A national reference for inactivated polio vaccine derived from Sabin strains in Japan.

As one aspect of its campaign to eradicate poliomyelitis, the World Health Organization (WHO) has encouraged development of the inactivated polio vaccine (IPV) derived from the Sabin strains (sIPV) as an option for an affordable polio vaccine, especially in low-income countries. The Japan Poliomyelitis Research Institute (JPRI) inactivated three serotypes of the Sabin strains and made sIPV preparations, including serotypes 1, 2 and 3 D-antigens in the ratio of 3:100:100. The National Institute of Infectious Diseases, Japan, assessed the immunogenic stability of these sIPV preparations in a rat

Immunologic characterization of a novel inactivated nasal mumps virus vaccine adjuvanted with Protollin.

An inactivated, mucosal mumps virus (MuV) vaccine would address many of the problems associated with current live-attenuated formulations. Protollin (Prl)-based adjuvants (containing TLR2 and TLR4 ligands) are well-suited for nasal administration. We sought to develop an inactivated whole-virus nasal vaccine for MuV using the Prl adjuvant/delivery vehicle and to test tolerability and immunogenicity in a mouse model. BALB/c mice exhibited signs of transient reactogenicity (hunched posture, erect fur, weight loss ≤10% of total body weight) following administration of intranasal MuV-Prl vaccines,

Immunogenicity and safety of an inactivated Rift Valley fever vaccine in a 19-year study.

An investigational, formalin-inactivated Rift Valley fever (RVF) vaccine, known as The Salk Institute-Government Services Division (TSI-GSD) 200 vaccine, was administered to 1860 at-risk subjects (5954 doses) between 1986 and 2004 as a three-dose primary series (days 0, 7, and 28) followed by booster doses as needed for declining titers. An initial positive serological response (PRNT(80)≥1:40) to the primary series was observed in 90% of subjects. Estimate of the PRNT(80) response half-life in initial responders to the primary series by Kaplan-Meier plot was 315 days after the primary series d

Nucleocapsid of rabies virus improve immune response of an inactivated avian influenza vaccine.

The purpose of this study was to determine if nucleocapsid of rabies virus could improve the immune response (humoral and protective) of chickens vaccinated against avian influenza with an inactivated avian influenza experimental vaccine (AIV). On the other hand, AIV with and without NC was compared with an inactivated oil emulsion avian influenza commercial vaccine (CV) virus, currently used in Mexico. Groups of 8 day old chickens were vaccinated intracutaneously with an AIV (group 1); group 2, AIV supplemented with 20 microg of nucleocapsid of rabies virus (NC); Group 3, commercial vaccine (

A Phase 2 study of a purified, inactivated virus vaccine to prevent Japanese encephalitis.

Japanese encephalitis (JE) is a serious disease caused by the JE virus. New generation JE vaccines are needed to prevent this disease. We conducted this Phase 2 randomized, open label, unblinded, single center study of a new, cell-culture derived, purified inactivated virus (JE-PIV) vaccine. The JE-PIV vaccine was administered in either two or three intramuscular (IM) doses (6.0 or 12.0 mcg each) with observation over 8 weeks. All volunteers completed the protocol without serious adverse reactions. Headache and transient tenderness at the injection site were the most common complaints. There w

Safety and efficacy in geese of a PER.C6-based inactivated West Nile virus vaccine.

Studies were performed with an inactivated vaccine against the mosquito-borne flavivirus, West Nile virus (WNV). The mammalian cell line, PER.C6, was selected as the platform for WNV growth since both the neurovirulent strains NY99 and ISR98 that cause epidemics in humans and high mortality in geese, respectively, could be propagated to high titers (10(9) to 10(10)TCID(50)/ml) on these cells. Based on the high DNA homology of the WNV envelope (E) protein and non-structural protein 5 (NS5), and identical neurovirulence in mice and geese, we concluded that NY99 and ISR98 viruses are closely rela

Safety and immunogenicity of a booster dose of inactivated poliovirus vaccine produced in vero-cells.

