Assessment of three large-scale depopulation methods for swine.

The threat of foreign animal disease outbreaks to U.S. swine herds warrants effective and readily available depopulation methods. Current American Veterinary Medical Association-recommendations using preferred physical methods for swine depopulation are unsuitable for large commercial swine herds. Our objectives were to assess and compare the efficacy and performance of three suggested large-scale depopulation methods: 1) medium-expansion water-based foam, 2) prototype high-expansion nitrogen foam and, 3) carbon dioxide gas for finisher pigs under field conditions. Out of 793 finisher pigs inc…

Potential pandemic risk of circulating swine H1N2 influenza viruses.

Influenza A viruses in swine have considerable genetic diversity and continue to pose a pandemic threat to humans due to a potential lack of population level immunity. Here we describe a pipeline to characterize and triage influenza viruses for their pandemic risk and examine the pandemic potential of two widespread swine origin viruses. Our analysis reveals that a panel of human sera collected from healthy adults in 2020 has no cross-reactive neutralizing antibodies against a α-H1 clade strain (α-swH1N2) but do against a γ-H1 clade strain. The α-swH1N2 virus replicates efficiently in human ai…

Suspected aerosol transmission of swine pathogens: A field case.

A swine production system had 3 sections located a few kilometers apart. Sections A and C contained several thousand sows and nursery and finishing pigs. Section B, located between the other 2 sections, was the smallest and had 6 finishing sites and 2 sow sites. The entire system was infected with porcine reproductive and respiratory syndrome virus, Mycoplasma hyopneumoniae, and Actinobacillus pleuropneumoniae. Section B was depopulated, cleaned, disinfected, and repopulated with negative gilts. Despite extreme measures, recontamination occurred for each pathogen, with aerosol considered the m…

Free PEG Suppresses Anaphylaxis to PEGylated Nanomedicine in Swine.

Covalent conjugation of poly(ethylene glycol) (PEG) is frequently employed to enhance the pharmacokinetics and biodistribution of various pr … ness of free PEG as a prophylaxis against anaphylaxis induced by PEG-specific allergic reactions in swine. Injection of PEG-liposomes (PL) resulted in anaphylactoid shock (pseudoanaphylaxis) within 1-3 min in both naïve and PL-sensitized swine. In contrast, repeated injection of free PEG alone did not result in allergic reactions, and injection of free …

Improving African Swine Fever Surveillance Using Fluorescent Rapid Tests.

African swine fever (ASF) is a viral disease of swine with a huge impact due to its high mortality. Lately, the disease has actively spread around the world, affecting new areas from which it had been eradicated long ago. To date, ASF control is carried out by the implementation of strict biosecurity measures such as the early identification of infected animals. In this work, two fluorescent rapid tests were developed to improve the sensitivity of point-of-care diagnosis of ASF. For antigen (Ag) detection in blood, a double-antibody sandwich fluorescent lateral flow assay (LFA) was developed, …

Swine enteric colibacillosis: Current treatment avenues and future directions.

Enteric colibacillosis is a common disease in nursing and weanling pigs. It is caused by the colonization of the small intestine by enterotoxigenic strains of Escherichia coli (ETEC) that make use of specific fimbria or pili to adhere to the absorptive epithelial cells of the jejunum and ileum. Once attached, and when both the immunological systems and the gut microbiota are poorly developed, ETEC produce one or more enterotoxins that can have local and, further on, systemic effects. These enterotoxins cause fluid and electrolytes to be secreted into the intestinal lumen of animals, which resu

Antiviral Drugs Screening for Swine Acute Diarrhea Syndrome Coronavirus.

Coronaviruses as possible cross-species viruses have caused several epidemics. The ongoing emergency of coronavirus disease 2019 (COVID-19) caused by SARS-CoV-2 has posed severe threats to the global economy and public health, which has generated great concerns about zoonotic viruses. Swine acute diarrhea syndrome coronavirus (SADS-CoV), an alpha-coronavirus, was responsible for mass piglet deaths, resulting in unprecedented economic losses, and no approved drugs or vaccines are currently available for SADS-CoV infection. Given its potential ability to cause cross-species infection, it is esse…

Toxoplasma gondii Infection in Swine: Implications for Public Health.

Toxoplasmosis, due to Toxoplasma gondii, is a parasitic disease with global importance. Among livestock, chronic T. gondii infection has been reported in higher rates in pigs and small ruminants, but with subclinical infections in case commonly encountered in pigs. Seroprevalence in the global pig population ranges according to the age or species of pigs, geographical distribution, production programs, and systems. Generally, T. gondii infections are noticed in low prevalence rates in conventional pig farms with high hygiene standards. In contrast, higher prevalence is common on free-ranging f

African swine fever vaccine: Turning a dream into reality.

