First cases of Angiostrongylus cantonensis infection reported in Martinique, 2002-2017.

Neuroangiostrongyliasis is a parasitic disease caused by the accidental ingestion of the nematode Angiostrongylus cantonensis in its larval form. Human infection can lead to eosinophilic meningitis, sometimes complicated by life-threatening radiculomyelitis or encephalitis. Although some cases have been reported from other Caribbean Islands, no cases have been diagnosed in Martinique so far. Here, we report the first eight laboratory-confirmed cases of neuroangiostrongyliasis on the island of Martinique, French West Indies, between 1 January 2002 and 31 December 2017. One case was fatal and fi

Toxoplasma Co-infection Prevents Th2 Differentiation and Leads to a Helminth-Specific Th1 Response.

Nematode infections, in particular gastrointestinal nematodes, are widespread and co-infections with other parasites and pathogens are frequently encountered in humans and animals. To decipher the immunological effects of a widespread protozoan infection on the anti-helminth immune response we studied a co-infection with the enteric nematode Heligmosomoides polygyrus in mice previously infected with Toxoplasma gondii. Protective immune responses against nematodes are dependent on parasite-specific Th2 responses associated with IL-4, IL-5, IL-13, IgE, and IgG1 antibodies. In contrast, Toxoplasm

Occurrence and zoonotic potential of endoparasites in cats of Cyprus and a new distribution area for Troglostrongylus brevior.

The present study investigated for the first time the occurrence of pulmonary and intestinal parasites of cats in Cyprus. Cats from five districts of Cyprus (Lefkosia, Lemesos, Larnaka, Pafos and Ammochostos) were examined by classical parasitological methods and the identity of lungworm larvae, whenever present, was confirmed by PCR-coupled sequencing. A total of 185 cats, 48 living exclusively indoors and 137 with outdoor access, were included in the study. Parasites were found in 66 (35.7%) of the examined cats, i.e. Toxocara cati (12%), Cystoisospora rivolta (12%), Joyeuxiella/Diplopylidiu

Reaction norms of host immunity, host fitness and parasite performance in a mouse--intestinal nematode interaction.

The outcome of the encounter between a host and a parasite depends on the synergistic effects of the genetics of the two partners and the environment (sensulato) where the interaction takes place. Reaction norms can depict how host and parasite traits vary across environmental ranges for different genotypes. Here, we performed a large scale experiment where three strains of laboratory mice (SJL, BALB/c and CBA) were infected with four doses of the intestinal nematode Heligmosomoides polygyrus. An increasing infective dose can be considered as a proxy for the environment-dependent risk incontra

Protein deficiency alters impact of intestinal nematode infection on intestinal, visceral and lymphoid organ histopathology in lactating mice.

Protein deficiency impairs local and systemic immune responses to Heligmosomoides bakeri infection but little is known about their individual and interactive impacts on tissue architecture of maternal lymphoid (thymus, spleen) and visceral (small intestine, kidney, liver, pancreas) organs during the demanding period of lactation. Using a 2 × 2 factorial design, pregnant CD1 mice were fed a 24% protein sufficient (PS) or a 6% protein deficient (PD) isoenergetic diet beginning on day 14 of pregnancy and were infected with 100 H. bakeri larvae four times or exposed to four sham infections. On day

The spatial ecology of free-ranging domestic pigs (Sus scrofa) in western Kenya.

BACKGROUND: In many parts of the developing world, pigs are kept under low-input systems where they roam freely to scavenge food. These systems allow poor farmers the opportunity to enter into livestock keeping without large capital investments. This, combined with a growing demand for pork, especially in urban areas, has led to an increase in the number of small-holder farmers keeping free range pigs as a commercial enterprise. Despite the benefits which pig production can bring to a household, keeping pigs under a free range system increases the risk of the pig acquiring diseases, either pro

Eosinophilic meningitis due to Angiostrongylus cantonensis in Switzerland.

