The spatial organization characteristics and redox status of the extracellular space (ECS) are crucial in the development of brain diseases. However, it remains a challenge to simultaneously capture dynamic changes in microstructural features and redox states at the submicron level within the ECS. Here, we developed a reversible glutathione (GSH)-responsive nanoprobe (RGN) for mapping the spatial organization features and redox status of the ECS in brain tissues with nanoscale resolution. The RGN is composed of polymer nanoparticles modified with GSH-responsive molecules and amino-functionaliz

The role of electrical activity in axon guidance has been extensively studied in vitro. To better understand its role in the intact nervous system, we imaged intracellular Ca(2+) in zebrafish primary motor neurons (PMN) during axon pathfinding in vivo. We found that PMN generate specific patterns of Ca(2+) spikes at different developmental stages. Spikes arose in the distal axon of PMN and were propagated to the cell body. Suppression of Ca(2+) spiking activity in single PMN led to stereotyped errors, but silencing all electrical activity had no effect on axon guidance, indicating that an acti

One of the most important properties of quantum dots (QDs) is their size. Their size will determine optical properties and in a colloidal medium their range of interaction. The most common techniques used to measure QD size are transmission electron microscopy (TEM) and X-ray diffraction. However, these techniques demand the sample to be dried and under a vacuum. This way any hydrodynamic information is excluded and the preparation process may alter even the size of the QDs. Fluorescence correlation spectroscopy (FCS) is an optical technique with single molecule sensitivity capable of extracti

Astrocytes play essential roles in brain function and disorders. Yet, compared to neurons, our knowledge of the physiological and pathological signaling mechanisms in astrocytes remains limited. As a major challenge, the ultrathin (~10-100 nm) processes of astrocytes render high-throughput quantitative molecular imaging within well-defined cellular contexts very difficult. Here, we introduce a single-molecule localization microscopy-based methodology that achieves unprecedented resolution of the intricate astrocytic arbor in intact brain circuits. Postnatal tagging of the plasma membrane by el

BACKGROUND: Neurofilament light is a marker of axonal degeneration, whose measurement from peripheral blood was recently made possible by new assays.
OBJECTIVE: We aimed to determine whether plasma neurofilament light chain (NfL) concentration reflects brain white matter integrity in patients with early Parkinson's disease (PD).
METHODS: 137 early PD patients and 51 healthy controls were included. Plasma NfL levels were measured using ultrasensitive single molecule array. 3T MRI including diffusion tensor imaging was acquired for voxelwise analysis of association between NfL and both fractio

Single molecule (SM) super-resolution microscopies bypass the diffraction limit of conventional optical techniques and provide excellent spatial resolutions in the tens of nanometers without overly complex microscope hardware. SM imaging using optical astigmatism is an efficient strategy for visualizing subcellular features in 3D with a z-range of up to ∼1 µm per acquisition. This approach however, places high demands on fluorophore brightness and photoswitching resilience meaning that imaging entire cell volumes in 3D using SM super-resolution remains challenging. Here we employ SM astigmatis

Radiopharmaceuticals are biologically active molecules labeled with radionuclides that have advanced the possibility of the nuclear medicine. They support non-invasive, high-resolution diagnostic imaging of molecular and physiological processes in vivo. The techniques such as positron emission tomography (PET) and single-photon emission computed tomography (SPECT) utilize short-lived β⁺ and γ-emitting isotopes to generate highly sensitive, three-dimensional assessments of biological function. In therapeutic applications, radionuclides that emit β⁻ particles, α particles, or Auger electrons ena

Rotational rainbows are a common feature in inelastic molecular collisions, which directly reflect the short-range anisotropic intermolecular repulsion. It is well-established that rotationally inelastic collisions of heteronuclear diatoms might exhibit double rainbows, due to different intermolecular interactions at the two ends of the diatom. This is in sharp contrast to single rainbows for homonuclear diatoms. Here, we present experimental observations of double rainbow structures in the collisional charge-transfer reaction between spin-orbit selected Ar+(2P3/2) ions and a representative ho

DNA degradation by nucleases is central to genome maintenance, immune defense, and the clearance of extracellular DNA, yet its execution at the single-molecule level remains poorly defined. Here, we use high-speed atomic force microscopy (HS-AFM) to directly visualize the real-time dynamics of DNA digestion by DNase I at nanometer resolution. DNase I repeatedly revisits structurally strained DNA regions before cleavage initiation, revealing a topology-sensitive mode of interaction that is inaccessible to ensemble biochemical approaches. Time-resolved imaging further uncovers a multistep degrad

Coacervates have been widely used to mimic membraneless organelles (MLOs). However, coacervates without a membrane or stabilizing surface do not feature the same level of stability as MLOs. This study shows that specifically engineered surface-active proteins can interact with the interface of polypeptide coacervates, conferring resistance to coacervate dissolution and fusion. Modulating the molecular characteristics of these coacervate stabilizing proteins highlighted that their dimerization aids in achieving effective interface stabilizers. Cryo-TEM imaging showed a densely packed protein mo

