Modulation of brain dopamine transmission by sex steroids.

Sex steroid hormones influence the dopaminergic systems of the hypothalamus as well as the extrahypothalamic regions of the brain in controlling movement and behavior in both humans and animals. This review focuses on the effects of sex steroids on dopaminergic activity in extrahypothalamic brain areas. Among sex steroids, estrogens have been most extensively investigated, and many studies report that estrogens affect behaviors mediated by the basal ganglia, such as in humans suffering from extrapyramidal disorders. Epidemiological and clinical evidence also suggests an influence of estrogens

Inferior olivary nuclear complex of the rat: morphology and comments on the principles of organization within the olivocerebellar system.

This study was undertaken (1) to provide a description of the normal morphology and anatomical interrelationships within the inferior olivary cell groups and (2) to determine the topographical organization of projections from this nuclear complex to the cerebellum. Conventional histological methods and the technique of retrograde transport of horseradish peroxidase (HRP) and WGA-HRP in conjunction with the sensitive chromogen tetramethyl benzidine were used in this series of experiments. In common with that of other animals, the inferior olivary nucleus of the rat consists of three subdivision

Synaptic high-frequency jumping synchronises vision to high-speed behaviour.

During high-speed behaviour, animals must synchronise perception and action despite rapid environmental and self-generated motion. How neural systems achieve such precision remains unclear. Here we show how the housefly (Musca domestica) maintains visual accuracy during fast motion. Using intracellular and photomechanical recordings during saccade-like stimulation, we traced information flow from photoreceptors to large monopolar cells (LMCs). Visual neurons achieved record-high information sampling (~2500 bits·s-1) and synaptic transmission (~4100 bits·s-1), far exceeding previous estimates. …

Synaptically silent sensory hair cells in zebrafish are recruited after damage.

Analysis of mechanotransduction among ensembles of sensory hair cells in vivo is challenging in many species. To overcome this challenge, we used optical indicators to investigate mechanotransduction among collections of hair cells in intact zebrafish. Our imaging reveals a previously undiscovered disconnect between hair-cell mechanosensation and synaptic transmission. We show that saturating mechanical stimuli able to open mechanically gated channels are unexpectedly insufficient to evoke vesicle fusion in the majority of hair cells. Although synaptically silent, latent hair cells can be rapi…

Structure-activity relationships of BnOCPA derivatives at native pre- and postsynaptic adenosine A1Rs.

The addition of a benzyloxy group to the adenosine A1R agonist cyclopentyladenosine (CPA) produces a compound (BnOCPA) that selectivity activates A1R signalling via the G protein alpha subunit Gob. In rat hippocampus, BnOCPA inhibits CA3-CA1 synaptic transmission (IC50 65 nM) likely via Gob, with no effect on Goa-mediated postsynaptic membrane electrophysiology at concentrations up to 1 μM. We have investigated how modifications of the benzyloxy group of BnOCPA affects signalling: addition of methoxy group, MBnOCPA, substituting a phenoxy group, PhOCPA; and addition of bromide to the phenoxy g…

GABAB receptor-mediated modulation in the developing dorsal nucleus of the lateral lemniscus.

The dorsal nucleus of the lateral lemniscus (DNLL) is a GABAergic, reciprocally connected auditory brainstem structure that continues to dev … 1, old gerbils GABAB receptor activation enhances glutamatergic and suppresses ionotropic GABAergic synaptic transmission. During development, this differential modulation becomes less distinct, because in P22-28, old animals Baclofen-activated GABAB receptors rather enhance ionotropic GABAergic synaptic transmission, whereas glutamatergic transmission is both enhanced …

BACE1 is necessary for experience-dependent homeostatic synaptic plasticity in visual cortex.

Alzheimer's disease (AD) is the most common form of age-related dementia, which is thought to result from overproduction and/or reduced clea … n and triggers AMPA receptor endocytosis to homeostatically downregulate the strength of excitatory synaptic transmission. We previously reported that Arc is critical for sensory experience-dependent homeostatic reduction of excitatory synaptic transmission in the superficial layers of visual cortex. Here we demonstrate that mice lacking the major neuronal β-secretase, …

Regulation of depotentiation and long-term potentiation in the dentate gyrus of freely moving rats by dopamine D2-like receptors.

Dopamine receptors are significantly involved in hippocampus-based cognitive processes. Whereas the involvement of D1-like receptors in hippocampal plasticity has been described, the role of D2-like receptors remains to be clarified. Therefore, we investigated the contribution of D2-like receptors to synaptic transmission, long-term potentiation (LTP) and depotentiation in the dentate gyrus of freely moving rats. Male Wistar rats underwent chronic implantation of a recording electrode in the granule cell layer, a stimulating electrode in the medial perforant path and a cannula in the ipsilater…

Deficits in synaptic function occur at medial perforant path-dentate granule cell synapses prior to Schaffer collateral-CA1 pyramidal cell synapses in the novel TgF344-Alzheimer's Disease Rat Model.

Alzheimer's disease (AD) pathology begins decades prior to onset of clinical symptoms, and the entorhinal cortex and hippocampus are among t … ale TgF344-AD rats and wildtype littermates, prior to reported behavioral deficits. Decreased basal synaptic transmission begins at medial perforant path-dentate granule cell (MPP-DGC) synapses prior to Schaffer-collateral-CA1 (CA3-CA1) synapses, in the absence of a change in paired-pulse ratio (PPR) or dendritic spine density. N-methyl-d-aspartate receptor (NMDAR)-dep …

Regulation of Nociceptive Glutamatergic Signaling by Presynaptic Kv3.4 Channels in the Rat Spinal Dorsal Horn.

