Cdc42/N-WASP signaling links actin dynamics to pancreatic β cell delamination and differentiation.

Delamination plays a pivotal role during normal development and cancer. Previous work has demonstrated that delamination and epithelial cell movement within the plane of an epithelium are associated with a change in cellular phenotype. However, how this positional change is linked to differentiation remains unknown. Using the developing mouse pancreas as a model system, we show that β cell delamination and differentiation are two independent events, which are controlled by Cdc42/N-WASP signaling. Specifically, we show that expression of constitutively active Cdc42 in β cells inhibits β cell de

Quantitative analysis of cell migration using optical flow.

Neural crest cells exhibit dramatic migration behaviors as they populate their distant targets. Using a line of zebrafish expressing green fluorescent protein (sox10:EGFP) in neural crest cells we developed an assay to analyze and quantify cell migration as a population, and use it here to characterize in detail the subtle defects in cell migration caused by ethanol exposure during early development. The challenge was to quantify changes in the in vivo migration of all Sox10:EGFP expressing cells in the visual field of time-lapse movies. To perform this analysis we used an Optical Flow algorit

Dynamic morphological staging of Drosophila pupal development.

This study provides a comprehensive analysis of the morphological-developmental characteristics of the pupal stage in Drosophila, a model organism for metamorphosis research. We have delineated the critical stages of pupal development, from the late third-instar larvae to adult emergence, through continuous photography and time-lapse imaging. Our findings include the identification of the head-movement phase and head-static phase as distinct transitional states preceding the pupal stage. We document pupal shell dynamics, including color transitions and spiracle development as staging markers,

Impact of Contrast Media on Cytological Evaluation of Urothelial Carcinoma: An In vitro Study with Relevance to Upper Tract Diagnostics.

BACKGROUND: Urinary cytology plays a pivotal role in the non-invasive diagnosis of upper tract urothelial carcinoma (UTUC), especially where histological sampling is limited or technically challenging. The second edition of the Paris System for Reporting Urinary Cytology standardizes for evaluating cytological specimens. However, in clinical practice, urinary samples obtained from the UT often contain residual contrast media (CM) due to prior imaging studies; hence, the effect of CM on urinary cytology accuracy remains unclear. AIMS: The aim of this study was to examine the impact of CM on di

Imaging-Based Reporter Systems to Define CVB-Induced Membrane Remodeling in Living Cells.

Enteroviruses manipulate host membranes to form replication organelles, which concentrate viral and host factors to allow for efficient replication. However, this process has not been well-studied in living cells throughout the course of infection. To define the dynamic process of enterovirus membrane remodeling of major secretory pathway organelles, we have developed plasmid-based reporter systems that utilize viral protease-dependent release of a nuclear-localized fluorescent protein from the endoplasmic reticulum (ER) membrane during infection, while retaining organelle-specific fluorescent

Sexual dimorphism in epidermal stem cell dynamics.

The epidermis is a stratified epithelium comprising proliferative basal cells and differentiated suprabasal cells that are continuously renewed throughout life. Conflicting theories have been proposed regarding the heterogeneity of the basal cell layer, with some studies proposing that all proliferative cells have self-renewal potential during homeostasis1-5. Meanwhile, others posit a model in which stem cell differentiation occurs through an intermediate transit-amplifying-like progenitor state with strictly limited proliferative potential6-10. Here, to determine the heterogeneity and pattern

Loss of Atp8a2 drives neurodegeneration through the dysregulation of spatiotemporal phosphatidylserine externalization in mature neurons.

Phosphatidylserine (PS) asymmetry in plasma membranes is critical for cellular functions and serves as an apoptotic signal in many cell types. However, in mature neurons, the molecular mechanisms governing PS distribution, its precise regulation, and its functional significance beyond apoptosis and development remain poorly understood - particularly in the context of neurodegeneration. Here, we mapped the spatiotemporal dynamics of PS exposure in mature hippocampal neurons under physiological and pathological conditions using time-lapse imaging, revealing specific PS externalization hotspots a

Mechanical polarity links adhesion-regulated protrusions to directional stability in glioblastoma cell migration.

