MHC class I of saltwater crocodiles (Crocodylus porosus): polymorphism and balancing selection.

Saltwater crocodiles are in high demand for the production of luxury fashion items. However, their susceptibility to disease incurs substantial losses and it is hoped to be able to genetically select these animals for disease resistance. So far, this has only been enabled by phenotypic selection. Investigating the major histocompatibility complex (MHC) could provide insight into the ability of an individual to respond to pathogens acting as a selective pressure on the host. Here, we assessed genetic diversity and a role of selection in shaping the diversity of MHC class I exon 3 among 42 saltw

A common genetic background for inflammatory bowel disease and ankylosing spondylitis: a genealogic study in Iceland.

OBJECTIVE: Patients with ankylosing spondylitis (AS) and approximately 50% of their first-degree relatives may have a genetic abnormality that results in subclinical intestinal inflammation. This study was undertaken to examine the familial occurrence and cosegregation of AS and inflammatory bowel disease (IBD) in order to determine whether there is a shared genetic risk factor in families. METHODS: The Icelandic genealogy database and population-wide data on all living Icelanders diagnosed as having AS (n = 205) and/or IBD (n = 1,352) were used to estimate the risk ratios of AS for relatives

Genetics and molecular biology of haemophilias A and B.

The development of rapid procedures for the characterization of mutations is advancing the knowledge of the molecular biology of the haemophilias and transforming the strategies for the diagnoses required for genetic counselling. In haemophilia B more than 300 mutants have been fully characterized. These comprise complete and partial deletions, rare insertions, and 'point' mutations. The latter may impair transcription (promoter mutations), RNA processing (splicing mutations) and translation (frameshifts and stop codons) or cause single amino acid (aa) changes. Eighty-four residues are involve

MAFF Suppresses Necroptosis in Pancreatic Cancer via the GATA4-MLKL Axis.

Pancreatic adenocarcinoma (PAAD) is a highly aggressive malignancy characterized by rapid progression. In this study, we identify the small MAF protein, MAFF, as a critical driver of PAAD tumorigenesis and a negative regulator of the necroptotic pathway. MAFF silencing significantly suppressed malignant phenotypes in PAAD cells in vitro and attenuated overall tumor growth in vivo, while ectopic expression promoted tumor progression. Mechanistically, RNA-sequencing and Gene Set Enrichment Analysis (GSEA) identified necroptosis as a key pathway suppressed by MAFF. We established that MAFF acts a

DNA methylation changes associated with Parkinson's disease in SNCA.

Parkinson's disease (PD) is a progressive neurodegenerative disorder influenced by both genetic and epigenetic mechanisms. Among these mechanisms, DNA methylation has emerged as a key regulatory process affecting gene expression without altering the DNA sequence. The synuclein alpha (SNCA) gene is a well-established contributor to Parkinson's disease pathogenesis, yet the extent and consistency of its methylation changes remain unclear. This scoping review aimed to map and summarise existing evidence on DNA methylation changes associated with PD in the SNCA gene. A scoping review methodology g
