Dracocephalum moldavica in the presence of salinity stress, and the effect was directly compared to free FA. Carboxymethyl cellulose was successfully used to prepare the FA-NC, which was verified by conventional physicochemical characterizat

A comparative framework for trichome and trichome-derived epidermal appendage development across model glycophytes and halophytes.

This review compares trichome and trichome-derived epidermal appendage development across model glycophytes and halophytes, revealing that conserved regulatory modules are independently rewired into lineage specific circuits to produce diverse epidermal structures. In halophytes, salt glands and epidermal bladder cells further integrate these developmental programs with ion transport and stress physiology, offering entry points for engineering crop stress resilience. Trichome-derived epidermal structures have arisen repeatedly across angiosperms, yet the mechanisms by which conserved epidermal

Xylem anatomy and pit architecture but not perforation-plate types shape hydraulic strategies against hypersaline and drought stress in eight woody mangroves.

Mangrove species mainly inhabit tropical and subtropical intertidal zones, where they suffer periodic tidal inundation. They display diverse perforation-plate types and pit structures; however, the extent to which these anatomical features contribute to their adaptation to coastal environments remains unclear. In this study, we quantified 27 traits, representing hydraulics, anatomy and pit characteristics, in eight woody mangrove species from Hainan, Southern China, including four simple-perforation (SI) and four scalariform-perforation (SC) plate species. All species exhibited high xylem embo

Compensatory root proliferation and physiological adjustments enable Velvet Ash (Fraxinus velutina) to tolerate high nonuniform salinity.

Partial desalination offering a promising approach in the saline soil reducing the costs of vegetation construction. However, the quantitative salinity thresholds for tree species in such zones remain unclear, and understanding plant responses to nonuniform salinity is critical for designing effective afforestation strategies via partial zone desalination. In this study, a split-root system was used to examine the growth, physiological responses, and salinity tolerance thresholds of velvet ash (Fraxinus velutina) seedlings exposed to uniform and nonuniform salinity. In the nonuniform treatment

Comparative analysis of DDR-related genes and microRNA expression during rice germination: Implications for salinity susceptibility screening.

Soil salinity poses a significant threat to the agri-food sector and particularly to rice cultivation. High salinity during germination induces overproduction of reactive oxygen species (ROS) that cause lesions in the DNA resulting in reduced vigor. MicroRNAs (miRNAs) are known to modulate stress response in plants, however, studies focusing on its relation with the expression of the DNA damage response (DDR)-related genes are not thoroughly explored. In this regard, the aim of this work was to investigate the link between the expression of miRNAs and putative targeted DDR-related genes in res

Analysis of Growth and Physiological Response Characteristics of Suaeda salsa Under Compound Stress.

This investigation examines the physiological and ecological adaptation mechanisms of Suaeda salsa, a halophytic plant, in response to multifaceted environmental stresses prevalent in coastal saline-alkali wetlands. Using a three-factor, three-level orthogonal experimental design, we evaluated the combined effects of temperature (15°C, 20°C, 25°C), light intensity (25, 50, 100 μmol m-2 s-1), and salinity (0, 200, 400 mM NaCl) on plant performance. Temperature emerged as the dominant factor influencing growth, photosynthetic efficiency, and betacyanin accumulation. The highest growth rate and b

4D-proteome analysis reveals the role of hydrogen sulfide-based priming in attenuating salinity-induced GABA accumulation in rice.

Soil salinity severely constrains rice productivity by inducing ionic imbalance, oxidative damage, and metabolic disruption. Hydrogen sulfide (H2S) has emerged as an important signaling molecule in plant stress responses, yet its mechanistic role in salinity tolerance remains incompletely understood. Here, we investigated the function of H2S in modulating salinity stress responses in rice using two cultivars with contrasting salinity tolerance, the sensitive Dongjin and the tolerant IR73. Salinity stress resulted in severe growth inhibition, particularly in Dongjin, accompanied by elevated mal

Effect of sodium on iron absorption, translocation and allocation in halophilic Amaranthaceae species.

In arid regions, alkaline conditions resulting from soil sodification make iron unavailable, causing plants to suffer from salt-alkali stress. We hypothesized that halophilic species possess an exceptional iron uptake mechanism involving Na. Here, we investigated the effects of medium pH (acidic or neutral), amounts of iron (0.358 μM or 35.8 μM) and NaCl (5 or 100 mM) on Fe uptake, translocation, and allocation in salt tolerant Mexican tea (Dysphania ambrosioides L.), halophilic mesophyte Swiss chard (Beta vulgaris L. spp. cicla), and euhalophyte romeritos (Suaeda edulis Flores Olv. & Noguez)

Role of non-coding RNAs against salinity stress in Oryza species: Strategies and challenges in analyzing miRNAs, tRFs and circRNAs.

Salinity is an imbalanced concentration of mineral salts in the soil or water that causes yield loss in salt-sensitive crops. Rice plant is vulnerable to soil salinity stress at seedling and reproductive stages. Different non-coding RNAs (ncRNAs) post-transcriptionally regulate different sets of genes during different developmental stages under varying salinity tolerance levels. While microRNAs (miRNAs) are well known small endogenous ncRNAs, tRNA-derived RNA fragments (tRFs) are an emerging class of small ncRNAs derived from tRNA genes with a demonstrated regulatory role, like miRNAs, in huma

Phenotypic and phylogenetic analyses of Listeria monocytogenes strains reveal enhanced bile tolerance in clinical isolates.

Introduction. Molecular epidemiological and phenotypic analyses of Listeria monocytogenes strains can inform our understanding of factors that influence onward transmission, virulence potential and ability to control the pathogen in foods or in clinical settings. Bile acids represent a host-specific barrier to microbial colonization of the gastrointestinal tract and are, therefore, not encountered in the external environment. We tested the hypothesis that tolerance of bile acids may be an evolutionary adaptation across L. monocytogenes clonal complexes (CCs), which varies with genotype and/or

Visible-light-induced multicomponent cascade cycloaddition involving N-propargyl aromatic amines, diaryliodonium salts and sulfur dioxide: rapid access to 3-arylsulfonylquinolines.

A visible-light-induced, Eosin Y catalyzed three-component synthesis of 3-arylsulfonylquinoline derivatives through N-propargyl aromatic amines, diaryliodonium salts and sulfur dioxide has been discovered. This transformation represents an efficient and attractive method for the straightforward synthesis of 3-arylsulfonylquinoline derivatives via the formation of C-S bonds and quinolines in one step. In addition, it exhibits good substrate scope and functional group tolerance. The use of easy-to-handle diaryliodonium salts, sulfur dioxide sources and the cheap photocatalyst Eosin Y together wi

From Plasma to Protection: Cold Plasma-Induced Biochemical and Anatomical Responses in Pea Plants under Salinity Stress.

Salinisation of agricultural land reduces crop productivity, often caused by poor irrigation practices and inadequate drainage that lead to salt accumulation in soil. High salinity disrupts water uptake, nutrient balance, promotes the formation of reactive oxygen species (ROS), induces stomatal closure, and damages photosynthesis, ultimately limiting plant growth and stress tolerance. Cold atmospheric pressure plasma (CAPP) has recently emerged as a promising method to mitigate salinity stress. Plasma-generated reactive particles (such as electrons, ions, metastables, reactive oxygen and nitro