The genome of African swine fever virus (ASFV) is characterized by a single linear double-stranded DNA molecule. However, the question of whether ASFV genomic DNA enters the nucleus has been a subject of debate. In this study, DNAscope in situ hybridization (ISH) technology was employed to trace the genomic DNA of ASFV. Specific probes targeting the DNA of the ASFV HLJ/18 strain were designed and synthesized, facilitating the successful detection of ASFV DNA in ASFV-infected porcine alveolar macrophages (PAMs). Detection of ASFV genomic DNA commenced as early as 2 h post-infection (hpi), with

Insoluble cytoplasmic aggregate formation of the RNA-binding protein TDP-43 is a major hallmark of neurodegenerative diseases including Amyotrophic Lateral Sclerosis. TDP-43 localizes predominantly in the nucleus, arranging itself into dynamic condensates through liquid-liquid phase separation (LLPS). Mutations and post-translational modifications can alter the condensation properties of TDP-43, contributing to the transition of liquid-like biomolecular condensates into solid-like aggregates. However, to date it has been a challenge to study the dynamics of this process in vivo. We demonstrate

Which isoforms of apolipoprotein E (apoE) we inherit determine our risk of developing late-onset Alzheimer's Disease (AD), but the mechanism underlying this link is poorly understood. In particular, the relevance of direct interactions between apoE and amyloid-β (Aβ) remains controversial. Here, single-molecule imaging shows that all isoforms of apoE associate with Aβ in the early stages of aggregation and then fall away as fibrillation happens. ApoE-Aβ co-aggregates account for ~50% of the mass of diffusible Aβ aggregates detected in the frontal cortices of homozygotes with the higher-risk AP

The wavelength control of photochemistry usually results from ultrafast dynamics following the excitation of different electronic states. Here, we investigate the CF3COCl molecule, exhibiting wavelength-dependent photochemistry both via (i) depositing increasing internal energy into a single state and (ii) populating different electronic states. We reveal the mechanism behind the photon-energy dependence by combining nonadiabatic ab initio molecular dynamics techniques with the velocity map imaging experiment. We describe a consecutive mechanism of photodissociation where an immediate release

Here, we introduce the full functional reconstitution of genetically validated core protein machinery (SNAREs, Munc13, Munc18, Synaptotagmin, and Complexin) for synaptic vesicle priming and release in a geometry that enables detailed characterization of the fate of docked vesicles both before and after release is triggered with Ca2+. Using this setup, we identify new roles for diacylglycerol (DAG) in regulating vesicle priming and Ca2+-triggered release involving the SNARE assembly chaperone Munc13. We find that low concentrations of DAG profoundly accelerate the rate of Ca2+-dependent release

In the adult ventricular-subventricular zone (V-SVZ), neural stem cells (NSCs) give rise to transit-amplifying progenitor (TAP) cells. These progenitors reside in different subniche locations, implying that cell movement must accompany lineage progression, but the dynamic behaviors of adult NSCs and TAPs remain largely unexplored. Here, we performed live time-lapse imaging with computer-based image analysis of young and aged 3D V-SVZ wholemounts from transgenic mice with fluorescently distinguished NSCs and TAP cells. Young V-SVZ progenitors are highly dynamic, with regular process outgrowth a

Targeting areas of inflammation offers potential therapeutic and diagnostic benefits by maximizing drug and imaging marker on-target effects while minimizing systemic exposure that can be associated with adverse side effects. This strategy is particularly beneficial in the management of inflammatory bowel disease (IBD). Here an inflammation-targeting (IT) approach based on heparin-coated human serum albumin nanoparticles (HEP-HSA NPs) that utilize the increased intestinal permeability and changes in electrostatic interaction at the site of intestinal inflammation is described. Using small-mole

The pathophysiology underlying abdominal aortic aneurysms (AAAs) remains unknown. In this study, we applied imaging mass spectrometry (IMS) to analyze the pathophysiology of the aneurysmal wall. Comparisons were performed between the tissue samples from the neck and the sac of the AAA, at a single time point, in 30 patients who underwent elective surgery of their AAAs. The localization of each lipid molecule in the aortic wall was assessed by IMS. Histopathological examination and IMS revealed a characteristic distribution of triglycerides (TGs) specifically in the aneurismal adventitia of the

Nucleation promoting factors (NPFs) initiate branched actin network assembly by activating Arp2/3 complex, a branched actin filament nucleator. Cellular actin networks contain multiple NPFs, but how they coordinately regulate Arp2/3 complex is unclear. Cortactin is an NPF that activates Arp2/3 complex weakly on its own, but with WASP/N-WASP, another class of NPFs, potently activates. We dissect the mechanism of synergy and propose a model in which cortactin displaces N-WASP from nascent branches as a prerequisite for nucleation. Single-molecule imaging revealed that unlike WASP/N-WASP, cortact

Matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) has been established as a powerful tool for the spatial characterization of molecules. Recent advances have been driven by improvements in the spatial resolution, acquisition time, and sensitivity. Although MALDI-MSI excels in the label-free, multiplexed detection of biomolecules, variations in sample preparation remain a significant source of experimental variability. Here, we present a workflow demonstrating the integration of a robust, completely automated tissue section processing system into a MALDI workflow