Statens Serum Institut has developed a new vero-cell culturing technique for the manufacturing of inactivated poliovirus vaccine (IPV). This technique implies that the cultivation of cells and poliovirus is performed in a medium free of materials of animal origin and free of antibiotics. In a double-blind randomised clinical trial, IPV(vero) manufactured by this new technique was compared to conventionally produced IPV(mkc). One hundred and twenty-nine (129) healthy adult volunteers were given booster vaccinations of IPV(vero) (65) or IPV(mkc) (64). Both vaccines were well tolerated and result

Immunoglobulin a as a serological marker for the (silent) circulation of poliovirus in an inactivated poliovirus-vaccinated population.

Poliovirus-specific immunoglobulin A (IgA) is detected after infection with wild-type virus or vaccination with live attenuated oral poliovirus (OPV) but not after vaccination with inactivated poliovirus (IPV). We examined whether the presence of IgA in serum can be used as a marker for poliovirus circulation in IPV-vaccinated populations in The Netherlands. In seronegative persons challenged with OPV, the sensitivity of this marker was 76%-86%. Results from a serosurvey showed a high seroprevalence (63%-73%) of IgA in the population born before vaccination was introduced in The Netherlands, w

Efficacy of an inactivated vaccine for equine herpesvirus type 1 in a novel hamster model.

A vaccine designated F.EHV1(S(-))BHK, which was prepared by formaldehyde treatment of equine herpesvirus type 1 (EHV1)-infected baby hamster kidney cells, stimulated neutralising antibodies in hamsters and rabbits and protected new-born hamsters against a lethal challenge with EHV1 by vaccination of their pregnant mothers. The preparative method was then modified to eliminate virus particles and intraparticulate DNA, producing a vaccine designated Bg.F.EHV1(S(-))BHK. this modification stimulated neutralising antibodies with evidence of in vivo inactivation of the virus above non-specific level

A Salmonella abortusovis inactivated vaccine protects mice from abortion after challenge.

Two types of Salmonella abortusovis vaccines were prepared, one with aluminium hydroxide (vaccine A) and the other with water in oil (vaccine B) adjuvants. They were compared in a pregnant mouse model, aiming at protecting them from abortions after challenge with a virulent strain of S. abortusovis. The protection for vaccine A was from 74% to 77.6% and that for vaccine B from 71% to 79.6%. Abortions occurred 5-10 days post challenge and S. abortusovis was isolated from all aborted fetuses and from the liver and the spleen of their mothers at the end of the experiment (18 days post challenge).

Immunogenicity of an inactivated Rift Valley fever vaccine in humans: a 12-year experience.

Rift Valley fever (RVF) virus causes serious and fatal disease in animals and man. To protect personnel who work with RVF virus in the laboratory, or troops who may be exposed to this virus, the US Army successfully developed an improved version of inactivated RVF vaccine, TSI-GSD-200. From early 1986 to late 1997, 598 at-risk workers at the US Army Medical Research Institute of Infectious Diseases (USAMRIID) were vaccinated as part of an occupational safety and health program. The subjects of this study received three subcutaneous doses (0, 7 and 28 days) of 0.5 ml of TSI-GSD-200. A total of

Cost-effectiveness of incorporating inactivated poliovirus vaccine into the routine childhood immunization schedule.

OBJECTIVE: To evaluate the economic consequences of introducing inactivated poliovirus vaccine (IPV) into the routine vaccination schedule in the United States to reduce vaccine-associated paralytic poliomyelitis (VAPP). DESIGN: Cost-benefit and cost-effectiveness models were formulated to compare the current national 4-dose live attenuated oral poliovirus vaccine (OPV) schedule with a 4-dose IPV schedule or a sequential schedule of 2 doses of IPV followed by 2 doses of OPV. Model assumptions were derived from the National Health Interview Survey (1994), current prices for OPV and IPV, a Delp

Good seroresponse to enhanced-potency inactivated poliovirus vaccine in patients on chronic dialysis.

The outbreak of paralytic poliomyelitis in Finland in 1984 was halted by nationwide oral poliovirus vaccination campaign. Immunocompromised patients, including those with chronic uraemia and on continuous dialysis, were excluded from the oral vaccination group and instead were given a dose of the new enhanced-potency inactivated poliovirus vaccine before the campaign. We studied the antibody response to this vaccine in 49 patients on chronic dialysis, using conventional antigen of all three serotypes and two additional type 3 strains. It was observed that 86% (42 of 49) of patients either had