Presynaptic voltage-gated K+ (Kv) channels in dorsal root ganglion (DRG) neurons are thought to regulate nociceptive synaptic transmission in the spinal dorsal horn. However, the Kv channel subtypes responsible for this critical role have not been identified. The Kv3.4 channel is particularly important because it is robustly expressed in DRG nociceptors, where it regulates action potential (AP) duration. Furthermore, Kv3.4 dysfunction is implicated in the pathophysiology of neuropathic pain in multiple pain models. We hypothesized that, through their ability to modulate AP repolarization, Kv3.…

Interaction between phorbol dibutyrate and anaesthetics on synaptic responses from olfactory cortex of rat.

Phorbol esters have been shown to enhance the release of transmitters and to potentiate the effect of local anaesthetics in olfactory cortex of the rat. This work examined the interaction between phorbol dibutyrate and a range of substances, which act on axonal conduction and synaptic transmission. Synaptically-evoked field responses were elicited by stimulation of the lateral olfactory tract of slices of olfactory cortex, maintained in vitro. Ketamine (0.2-1.0 mM), benzocaine (0.2-1 mM), atropine (0.1-2 mM) and tetrodotoxin (20-200 nM) depressed synaptic transmission and, in the presence of p…

Anaesthetics depress the sensitivity of cortical neurones to L-glutamate.

1 The effects of general anaesthetics on the responses of neurones to iontophoretically applied L-glutamate have been examined in slices of the guinea-pig olfactory cortex in vitro. 2 Concentrations of pentobarbitone, ether, methoxyflurance, trichloroethylene and alphaxalone that are known to depress synaptic transmission in the prepiriform cortex also depressed the sensitivity of prepiriform neurones to L-glutamate. 3 Halothane, in concentrations that depress synaptic transmission (less than 1%) did not alter sensitivity of neurones to glutamate. Higher concentrations (greater than 1% produce…

Early defects of GABAergic synapses in the brain stem of a MeCP2 mouse model of Rett syndrome.

Rett syndrome is a neurodevelopmental disorder caused by mutations in the transcriptional repressor methyl-CpG-binding protein 2 (MeCP2) and … disease. Here, we report that MeCP2 KO mice present an imbalance between inhibitory and excitatory synaptic transmission in the ventrolateral medulla already at postnatal day 7. Focusing on the inhibitory synaptic transmission we show that GABAergic, but not glycinergic, synaptic transmission is strongly depressed in MeCP2 KO mice. These alterations are presumably due …

Synaptic AMPA receptor exchange maintains bidirectional plasticity.

Activity-dependent synaptic delivery of GluR1-, GluR2L-, and GluR4-containing AMPA receptors (-Rs) and removal of GluR2-containing AMPA-Rs mediate synaptic potentiation and depression, respectively. The obvious puzzle is how synapses maintain the capacity for bidirectional plasticity if different AMPA-Rs are utilized for potentiation and depression. Here, we show that synaptic AMPA-R exchange is essential for maintaining the capacity for bidirectional plasticity. The exchange process consists of activity-independent synaptic removal of GluR1-, GluR2L-, or GluR4-containing AMPA-Rs and refilling

[Effects of acetylcholinesterase inhibitors on the process of reinnervation of the mouse skeletal muscle].

The effect of chronic in vivo injections of anticholinesterases drugs (phosphacol, proserine) on the reinnervation of m. extensor digitorum longus were studied in adult mice after motor nerve crushing. An electrophysiological technique was used to register synaptic transmission. The chronic anticholinesterases injections were shown not to change the time of initial neuro-muscular transmission reestablishment. The potentiating effects of investigated anticholinesterase agents were poorly manifested in the early period of transmission recovery. Later in the course of reinnervation the potentiati…

Angiotensin-Converting Enzyme 2 in the Rostral Ventrolateral Medulla Regulates Cholinergic Signaling and Cardiovascular and Sympathetic Responses in Hypertensive Rats.

The rostral ventrolateral medulla (RVLM) is a key region in cardiovascular regulation. It has been demonstrated that cholinergic synaptic transmission in the RVLM is enhanced in hypertensive rats. Angiotensin-converting enzyme 2 (ACE2) in the brain plays beneficial roles in cardiovascular function in hypertension. The purpose of this study was to determine the effect of ACE2 overexpression in the RVLM on cholinergic synaptic transmission in spontaneously hypertensive rats (SHRs). Four weeks after injecting lentiviral particles containing enhanced green fluorescent protein and ACE2 bilaterally …

A Key Role for Neurotensin in Chronic-Stress-Induced Anxiety-Like Behavior in Rats.

Chronic stress is a major cause of anxiety disorders that can be reliably modeled preclinically, providing insight into alternative therapeutic targets for this mental health illness. Neuropeptides have been targeted in the past to no avail possibly due to our lack of understanding of their role in pathological models. In this study we use a rat model of chronic stress-induced anxiety-like behaviors and hypothesized that neuropeptidergic modulation of synaptic transmission would be altered in the bed nucleus of the stria terminalis (BNST), a brain region suspected to contribute to anxiety diso…

The synaptic availability of GluA1 is reduced in hippocampal neurons of a murine model of dynamin-2 linked autosomal dominant centronuclear myopathy.

ObjectiveAutosomal dominant centronuclear myopathy (AD-CNM) is a neuromuscular congenital disease caused by mutations in the DNM2 gene that encodes dynamin-2 (DNM2). The main clinical features of AD-CNM are progressive weakness and atrophy of skeletal muscles. However, cognitive defects have also been reported, suggesting that AD-CNM-causing mutations in DNM2 might also affect central nervous system (CNS). We recently demonstrated that defects in excitatory synaptic transmission occur in the brain of transgenic knock-in (KI) mice harboring the DNM2 p.R465W mutation, the most common causing AD-…
