Synthesis of specific IgG idiotypes by rheumatoid synovium.

Synovial tissue samples from 6 patients with rheumatoid arthritis were cultured, and the IgG antibodies isolated from the synovial culture supernatants were used to immunize rabbits to make 6 antiidiotypic (anti-Id) antibody preparations. After extensive adsorption, the rabbit anti-Id were tested in a solid-phase enzyme-linked immunosorbent assay (ELISA). Each anti-Id reacted predominantly with the immunizing synovial IgG and showed almost no reactivity with either pooled normal human serum IgG or with IgG from 50 normal donors. When identical amounts of matched rheumatoid arthritis serum IgG

[Ovine salmonellosis caused by Salmonella abortus ovis].

Salmonella enteritidis subsp. enteritidis ser. abortus ovis, a sheep-adapted serotype, causes a contagious infectious disease with abortion as the main symptom, sometimes accompanied with mortality of lambs. This auxotrophic gram-negative bacterium ranks among the main causes of ovine abortions in some countries of Europe and of western Asia. The disease tends toward an endemic pattern with a multi-annual rhythm in the frequency of abortions. Bacteremia leads to bacterial colonization of the feto-placental unit, the principal site of salmonella multiplication. Peripartum vaginal excretion, com

Development of monoclonal antibodies directed against different epitopes of human chorionic gonadotropin.

Human chorionic gonadotropin (HCG) is a 40,000 dalton glycoprotein composed of two non-identical alpha- and beta-subunits. HCG and other related pituitary hormones, such as human luteinizing hormones (HLH), human follicle stimulating hormone (HFSH), and human thyroid stimulating hormone (HTSH), consist of nearly identical alpha-chains. However, their beta-chains show a variable degree of amino acid sequence homology. Detection and subsequent quantitative determination of HCG in human biological fluids is useful in early diagnosis of pregnancy and in monitoring of tumor patients. For these appl

Some zoonotic infectious diseases are prevalent among children under 15 years of age in Ardabil Province, northern Iran.

BACKGROUND: This study sought to investigate the seroprevalence of visceral leishmaniasis (VL), toxocariasis, brucellosis, and salmonellosis, as well as their co-infection and potential cross-reaction, in children under 15 years referred to health centers in Ardabil province, Iran, from 2019 to 2021. METHODS: The current study examined 1,550 serum samples using direct agglutination test (DAT), Toxocara canis ELISA, Wright, and Widal tests to detect antibodies against Leishmania, Toxocara, Brucella, and Salmonella, respectively. We also compared the test results to determine the possibility of

Maximizing the chances of detecting pathogenic leptospires in mammals: the evaluation of field samples and a multi-sample-per-mammal, multi-test approach.

Identification of wild animals that harbour the causative leptospires, and the identification of the most important of these 'wild reservoirs' (in terms of threat to human health), are key factors in the epidemiology of human leptospirosis. In an epidemiological investigation in the Australian state of Queensland, in 2007-2008, samples were collected from fruit bats (Pteropus conspicillatus) and rodents (to investigate the potential role of fruit bats in the maintenance and transmission of leptospires to ground-dwelling rodents) and checked for pathogenic leptospires. The results of these stud

A rapid and reliable method for early Legionella pneumophila identification and characterization in support of the epidemiology study.

INTRODUCTION: Legionnaires' disease is a severe pneumonia predominantly caused by Legionella pneumophila (Lp), whose major reservoirs are artificial water systems. As most human infections are caused by L. pneumophila serogroup 1 (Lp1), a reliable method for Lp distinction can be crucial for bacterial spread prevention. As the ability to withstand in environments and to cause the waterborne disease is strongly related to specific genes, the identification of virulent strains can be of great relevance to implement water environmental monitoring and to contain harmful outbreaks to public health.

Molecular and serological diagnosis of multiple bacterial zoonoses in febrile outpatients in Garissa County, north-eastern Kenya.

Bacterial zoonoses are diseases caused by bacterial pathogens that can be naturally transmitted between humans and vertebrate animals. They are important causes of non-malarial fevers in Kenya, yet their epidemiology remains unclear. We investigated brucellosis, Q-fever and leptospirosis in the venous blood of 216 malaria-negative febrile patients recruited in two health centres (98 from Ijara and 118 from Sangailu health centres) in Garissa County in north-eastern Kenya. We determined exposure to the three zoonoses using serological (Rose Bengal test for Brucella spp., ELISA for C. burnetti a

Leptospira Seropositivity in Humans, Livestock and Wild Animals in a Semi-Arid Area of Tanzania.

BACKGROUND: Leptospirosis is among the major neglected zoonoses in developing countries. The prevalence of leptospirosis remains underestimated in many African countries because of limited diagnostic facilities. We studied Leptospira seropositivity prevalence in humans, sheep, goats and rodents in a semi-arid region of central Tanzania and compared findings with reports from humid tropical areas. The aims were to establish the disease burden in different settings; understand circulating Leptospira serovars and potential major reservoirs for establishing appropriate control measures. METHODS:

Infection exposure, detection and causes of death in perinatal mortalities in Polish dairy herds.

