Tsetse-trypanosomiasis challenge to village N'Dama cattle in The Gambia: field assessments of spatial and temporal patterns of tsetse-cattle contact and the risk of trypanosomiasis infection.

The severity of the trypanosomiasis problem in a particular location is traditionally assessed in terms of a challenge index-the product of some measure of tsetse abundance and infection-rate-which is assumed to be proportional to the force of infection. However, this index masks variation in the force of infection between herds and among individuals within herds. It is also not comparable between sites since the relative abundance of tsetse to hosts may vary. We have studied spatial distribution of herds of cattle in relation to tsetse in The Gambia and calculated an index of challenge based …

Feeding site selection in Rhodnius prolixus nymphs, American trypanosomiasis vector.

Thermal cues are fundamental for host-seeking in kissing bugs, primary vectors of American trypanosomiasis. Host surface temperature may reflect underlying vascular richness, potentially guiding feeding site selection. In this study, we investigated whether Rhodnius prolixus nymphs preferentially select specific regions of a mammalian host and whether local skin temperature influences this choice. Nymphs from first to fifth instars were allowed to feed on anesthetized mice, and insect densities were recorded across distinct body regions (head, lateral abdomen, paws and tail). Mouse body temper…

Host Immune Responses and Immune Evasion Strategies in African Trypanosomiasis.

Parasites, including African trypanosomes, utilize several immune evasion strategies to ensure their survival and completion of their life c … cells (MDSCs), and regulatory T cells have been shown to contribute to immunosuppression in African trypanosomiasis. In this review, we discuss the key factors that contribute to immunity and immunosuppression during T. congolense infection, and how these factors could aid immune evasion by African trypanosomes. Understanding the regulatory mechanisms that influe …

Inhibitors of glycosomal protein import provide new leads against trypanosomiasis.

Vector-borne trypanosomatid parasite infections in tropical and sub-tropical countries constitute a major threat to humans and livestock. Trypanosoma brucei parasites are transmitted by tsetse fly and lead to African sleeping sickness in humans and Nagana in cattle. In Latin American countries, Trypanosoma cruzi infections spread by triatomine kissing bugs lead to Chagas disease. Various species of Leishmania transmitted to humans by phlebotomine sandflies manifest in a spectrum of diseases termed Leishmaniasis. 20 million people are currently infected with trypanosomatid parasites, leading to

Bioluminescence Imaging to Detect Late Stage Infection of African Trypanosomiasis.

Human African trypanosomiasis (HAT) is a multi-stage disease that manifests in two stages; an early blood stage and a late stage when the parasite invades the central nervous system (CNS). In vivo study of the late stage has been limited as traditional methodologies require the removal of the brain to determine the presence of the parasites. Bioluminescence imaging is a non-invasive, highly sensitive form of optical imaging that enables the visualization of a luciferase-transfected pathogen in real-time. By using a transfected trypanosome strain that has the ability to produce late stage disea…

Discovery of Infection Associated Metabolic Markers in Human African Trypanosomiasis.

Human African trypanosomiasis (HAT) remains a major neglected tropical disease in Sub-Saharan Africa. As clinical symptoms are usually non-specific, new diagnostic and prognostic markers are urgently needed to enhance the number of identified cases and optimise treatment. This is particularly important for disease caused by Trypanosoma brucei rhodesiense, where indirect immunodiagnostic approaches have to date been unsuccessful. We have conducted global metabolic profiling of plasma from T.b.rhodesiense HAT patients and endemic controls, using 1H nuclear magnetic resonance (NMR) spectroscopy a…

Novel naphthoquinone derivatives: synthesis and activity against human African trypanosomiasis.

A series of naphthoquinone derivatives has been synthesized and tested for its biological activity against human African trypanosomiasis. The use of reverse micellar medium not only enhanced the conversion rate, but also showed selectivity towards mono-coupled product in aryl chloride-aniline coupling reactions. Two derivatives of naphthoquinone (9b and 9c) exhibited potent activity against Trypanosoma brucei in vitro with low cytotoxicity.

Research priorities for Chagas disease, human African trypanosomiasis and leishmaniasis.

This report provides a review and analysis of the research landscape for three diseases - Chagas disease, human African trypanosomiasis and leishmaniasis - that disproportionately afflict poor and remote populations with limited access to health services. It represents the work of the disease reference group on Chagas Disease, Human African Trypanosomiasis and Leishmaniasis (DRG3) which was established to identify key research priorities through review of research evidence and input from stakeholders' consultations. The diseases, which are caused by related protozoan parasites, are described i…

How can tsetse population genetics contribute to African trypanosomiasis control?

In sub-Saharan Africa, tsetse transmitted Trypanosomiases have an enormous impact on human health and economic development. Both the World Health Organisation and African countries through the Pan African Tsetse and Trypanosomiasis Eradication Campaign (PATTEC) have recently asserted their determination to rid the sub-continent of these diseases, and it is increasingly recognised that vector control should play an important role. This review mainly focuses on population genetics of tsetse of the palpalis group, the main vectors of sleeping sickness, and reports recent results on tsetse populat…

Increased CXCL-13 levels in human African trypanosomiasis meningo-encephalitis.

OBJECTIVES: To determine the role of the B-cell attracting chemokine CXCL-13, which may initiate B-cell trafficking and IgM production in diagnosing HAT meningo-encephalitis. METHODS: We determined CXCL-13 levels by ELISA on paired sera and CSF of 26 patients from Angola and of 16 controls (six endemic and ten non-endemic). Results were compared to standard stage determination markers and IgM intrathecal synthesis. RESULTS: CXCL-13 levels in patients' sera had a median value of 386.6 pg/ml and increased levels were associated with presence of trypanosomes in the CSF but not with other stage

Serum antibodies against human brain myelin proteins in Gambian trypanosomiasis.

Human brain myelin was obtained by flotation centrifugation on 0.88 M sucrose. The myelin proteins were extracted with 0.1% SDS-0.01 M Tris, pH 7.4 from the myelin floating on 0.88 M sucrose following centrifugation. Using ELISA and immunoblotting, we have shown that antibodies against the myelin proteins exist in sera collected from Gambian trypanosomiasis patients. The antibodies were more prevalent in patients with CNS involvement since of 21 sera from patients with more than 4 cells/ml cerebrospinal fluid (CSF), 15 (71.4%) had antimyelin antibodies, as compared to 3 of 13 (23.1%) from pati…

Immunization of mice against African trypanosomiasis using anti-idiotypic antibodies.

Anti-idiotypic (anti-Id) antibodies were raised against three protective monoclonal antibodies, each with specificity for the variable antigen type (VAT) of a clone of Trypanosoma rhodesiense. The IgG1 fractions of each were pooled and administered to BALB/c mice 3-4 wk before homologous challenge. The course of primary parasitemia was altered in 19 of 30 anti-Id-treated animals. The immunity was manifested as either: (a) complete protection, (b) reduced parasitemia, or (c) selection against parasites bearing the original VAT. The three idiotypes (Id) were found in variable levels in serum dur
