Condensate-driven triglyceride reduction links α-synuclein to mitochondrial dysfunction.

α-Synuclein (αSyn) inclusions characterize multiple age-related neurodegenerative diseases, including Parkinson's disease (PD). While interactions between αSyn and lipids are known to contribute to αSyn pathobiology, the precise cellular mechanisms linking lipids to αSyn toxicity have yet to be elucidated. Through lipidomic profiling of Caenorhabditis elegans, we find that αSyn progressively alters lipid metabolism in aging worms. αSyn reduces the overall content of triacylglycerols (TAG) and disrupts the structure of lipid droplets (LD) and mitochondria. These pathological changes depend on α

N-terminal acetylation reduces synuclein pathology in models of Parkinson's disease.

α-synuclein protein is a major constituent of pathological intracellular inclusions such as Lewy bodies found in brains of patients with Parkinson's disease and other synucleinopathies. Whereas α-synuclein phosphorylation has been much studied, comparatively less work has been devoted to other post-translational modifications such as acetylation, especially given that N-terminally acetylated α-synuclein is the most abundant endogenous form of the protein in the brain. In this study, using multiple in vitro and in vivo models of Parkinson's disease, we sought to understand the pathological cons

Clearance and secretion of α-synuclein by RTN3L-mediated endoplasmic reticulum autophagy.

The misfolding and aggregation of α-synuclein (α-syn), an abundant synaptic protein, leads to the pathogenesis of Parkinson's disease and related synucleinopathies. The cell-to-cell propagation of seeding-competent α-syn is initiated by unconventional protein secretion, yet the physiological pathway(s) underlying this process remain poorly defined. Here we show that α-syn secretion in human cells is mediated by Reticulon-3L (RTN3L)-dependent endoplasmic reticulum autophagy (ER-phagy), a conserved protein quality-control pathway that safeguards ER protein homeostasis. We also demonstrate that R

Systematic review of alpha-synuclein seed amplification assays for the detection of synucleinopathies.

Parkinson's disease and other synucleinopathies are neurodegenerative disorders defined by the pathological aggregation of alpha-synuclein (α-syn). The alpha-synuclein Seed Amplification Assay (α-syn SAA), also known as real-time quaking-induced conversion (RT-QuIC), has gained recognition as a high-sensitivity, high-specificity diagnostic tool capable of detecting misfolded α-syn seeds in biological samples through cyclic amplification and thioflavin T-based fluorescence readout. Despite its diagnostic promise, the lack of standardized protocols across laboratories remains a critical obstacle

TLR2 Regulates α-Synuclein Phosphorylation and Release in an Enteroendocrine Cell Model.

Parkinson's disease (PD) is characterized by the progressive accumulation and prion-like spread of pathological α-synuclein (α-syn). Increasing evidence suggests that α-syn pathology may originate in the gastrointestinal tract before spreading to the central nervous system; however, the intestinal mechanisms that initiate pathological α-syn phosphorylation and intercellular transfer remain poorly understood. Here, we investigated the role of Toll-like receptor 2 (TLR2) in regulating α-syn phosphorylation using intestinal epithelial cell model. We found that pSer129-positive α-syn immunoreactiv

Aβ42-Driven α-synuclein Fibril Polymorphism and Distinct Intracellular Aggregation.

The frequent coexistence of α-synuclein (α-syn) and amyloid-β (Aβ) aggregates in neurodegenerative diseases suggests that heterotypic interactions between these amyloidogenic proteins may influence disease progression, yet their molecular consequences remain poorly understood. Here, we investigated how distinct aggregation states of Aβ42, monomers and preformed fibrils (PFFs), modulate α-syn fibril formation, structure, and downstream neuronal pathology. Thioflavin T kinetics showed that Aβ42 monomers delayed α-syn fibril formation, whereas Aβ42 PFFs exhibited a trend toward accelerated aggreg

Cholesterol dysregulation in APOE4 astrocytes promotes α-synuclein pathology in miBrains.