African swine fever (ASF) is currently threatening the swine industry at a global level. The disease originated in Africa has spread to Europe, Asia and Oceania, since 2007, reaching a pandemic dimension. Currently, the spread of ASF is unstoppable and that the development of a safe and effective vaccine is urgently required. The objective of this paper is to review the vaccine candidates tested during the 20th and 21st centuries, to identify the strengths and weaknesses of these studies and to highlight what we should learn. Several strategies have been explored to date, some of which have sh…

African swine fever virus genotype II in Mongolia, 2019.

African swine fever (ASF) is a severe haemorrhagic disease of domestic and wild pigs caused by the African swine fever virus (ASFV). In recent years, ASF has steadily spread towards new geographical areas, reaching Europe and Asia. On January 15th, 2019, Mongolia reported its first ASF outbreak to the World Organization for Animal Health (OIE), becoming, after China, the second country in the region affected by the disease. Following an event of unusual mortality in domestic pigs in Bulgan Province, a field team visited four farms and a meat market in the region to conduct an outbreak investig…

Novel reassortant swine H3N2 influenza A viruses in Germany.

Analysis of 228 H3N2 swine influenza A virus isolates collected between 2003 and 2015 in Germany revealed important changes in molecular epidemiology. The data indicate that a novel reassortant, Rietberg/2014-like swine H3N2, emerged in February 2014 in Northern Germany. It is comprised of a hemagglutinin gene of seasonal H3N2 (A/Denmark/129/2005-like), a neuraminidase gene of Emmelsbuell/2009-like swine H1N2 and the internal gene cassette of pandemic H1N1 viruses. Together with Danish swine H3N2 strains of 2013-2015 with identical genome layout, the Rietberg/2014-like viruses represent a seco…

Copper Speciation Evolution in Swine Manure Induced by Pyrolysis.

Swine manures generally contain high levels of copper (Cu) resulting from its use as a growth promoter in feedstuff. Pyrolysis can further concentrate Cu whereas decrease its available fraction in swine manures. Here we investigated the speciation transformation of Cu and associated elements in swine manures induced by pyrolysis using multiple X-ray absorption spectroscopies. Results showed that over 82% of Cu existed as Cu(I)-S and Cu(I)-thiolate complexes in swine manures, which were transformed into stable Cu(I)2S during pyrolysis at a low temperature of 300 °C and partially oxidized and de…

Clonality of Mycoplasma hyopneumoniae in swine farms from Brazil.

Mycoplasma hyopneumoniae causes enzootic pneumonia (EP) in swine, a disease related to high economic losses in production systems. Epidemiological spread of M. hyopneumoniae clones was studied by multi-locus sequence typing (MLST) in several swine production regions but so far not in South America. Using MLST, we have therefore investigated M. hyopneumoniae clones circulating in farms from three main swine production regions in Brazil. Porcine lungs samples were collected between 2015 and 2016 in farms with EP outbreaks. Three geographically distant regions were selected, and 67 M. hyopneumoni…

Inhibition of African swine fever virus infection by genkwanin.

African swine fever virus (ASFV) is the causative agent of an economically important disease of pigs for which no effective vaccines or antiviral drugs are available. Recent outbreaks in EU countries and China have highlighted the critical role of antiviral research in combating this disease. We have previously shown that apigenin, a naturally occurring plant flavone, possesses significant anti-ASFV activity. However, apigenin is practically insoluble in highly polar solvents and it occurs typically in derivative forms in plants. Here we screened several commercially available apigenin derivat…

Identification and Immunogenicity of African Swine Fever Virus Antigens.

African swine fever (ASF) is a lethal haemorrhagic disease of domestic pigs for which there is no vaccine. Strains of the virus with reduced virulence can provide protection against related virulent strains of ASFV, but protection is not 100% and there are concerns about the safety profile of such viruses. However, they provide a useful tool for understanding the immune response to ASFV and previous studies using the low virulent isolate OUR T88/3 have shown that CD8+ cells are crucial for protection. In order to develop a vaccine that stimulates an effective anti-ASFV T-cell response we need …

Swine influenza viruses in Northern Vietnam in 2013-2014.

Swine are an important intermediate host for emergence of pandemic influenza. Vietnam is the largest swine producer in South East Asia. Systematic virological and serological surveillance of swine influenza viruses was carried out in Northern Vietnam from May 2013 to June 2014 with monthly sampling of pigs in local and large collective slaughterhouses and in a live pig market. Influenza A seroprevalence in the local slaughterhouses and in the large collective slaughterhouse was 48.7% and 29.1%, respectively. Seventy-seven influenza A viruses were isolated, all from the large collective slaught…

Altered splenic miRNA expression profile in H1N1 swine influenza.

Previous studies have demonstrated the key regulatory roles played by microRNAs (miRNAs) in influenza virus-host interactions. To gain more insight into the contribution of miRNAs to the host immune response, spleen tissues from mice infected with A/Swine/GD/2/12 (H1N1) virus were harvested 5 days postinfection, and miRNA deep sequencing was performed. The results showed that 50 miRNAs were modulated. Interestingly, pathway analysis of miRNAs and targets showed that upregulated miR-124-3p interacts with innate immune-related pathways such as the Toll-like receptor pathway, RIG-I-like receptor …