Eosinophilic meningitis is a well-known disease in areas where the rat lung worm Angiostrongylus cantonensis is endemic, i.e. in Southeast Asia and in the Caribbean. We report a case of eosinophilic meningitis due to infection with Angiostrongylus cantonensis in a traveler returning from Cuba. The diagnosis was based upon symptoms of chronic meningitis associated with hyperesthesia and presence of eosinophilia in the cerebrospinal fluid. With increasing mobility rare infections like eosinophilic meningitis due to A. cantonensis may be encountered in the western hemisphere. A Giemsa stain of th

Detection of circulating antigens of Parastrongylus cantonensis in human sera by sandwich ELISA with specific monoclonal antibody.

A specific monoclonal antibody (AW-3C2) as revealed by ELISA was produced against the adult worm antigens of Parastrongylus cantonensis and used in a sandwich ELISA for the detection of circulating antigens in the sera of parastrongyliasis patients and those with other parasitic diseases. A total of 60 sera was used in this study. Of these, 10 each were from patients with parastrongyliasis, cysticercosis, filariasis, gnathostomiasis, malaria and toxocariasis. The control group consisted of 53 serum samples from normal healthy Thais and Malaysians. The mean +/- optical density (OD) values for t

Prevalence of Angiostrongylus vasorum in southern Belgium, a coprological and serological survey.

BACKGROUND: Canine angiostrongylosis, a gastropod-borne helminthic infection, is increasingly being described in North America and is now reported in many European countries. In dogs, Angiostrongylus vasorum may cause a wide spectrum of clinical signs. Respiratory distress such as coughing and dyspnoea are the most frequently described manifestations. The aim of the present study was to gain additional information on the distribution, prevalence and risk factors associated with A. vasorum infection in dog from southern Belgium through the combined used of a commercially available in-clinic ass

Dynamics of Parascaris and Strongylus spp. parasites in untreated juvenile horses.

Parasite control in foals is of utmost importance due to the high susceptibility to parasitic infection and disease in this age group. Foals are commonly co-infected with strongyle and ascarid parasites, which complicate parasite control strategies. The present study retrospectively investigated necropsy records of foals born into a university herd kept without anthelmintic treatment since 1979. The aims were to statistically analyze the relationship between fecal egg counts, worm burdens, foal age, sex, and season with specific focus on Parascaris and Strongylus spp. A total of 83 foals born

Effects of albendazole combined with TSII-A (a Chinese herb compound) on optic neuritis caused by Angiostrongylus cantonensis in BALB/c mice.

BACKGROUND: Angiostrongylus cantonensis (A. cantonensis) infection can lead to optic neuritis, retinal inflammation, damage to ganglion cells, demyelination of optic nerve and visual impairment. Combined therapy of albendazole and dexamethasone is a common treatment for the disease in the clinic, but it plays no role in vision recovery. Therefore, it has been necessary to explore alternative therapies to treat this disease. Previous studies reported the neuro-productive effects of two constituents of Danshen (a Chinese herb)-tanshinone II-A (TSII-A) and cryptotanshinone (CPT), and this study a

Detection of gastrointestinal nematodes by a coproantigen capture ELISA.

An assay is described for the quantitative detection of excretory/secretory antigens liberated by the intestinal nematode Heligmosomoides polygyrus and voided in the host's faeces. A rabbit polyclonal antiserum to the antigens detected them in a dose-dependent manner and was sufficiently sensitive to recognise infection in mice carrying a mean worm burden of nine worms. The assay was specific, giving higher optical density readings with the faeces of mice infected with H polygyrus than with faeces from mice carrying patent infections with Trichuris muris, Hymenolepsis microstoma or the intesti

IL-33-induced ILC2 effector cytokine responses promote the expansion of red pulp macrophages.

Red pulp macrophages (RPMs) remove senescent erythrocytes from the circulation and recycle their iron for erythropoiesis. The development of RPMs is guided by signals from the microenvironment, which promote expansion and tissue adaptation through the induction of transcription factors, including Spi-C and PPARγ. Here, we show that infection with the nematode Nippostrongylus brasiliensis or treatment with IL-33 results in the accumulation of activated group 2 innate lymphocytes (ILC2s) in the spleen. ILC2s activated by IL-33 drive the expansion of RPMs by secretion of the effector cytokines IL

Helminth exposure influences Th17 plasticity, suppressing inflammatory and promoting regulatory activity by Th17 lineage cells.