Glioblastoma invasion critically limits therapeutic outcomes, yet the physical principles that govern directional cell migration remain poorly understood. In particular, the degree to which protrusive forces are aligned or cancel each other, and how this governs migration efficiency, have remained unquantified. Here, we introduce mechanical polarity, a quantitative descriptor that captures the alignment of protrusive and adhesive forces driving migration. By integrating time-lapse imaging of the glioblastoma-derived cells with a coarse-grained biophysical model incorporating catch- and slip-bo

DRP1 mutations associated with EMPF1 encephalopathy perturb the transcriptional profile and maturation of cortical neurons.

With the advent of exome sequencing, a growing number of children are being identified with de novo loss-of-function mutations in the dynamin 1-like (DNM1L) gene, which encodes the large GTPase essential for mitochondrial fission, dynamin-related protein 1 (DRP1). Mutations in DRP1 result in severe neurodevelopmental phenotypes, such as developmental delay, optic atrophy, and epileptic encephalopathies. Though it is established that mitochondrial fission is an essential precursor to the rapidly changing metabolic needs of the developing cortex, it is not understood how identified mutations in

YB-1 Knockdown Inhibits the Proliferation of Mesothelioma Cells through Multiple Mechanisms.

Y-box binding protein-1 (YB-1) is a multifunctional oncoprotein that has been shown to regulate proliferation, invasion and metastasis in a variety of cancer types. We previously demonstrated that YB-1 is overexpressed in mesothelioma cells and its knockdown significantly reduces tumour cell proliferation, migration, and invasion. However, the mechanisms driving these effects are unclear. Here, we utilised an unbiased RNA-seq approach to characterise the changes to gene expression caused by loss of YB-1 knockdown in three mesothelioma cell lines (MSTO-211H, VMC23 and REN cells). Bioinformatic

Migration of immortalized nasopharyngeal epithelia and carcinoma cells through porous membrane in 3D platforms.

In the present study, 3D biomimetic platforms were fabricated with guiding grating to mimic extracellular matrix topography, porous membrane to resemble the epithelial porous interface and trenches below to represent blood vessels as an in vitro tissue microenvironment. Fabrication technologies were developed to integrate the transparent biocompatible polydimethylsiloxane platforms with preciously controlled dimensions. Cell migration behaviors of an immortalized nasopharyngeal epithelial cell line (NP460) and a nasopharyngeal carcinoma cell line (NPC43) were studied on the 2D and 3D platforms

Zebrafish Pax1a and Pax1b are required for pharyngeal pouch morphogenesis and ceratobranchial cartilage development.

Pharyngeal arches are derived from all three germ layers and molecular interactions among the tissue types are required for proper development of subsequent pharyngeal cartilages; however, the mechanisms underlying this process are not fully described. Here we report that in zebrafish, Pax1a and Pax1b have overlapping and essential functions in pharyngeal pouch morphogenesis and subsequent ceratobranchial cartilage development. Both pax1a and pax1b are co-expressed in pharyngeal pouches, and time-lapse imaging of a novel Tg(pax1b:eGFP) enhancer trap line further revealed the sequential segment

Plasticity of Lh cells caused by cell proliferation and recruitment of existing cells.

Luteinizing hormone (Lh) and follicle-stimulating hormone (Fsh) control reproduction in vertebrates. Using a transgenic line of medaka, in which green fluorescent protein expression is controlled by the endogenous lhb promotor, we studied development and plasticity of Lh cells, comparing juveniles and adults of both genders. Confocal imaging and 3D reconstruction revealed hypertrophy and hyperplasia of Lh cells in both genders from juvenile to adult stages. We show that Lh cell hyperplasia may be caused by recruitment of existing pituitary cells that start to produce lhb, as evidenced by time

A dynamic cell entry pathway of respiratory syncytial virus revealed by tracking the quantum dot-labeled single virus.