The pathological hallmarks of neurodegeneration are the aberrant post-translational modification and aggregation of proteins. Genetic factors, like APOE4, increase the prevalence and severity of tau, amyloid, and α-synuclein pathologies. However, the human brain is largely inaccessible during this process, limiting mechanistic understanding. Here, we developed an iPSC-based 3D model that integrates neurons, glia, myelin, and cerebrovascular cells into a human brain-like tissue ("miBrain"). Single-nucleus RNA sequencing of miBrains confirmed the presence of diverse cell populations and revealed

Glial control of extracellular α-synuclein propagation in LRRK2 Parkinson's disease.

Parkinson's disease (PD) pathology extends well beyond dopaminergic neuronal loss, with glial cells-microglia and astrocytes-emerging as active architects of α-synuclein spread rather than passive bystanders. This review synthesises current evidence on how mutations in Leucine-Rich Repeat Kinase 2 (LRRK2), the most common genetic cause of familial PD, fundamentally corrupt glial handling of extracellular α-synuclein. We first outline the biology of extracellular α-synuclein-its cellular sources, conformational spectrum, prion-like propagation mechanisms, and principal glial clearance routes-be

Lipid Packing Defects are Necessary and Sufficient for Membrane Binding of α-Synuclein.

α-Synuclein (αSyn), an intrinsically disordered protein implicated in Parkinson's disease, is thought to initiate aggregation by binding to cellular membranes. Previous studies suggest that anionic lipids are necessary for this binding. However, these studies largely focused on unmodified αSyn, while physiological αSyn is N-terminally acetylated (NTA). Our work challenges the long-standing paradigm that anionic lipids are necessary for αSyn binding by demonstrating that NTA diminishes αSyn's reliance on anionic membrane charge, revealing that membrane packing defects (i.e., interfacial hydroph

Aggregation of α-synuclein splice isoforms through a phase separation pathway.

The aggregation of α-synuclein (αSyn) is associated with Parkinson's disease and other related synucleinopathies. Considerable efforts have thus been directed at understanding this process. However, the recently discovered condensation pathway, which involves the formation of phase-separated liquid intermediate states, has added further complexity. In parallel, it has been reported that different αSyn splice isoforms may be implicated in aggregate formation in disease. In this study, we compare the phase behavior of four αSyn isoforms (αSyn-140, αSyn-126, αSyn-112, and αSyn-98). Using differen

The interplay of iron, oxidative stress, and α-synuclein in Parkinson's disease progression.

The irreversible degeneration of dopamine neurons induced by α-synuclein (α-syn) aggregation in the substantia nigra is the central pathological feature of Parkinson's disease (PD). Neuroimaging and pathological autopsy studies consistently confirm significant iron accumulation in the brain of PD patients, suggesting a critical role for iron in disease progression. Current research has established that iron overload induces ferroptosis in dopaminergic neurons, evidence indicates that the impact of iron on PD pathology extends beyond ferroptosis. Iron also plays a regulatory role in modulating

Criterion for Assessing Accumulated Neurotoxicity of Alpha-Synuclein Oligomers in Parkinson's Disease.

The paper introduces a parameter called "accumulated neurotoxicity" of α-syn oligomers, which measures the cumulative damage these toxic species inflict on neurons over time, given the years it typically takes for such damage to manifest. A threshold value for accumulated neurotoxicity is estimated, beyond which neuron death is likely. Numerical results suggest that rapid deposition of α-syn oligomers into fibrils minimizes neurotoxicity, indicating that the formation of Lewy bodies might play a neuroprotective role. Strategies such as reducing α-syn monomer production or enhancing degradation

Imbalanced mitochondrial dynamics in human PD and α-synuclein mouse brains.

Emerging studies have shown that dysregulation in mitochondrial dynamics has a major negative impact on mitochondria. Partial genetic and pharmacological inhibition of the mitochondrial fission dynamin-related protein 1 (DRP1) has been demonstrated to be beneficial in models of neurodegenerative disorders, including Parkinson's disease (PD). However, the expression of DRP1 and other mitochondrial fission/fusion mediators have not been investigated in the brains of Parkinson's patients. This information is critical to strengthening mitochondrial dynamics as a potential therapeutic target for PD

Cerebrospinal Fluid Phosphorylated Alpha-Synuclein in Newly Diagnosed Parkinson's Disease.