INTRODUCTION: Many autoimmune and inflammatory-mediated diseases are driven by pathogenic Th17 responses. Infection with parasitic worms (helminths) alters host immune responses, suppresses Th17 activity and can inhibit pathogenic inflammation. Instead of being terminally differentiated, Th17 cells are plastic and can assume highly pathogenic Th1-like function or more regulatory Tr1/Treg-like function. We investigated if helminth infection influences this Th17 plasticity. METHODS: Lymphocytes from Th17-reporter mice permanently express eYFP if they previously transcribed IL-17. Using these mi

Human Neuroangiostrongyliasis Caused by Angiostrongylus cantonensis in Northern Vietnam: a Clinical-Epidemiological Case Series of Eosinophilic Response and Disease Manifestations.

PURPOSE: Angiostrongylus cantonensis is a major cause of eosinophilic meningitis in Southeast Asia, but clinical data from northern Vietnam remain limited. We describe the clinical spectrum, eosinophilic response and outcomes of patients treated at a national tertiary referral centre. METHODS: We retrospectively reviewed 30 consecutive patients admitted to Bach Mai Hospital, Hanoi, between January 2018 and December 2021 who met a composite case definition of probable neuroangiostrongyliasis: clinical meningitis, peripheral and/or cerebrospinal-fluid (CSF) eosinophilia, and a reactive serum EL

Aging impairs type 2 immune responses to nematodes associated with reduced gut microbiota responsiveness.

Gastrointestinal nematode infections elicit robust type 2 immune responses that facilitate rapid parasite expulsion. Our previous studies demonstrated that 18-month-old mice exhibit both impaired nematode clearance and reduced type 2-cytokine production, suggesting that aging diminishes the host's potential to mount effective immune defenses. To further investigate the underlying mechanisms, we compared young (3 months old) and aged mice (18 months old) infected with the nematode Heligmosomoides polygyrus (Hp), focusing on the interplay between type 2 immune responses and intestinal ecology. First Molecular Identification and Clinical Presentation of Crenosomosis in a Dog from Slovakia.

PURPOSE: Crenosoma vulpis (Dujardin,1845) is a lungworm which has spread worldwide in canines and is associated with upper respiratory infections. In a majority of cases, the infections are accompanied with chronic cough. Diagnosis of lungworms is often underdiagnosed and can be misinterpreted as other respiratory diseases. METHODS: The Small Animal Clinic of the University Veterinary Hospital admitted an 11-month-old dog presented with persistent cough associated with difficulty in breathing and even asphyxia. Based on clinical symptoms, the patient underwent radiological and bronchoscopic e

Identification and coregulation pattern analysis of long noncoding RNAs in the mouse brain after Angiostrongylus cantonensis infection.

BACKGROUND: Angiostrongyliasis is a highly dangerous infectious disease. Angiostrongylus cantonensis larvae migrate to the mouse brain and cause symptoms, such as brain swelling and bleeding. Noncoding RNAs (ncRNAs) are novel targets for the control of parasitic infections. However, the role of these molecules in A. cantonensis infection has not been fully clarified. METHODS: In total, 32 BALB/c mice were randomly divided into four groups, and the infection groups were inoculated with 40 A. cantonensis larvae by gavage. Hematoxylin and eosin (H&E) staining and RNA library construction were pe

Real-time qPCR coupled with high-resolution melting curve analysis for the detection of the internal transcribed spacer 1 of Angiostrongylus costaricensis.

Abdominal angiostrongyliasis (AA) is a zoonotic and severe parasitic infection caused by Angiostrongylus costaricensis. AA is currently diagnosed by the observation of A. costaricensis-compatible structures in biopsies or the detection of antibodies in serological tests. However, molecular methods targeting homologous sequences of A. costaricensis have not been designed before, and therefore, an HRM-coupled qPCR was developed to detect the internal transcribed spacer 1 (ITS1) of the parasite. The present assay successfully amplified DNA of A. costaricensis obtained from different hosts and ide
