Efficient Genome Editing Using the T2A-Coupled Co-Expression of Two ZFN Monomers.

Genome editing is commonly used in biomedical research. Among the genome editing tools, zinc finger nucleases (ZFNs) are smaller in size than transcription activator-like effector nucleases (TALENs) and CRISPR-Cas9. Therefore, ZFNs are easily packed into a viral vector with limited cargo space. However, ZFNs also consist of left and right monomers, which both need to be expressed in the target cells. When each monomer is expressed separately, two expression cassettes are required, thus increasing the size of the DNA. This is a disadvantage for a viral vector with limited cargo space. We herein

Analysis of Cushioned Landing Strategies of Cats Based on Posture Estimation.

This article addresses the challenge of minimizing landing impacts for legged space robots during on-orbit operations. Inspired by the agility of cats, we investigate the role of forelimbs in the landing process. By identifying the kinematic chain of the cat skeleton and tracking it using animal posture estimation, we derive the cushioning strategy that cats use to handle landing impacts. The results indicate that the strategy effectively transforms high-intensity impacts into prolonged low-intensity impacts, thereby safeguarding the brain and internal organs. We adapt this cushioning strategy

Clinical Insights Into Lower Incisor Extraction for Orthodontic Treatment: A Case Report.

Extracting a single lower front tooth can be an appropriate treatment for class I malocclusions when the alignment of the upper teeth is normal and there is an adequate overlap of the teeth. This approach is particularly effective in cases of significant crowding in the lower front teeth, especially when the space deficiency exceeds 4-5 mm and the combined width of the lower front teeth surpasses 83 mm. It is also a viable option for malocclusions resulting from discrepancies in tooth size, such as narrower upper front teeth or larger lower front teeth. Research suggests that this method leads

Clinical Challenge: Unusual Presentation of a Macular Hemorrhage Extending From the Internal Limiting Membrane to the Subretinal Space on Optical Coherence Tomography (OCT) in a Patient With Branch Retinal Vein Occlusion (BRVO).

Branch retinal vein occlusion (BRVO) is a common retinal vascular disorder typically characterized by distinct findings on fundus examination and optical coherence tomography (OCT). An unusual and challenging presentation involves a full-thickness macular hemorrhage extending from the internal limiting membrane (ILM) to the subretinal space. Here, we present the case of a 52-year-old male with type 2 diabetes mellitus who experienced sudden vision loss in the right eye and progressive vision decline in the left eye. Fundus examination revealed superotemporal BRVO in the right eye and old infer

The Effect of Low Temperatures on Environmental Radiation Damage in Living Systems: Does Hypothermia Show Promise for Space Travel?

Low-temperature treatments (i.e., hypothermia) may be one way of regulating environmental radiation damage in living systems. With this in mind, hibernation under hypothermic conditions has been proposed as a useful approach for long-term human space flight. However, the underlying mechanisms of hypothermia-induced radioresistance are as yet undetermined, and the conventional risk assessment of radiation exposure during hibernation remains insufficient for estimating the effects of chronic exposure to galactic cosmic rays (GCRs). To promote scientific discussions on the application of hibernat

Mounier-Kuhn Syndrome Mimicking Lymphangioleiomyomatosis.

We present the case of a man with Mounier-Kuhn syndrome (MKS), or tracheobronchomegaly, who was referred to the National Institutes of Health Clinical Research Center because of a potential diagnosis of lymphangioleiomyomatosis (LAM), a rare condition in men. The patient was evaluated using ongoing protocols and provided written informed consent. The case demonstrates the presence of chronic inflammation surrounding the dilated airways and histologic changes of the lung parenchyma with emphysematouslike disruption in areas adjacent to the dilated airways. This finding suggests that damage to t

Space motion sickness and motion sickness: symptoms and etiology.

The adverse symptoms of space motion sickness (SMS) have remained problematic since the beginning of manned spaceflight. Despite over 50 yr of research SMS remains a problem that affects about half of all space travelers during the first 24-72 h of a spaceflight. SMS has been treated as another form of motion sickness (MS) despite distinct differences in symptomology. In this review SMS and MS differences are examined and documented based on available data. Vestibular biomechanics that occur during weightlessness coupled with theoretical assertions regarding human evolution have led us to prop

Development of a rabbit pleural cancer model by using VX2 tumors.

Primary and secondary pleural cancer remains an important clinical problem, with research progress limited by the lack of a suitable moderate- to large-sized (3 to 4 kg) animal model of pleural cancer. Many potential pleura-based imaging and treatment modalities cannot be investigated sufficiently by using currently available small murine animal models because their pleural space is not comparable to that of humans and therefore does not allow for the use of standard thoracoscopic techniques. Here we describe the development of a reproducible model of pleural malignancy in moderate-sized immun

Microcapsule-gel formulation of promethazine HCl for controlled nasal delivery: a motion sickness medication.