BACKGROUND: Alpha-synuclein (α-syn), phosphorylated at serine 129 (pS129-α-syn), is a potential biomarker for Parkinson's disease (PD) because it is the predominant α-syn species found in Lewy bodies. METHODS: We developed an in-house SIMOA assay, using commercially available components, to quantify pS129-α-syn in CSF. The clinical utility of the assay was tested in CSF from 120 patients with PD from the Norwegian ParkWest longitudinal study and 29 normal controls. Prior measurements of CSF total (t)-α-syn and the pS129-α-syn/t-α-syn ratio were included for comparison. RESULTS: The lower lim

[Research progress on the effect of α-synuclein in acupuncture treatment for Parkinson's disease].

Parkinson's disease (PD) is a chronic progressive neurological degenerative disease caused by the degeneration of dopaminergic neurons in the substantia nigra. α-synuclein (α-Syn) misfolding and aggregation is the crucial pathogenesis of PD, and is closely related to the other pathogenesis, such as brain-gut axis dysfunction, mitochondrial dysfunction, oxidative stress, neuroinflammation, iron and lipid metabolic disorders, and autophagy lysosomal dysfunction. Acupuncture plays a neuroprotective role by attenuating neuroinflammation, regulating brain-gut axis, repairing ubiquitin-proteasome sy

Secretory autophagy mediates lysosomal and autophagic degradation for α-synuclein proteostasis.

Autophagy has two distinct pathways, degradation and secretion. Autophagic degradation plays a pivotal role in proteostasis. However, the role of autophagic secretion in proteostasis maintenance is not fully understood. Here, we investigate how the blockade of autophagic secretion impairs proteostasis in SH-SY5Y cells. siRNA-mediated knockdown (KD) of a modulator for autophagosome formation, ATG5, BECN1, or FIP200 inhibited autophagic flux and secretion, causing accumulation of Triton X-100-insoluble α-synuclein (α-syn), which is an aggregate-prone protein responsible for neuronal loss in Park

Differentiating α-synuclein aggregates using charge-sensitive gold nanoclusters.

The accumulation of α-synuclein (α-syn) aggregates in the brain is associated with Parkinson's disease, making it a promising biomarker for understanding the disease's pathogenesis. Distinguishing between oligomeric forms of amyloidogenic proteins is crucial, as their toxicity depends on conformation. This study leverages surface charge differences to distinguish α-syn monomers from amyloids. We employed label-free gold nanoclusters (AuNCs) with distinct surface charges to differentiate between wild-type and mutant [A30P]-α-Syn based on their charge characteristics. Fluorescence spectroscopy a

BACKGROUND: Parkinson's disease (PD) is a complex multifactorial disorder with a genetic component in about 15% of cases. Multiplications and point mutations in SNCA gene, encoding α-synuclein (aSyn), are linked to rare familial forms of PD. OBJECTIVE: Our goal was to assess the clinical presentation and the biological effects of a novel K58N aSyn mutation identified in a patient with PD. METHODS: We describe the clinical presentation associated with the novel mutation, together with genetic testing through whole exome sequencing (WES). Furthermore, we conducted extensive biophysical and cel

ATP Hydrolysis by α-Synuclein Amyloids is Mediated by Enclosing β-Strand.

Pathological amyloids, like those formed by α-synuclein in Parkinson's disease, are recently found to catalyze the hydrolysis of model substrates in vitro. Here it is reported that the universal energy molecule ATP is another substrate for α-synuclein amyloid chemical catalysis. To reveal the underlying mechanism, the high-resolution cryo-EM structure of the amyloids in the presence of ATP is solved. The structure reveals a type 1A amyloid fold with an additional β-strand involving residues 16-22 that wraps around the ATP, creating an enclosed cavity at the interface of the protofilaments. Mut

Chromogranin A promotes the pathological conversion of α-synuclein at the synapse in Parkinson's disease.

α-Synuclein (αSyn) aggregation is a prominent hallmark of Parkinson's disease (PD), yet the initial cellular mechanisms are not well understood. In this study, we show that a single day of αSyn preformed fibril (PFF) administration leads to prominent localization of phosphorylated αSyn (p-αSyn) within the pre-synapse of primary neurons. Overexpressing chromogranin A (CgA), which is found in large dense-core vesicles (LDCVs), enhances αSyn aggregation in various neuronal and PD mouse models. Subsequently, by incubating αSyn with CgA, we create a unique strain of fibrils displaying smaller lengt