The objective of this study was to determine the prevalence and types of infections in perinatal mortality (PM) cases from Polish dairy farms and the relevance of the presence of infection to the cause of death. This prospective longitudinal study was carried out on 121 PM and 21 control calves with a gestation of ≥260 days. Six control calves were euthanized and examined using the same protocol as for PM calves. Material was collected over a 20-month period between November 2013 and June 2015. The PM and control calves were collected from 29 to 5 herds, respectively. Blood samples from calves

A Retrospective Survey of the Abortion Outbreak Event Caused by Brucellosis at a Blue Fox Breeding Farm in Heilongjiang Province, China.

Brucellosis is a common zoonosis in China, resulting in abortion in animals. Outbreaks of abortion in blue foxes caused by Brucella infection have rarely been reported. In the present study, 3-5 mL blood samples collected from the femoral veins of 10 abortuses of blue foxes were assessed by RBPT (Rose Bengal plate test) and SAT (serum tube agglutination test) to preliminarily investigate the source of infection for the clustering of abortion events at a blue fox farm in Heilongjiang Province. Screening experiments showed that all 10 blood samples were positive in the RBPT, while only eight blo

Emerging Leptospira strain poses public health risk for dairy farmers in New Zealand.

Leptospira infection in dairy cattle and leptospirosis in dairy farm workers were common in New Zealand prior to the introduction of dairy cattle vaccination in the 1980s. Despite widespread vaccination within the dairy industry, the long-term effectiveness of vaccination and current Leptospira exposure status remained unknown. A cross-sectional study was conducted from January-April 2016 to investigate the prevalence of pathogenic Leptospira spp. DNA in urine at cow and herd level, and its relationship to five Leptospira serovars known to be endemic. Two hundred dairy farms were randomly sele

Disease burden and antimicrobial resistance of invasive group B streptococcus among infants in China: a protocol for a national prospective observational study.

BACKGROUND: Group B Streptococcus (GBS) is a cause of neonatal sepsis, pneumonia, and meningitis that can lead to neurological sequelae in infants less than 3 months of age. The GBS disease burden is not known in China, therefore it cannot receive major attention. The main objectives of this study are the evaluation of the incidence of neonatal GBS infection, GBS case-fatality ratio, its serotypes and genotypes, bacterial resistance, clinical treatment and outcomes in China. METHODS: We are conducting a nation-wide, population-based, multi-center, prospective, observational cohort study in Ch

Serological findings obtained in cattle herds immunised with the Brucella melitensis Rev.1 and the B. abortus B19 vaccine in Mongolia.

To reduce the economic losses caused by Brucella abortus infection under the conditions of nomadic cattle breeding, more than half million cattle over 3 months of age (excluding male animals) were immunised with the B. melitensis Rev.1 vaccine in 7 provinces of Mongolia in the first year of a programme (1987). In the second year, only heifer calves over 3 months of age, yearling animals not vaccinated in the first year, as well as heifers over 2 years of age and cows were vaccinated. The vaccine was administered subcutaneously at a dose of 5 x 10(9) viable organisms. In co-operatives that had

Cryptococcosis associated with HIV negative Indian patients and HIV positive Indian blood donors.

Cryptococcosis, particularly cryptococcal meningitis (CM), has become an increasing problem globally in the AIDS era. In the present investigation we have made an effort for the first time to study Indian cases (100) both HIV-positive (23 cases, male, mostly Indian professional blood donors, PBDs') confirmed by an ELISA test and Western Blot but asymptomatic for CM and HIV-negative (77:49 male and 28 female) asymptomatic or symptomatic. These subjects were patients from the Lucknow hospitals admitted during the period between February, 1991 to February, 1994, for suspected cryptococcosis or CM

Longitudinal evaluation of asymptomatic Leishmania infection in HIV-infected individuals in North-West Ethiopia: A pilot study.

BACKGROUND: In endemic regions, asymptomatic Leishmania infection is common. In HIV patients, detection of asymptomatic Leishmania infection could potentially identify those at risk of visceral leishmaniasis (VL). However, data on the prevalence, incidence, and determinants of asymptomatic infection, and the risk of VL are lacking. METHODS: We conducted a cross-sectional survey at a single ART centre, followed by a prospective cohort study amongst HIV-infected adults in HIV care in a district hospital in a VL-endemic area in North-West Ethiopia (9/2015-8/2016). Asymptomatic Leishmania infecti

Seroprevalence studies of human brucellosis in pyrexia of unknown origin (PUO) patients by various diagnostic modalities.

BACKGROUND: Brucellosis is a re-emerging zoonosis with global distribution. In India, it remains common but often neglected. In Rajasthan, where agriculture is prevalent, frequent contact with infected animals such as goats, sheep, and cattle leads to brucellosis-like clinical symptoms. This study aims to determine the seroprevalence of brucellosis in patients presenting with PUO (Pyrexia of Unknown Origin). MATERIALS AND METHODS: This cross-sectional observational study was conducted on 170 suspected PUO patients from January 2019 to December 2019 in Western Rajasthan to detect the seropreva

Demographic characteristics and prevalence of asymptomatic Leishmania donovani infection in migrant workers working in an endemic area in Northwest Ethiopia.

INTRODUCTION: Visceral leishmaniasis (VL), a neglected tropical disease that causes substantial morbidity and mortality, is a serious health problem in Ethiopia. Infections are caused by Leishmania (L.) donovani parasites. Most individuals remain asymptomatic, but some develop VL, which is generally fatal if not treated. We identified the area of Metema-Humera in Northwest Ethiopia as a setting in which we could follow migrant workers when they arrived in an endemic area. The demographic characteristics of this population and factors associated with their risk of asymptomatic infection are poo