Studying the cell entry pathway at the single-particle level can provide detailed and quantitative information for the dynamic events involved in virus entry. Indeed, the viral entry dynamics cannot be monitored by static staining methods used in cell biology, and thus virus dynamic tracking could be useful in the development of effective antiviral strategies. Therefore, the aim of this work was to use a quantum dot-based single-particle tracking approach to monitor the cell entry behavior of the respiratory syncytial virus (RSV) in living cells. The time-lapse fluorescence imaging and traject

Impaired glomerulogenesis and endothelial cell migration in Pkd1-deficient renal organ cultures.

The PKD1 gene is essential for a number of biological functions, and its loss-of-function causes autosomal dominant polycystic kidney disease (ADPKD). The gene is developmentally regulated and believed to play an essential role in renal development. Previous studies have shown that manipulating murine renal organ cultures with dominant-negative forms of the Pkd1 gene impaired ureteric bud (UB) branching. In the current study, we analyzed different stages of renal development in two distinct mouse models carrying either a null mutation or inactivation of the last two exons of Pkd1. Surprisingly

P2X7 receptor activation regulates microglial cell death during oxygen-glucose deprivation.

Brain-resident microglia may promote tissue repair following stroke but, like other cells, they are vulnerable to ischemia. Here we identify mechanisms involved in microglial ischemic vulnerability. Using time-lapse imaging of cultured BV2 microglia, we show that simulated ischemia (oxygen-glucose deprivation; OGD) induces BV2 microglial cell death. Removal of extracellular Ca(2+) or application of Brilliant Blue G (BBG), a potent P2X7 receptor (P2X7R) antagonist, protected BV2 microglia from death. To validate and extend these in vitro findings, we assessed parenchymal microglia in freshly is

Tau/PTL-1 associates with kinesin-3 KIF1A/UNC-104 and affects the motor's motility characteristics in C. elegans neurons.

Tauopathies are neurodegenerative diseases based on pathological tau-aggregation including Alzheimer's disease, frontotemporal dementia (FTD) and Pick's disease. In general, cargo (e.g., β-amyloid precursor protein, tau, neurofilaments) accumulation is a commonly observed phenomenon in degenerated neurons. Therefore, it is crucial to investigate the interaction between cargo, microtubule-binding proteins and molecular motors. We report the effect of tau/PTL-1 (protein with tau-like repeats) on the transport characteristics of the major axonal transporter kinesin-3 KIF1A/UNC-104 in the nervous

CellTrap: an instrument-free microfluidic platform for cell-cell interactions at stochastically generated effector-to-target ratios.

Immune-cancer cell interactions play a central role in understanding antitumor responses and evaluating immunotherapies. However, long-term, single-cell-level analysis of these interactions remains challenging. To address this, we developed CellTrap, an instrument-free, perfusion-capable microfluidic device featuring 1024 parallel traps. Each trap is equipped with a filter constriction to stably retain cells under hydrostatic flow, sustain continuous medium perfusion, and minimize trap-to-trap crosstalk. By intentionally leveraging stochastic Poisson loading, a single seeding step simultaneous

A Tale of Two Cell Lines: Characterization of Differential Efficacy of Small Molecule Drugs Cediranib and NU-7441 on Primary Versus Metastatic Colorectal Cancer.

Colorectal cancer is recognized as one of the leading causes of cancer death amongst both sexes in the U.S. Despite rigorous screening, those diagnosed with colon cancer often face poor prognosis, and approximately 70% of affected patients will develop metastatic relapse. The investigation of colon carcinoma cell lines' genetic variability and response to chemotherapy panels may aid in targeting therapies to improve outcomes. This study aims to find correlations between metastasis status, gene variability, and drug response. We used two cell lines that were isolated from the same 51-year-old m
