A novel explainable online calculator for contrast-induced AKI in diabetics: a multi-centre validation and prospective evaluation study.

BACKGROUND: In patients undergoing percutaneous coronary intervention (PCI), contrast-induced acute kidney injury (CIAKI) is a frequent complication, especially in diabetics, and is connected with severe mortality and morbidity in the short and long term. Therefore, we aimed to develop a CIAKI predictive model for diabetic patients. METHODS: 3514 patients with diabetes from four hospitals were separated into three cohorts: training, internal validation, and external validation. We developed six machine learning (ML) algorithms models: random forest (RF), gradient-boosted decision trees (GBDT)

Prediction of the position of external markers using a recurrent neural network trained with unbiased online recurrent optimization for safe lung cancer radiotherapy.

BACKGROUND AND OBJECTIVE: During lung cancer radiotherapy, the position of infrared reflective objects on the chest can be recorded to estimate the tumor location. However, radiotherapy systems have a latency inherent to robot control limitations that impedes the radiation delivery precision. Prediction with online learning of recurrent neural networks (RNN) allows for adaptation to non-stationary respiratory signals, but classical methods such as real-time recurrent learning (RTRL) and truncated backpropagation through time are respectively slow and biased. This study investigates the capabil

Prediction Aided Tapering In rheumatoid arthritis patients treated with biOlogicals (PATIO): protocol for a randomized controlled trial.

BACKGROUND: Biological disease-modifying anti-rheumatic drugs (bDMARDs) are effective in the treatment of rheumatoid arthritis (RA) but are expensive and increase the risk of infection. Therefore, in patients with a stable low level of disease activity or remission, tapering bDMARDs should be considered. Although tapering does not seem to affect long-term disease control, (short-lived) flares are frequent during the tapering process. We have previously developed and externally validated a dynamic flare prediction model for use as a decision aid during stepwise tapering of bDMARDs to reduce the

Structure-activity relationship-based chemical classification of highly imbalanced Tox21 datasets.

The specificity of toxicant-target biomolecule interactions lends to the very imbalanced nature of many toxicity datasets, causing poor performance in Structure-Activity Relationship (SAR)-based chemical classification. Undersampling and oversampling are representative techniques for handling such an imbalance challenge. However, removing inactive chemical compound instances from the majority class using an undersampling technique can result in information loss, whereas increasing active toxicant instances in the minority class by interpolation tends to introduce artificial minority instances

Presence of early CKD-related metabolic complications predict progression of stage 3 CKD: a case-controlled study.

BACKGROUND: Only a subset of patients who enter stage 3 chronic kidney disease (CKD) progress to stage 4. Identifying which patients entering stage 3 are most likely to progress could improve outcomes, by allowing more appropriate referrals for specialist care, and spare those unlikely to progress the adverse effects and costliness of an unnecessarily aggressive approach. We hypothesized that compared to non-progressors, patients who enter stage 3 CKD and ultimately progress have experienced greater loss of renal function, manifested by impairment of metabolic function (anemia, worsening acido

Advancing Gene Feature Selection: A Synergistic Approach with Co-expression Networks and Genetic Algorithms.

INTRODUCTION: Gene feature selection is essential in bioinformatics and medical research, as it identifies gene subsets closely associated with specific diseases or biological traits from high-dimensional gene datasets. High-dimensional gene data causes the curse of dimensionality, leading to sparsity, complex inter-feature relationships, and significant noise. These issues undermine the reliability of conventional statistical methods in capturing underlying biological information. While gene feature selection can enhance classification model accuracy and reduce computational complexity, exist

A systematic analysis of the impact of data variation on AI-based histopathological grading of prostate cancer.

The histopathological evaluation of biopsies by human experts is a gold standard in clinical disease diagnosis. While recent artificial intelligence-based (AI) approaches have reached human expert-level performance, they often display shortcomings caused by variations in sample preparation, limiting clinical applicability. This study investigates the impact of data variation on AI-based histopathological grading and explores algorithmic approaches that confer prediction robustness. To evaluate the impact of data variation in histopathology, we collected a multicentric, retrospective, observati

Toward FOXM1 inhibitors based on supervised machine learning classification models and similarity-based screening.

Ovarian cancer remains among the deadliest gynaecological malignancies, with aberrant activation of the Forkhead Box M1 (FOXM1) transcription factor strongly implicated in tumour progression, therapy resistance, and poor prognosis. Given the limited availability of validated FOXM1 inhibitors and the lack of prior dedicated machine learning-based QSAR studies identified in our literature survey, this study employed a supervised machine learning framework to predict the bioactivity of structurally diverse small-molecule inhibitors. A curated dataset of 101 compounds with reported IC50 values was

Artificial intelligence-based prediction of radio-cephalic arteriovenous fistula maturation using preoperative duplex examination: a retrospective multicenter cohort study.

Establishing a functional arteriovenous fistula (AVF) is crucial to ensure effective dialysis treatment. However, there are currently no definitive criteria for predicting AVF maturation using preoperative ultrasound. This study aims to assess the efficacy of AI-driven models in analyzing comprehensive ultrasonographic variables across multiple forearm locations to predict successful AVF maturation. A retrospective analysis was conducted on 492 cases within a cohort of 494 individuals who underwent radiocephalic arteriovenous fistula (AVF) creation across three centers over a span of four year