The current method of choice for astronauts to treat space motion sickness is an intra-muscular injection of promethazine hydrochloride (PMZ HCl) which is invasive and causes considerable local irritation and discomfort at the site of injection. Intra-nasal delivery is considered a feasible alternative route for administration of medications to treat space motion sickness. The purpose of this research is to develop a PMZ HCl formulation that can be administered intra-nasally without irritation (i.e. leukocyte infiltration) in the nasal epithelium when dosed at PMZ HCl concentrations greater th

Preflight adaptation training for spatial orientation and space motion sickness.

Two part-task preflight adaptation trainers (PATs) are being developed at the NASA Johnson Space Center to preadapt astronauts to novel sensory stimulus conditions similar to those present in microgravity to facilitate adaptation to microgravity and readaptation to Earth. This activity is a major component of a general effort to develop countermeasures aimed at minimizing sensory and sensorimotor disturbances and Space Motion Sickness (SMS) associated with adaptation to microgravity and readaptation to Earth. Design principles for the development of the two trainers are discussed, along with a

Is this scapholunate joint and its ligament abnormal?

There is confusion in the literature with respect to evaluation of the scapholunate joint space and ligament. Because routine x-ray films of the wrist commonly do not profile the scapholunate joint perfectly, determination of the joint space width often is inaccurate. One method that invariably will allow a good profile of the scapholunate joint involves the use of fluoroscopically controlled views in different positions of the wrist. However, when abnormal width is suspected, in all examinations the wrist in question should be compared with the opposite wrist and the findings carefully correl

Brain and aviation: on the 80th anniversary of Constantin von Economo's (1876-1931) death.

2011 marks the 80th anniversary of the death of Constantin Alexander von Economo who conducted advanced research on the cytoarchitectonics of the brain. This Austrian neurologist and the pioneer of aviation described encephalitis lethargica, discovered the spindle neurons, and postulated the existence of the sleep and wakefulness centre in the brain. What is more he realized two of the biggest dreams of humankind: conquering space and getting to know the secrets of the human brain.

Reciprocal Quantum Electrodynamics With Bound States in the Continuum.

Quantum electrodynamics (QED) accurately describes all known forms of modern optics and photonics regarding interactions between photons and matter. Although matter ranges widely from atoms and particles to solids, photons are predominantly confined within a physical space, such as between a pair of mirrors, for enhanced photon-matter interactions known as cavity QED. Because position and momentum are canonically conjugate variables governed by Heisenberg's uncertainty principle, a fundamental question arises-what if light confinement is in the not-so-intuitive momentum or reciprocal space? Th

Study of stimulation mechanism of hydraulic fracturing induced thermal shock on joint system of geothermal reservoirs.

Hydraulic fracturing has been prevailingly applied to induce thermal shock in geothermal reservoirs for the utilization of geothermal energy. With natural joints commonly distribute in the reservoirs, the stimulation mechanism of thermal shock on joint system is inevitably investigated at fracturing initiation stage to enlarge heat transfer space. In this paper, the interaction of hydraulic fractures (HFs) and joints was simulated by lattice-spring method (LSM). The factors of fluid viscosities, shock times, approaching angles, and joint sets were analyzed to reveal the stimulation effect of t

0.33g mitigates muscle atrophy while 0.67g preserves muscle function and myofiber type composition in mice during spaceflight.

As human space exploration advances, understanding how different gravity levels affect skeletal muscle is critical for long-term health. Among the major organ systems, skeletal muscle is particularly sensitive to gravitational unloading, yet the gravity threshold required to maintain homeostasis remains unclear. Using the Multiple Artificial-gravity Research System aboard the International Space Station, mice were exposed to graded gravity levels, microgravity, 0.33g, 0.67g, and 1g, and their muscles were analyzed postflight. In the gravity-sensitive soleus, the cross-sectional area was preser

A call for neurovascular monitoring in an era of longer missions and broader spaceflight participation. Commentary.

Spaceflight-Associated Neuro-ocular Syndrome (SANS) has emerged as a critical neuro-ophthalmic risk for human space exploration, particularly as mission duration increases and access to space expands. Current spaceflight ocular surveillance and research protocols have prioritized structural imaging and selected neuroimaging/physiological assessments. However, accumulating evidence suggests that SANS is not confined to the posterior pole as a purely structural optic nerve head phenomenon but may also involve vascular and hemodynamic alterations. At the same time, structural changes at the optic
